Abstract

Repeat breeding is a reproductive disorder that harms dairy farmers because of the unachieved target to obtain one calf per year and decreased milk production. The repeat breeding syndrome can be caused by malnutrition. This study aimed to determine the effect of feeding silage, complete feed, and growth promoters in repeat breeder cows to restore the services per conception (S/C) and daily milk production. Thirty repeat breeder Holstein Friesian cows were divided randomly into three groups. The control group (P0) was fed 30-40 kg forage and 3-4 kg concentrate/head/day. Group P1 was given 8-10 kg silage or complete feed alternately every day; whereas P2 was given the same as for the P1 group, added with 10 ml growth promoter/head/day. Milk production and S/C were measured after six months of treatment. The results showed that the P1 and P2 groups returned S/C to normal (1.40 ± 0.52 and 1.30 ± 0.48), while the S/C of P0 group was 3.20 ± 0.63 (p <0.05). Daily milk production in the P1 and P2 groups (14.20 ± 1.40 and 14.60 ± 1.43 l/d) was also higher (p <0.05) than the control group (8.30 ± 1.06 l/d). It could be concluded that feeding silage and complete feeds alternately daily, with or without the addition of growth promoter, improved the S/C to the normal range and increased milk production.

Abstract

Artificial Insemination is used to conserve genetic resources. It is necessary to preserve spermatozoa in a certain diluent at certain temperature before artificial insemination. Information regarding the use of honey in the spermatozoa diluent is still less in number. This research aims to determine the effect of Sumbawa honey and processed honey in various concentrations on the quality of spermatozoa after storage in refrigerator. The main materials used were five ram testicles. Spermatozoa was collected from cauda epididymis by mincing and adding 5 ml PBS. The sample were divided into 7 groups of treatment: 1% Sumbawa honey, 2% Sumbawa honey, 3% Sumbawa honey, 1% processed honey, 2% processed honey, 3% processed honey, and control using 0.5 gram glucose in 100 ml Tris diluent. The assessment was carried out microscopically including motility and viability. Data were analyzed statistically using one-way analysis of variance (Anova) on the SPSS statistics 16.0 program. The results showed no significant difference (p > 0.05) in the motility and viability of spermatozoa in all treatment of Sumbawa honey (1%, 2%, 3%), processed honey (1%, 2%, 3%), and control. The conclusion of this study is that honey in spermatozoa diluent has no effect on the quality of spermatozoa.

Abstract

The process of spermatogenesis is sensitive to nicotine, a main component of cigarette smoke. This study aimed to observe the effect of nicotine injection on motility, viability, and membrane integrity of the spermatozoa of mice (Mus musculus). The mice were 10 weeks old, weighing about 20 grams, and in healthy and fertile conditions. Mice were adapted for 1 week by being given feed in the form of pellets and drinking water every day. Twenty of the mice were divided into four groups. Group K as a control was injected with 0.9% NaCl at 2.5 mg/Kg bw. Group P1 was injected with nicotine at 2.5 mg/Kg bw. P2 was injected with nicotine at 5 mg/Kg bw. Group P3 was injected with nicotine at 10 mg/Kg bw. The injection was carried out subcutaneously as much as 0.5 ml once a day for 35 days. Data analysis used the Analysis of Variances (ANOVA), followed by Duncan's Multiple Range Test. The results showed that the viability, motility, and membrane integrity of the spermatozoa decreased significantly (p <0.05) according to the increase in the dose of nicotine injected. It could be concluded that subcutaneous nicotine injection linearly decreased the motility, viability, and membrane integrity of the spermatozoa of mice.

Abstract

This study was conducted to determine the relationship of birth weight of calves, sex ratio of the calves, and the age of the dam to the length of pregnancy in buffaloes resulting from artificial insemination in Kampar District. The method used in this research is the descriptive method by displaying the average and standard deviation. The total number of buffaloes that were artificially inseminated in 2018 and gave birth in 2019 was 21 buffalo-cows. Data were analyzed with the Pearson Product Moment (PPM) correlation. The parameters measured in this study were the length of pregnancy, calf birth weight, calf sex ratio, and dam's age. The results showed a positive correlation between the birth weight of the calves, sex ratio of the calves, and the age of the dam to the length of pregnancy of 1.4%, with an average length of pregnancy, calf birth weight, and sex ratio of 351.81 days, 27.38 kg, and 1: 1 respectively.

