Abstract

Abstrak. Salmonella typhi merupakan bakteri patogen yang berbahaya bagi kesehatan manusia yang dapat menyebabkan Salmonellosis. Salmonellosis bersifat zoonosis, sehingga penyakit ini dapat ditularkan dari hewan ke manusia. Salmonella typhi menular kemanusia melalui berbagai makanan asal ternak yang terkontaminasi oleh bakteri Salmonella typhi. Gejala dari terkontaminasi bakteri Salmonella typhi yaitu demam tinggi, mual, muntah, pusing dan nyeri abdomen. Tujuan dilakukan penelitian ini adalah untuk mengetahui adanya bakteri Salmonella typhi pada makanan sushi yang mentah dan setengah matang. Penelitian ini menggunakan metode deskriptif dengan pengambilan sampel secara simple random sampling. Sampel yang digunakan makanan sushi yang mentah dan yang dimasak setengah matang. Sampel makanan diambil sebanyak 3,5 gram lalu di haluskan menggunakan aquadest sebanyak 25 ml, dan sampel diambil 1 ml. uji laboratorium yang dilakukan adalah pembiakan bakteri pada media Salmonella Shigella Agar, lalu dilakukan pewarnaan gram dan selanjtnya penanaman pada media TSIA dan dilakukan pewarnaan gram. Hasil penelitian dari 8 sampel makanan sushi yang diteliti, menunjukkan pertumbuhan bakteri Salmonella typhi sebanyak 7 sampel. 1 sampel tidak ditemukan pertumbuhan bakteri. Ciri ciri media yang terdapat bakteri Salmonella typhi koloni berwarna hitam. Saran penelitian ini adalah pada saat mengonsumsi makanan harus dilihat apakah makanan itu baik dan benar, agar makanan yang dikonsumsi terhindar dari kontaminasi bakteri. Kata Kunci : Sushi, Salmonella typhi Absctract Salmonella typhi is a pathogenic bacteria that is harmful to human health which can cause Salmonellosis. Salmonellosis is zoonotic, so this disease can be transmitted from animals to humans. Salmonella typhi is transmitted to humans through a variety of livestock-based foods contaminated with Salmonella typhi bacteria. Symptoms of being contaminated with Salmonella typhi bacteria are high fever, nausea, vomiting, dizziness and abdominal pain. The purpose of this study was to determine the presence of Salmonella typhi bacteria in raw and undercooked sushi. This study used a descriptive method with simple random sampling. The food samples used were raw and undercooked sushi. Food samples were taken as much as 3.5 grams and then refined using 25 ml of distilled water, and 1 ml of samples were taken. Laboratory tests carried out were culturing bacteria on Salmonella Shigella Agar media, then gram staining was carried out and then planting on TSIA media and gram staining was carried out. The results of the 8 samples of the studied sushi food showed the growth of Salmonella typhi bacteria in 7 samples. 1 sample found no bacterial growth. The characteristics of the media containing Salmonella typhi are black colonies. The suggestion of this research is that when consuming food, it must be seen whether the food is good and correct, so that the food consumed is protected from bacterial contamination. Abstract

Diabetes Mellitus Type 2 is a metabolic disorder condition characterized by hyperglycemia, due to decreased insulin secretion. Many cytokines play a role in the acute phase response, so that markers such as CRP increase in patients with type 2 DM. The type of research used in this study is descriptive research, which describes the results of CRP examination in patients with type 2 diabetes. Examination of respondents' serum C-Reactive Protein (CRP) levels was carried out qualitatively using the Latex Agglutination method and followed by a semi-quantitative method, namely dilution to determine the level of inflammation. The results of CRP examination on 30 serum samples that have been carried out on patients with Type 2 Diabetes Mellitus found that 29 (96.7%) were positive and 1 (3.3%) of respondents were negative. The group of CRP levels for titer 12 was 5 samples (16.7%), titer 24 was 17 samples (56.7%), titer 48 was 5 samples (16.7%), and titer 96 was 2 samples (6.7). %). The results of the four groups with the highest CRP levels, namely a titer of 24 mg/L as many as 17 samples (56.7%) showed that some of the CRP levels were higher than normal. Conclusion Almost all respondents with type 2 Diabetes Mellitus had positive C-Reactive Protein examination results and a small proportion of type 2 Diabetes Mellitus patients had negative C-Reactive Protein examination results.

