Abstract

The study was conducted to know conception rate and service per conception of Limousin Cross breed and Ongole Cross Breed in Pilangkenceng, Madiun. Data were collected for the periode of Oktober 2016 to February 2017 from inseminators in Pilangkenceng district. The data were taken by primary and secondary. Primary data retrieval were done by direct observation, which includes several variables, where the variables include: the identity of the breeder, feeding and drinking. The secondary data were obtained by recording the farmer's artificial insemination card. All data was analyzed to find a percentage of conception rate (CR) and find number of service per conception (S/C). Then S/C were analyzed with T-test adn CR were analyzed with chi-square test. The results of this research showed that the service per conception's average of limousin cross breed and ongole cross breed is 1,61 and 1,72. The average of conception rate limousin cross breed and ongole cross breed is 53% % and 59 %. The result showed no significant difference (p>0,05) on CR and S/C between limousin cross breed and ongole cross breed. The conclusion of this research showed that service per conception and conception rate of ongole cross breed in Pilengkenceng Madiun is better than limousin cross breed but there's no significant difference (p>0,05).

Abstract

This research was conducted to find out the Body Condition Score (BCS, in nine scale) of 30 Limousine crossbred cows in Balongpanggang, Gresik regency at the time of  Artificial Insemination,  21 and 60 days later. Questionnaires obtained to identify the knowledge of farmers and inseminators in estrous detection. Conception Rate (CR) and Service per Conception (S/C) counted based on pregnancy diagnosis through rectal palpation 60 days post insemination. The result showed that the average of BCS in pregnant cows at 60 days post insemination was highest compared to the others. Conception rate and service per conception of the cows were 60% and 1.63 respectively, meanwhile the knowledge of estrous detection of farmers was lower compared to those of inseminator. It could be concluded that implementation of AI technique in Limousine crossbred cows at Balongpanggang, Gresik regency was good enough, but the knowledge of farmers about estrous detection needs to be improved.

Abstract

Cross-breeding between Madura cattle and exotic breeds (exotic cows) occurs in Madura area. The reality in the field shows that the cross-breeding occurs between Limousin cow and Madura cow and the result of the breeding is called as Madrasin cow. One of factor that influences the animal's growth is Growth Hormone Receptor (GHR).  GHR is needed by Growth Hormone (GH) to carry out its effects on target tissues. The purpose of this study is to identify the profile of Growth Hormone Receptors in Madrasin cow. This research was conducted on farmers in Bangkalan, East Java and the Center of Veterinary, Denpasar in MayJune 2017. This study used 14 cows Madrasin. The samples of this study were blood which were taken and used for analyzing DNA that includes DNA isolation, DNA amplification by using Polymerase Chain Reaction (PCR) and Restriction Fragment Length Polymorphism (RFLP) method to determine the genotype. An identification of Growth Hormone Receptor gene polymorphism is done through digesting the DNA fragment of 298 bp by the restriction enzyme AluI. The results shows that the frequency of Growth Hormone Receptor Gene of  cow Madrasin allele A was 0.92 and the G allele was 0.08 with frequency of the A allele is higher than G allele.The genotype changing was found in cow Madrasin as much as 0.85 to 0.14 for the AA and AG. As a result of the study, the researcher concluded that Growth Hormone Receptor gene experienced a changing or polymorphisms in Madrasin cow and information which stated in this study be able to used as a basic for selection.

Abstract

The purpose of this research is to determine the effect of mucos acidity of vaginal mucous when estrus towards conception rate at KUD Tani Wilis Tulungagung District and KSU Tunas Setia Baru Pasuruan District. Measured samples were taken from 30 dairy cattles in each KUD. Mucos acidity of vaginal mucous were obtained by using a digital acidity meter before insemination was done on dairy cattle. After measuring the acidity from mucosal vagina mucous, artificial insemination has done on estrus dairy cattle. On the 60th day after insemination, rectal palpation was done to check the conception rate. Research result from 30 dairy cattle on each KUD showed that mucos acidity of vaginal mucous has signifficant effect towards conception rate. Mucos acidity of vaginal mucous range 6.5-8.5 with the highest range of conception rate in KSU Tunas Setia Baru Pasuruan District 7.5-7.7 and in KUD Tani Wilis Tulungagung District with the highest range of conception range 7.2-7.4 and 7.5-7.7.

Abstract

Aims of this research was to determine the effect of distance and the duration of straw count above liquid nitrogen to the Quality of Frozen Simental Cattle. This study used a sample derived from simmental cattle from BBIB singosari as much as 48 straw divided into 8 treatments. P1(0,5cm 15 sec), P2(0,5cm 30 sec), P3( 0,5cm 45 sec), P4(0,5cm 60 sec), P5(1,5cm 15 sec), P6(1,5cm 30 sec), P7(1,5cm 45 sec), P8(1,5cm 60 sec) and repeated as much as 6 times then observed for motility and viability of post thawing spermatozoa. The data obtained were analyzed using SPSS 16.0. The data obtained were analyzed using SPSS 16.0. The results showed that the duration of straw count and the position of straw from liquid nitrogen  not significant (P>0,05) on post thawing motility. The position of straw from liquid nitrogen not signifant (P>0,05) on post thawing viability. The conclusion of this research showed that 0,5 cm 15 sec give the higher post thawing motility.

