Abstract

The purpose of this study was to evaluate the characteristics of carcass yields and internal and external organs of the broilers fed the diets containing the fermented moringa leaf meal (FMOL) + yellow corn (YC), the moringa-corn (MC) diet formulated to replace parts of the commercial diet (CD). As many as 100 DOC of broiler chicken strain CP 707 were reared for up to 5 weeks to feed experimental diets. A Block Randomized Design (BRD) with subsamples consisted of 4 treatments, 5 blocks, and 2 subsamples were applied in the recent study. The results of studies indicated that removing parts of the CD diet to substitute with the mixture of FMOL + YC caused a significant effect (P0.05) in declining the weights of the whole carcass and the cut-ups but in percentage no significant differences. The weights and percentages of all examined organs were not significantly affected (P0.05). It was concluded that using the moringa mixed with yellow corn reduced the weight of the whole carcass and cut-up carcasses but it could use up to 7.5% in the diet without detrimental effects on the internal and external organs of the broilers.

Abstract

The objective of this research was to examine the correlation between estrogen level and estrous cycle of rat fed with soybean flour and tempeh flour. A completely randomized design was applied in this study with 6 treatment groups and 5 replications. Rats in group 1, 2, and 3 were non-ovariectomized rats which were fed with pellet, 10 g soybean flour/100 g body weight/day, and 10 g tempeh flour/ 100 g body weight/day, respectively. The rats in group 4, 5, qnd 6 were ovariectomized rat which were fed with pellet, 10 g soybean flour/100 g body weight/day, and 10 g tempeh flour/100 g body weight/day, respectively. The rats were after fed with pellet, soybean and tempeh flour for 4 weeks, the estrous cycle phases of rats were examined based on the presence of vaginal epithelial cells and the number of qualitative vaginal epithelial cells, while the estrogen concentrations in serum were measured by radioimmunoassay method. The data was analyzed using analysis of variance and followed by Duncan multiple range test at 95% confidence. Non-ovariectomized rats given tempeh flour have longer estrus cycle than the non-ovariectomized rats which given soybean flour. The isoflavon contained in tempeh flour and soybean flour induced proliferation and cornification of vaginal epithelial cell. Feeding the soybean flour and tempeh flour to ovariectomized rats could optimize the estrogen hormone in initiating the estrus phase, in which of the tempeh flour addition had better effect than soybean flour. Correlation between the level of estrogen to the length of estrous cycle in both nonovariectomized rats and ovariectomized rats was positive, in which the higher the levels of estrogen the longer the length of estrous cycle.

Abstract

The objective of this research was to determine the allergy hyperplasma pathologic antibody and to evaluate the immunogenic characteristics. Blood and serum were collected from human suffering rhinitis, skin, eye and asthma allergies. To obtain a cloned allergen plasma protein (protein allergen that had been induced with the serum from human with allergic rhinitis, skin, eye and asthma), the goat was used as an intermediate animal. Hematological analysis showed that the leukocyte cell such as neutrophil, lymphocyte, monocyte, and eosinophil increase in allergic-suffered human. The blood smear test exhibited that the mastocyte cell was dominant which contributes to allergy activities in human body. The sodium dodecyl sulphate-polyacrylimide gel electrophoresis (SDS-PAGE) assay detected protein allergens with molecular weight of 188 kDa (IgE) and 60-62 kDa (mastocyte cell). The reactivity assay using enzyme linked immunosorbent-assay (ELISA) revealed that cloned-allergens (whole hyperplasma allergen from goat isolates) express the best reactivity at various concentrations of IgE than the leukocyte cells. This research concluded that the clones of protein allergen have better immunogenic characteristic and those proteins can be recommended as the candidate of allergen to induce the humoral immunity on host and deliver specific product of anti-allergy such as milk.

