Abstract

Preservation of ram semen has an impact on the exposure of artificial conditions to the spermatozoa which was followed by overproduction of reactive oxygen species (ROS) and resulted in functional damages of spermatozoa. This study aimed to determine the effect of the combination of chicken, quail, and duck egg yolks with green tea extract (GTE) on the quality of Sapudi ram spermatozoa at 5°C storage. Ejaculates of Sapudi ram with more than 70% spermatozoa motility were divided into four groups. Semen were extended 1 : 10 in citrated-chicken egg yolk extender (control group, T0), and citrated yolk of chicken (T1), quail (T2), and duck (T3) egg added with 0.05 mg GTE per 100 mL of extender. Extended semen was kept in a tube, which was put in a beaker containing distilled water and stored in a refrigerator (5°C). Spermatozoa motility, viability, and plasma membrane integrity (PMI) were evaluated daily for five days. The results showed that GTE in duck egg yolk citrate extender could maintain the highest spermatozoa motility, viability, and PMI for five days (p <0.05). It could be concluded that the duck egg yolk citrate extender with the addition of GTE (Camellia sinensis) was beneficial for maintaining the semen of Sapudi ram spermatozoa at chilled temperatures. Further study is needed to use the extender for freezing Sapudi ram semen.

Abstract

This study aims to determine the motility and viability of kampung rooster (Gallus domesticus) spermatozoa in 5% Dextrose Ringer's solution as an extender with or without the addition of various egg yolk concentrations at 5ºC during storage. Semen was collected by massage in the morning. In this study, four types of extenders were used, i.e., 5% Dextrose Ringer's extender without egg yolk, 5% Dextrose Ringer's extender with 5, 7.5, and 10% egg yolk for T0, T1, T2, and T3, respectively. Then, 1000 IU Penicillin and 1 mg Streptomycin were added per mL extender. Fresh semen from each rooster was evenly divided into four volumes to be diluted 11 times in each group extender. Extended semen from all groups was stored at 5°C. Data were analyzed using ANOVA and Duncan's new multiple range test. The result showed a significant difference (p <0.05) between each treatment group for spermatozoa motility and viability. In conclusion, this study revealed that the addition of various concentrations of egg yolk into 5% Dextrose Ringer's solution as an extender maintained the motility and viability of kampung rooster spermatozoa.

Abstract

Predicting the turtle's reproductive status and sexual maturity is necessary for ex-situ breeding and conservation program. The histology of the seminiferous tubules of the Indonesian box turtle (Cuora amboinensis couro) at various ages has not been studied much. This study aims to develop equations to predict the sexual maturity of Indonesian box turtles based on straight carapace length (SCL). Six turtles with SCL sizes 10-21 cm were sacrificed and their testes were taken for morphometric measurements and histological preparations for Hematoxylin Eosin staining. Pearson's correlation of SCL with testicular maturity parameters was analyzed, followed by Anova Regression. The results showed that there were differences in the morphometry and topographic anatomy of the Indonesian box turtle testes between SCL below 15 cm and above 15 cm. Six turtles with SCL size of 10-21 cm were sacrificed and their testes were taken for morphometric measurements and histological preparations for Hematoxylin Eosin staining. Abstract

Lead is a harmful pollutant from engine exhaust that causes free radicals and has detrimental effects on the testicular tissue. This study aimed to determine the effects of tomato juice on the number of Leydig cells and the diameter of the seminiferous tubules of mice exposed to lead acetate. Twenty-five male mice were divided into five groups. Mice in the control (C-) group were given placebos. Meanwhile, mice in C+, T1, T2, and T3 groups were exposed to lead acetate at a dose of 100 mg/kg BW/day for 14 days and given tomato juice respectively at 0, 0.16, 0.32, and 0.64 mL/day from day 8 to day 35. On day 36, all mice were sacrificed, and the testes were collected for histological preparation. The result showed that lead exposure in the C+ group decreased (p <0.05) in the number of Leydig cells and the diameter of the seminiferous tubules compared to the mice in group C-. Administration of tomato juice in groups T1, T2, and T3 increased (p <0.05) the number of Leydig cells and the diameter of seminiferous tubule compared to the mice in the C+ and C- groups. However, tomato juice administration to the T3 group decreased the number of Leydig cells and the diameter of the seminiferous tubules (p <0.05) compared to the T2 group. In conclusion, an effective dose of 0.32 mL/day of tomato juice restored Leydig cell number and seminiferous tubules diameter in mice exposed to lead acetate.