Abstract

Diabetes melitus adalah penyakit kelainan metabolisme yang ditandai dengan hiperglikemia dengan penurunan insulin. Menurut data kesehatan dunia (WHO), penderita diabetes melitus mencapai 422 juta jiwa diperkirakan meningkat sebesar 11,8% pada tahun 2030 dan 2045. Lama mengonsumsi obat diabetes di Puskesmas (X) yaitu 5-11 tahun. Rumusan masalah dalam penelitian ini apakah ada hubungan kadar kreatinin dan lama Mengonsumsi obat diabetes pada penderita DM tipe 2 di Puskesmas(X), yang bertujuan untuk mengetahui hubungan kadar kreatinin dan lama mengonsumsi obat diabetes pada penderita DM  tipe 2 di Puskesmas(X). Rancangan penelitian ini menggunakan kuantitatif dengan desain penelitian metode cross-sectional. Populasi data rekam medis bulan Juni 2023 sebanyak  30 sampel pasien DM tipe 2, dengan teknik sampel dalam penelitian ini menggunakan random sampling/probability. Variabel penelitian ini, variabel independen lama mengonsumsi obat diabetes dan variabel dependen kadar kreatinin. Pengumpulan data berupa data sekunder dari catatan rekam medis dan analisis data dilakukan menggunakan SPSS. Hasil nilai kadar kreatinin yang abnormal didapatkan sebanyak 8 penderita dan nilai kadar kreatinin yang normal didapatkan 22 penderita dengan lama mengonsumsi obat sekitar 5-11 tahun. Penelitian ini menunjukkan bahwa kadar kreatinin penderita diabetes setelah mengkonsumsi obat tidak terdapat korelasi yang signifikan p > 0,05 (0,998 > 0,05). Oleh sebab itu dapat diartikan bahwa kadar kreatinin tidak berkorelasi dengan Lama mengonsumsi obat diabetes melitus.

Abstract

Coconut dregs are waste produced after processing coconuts into coconut oil. Coconut pulp has a high nutritional content which can be used for bacterial growth. The nutritional content of coconut dregs is 38.1% carbohydrates, 5.6% crude protein, 16.3% crude fat, 31.6% crude fiber, 2.6% ash content and 5.5% water content. The research used coconut dregs as an alternative medium for the growth of gram-negative and gram-positive bacteria. In the trials, B. subtilis and E. coli bacteria were used. The type of research used was a true experiment with a completely randomized design. The results of the research showed that the highest average number of colonies on the coconut dregs alternative media was in treatment A1, namely the average of B.subtilis colonies was 50.6 x 10² CFU/mL and E.coli 36 .6 x 10² CFU/mL while the control media averaged B.subtilis colonies 75 x 10² CFU/mL and E.coli 86.3 x 10² CFU/mL. The results of statistical tests using anova p<0.05 with a sig value of 0.001 show that there is an influence of differences in the concentration of coconut dregs and agar as an alternative medium for bacterial growth. The post hoc test compares the concentration between treatment A1 and control with a sig p value of >0.05 (0.250), namely There is no significant difference, while the concentration between treatments A2, A3 and A4 with control has a sig p value <0.05, namely 0.000, meaning there is a significant difference. So treatment A1 is the best treatment for the growth of B.subtilis and E.coli bacteria.