Abstract

To examine whether Javanese ginseng root has an antihyperglycaemic effect, twenty-one male Rattus norvegicus rats were randomly divided into four groups: negative control (n= 3), positive control (n= 4), treatment (n= 7), and placebo (n= 7). Alloxan was injected intraperitoneally at a single dose of 80 mg/kg BW to rats to induce hyperglycaemia. Infusion of Javanese ginseng root or distilled water was given at a dose of 2% w/v solution with a volume of 1.8 mL/200 g BW per day for 14 days through an enteral feeding tube to either the treatment or placebo group, respectively. Blood glucose levels were measured using the colorimetric method. One-way analysis of variance (ANOVA) was used to examine differences in the mean of delta blood glucose (post intervention minus post alloxan blood glucose levels) among groups, followed by Tukeys post hoc analysis. A P value 0.05 was considered statistically significant. There was a significant difference (P= 0.0001) in delta blood glucose among groups. Post hoc analysis revealed that delta blood glucose in the treatment group (-102.992.26 mg/dL) was significantly (P= 0.0001) greater than in the placebo (2.450.29 mg/dL), positive control (3.050.70 mg/dL) and negative control (1.600.17 mg/dL) grou p. In conclusion, Javanese ginseng root has potential as a hypoglycemic agent in alloxan-induced diabetic animal models

Abstract

This study aimed to estimate the gestational age of Etawa crossbred goats based on ultrasound images of embryo and fetal usingtranscutaneous ultrasonography. Etawa crossbred does (n= 5) were synchronized using double PGF2 injection with 10 days interval and matednaturally. Early pregnancy was diagnosed on 20th day after mating base on isoechogenic image of the embryo (2.650.75 mm) andhypoechogenic appearance of gestational sac. On 30th day of the first trimester, the embryo size was 8.310.42 mm. The embryo reached10.440.51 mm on 35th day. On 60th day, the existence of the fetal were clearly appeared along with isoechogenic of umbilical cord,hyperechogenic os cranium (25.50.1 mm), and body length (51.831.59 mm). On 75th day, the diameter of the os cranium was 33.030.20 mm,body length of 1101.73 mm, and bones structures appeared hyperechogenic for os thoracalis and os vertebrae. The placenta was isoechogenoicwith diameter of 23.30.2 mm. On 90th day, the placenta dominated the uterus (22.671.70 mm) and reached its maximum diameter on 145th day(33.732.91 mm). In conclusion, early pregnancy detection of Etawa crossbred goats using transcutaneous ultrasonography could be performedon 20th day after mating

Abstract

Penelitian dengan tujuan mengidentifikasi protein penentu implantasi embrio pada tikus Wistar (Rattus norvegicus) dilakukan sebagai upayaeksplorasi agen imunokontrasepsi baru untuk regulasi fertilitas hewan liar. Hewan uji yang digunakan adalah tikus Wistar betina dewasa kelamin dengan bobot badan berkisar 280-300 g. Tikus dibagi dalam dua kelompok, satu kelompok sebagai kontrol (tidak dikawinkan) sedangkankelompok lain adalah kelompok yang dikawinkan. Tikus dipelihara dan dikawinkan dalam kondisi laboratorium yang terkontrol. Kebuntingan ditentukan dengan adanya vaginal plug pada tikus betina setelah dikawinkan. Protein diisolasi dari sampel uterus yang diambil pada umur kebuntingan 3-5 hari. Isolasi dan separasi protein dilakukan berdasarkan manual dan standar protein produk Bio-Rad. Hasil penelitian menunjukkan pita protein yang terekspresi pada hari ke-4 kebuntingan dengan berat molekul berkisar 45 kDa diduga leukemia inhibitory factor.