Abstract

High temperatures lead to oxidative stress, which can disturb spermatogenesis process. Watermelon (Citrullus lanatus) peel contain antioxidant expected to compensate oxidative stress due to heat stress exposure. This study aimed to determine the effect of watermelon rind ethanolic extract on the number of Leydig, Sertoli, and spermatogenic cells of rats exposed to heat (40°C). Twenty rats (Rattus norvegicus) were divided randomly into five groups. In the control group (T0) rats were not exposed to heat nor given the watermelon rind extract. T1, T2, T3, and T4 groups were exposed to heat for an hour daily and orally given placebo (1% Na CMC), 100, 200, and 400 mg/kg BW of watermelon rind extract (in 1% Na CMC). Rats were treated for 52 days, and sacrificed for the testicle collection. Hematoxylin-eosin stained histological slides were prepared for the examination of Leydig, Sertoli and spermatogenic cells. The results showed no significant difference (p >0.05) in the average number of Leydig cells in rats among groups. The number of Sertoli cells and spermatogenic cells of rats exposed to heat (T1) was lower than those of the normal rats (T0 group). The dose of watermelon rind ethanolic extract at 200 mg/kg BW (T3 group) and 400 mg/kg BW (T4) increased (p <0.05) the number of Sertoli and spermatogenic cells. It could be concluded that a dose of 400mg/kg BW of watermelon rind ethanolic extract maintained the number of Leydig cells, Sertoli cells, and spermatogenic cells of rats exposed to heat.

Abstract

This study was conducted to determine the relationship between the estrous detector's reading value (Draminski®, DramiÅ„ski S.A, Poland) and serum progesterone levels on Etawa crossbred does. Eighteen healthynon-prehnant Etawa crossbreds does weighing 20-25 kg, in their first parity were used in this study. The does were divided equally into two groups. In the two groups, the does were synchronized their estrus by means of intramuscular and intravulvar injection of 7.5 mg prostaglandin FÉ‘ (PGF2É‘) twice in 11-day intervals. After the second injection, the estrous detector value measurement, visual observation of the estrus sign, and blood sampling were conducted for five days. The blood sample was processed into the serum to examine for progesterone levels by ELISA. The data analysis was using the simple correlation method on SPSS type 23 for windows. The result showed that a positive relationship (p <0.05) with coefficient correlation (r) was 0.609 between the values of the estrous detector measurement with serum progesterone levels. It could be concluded that the estrous detector's low value indicates that the Etawa crossbred was in the estrus phase and followed by the decreasing of progesterone level but not reaching the basal level.

Abstract

This study was aimed to determine the effect of urea in maturation medium on in vitro oocyte maturation rate. The medium used was TCM-199 added with Hepes, NaHCO3, Kanamycin 0.15 IU/mL, PMSG, 0.15 IU/mL hCG, and 10% FBS. Cumulus oocyte complexes (COCs) of cows derived from follicle aspiration were divided into three groups. In control group (P0), the COCs were matured in vitro in a maturation medium without urea addition, meanwhile in the P1 and P2 groups, the medium was added with urea 20 and 40 mg/dL, respectively. Each petri dish contained three drops of maturation medium (300 µl/drops) according to the groups. Microdrops were coated with mineral oil and then incubated in a 5% CO2 incubator, at 39 ËšC with maximum humidity. Aceto-orcein staining was conducted to evaluate the maturation of oocytes based on the achievement of metaphase II phase that is indicated by the presence of metaphase plate and/or first polar body. The result showed that the oocyte maturation rates of P0, P1, and P2 were 51.25, 52.43 (p >0.05), and 46.88 % (p <0.05) respectively. It could be concluded that the presence of urea at 40 mg/dL in maturation medium reduced the percentage of bovine oocyte maturation in vitro.