Abstract

Soil Transmitted Helminths are parasitic worms found in tropical and subtropical countries that can cause helminthic infections in humans through contact with eggs, parasite larvae spread through the soil. Detection of STH infection is carried out by sedimentation and flotation methods. Garbage carriers are vulnerable to STH infection because they work directly with garbage and are in a sanitary environment that is not clean. The purpose of the study was to determine the results of STH examination on the feces of waste transport officers in Tawangsari Village, Malang Regency. Type of Descriptive Analytical research, conducted in July 2023. The sampling technique uses Total Sampling of a sample of 8 respondents. The results of the sedimentation method found 2 heads of Ascaris lumbricoides, respondents A1, A2 (1tail), the flotation method was not found helmints. The sedimentation method found eggs of Ascaris lumbricoides respondents A1, A7(1 grain) and A2, A3(2 eggs). The flotation method found eggs of Ascaris lumbricoides respondents A2(5eggs), A3(2eggs) and A6(1eggs). The results of the pearson correlation test show a relationship between STH infection and personal hygiene, the use of PPE is 0.875 > 0.01 with a value of r = -0.67 meaning that there is no relationship between personal hygiene and the use of PPE with STH infection means better personal hygiene, the use of PPE while working does not require STH infection, the relationship between STH infection and Environmental Sanitation of 0.000 < 0.01 with a value of r = -1 means that there is a strong relationship inversely proportional between Environmental sanitation with STH infection in waste haulers, means that the better the environmental sanitation, the lower the risk level of STH.

Abstract

Pembuatan preparat awetan permanen meliputi proses fiksasi menggunakaan KOH 10%, dehidrasi, clearing dan mounting. Clearing bertujuan untuk menjadikan struktur morfologi sampel atau objek menjadi lebih jernih, transparan dan jelas.. Pada penelitian ini larutan yang digunakan yaitu larutan Xilene dan Toluena. Xilene merupakan larutan yang tidak ramah lingkungan, harga lebih mahal dan mudah terbakar,. Sedangkan Toluena merupakan larutan yang terbuat dari pohon tolu yang ramah lingkungan, digunakan sebagai minyak bumi mentah, harga terjangkau, Xilene dan Toluena bersifat menjernihkan karena memiliki kandungan karbon yang sama. Penelitian ini bertujuan untuk mengetahui ada atau tidaknya perbedaan kualitas hasil preparat awetan permanen Aedes albopictus (Stegomyia albopictus) pada proses clearing dengan menggunakan larutan Xilene dan Toluena. Sampel penelitian ini adalah 32 nyamuk Aedes albopictus (Stegomyia albopictus) dewasa. Hasil penelitian menunjukkan bahwa preparat awetan menggunakan Toluena didapatkan hasil 12 preparat baik, 4 preparat cukup baik. Sedangkan hasil preparat awetan menggunakan Xilene didapatkan hasil 5 preparat baik, 8 preparat cukup baik, 3 preparat buruk. Pada penelitian ini peneliti menggunakan uji statistik Mann-Whitney dan diperoleh hasil yang signifikan yaitu nilai 0,000 < 0,05. Simpulan, pembuatan preparat awetan permanen Aaedes albopictus (Steggomyia albopictus) lebih baik menggunakan Toluena dibandingkan dengan menggunakanXilene.Kata Kunci: Aaedes Albopictus, Clearing, Toluena, Xilene