Abstract

The aim of this research was to determine the anatomy and histology of pancreas of water monitor lizard (Varanus salvator). Two pancreas were used in this research (a male and a female). Water monitor lizard was euthanized using chloroform and subsequently necropsied to collect the pancreas. The pancreas was washed in 0.9 % NaCl solution. Anatomical observations were carried out including location, shape, and color of pancreas, also the length and weight measurements. Then, pancreas was fixed with 4% paraformaldehyde for 7 days to proceed to histological preparation and hematoxylin eosin (HE) staining. The results of the anatomical observations showed that pancreas of water monitor lizard have two lobes, dorsal lobe (splenic lobe) and ventral lobe (duodenal lobe). Dorsal lobe which was in oval form adhered with the spleen, and ventral lobe was located in the cranial of intestines with the form of long spherical. Both of pancreas lobes were white-yellow color with the length of 1.450.64 cm (dorsal lobe) and 7.000.42 cm (ventral lobe). Weight of dorsal lobe was 0.150.01 g and ventral lobe was 6.352.30 g. The histological observations found the acinar cells, ducts, and blood vessels in both of pancreas lobes. Additionally, the similar structure of Langerhans islet only found at the ventral lobe of pancreas. In conclusion, pancreas of water monitor lizard consists of two lobes which located differently. Ventral lobe is larger than the dorsal lobe. Both lobes have similar histological structure except the Langerhans islets are only appeared in the ventral lobe.

Abstract

Penelitian ini bertujuan mengevaluasi status virulensi 22 isolat Bacillus anthracis (B. anthracis) asal Nusa Tenggara dan Papua menggunakan metode polymerase chain reaction (PCR) multiplex dengan dua pasang primer nukleotida yang memiliki target amplifikasi gen spesifik pada kedua plasmid. Ektraksi DNA dilakukan dengan metode lisis panas. Pasangan primer PA5 dan PA8 digunakan untuk mengamplifikasi gen pagA pada pXO1, sedangkan pasangan primer 1234 F dan 1301 R mengamplifikasi gen capABC pada pXO2. Hasil reaksi PCR menghasilkan dua pita DNA berukuran sekitar 600 dan 800 bp pada 20 isolat. Namun, dua isolat lain, masing-masing hanya memiliki salah satu dari kedua ukuran pita DNA tersebut. Sebagian besar koleksi isolat asal Nusa Tenggara dan Papua (91%) masih memiliki kedua plasmid secara lengkap (pXO1+/2+) dan karena itu bersifat virulen, sedangkan dua isolat lain (9%) telah kehilangan salah satu plasmid virulennya sehingga bersifat avirulen. Disimpulkan bahwa PCR multiplex dengan dua pasang primer dengan target amplifikasi pada plasmid dapat digunakan untuk evaluasi status virulensi isolat B. anthraci.

Abstract

This study aims to measure the level of antibiotic resistance to Klebsiella pneumoniae isolated from clinical cats in Bogor. Samples were isolated and identified macroscopically, microscopically, and biochemically. Positive isolates were tested for antibiotic sensitivity using the Kirby-Bauer disk diffusion method. The results showed that Klebsiella pneumoniae isolated from sputum and laryngeal swabs of clinic cats in Bogor had experienced Multidrug Resistance (MDR). The highest level of resistance to Klebsiella pneumoniae occurred in the -lactam group (amphicillin 76%) followed by the tetracycline group (oxytetracycline 72% and tetracycline 68%), then the quinolone group (enrofloxacin 52%), and finally the aminoglycoside group (gentamicin 44%). The results of this study are expected to be taken into consideration in the use of antibiotics for the treatment of cases related to the Klebsiella pneumoniae bacteria.

Abstract

Brucellosis is among the important diseases in livestock because the disease infects multiple species of animals and causes economic loss. Brucellosis in sheep is generally caused by Brucella melitensis and/or Brucella ovis. This study aimed to detect seropositive brucellosis in sheep. Serological tests used in this study was a parallel test between Rose Bengal Test (RBT) and Complement Fixation Test (CFT). Samples were collected from 150 sheep slaughtered in small ruminant slaughterhouse, Sentul, Bogor Regency. Seropositive proportion of brucellosis in sheep based on parallel test RBT and CFT was 52% (78/150).