Abstract

The aim of this research was to know the correlation between heart girth, body length, and wither's height with semen volume and sperm concentration of Madura Bulls. Ten Madura bulls aged three to four years from Madura  were measured. Variables observed were heart girth, body length, wither's height, semen volume and sperm concentration. Data were analyzed by multiple linear regression analysis. The result of this research showed that there was no correlation between heart girth, body length, and wither's height with semen volume Y = 25,212 + 0,190 X1+ (-0,131) X2 + 0,136 X3, so as there was no correlation between heart girth, body length and wither's height with sperm concentration Y = 8739,559 + (-36,663) X1+ (19,714) X2 + 6,2960 X3. And than was no correlation between semen volume and sperm concentration Y = 7,303 + (-0,002) X1.

Abstract

The aim of this study was to find out protein tyrosine kinase profile in Merino Sheep seminal plasma. This study consist of collecting semen of Merino Sheep containing plasma and spermatozoa, the separation between seminal plasma and spermatozoa, then tyrosine kinase analysis using Sodium Dodecyl Sulphate Polyacrilamide Gel Electrophoresis (SDS-PAGE). Semen was collected by artificial vagina and then centrifugated for 40 minutes at 4000 rpm to separated seminal plasma and spermatozoa. Protein was analyzed using SDS-PAGE, to separate each protein based on their molecular weight. The result showed that there were 13 protein bands in 3 seminal plasma samples with an average 149,63 kDa, 139,7 kDa, 114,97 kDa, 109,3 kDa, 97,33 kDa, 93,83 kDa, 86,23 kDa, 77,6 kDa, 64,6 kDa, 52,3 kDa, 41,93 kDa, 38,13 kDa, and 34,5 kDa. In conclusion, it is believed that tyrosine kinase located on the sixth band with molecular weight 93,83 kDa.

Abstract

The aim of this research was to show the influence of seminiferous tubules histopathology of mice (Mus musculus) caused Fusarium graminearumexposure. This research was done in April-May 2017 in Microbilogy Laboratory Faculty of Science and Technology, Animal Laboratory, and Microbilogy Laboratory Faculty of Veterinary Medicine UniversitasAirlangga. This research used 20 male mice (Mus musculus) aged 6 weeks with 18-30 gram body weight. The mice devided into four groups: P0 given 0,25 ml Sodium chloride without Fusarium graminearum exposure by oral; P1 given 0,25 ml Fusarium graminearum exposure with dilution 102 by oral; P2 given 0,25 ml Fusarium graminearum exposure with dilution 103 by oral; and P3 given 0,25 ml Fusarium graminearum exposure with dilution 104 by oral. This treatment carried out for 21 days. Each milliliter dilution containing 228x106 spore for P1, 228x107 spore for P2, and 228x108 spore for P3. Then do the surgery and harvesting the testes then performed histopathological examination by scoring of seminiferous tubules. For data analyzing used non parametric difference Kruskall-Wallis and continued with Mann-Whitney. The result of this research was showed that decreased the spermatogenic cells in the process of spermatogenesis significant.

Abstract

This study aimed to determine the relationship between heat detector reading results and the feature of vaginal swab preparations as a parameter for determining estrus in Peranakan Etawa goats. Five healthy non-pregnant goats weighing 20-30 kg, were synchronized using PGF2α (7.5 mg Dinoprost, Enzaprost) intramuscularly twice with 11 days interval. Vaginal cytology examination began the day after the second PGF2α injection. The heat detector (Draminski®, DramiÅ„ski SA, Poland) is an estrus detecting device based on the amount of electric current flowing through the vaginal mucus. Spearman correlation analysis showed a relationship (p <0.05) between heat detector reading and estrus detection based on vaginal cytology. The heat detector readings were very significantly correlated positively (p <0.01) with the number of epithelial cells (coefficient correlation, r= 0.564), but negatively correlated with the number of cornified cells (r = –0.854), and also significantly correlated positively (p <0.05) to the number of leukocytes (r = 0.487). The estrus stages were significantly correlated positively (p <0.05) only to the number of leukocytes (r = 0.918). Likewise, the number of epithelial cells was highly correlated negatively (p <0.01) only to cornified cells (r = –0,643), and the number of cornified cells was significantly correlated negatively (p <0.05) only to the number of leukocytes (r = –0.472). It could be concluded that the features of the proestrus, estrus and metestrus stages based on the goat vaginal cytology was correlated with the reading results of the heat detector.

Abstract

The purpose of this research was to determine the effect of pomegranate (Punica granatul l) fruit extract for spermatozoa quality including the motility, viability and membrane integrity of spermatozoa. The researchused 20 male white rats  (Rattus norvegicus) with 200-300 gram average body weight. P0 as control was given with CMC Na 0.5%.  P1 used 75 mg/kg BW dose of extract,  P2used 150 mg/kg BW dose of extract,  P3 used 300 mg/kg BW dose of extract. The treatment was given perorally for 14 days. The experiment was used completely random design (CRD). The result of this study was analyzed with ANOVA followed by Tukey Test (p<0.05). The result showed that there was significant effect from therapy on the quality of rats spermatozoa. The difference of spermatozoa's concentration between first treatment group P1 and control treatment group is not significant, while the treatment other result showed between the second treatment group and the control group P0. A significant difference occurs in the percentage of motility, viability and  membrane integrity. The treatment from P2 group showed the best result in maintaining the quality of rats spermatozoa.