Abstract

Cigarette smoke chemicals caused oxidative stress by increasing Reactive Oxygen Species (ROS). ROS exposure could be neutralized by antioxidants, such as green tea (Camellia sinensis) leaf extract. This study aimed to determine the effect of green tea extract (GTE) on spermatozoa plasma membrane integrity, spermatozoa viability, and spermatozoa motility of mice (Mus musculus) exposed to cigarette smoke. Twenty-five mice were randomly divided into five groups. Group C- mice were given a placebo (1% Sodium carboxymethyl cellulose, Na-CMC). Group C+, T1, T2, and T3 mice were exposed to cigarette smoke and given 0, 20, 40, and 60 mg/kg BW GTE respectively. Cigarette smoke exposure used a clove cigarette per day. GTE in 1% Na-CMC solution was administered at 0.5 mL orally using a gastric probe. The treatment was conducted daily for 36 days, and on day 37, all mice were euthanized for spermatozoa evaluation. The results showed that all parameters evaluated in the C+ group were lower (p <0.05) than in C- group. Administration of GTE in the T2 group increased (p <0.05) all parameters compared to mice in the C+ group. However, administration of GTE to mice in the T3 group caused a decrease (p <0.05) in all parameters than those of the T2 group and was not significantly different (p >0.05) compared to those of the C+ group. It could be concluded that the administration of GTE at 40 mg/kg BW has maintained the spermatozoa plasma membrane integrity, spermatozoa viability, and spermatozoa motility of mice exposed to cigarette smoke.

Abstract

Accurate determination of estrus status in an animal based on an appropriate estrus detection method can guarantee the cows' pregnancy after mating. This study aims to determine the estrus status of Aceh cows using the van Eerdenburg estrus behavior scoring method. This study was conducted from 22 January to 12 February 2021, at the Technical Implementation Unit for Animals of the Faculty of Veterinary Medicine, Universitas Syiah Kuala, Banda Aceh, Indonesia. The sample used was eleven Aceh cows aged 4-6 years old. Parameters observed were the number of cows exhibiting signs of estrus using the estrus behavior scoring method. The accuracy of estrus detection was confirmed by palpation per rectum to detect the presence of uterine tone. By observing signs of estrus using the method studied, 90.9% (10/11) of the cows were determined to be in estrus. Confirmation by the presence of uterine tone showed a 100% accuracy rate. The most effective sign of estrus was the passivity of the cow when being ridden by another cow or bull. This study concluded that estrus status in local cows could be determined using the estrus behavior scoring of the van Eerdenburg method.