Abstract

This study aimed to determine the progesterone concentrations of Ettawa crossbred does at day-0, -21, -42, -63, and -84 after mating. Five Ettawa crossbred does were injected with PGF2α twice with an interval of 11 days for estrous synchronization followed by natural mating on day-3 after the second PGF2α injection. Blood was collected through the jugular vein for the measurement of serum progesterone concentrations using ELISA, whereas pregnancy was diagnosed at 90 days after mating by abdominal palpation. The results showed that all does were on estrus onthethird day after the second PGF2α, three of them diagnosed to be pregnant following mating. The progesterone concentration at day-0 was 1.52 ± 0.33 ng/mL, increased (p <0.05) to 7.08 ± 1.02 and 5.50 ± 2.82 ng/mL at day-21 and day-42, and increased (p <0.05) again to 12.01 ± 5.30 and 12.04 ± 0.30 ng/mL at day 63 and day 84, respectively. There was no significant difference in progesterone concentrations between day 21 and day 42 and between day 63 and day 84. It could be concluded that there was a biphasic increase of progesterone concentrations of the pregnant Ettawa crossbred does, i.e. from day-0 to day-21/day-42 and from day-21/day-42 to day-63/day-84.

Abstract

This case study aims to determine the effect of a combination of prostaglandin F2α (PGF2α) with Gonadotropin (PG-600), and human chorionic gonadotropin (hCG) on the estrus and pregnancy rates in Holstein Friesian (HF) cows with persistent corpus luteum (PCL). A total of 15 Holstein Friesian (HF) dairy cows aged 3-5 years with body condition score (BCS) 5-7 (scale 1-9) diagnosed with PCL, were injected with 7.5 mg PGF2α intra-vulvar submucosa. Cows were then divided into three treatment groups. Group P0 was not given the combination treatment, while P1 group was injected with a  combination of 300 IU PG-600 and 150 IU hCG, meanwhile P2 group was was injected with a combination of 300 IU PG-600 and 300 IU hCG intra-muscularly. The PGF2α and PG-600 injections were carried out on the same day PCL was diagnosed. Meanwhile, the injection of hCG was carried out at the same time as the AI. The Pregnancy examination was performed 90 days after AI through rectal palpation. The results showed that all treated cows were estrus  observed with the fastest onset of estrus was the P2 group, that was significantly different (p <0.05) compared to the P0 and P1 groups. All treated cows were diagnosed as pregnant. It can be concluded that the administration of PGF2α or its combination with gonadotropins in HF cows with PCL could result in 100% estrus and pregnancy rates.

Abstract

This study aimed to determine the effect of binahong (Anredera cordifolia) leaf extract on the recovery of the estrous cycle of mice (Mus musculus) which has been induced for the extension of the estrous cycle using angsana (Pterocarpus indicus) bark extract. In this study vaginal smear was performed daily at 10.00 WIB to determine the phase of the estrous cycle for 26 days. Mice were divided into 5 groups, each with 6 replicates. The treatments were T1 (positive control), T2 (negative control), T3 (0.8 mg/head/day), T4 (2.4 mg/head/day) and T5 (4 mg/head/day). The data obtained were processed using Analysis of Variance (ANOVA) and followed by the Duncan's New Multiple Range Test (DNMRT) at a 95% level of significance.  The results showed that the angsana (Pterocarpus indicus) bark extract adminsitration (T2) lengthen (p <0.05) the estrous cycle of mice compared to those of normal mice (T1). The administration of binahong (Anredera cordifolia) leaf extract on T3 and T4 groups mice were followed by shorter (p <0.05) estrous cycle than those of T2, and it were not significantly different (p >0.05) compared to T1 group. However, the higher dose of binahong (Anredera cordifolia) leaf extract (T5) revealed the length of estrous cycle was not significantly different (p >0.05) compared to T2 group. It can be concluded that the administration of binahong leaf extract was able to recover the normal estrous cycle of mice.