Abstract

Dengan semakin berkembangnya institusi pendidikan terutama bidang kesehatan membuat kondisi limbah cair meningkat dari sifat fisik, kimia dan biologi. Suatu laboratorium idealnya mempunyai unit pengelolaan limbah cair, sehingga buangan yang dihasilkan aman bagi lingkungan. Limbah cair yang tidak dikelola dengan baik akan menghasilkan adanya perombakan yaitu bakteri patogen. Tujuan penelitian ini adalah untuk mengetahui gambaran bakteri gram negatif pada limbah cair Laboratorium Klinik STIKes Maharani. Jenis penelitian ini secara deskriptif kualitatif. Teknik pengambilan sampel menggunakan purposive sampling. Tahapan dalam penelitian ini meliputi pengambilan limbah cair sebanyak 250 ml, pembuatan isolasi koloni bakteri dengan media MCA, pembuatan biakan murni media agar miring MCA, pengamatan mikroskopis dan uji biokimia sesuai dengan Bergey manual. Bakteri ditumbuhkan pada MCA 2x24 jam. Hasil penelitian dengan pewarnaan gram menunjukkan bakteri gram negatif, bentuk batang, menyebar, berwarna merah. Uji biokimia menunjukkan glukosa (+), laktosa (-), mannitol (-), maltosa (-), sukrosa (-), H2S (+),indol (+), Mr (+), Vp (-), citrat(+), ditemukan spesies bakteri Pseudomonas aeruginosa. Saran penelitian ini adalah mengidentifikasi bakteri temuan pada limbah menggunakan media agar lainnya untuk mendapatkan bakteri selain gram negatif.Kata kunci: Limbah cair laboratorium, Bakteri gram negatif

Abstract

Soil transmitted helminths are parasitic groups of worms that live in countries with subtropical or tropical climates. In humans, when exposed to parasitic eggs or larvae, they can cause infection. Stool examination is divided into qualitative and quantitative examination. Qualitative are direct examination, flotation, tape, deposition, and thick preparations. Kato-Katz and Stoll are quantitative. The purpose of this study was to compare the results of STH identification using the Formol Ether Sedimentation Method and ZnSO4 Solution Flotation with a total sample of 30 samples using purposive sampling. Data analysis with percentage tables and the Chi Square statistical correlation test The results of the analysis prove that the number of samples infected with soil transmitted Helminth with the Formol Ether Sedimentation Method and ZnSO4 solution flotation is the same. With p value = 0.850, which means there is no significant difference between Formol Ether Sedimentation and ZnSO4 solution flotation.

Abstract

The preparation of preserved preparations is one of the useful methods to determine the morphology of ectoparasites and insects whose manufacturing process goes through four stages starting with 10% KOH fixation, dehydration process, clearing process, and mounting process. The 10% KOH immersion serves to thin out the chitin layer that forms the exoskeleton, which is a thick covering on the body surface of Pediculus humanus capitis (human lice). The purpose of this study was to determine the quality of the preserved preparations of Pediculus humanus capitis by immersing them in 10% KOH at temperatures of 60°C, 70°C, 80°C, and 90°C for 30 minutes using the whole mount method, in order to observe the entire morphology of the species clearly. The sample used is Pediculus humanus capitis adult stage, as many as 24 tails. Observation of the quality of preserved preparations was assessed based on clarity, color quality, and morphological integrity. At a temperature of 60°C, two good qualities are obtained. Three good qualities are obtained at 70oC. At 80°C, it is of good quality, and at 90°C, there is no good quality. The results of the study were then subjected to statistical tests using the Kruskall-Wallis test, which obtained a p value of 0.040<0.05, so that it could be interpreted that there was an influence of variations in heating temperature and 10% KOH immersion on the quality of Pediculus preserved preparations. humanus capitis. namely, by immersing 10% KOH at 70°C for 30 minutes, with complete morphological characteristics, transparent color, and a clear appearance.

Abstract

In the period of growth and development, adolescents need more nutrients. Adolescents face many health problems due to biological and psychological changes as well as malnutrition. Anemia is a common health problem. Riskesdas data in 2018, in Indonesia there is a prevalence of anemia of 48.9%. Cases of anemia often occur in the age group of 15-24 years and 25-34 years. This study aims to determine the state of anemia with a lean body mass index in adolescent girls at SMK Madani Turen, Malang Regency. The research design used is descriptive quantitative with a cross sectional approach and a total of 101 respondents. The samples obtained had a lean body mass index of 24 people (23.76%) and the majority were known not to be anemic, namely 17 people (70.83%) and those who had anemia were 7 people (29.16%). The p value of the Fisher's exact test is 0.000 if the p value is <0.05, which means Ho is rejected, it can be concluded that there is a relationship between body mass index and Hb levels in female students at SMK Madani Turen.
