Abstract

Drug interactions can occur in the use of two or more drugs simultaneously, including the use of synthetic drug with herbal medicine. Combination of glibenclamide with noni juice (Morinda citrifolia Linn.) often used by diabetic patient to decrease their blood glucose level. lhe aim of this research was to know the interaction between glibenclamide and noni juice administration on blood glucose level. This research used 24 Sprague-Dawley male rats which were divided into 6 groups. Before the experiment, the rats were first induced by alloxan, except group 1, which was the normal control. Group 2 was the control of diabetic without given any drugs. Group 3 and 4 were the control of glibenclamide (0.9 mg/200g body weight of rat) and of noni juice (2.5 ml/200g body weight of rat) respectively. Group 5 and 6 were the interaction test group which were given glibenclamide (0.9 mg/ 200 g body weight rat) and noni juice (2.5 ml or 5.0 ml/200 g body weight of rat) with an hour interval. The blood glucose level was measured by using spectrophotometer with o-toluidine as reagent. The result of this research shows that noni juice at a dose 5.0 ml/200 g body weight rat was able to increased the reduction of blood glucose levels by glibenclamide after two weeks administration

Abstract

Archaea have phytanyl ether lipids which is one of the characteristics that separates them from bacteria and eukaryotes. Some archaea have also unique membrane spanning tetrae ther lipids (T EL); in Sulfolobus and Ihermoplasma species these TELmake up the majority of total membrane lipids. Archaeal lipids are able to form stable liposomal structures, both from membrane factions, mainly the polar membrane fraction, i.e. archaeosomes, or from highly purified TEL (tetraether lipid liposomes). Liposomes of the main polar lipid (MPL) from thermoacidophilic archaeon Ihermoplasma acidophilum were thoroughly inevstigated. Archaeosomes and TEL liposomes exhibit extremely low proton permeability and high stability at low pH, which makes them suitable to delivery of therapeutics and vaccines via the gastro-inetestinal route. Liposomes from egg lecithin can be stabilized by incorporation of 11-12 mole% MPL. MPL liposomes show long-term shelf stability even at high temperatures without conservation. The size of MPL liposomes between 100 nm (unilamellar) and several gm (multilamellar) depends mainly on the method of preparation. Toxicity and mutagenicity have not been detected in toxicological screening. This is a review report on the lipid from archaea to be developed as an alternative to conventional liposome for various industrial applications, especially for pharmaceutical industry.

Abstract

alpha-Glucosidase inhibitor has known to be a therapeutic agent for diabetes mellitus (DM) treatment, especially type 2 DM. Based on previous studies. There are various plants that have the effect of inhibiting the activity of a-glucosidase, one of which is garu leaves (Antidesma montanum Blume). This research aimed to get the fraction which had the highest Il-glucosidase inhibiting activity from ethanol extract of garu leaves and identify the chemical compounds from the most active fraction. Simplisia powder was extracted by maseration using 80% ethanol then fractionated using n-hexane, ethyl acetate, and methanol. Inhibitory activity test was performed by measuring absorbance of p-nitrophenol, which produced by reaction between Il-glucosidase and p-nitrophenyl-u-l)-glucopyranoside, using microplate reader at 405 nm. The result showed that ethyl acetate fraction have the best Il-glucosidase inhibitory activity with IC50 values 138.38 ppm. The test of enzyme kinetics showed that ethyl acetate fraction inhibited competitively. The phytochemical screening showed that ethyl acetate fraction of garu leaves contained glycosides, tannins, and terpenes.