Abstract

Peripheral progesterone titers characterizing the reproductive cycle of the bitch are fairly well-documented in literature. Progesterone titers obtained via fluorescence-immunoassay technique at different points to the appearance-of-bloody-vulva-discharge (ABVD) in apparently healthy, exotic, multi-breed bitches (n = 76) that were subsequently inseminated were summarized using descriptive statistics. The recommended/predictable progesterone titer-ranges- PPTRs (ng/mL) and modality for insemination(s) were: 7.6-8.5 (inseminate-after-24-hours), 8.6-9.9 (inseminate-after-12-hours) and 10.0-20.0 (inseminate-immediately). The length of days from ABVD to the point of obtaining a PPTR (Mean ± SD) was 12.43 ± 3.38 days (range 8-17 days). The frequencies of progesterone assay per bitch were: one-fold (39.47%), two-fold (39.47%). three-fold (18.43%) and four-fold (2.63%). Observations on pregnancy statuses of bitches based on frequency of progesterone assay(s) were: 57.90% (pregnant, multiple-assay), 5.26% (pregnant, one-fold-assay), 26.32% (not-pregnant, multiple-assay) and 10.52% (not-pregnant, single-assay). The litter sizes at the PPTRs (Mean ± SD) i.e., 7.83 ± 1.86 (7.6-8.5 ng/mL), 7.86 ± 1.73 (8.6-9.9 ng/mL) and 8.83 ± 1.86 (10.0-20.0 ng/mL) were comparable with that for the entire study i.e., 8.16 ± 1.87. Similarly, the range of litter sizes at the three PPTRs as well as that for the entire study were same i.e., 5-10 puppies. It is concluded as follows: multiple progesterone assays resulted in higher pregnancy outcome and, litter sizes were not affected as long as insemination(s) occurred within the recommended progesterone titer-ranges. We recommend a comprehensive investigation of all pregnancy determinants, even for apparently healthy dogs, by dog-lovers and, regarding progesterone, multiple-assays, rather than one-fold assay, will upscale their breeding business.

Abstract

This study aims to determine the effect of adding melon (Cucumis melo L.) juice to Lactated Ringer's-egg yolk extender on the intact plasma membrane (IPM) and morphological abnormalities of native rooster spermatozoa. Five ejaculate each from two native roosters (Gallus gallus domesticus) were divided into four equal volumes. Ejaculate was extended 1:10 (v/v) in Lactated Ringer's-egg yolk without or with the addition of of 2, 4, and 8% (v/v) melon flesh juice, respectively for groups T0, T1, T2, and T3. Extended semen was stored in a refrigerator (5°C) and evaluated every two hours. The results showed that IPM of all groups decreased (p <0.05) consistently, lasting more than 40% for six hours in the T0 and T1 groups and four hours in the T2 and T3 groups. The percentage of spermatozoa morphological abnormalities in all groups stored for six hours was higher (p <0.05) than before storage. However, it was not significantly different (p >0.05) from that which had been stored for two and four hours. It was concluded that native rooster semen extended in Lactated Ringer's-egg yolk without (group T0) or with 2% melon flesh juice (group T1) and stored at 5°C retained spermatozoa plasma membrane integrity of more than 40% and morphological abnormalities of less than 20% for six hours. While the addition of 4 % (T2) and 8% (T3) melon flesh juice maintained the percentage of IPM and spermatozoa morphological abnormalities for only four hours.

Abstract

Yoruba Ecotype Chicken (YEC) is characterized by hardiness, and better adaptation to prevailing tropical environment. However, its poor egg production has hindered its utilization for commercial production. Crossbreeding is a major tool used in improving desired traits in animals. The objective of the study was to evaluate egg production and quality characteristics in YEC and its crosses with Goliath (GG) and Sussex (SS). The study investigated egg production and egg quality characteristics using 120 sixteen-week-old YEC and its crosses. The findings of the study showed that egg parameters were significantly higher (p<0.05) and maturity occurred earlier (154 days) in YECxSS than in YECxYEC (146 days) and other YEC crosses. The study concluded that cross breeding improved egg production of YEC. Crossbred YEC x SS is therefore recommended for egg production in Nigeria.

Abstract

Bali cattle have advantages in terms of adaptation and fertilization rates, but tend to have slow growth rate because the rearing system is still grazing so that livestock growth becomes uncontrollable. This study aims to determine the peak of mating and calving estimation of Bali cattle (Bos sondaicus) that did not experience reproductive disorders, aged 4-7 years, belonging to smallholder farmers. The cattle were reared in semi-arid area of Netemnanu village, South Amfoang district, Kupang regency, Nusa Tenggara Timur. The method used was interviews with farmers, diagnosis of pregnancy through rectal palpation and ultrasonography. The results showed that the pregnancy rate was 86.20% (50/58).  Based on pregnancy data, it was known that the peak of mating would occur in December 2021 and estimated calving would reach its peak in September 2022.