Abstract

Delima (Punica granatum L) merupakan salah satu buah memiliki aktivitas antioksidan yang kuat karena mengandung senyawa flavanoid dan tannin seperti asam elagic, asam gallat, punicalin, punicalagin, anthocianin, elligatanin, gallotanin, kuersetin, katekin. Senyawa—senyawa ini diketahui dapat mencegah dan menghambat terbentuknya radikal bebas yang penyebabkan penuaan dini dan penyakit kronis. Dalam penelitian ini ekstrak kulit buah delima diformulasikan dalam bentuk krim yang dibedakan kandungan nya yaitu konsentrasi 0, 75%, 1%, 2%. Uji kestabilan fisik dilakukan dengan penyimpanan sediaan pada tiga suhu yaitu suhu kamar; suhu 40C; 40,2 C, uji mekanik dan cycling test. Hasil penelitian ini menunjukkkan bahwa krim ekstrak kulit buah delima 0,75%,1% dan 2% memiliki kestabilan setelah pengujian suhu kamar; suhu 40C; 40,2 C, uji mekanik dan cycling test. Penentuan aktivitas antioksidan dilakukan dengan metode peredaman DPPH berdasarkan nilai penghambatan (IC50) yang didapat. Dengan demikian diperoleh hasil bahwa krim ekstrak kulit buah delima dengan konsentrasi 0, 75%, 1 % dan 2% memiliki aktivitas antioksidan dan masih memenuhi nilai minimum IC50. Uji statistik Anova menunjukkan bahwa aktivitas antioksidan pada krim ekstrak kulit buah delima dengan waktu peyimpanan to sampai t8 mengalami penurunan yang tidak bermakna dan penurunan aktivitas antioksidan sebelum dan sesudah penyinaran UV A dengan uji Wilcoxon pada krim ekstrak kulit buah delima juga tidak bermakna

Abstract

In the previous study the analgesic effect of Cinnamommum zeylanicum Breyn had been investigating. The aim of this study was to investigate analgesic effect of the 70% ethanol extract of cinnamon bark (Cinnamommum zeylanicum Breyn.). This study used Tail Flick method at 25 male mice which have passed sensitivity test, divided into five groupes. Group I as negative control was administered 0,5% CMC, group Il as positive control was administered tramadol HCI, group Ill, IV and V was administered extract of cinnamon bark at 8; 16 and 32 mg/20 g BW. Drugs were orally administered to mice. The reaction of time exhibited by each animal was counted for two hour with interval thirty minutes. The Result revealed that the prepared extract in at the all dose, had analgesic effect significantly in all groups which have been studied (p< 0,05) in comparison to the negative control, to the highest analgesic effect by dose (32 mg/ 20 g BW). Our results showed that the analgesic effect of extract (32 mg/20 g BW) the same with tramadol hidroclorida.

Abstract

Jam is a semi-solid food products made of fruit cooked with sugar that used as aflavoring on bakery including strawberry jam. Strawberry jam is added to food additives such as food dyes. The purpose of this study is to know about ponceau 4R, allura red, rodamin B, and amaran in the sample of strawberry jam in the traditional market at Depok City and to determine the levels of synthetic red dyes that are permitted on the sample strawberry jam. The method applied was dye isolation with wool and followed by analysis using a color reaction, then followed by paper chromatography using mobile phase n-butanol-eth-anol-water (3:4:4) and isobutanol-ethanol-water (3:2:4 ) and also TLC Densitometri using eluent ethanol-n-butanol-water (3:7:1). The result of study that from eight that has been investigated, it was found that six of them was contained ponceau 4R with levels of each sample at 0, 01164; 0, 00469; 0, 00974; 0, 00283; 0, 00482 and 0, 00435% still safe to consume.

Abstract

Lactid Acid Bacteria (LAB) are known to produce bacteriocins which have antimicrobial activity, and possessed to be developed as antibiotic complement. This study aimed to characterize bacteriocins activity from Leuconostoc strains isolated previously from local sources, and to optimize pH and incubation temperature as well. A well diffusion agar assay for zone inhibition method and bacteriocin potency assay performing minimum inhibition concentration (MIC) have been done. Bacterial indicators used in this study are Leu. mesenteroides TISTR 120, and JCM 6124, Staphylococcus aureus FNCC 0047, Listeria monocytogenes FNCC 0156, Escherichia coli FNCC 0183, Pseudomonas aeruginosa FNCC 0063, Salmonella typhi FNCC 0165 and Bacillus subtilis FNCC 0061. Catalase, Trypsin and Protease K were also used for confirmation test. Results revealed that both Leu. mesenteroides MBF2-5 and MBF7-17 possessed bacteriocin activity although against Leu. mesenteroides TISTR 120 and JCM 6124 indicators strains. The optimum pH for bacteriocin potency assay for both Leuconostoc strains MBF2-5 and MBF7-17 was pH 6, whereas the optimum incubation temperature was 32 oc with MIC value of 90% and 80%, respectively.

Abstract

The cortex of jackfruit (Artocarpus heterophyllus) contains some flavonoids which have activity as tyrosinase inhibitors. This compound can inhibit the oxidation of l-tyrosine and levodopa in the mechanism of melanogenesis. The extract of jackfruit cortex formulated into creams differentiated by the extract concentration of 1,5% and 2,0%. Physical stability test was conducted with storing the creams at three different temperatures, 7 ± 2°, 27 ± 2°, and 40±2°C respectively. Centrifugal tests and cycling test was also performed on both cream. Tyrosinase inhibitory activity measurement was done by in vitro studies with measuring dopachrome. The result showed that both of formulations which stored at 40±2°C and centrifugated at 3800 rpm for 5 hours were not stable. The result of tyrosinase inhibiton activity measurement of creams containing extract of 1,5% and 2,0 % were 10,64% and 11,34%, respectively. Tyrosinase inhibition activity of creams decreased after two month stored. Tyrosinase inhibition activity of cream containing 1,5%extract decreased into 6,93%, and cream containing 2,0%extract decreased into 7,74%. The decreasing of tyrosinase inhibition activity is caused by the small amount of antioxidant is not enough to prevent oxidation of active ingredient.

Abstract

Lipid metabolism disorder is one of common condition among HIV (Human Immunodeficiency Virus)/AIDS (Acquired Immune Deficiency Syndrome) patients who used first line antiretroviral efavirenz and nevirapine. This study was conducted to compare LDL (Low Density Lipoprotein) cholesterol between two group HIV/AIDS patients with efavirenz and nevirapine. The prospective study was performed in 2 months. HIV/AIDS patients who used first line antiretroviral at least for 6 months, aged ≥18 years old, not pregnant, without Mycobacterium ovium complex opportunistic infection and antihyperlipidemic, corticosteroid, retinoid in last 3 months were included. The 70 patients who enrolled in this study were divided into two groups. The independent T test was carried out to analyze the different of LDL cholesterol between two group. The group of patients using efavirenz showed 142.31 ±41,461 mg/dL and the group of nevirapine 126.83 ±40,55 mg/dL LDL cholesterol. This study showed that patient with efavirenze had higher LDL than nevirapine, but not statistically different.

Abstract

Silver nanoparticles have been studied for its application in wound healing therapy, while aqueous extract of Plantago major L has been studied also for its antibacterial and wound healing activity. This study aimed to obtain silver nanoparticles (AgNPs) through a green synthesis pathway using aqueous extract of Plantago major L. The initial study was performed to optimize the three concentration points of Plantago major L. aqueous extract, namely 0.125%, 0.25% and 0.5% and three synthetic temperature points (60 °C, 70 °C, and 80 °C). The formation of AgNPs were confirmed using a UV-Vis spectrophotometer which shows a peak at a wavelength of 445 nm. The obtained silver nanoparticles were then characterized using Dynamic Light Scattering (DLS Nano), Scanning Electron Microscope (SEM) dan X-Ray Diffraction (XRD). The study showed that the extract concentration of 0.25% with an optimum temperature of 70 °C was the optimal combination to produce the best AgNPs, based on the organoleptic parameters (as the color of silver was produced), the highest yield with weight of 9.13 mg and particle size of 129.20 nm with Polydispersity Index (PDI) of 0.25. The morphology of AgNPs showed the spherical shape and the crystallization of AgNPs showed a diffractogram pattern similar to the silver standard using X-Ray Diffraction (XRD). Antibacterial activity of AgNPs was tested against three bacterial strains, namely Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa and was measured with diffusion method using paper disc. The antibacterial activity testing showed that AgNPS has better antibacterial activity than silver nitrate and is equivalent to its comparative antibiotics, gentamicin sulfate for gram-negative bacteria and Chloramphenicol for gram-positive bacteria.
