Abstract

The aim of the research was to analyze the nitrogen and phosphorus content in sediments in the cultivation area of Toulimembet Village. This research method is "survey" determining sampling points "purposive sampling" taking into account that the research location is a fish cultivation area with a floating net system, and data is obtained using primary data and secondary data. Primary data collected were in situ measurements of water quality parameters (temperature, turbidity, TDS, pH, DO) in the Toulimembet Village cultivation area and sediment analysis (Nitrogen and Phosphorus) at the Manado Standardization Research Center Laboratory (BARISTAND). Secondary data was obtained from various sources such as journals and previous research reports. Based on research results in Toulimembet Village, Kakas District, Minahasa Regency, North Sulawesi Province, nitrogen (0.78-1.16%), phosphorus (0.04-0.08%), temperature (25.73-27.01°C), turbidity (4.03-4.90 NTU), TDS (0.160-0.164 g/l), pH (7.89-10.52), DO (7. 20-12.67 mg/L), ammonia (0.07 mg/L), phosphate (0.0030-0.0158 mg/L), nitrite (0.069-0.295 mg/L) are still suitable for cultivation activities. Abstract

The fishery sector which has great profit prospects can still be developed by cultivating fish better by paying attention to the condition of the water quality. In fish farming, it is very important to study water quality conditions because water quality parameters are a limiting and life-supporting factor for the types of aquatic biota cultivated in a waters including tilapia (Oreochromis niloticus. This research was conducted to assess water quality, both physical, chemical, and aquatic biology of Lake Tondano to support the feasibility test activities of Lake Tondano waters for Cage Netting Cage cultivation media. The research method used was purposive sampling. Primary data comes from primary data which is the result of measurements of physico-chemical parameters of water quality directly in the field (in situ) and supported by secondary data on several physical and chemical parameters of Lake Tondano waters as well as data on water quality of the cultivation media with the Netting Cage system. The physical and chemical parameters of Lake Tondano waters at the three stations were analyzed using the suitability scoring method for the location of Tondano Lake waters to be used as a maintenance medium fish with a Stuck Net Containment system. The results of the study concluded that the physical and chemical quality of water in Urongo waters was suitable for net cage culture. Abstract

The aim of this research was to determine the physical and chemical parameters of water quality in vanname shrimp culture in Pond PT. Pillar Persada Parigi, Bajo Village, South Minahasa. This research uses direct measurement methods in the field (In situ) and sample analysis in the laboratory. Water quality parameters measured in the field included temperature, degree of acidity (pH), salinity, dissolved oxygen, while ammonia, nitrite, nitrate, phosphate was analyzed at the Manado Industrial Research and Standardization Center. Water sampling and in-situ measurements were carried out in 4 ponds selected randomly from the ponds that were temporarily operating at the time of sampling. Based on the results of measurements of several physical and chemical parameters of water quality, both directly in the field (in situ) and the results of laboratory analysis of pond water samples from PT Pillar Persada Parigi, Bajo Village, South Minahasa Regency, a temperature range of 27 - 32˚C, salinity of 36 ppt was obtained. , dissolved oxygen 3.63 - 7.8 ppm, acidity degree (pH) 7.6 - 8.6, ammonia content 1 mg/L, nitrite <0.2757 mg/L, nitrate 5 mg/L, phosphate <0 .0027 – 1 mg/L. Abstract

Organochlorine pesticides are one type of pesticide that is often used and some organochlorine pesticides are included in the group of Persistent Organic Pollutants, this type of pesticide is a pollutant that is a problem because it is resistant and bioaccumulative. Organochlorine characteristics that can adversely affect the environment are; 1). Decomposes very long in soil, air, water, and in living things, and can also stay in the environment for a long time. 2). It can enter the food chain and accumulate in fat tissue, making it difficult to dissolve in water. 3). Can be carried far through air and water. Seeing the nature of organochlorine pesticides that can last a long time in the environment and can be carried away by air and water, and the discovery of organochlorines in Manado Bay does not rule out the possibility that organochlorine pesticides can also exist in the waters of Nain Island. The objectives of this study are: (1) Measuring the physical parameters of water chemistry (temperature, DO, pH, and salinity) and organochlorine pesticide levels in Nain Island waters, (2) Analyzing organochlorine levels in Nain Island seawater samples. Measurements and sampling were carried out on Nain Island, Wori District, North Minahasa Regency on May 22, 2023. The results showed that there were organochlorines of endrin and DDT types in the waters of Nain Island, the concentration of endrin and DDT did not exceed the threshold of seawater quality standards PP RI No. 22 of 2021. Abstract

Abstract Bahoi Waters is located in West Likupang District, known as a coastal village that has a lot of fishing and marine potential. Utilization of Bahoi waters for seaweed cultivation requires good seawater quality to meet the requirements for cultivation. Water quality is one of the factors that plays an important role in the success of seaweed cultivation. The aim of the research was to determine the suitability of water in Bahoi Village for  land for seaweed (Caulerpa sp.) cultivation. The research used in situ measurements at 3 stations, namely: Station A (Existing Cultivation Location), Station B (Tanjung Kamala Waters), and Station C (Tamperong Island Waters). Data collection was carried out 2 times with 3 repetitions for measurement of water parameters. Measurement time was carried out at 07:00 – 09:00; 12:00-14:00, and 17:00- 19:00. Data was analyzed by comparing physical parameters and chemical parameters at each station with a scoring table and weights in the land suitability matrix. The conformity class determination obtained was: Very Suitable with a value range of 37 – 48 (S1), Suitable 25 – 36 (S2), and Not Suitable 16 – 24 (S3). The scoring value for suitability of water quality parameters at Station A was 46, Station B 47, and Station C 44. In conclusion, all stations are in the Very Suitable class for cultivating Caulerpa sp.  Keywords: Caulerpa sp., water quality, seaweed

Abstract

The purpose of this study was to determine the distribution and the diversity of ascidians in Manado Bay including species composition, density, diversity, and dominance. This study used the quadrat transect method. This study found differences in the number of ascidian species with water depth, 11 species of 5 families at 15 M depth, and 8 species of 3 families at 7 M depth. The diversity index ranged from 0.868 to 1.844 at 15 M depth and 0.965 to 1.864 at 7 M depth, the evenness index was 0.533 – 0.839 at 15 M depth and 0.600 – 0.897 at 7 M depth, the dominance index was 0.254 – 0.745 at 15 M depth and 0.254 – 0.708 at 7 M depth. Ascidian in Manado Bay had two distribution patterns, a uniform distribution pattern and a clustered distribution pattern. Environmental parameters had a water temperature of 27 oC – 31 oC, the salinity of 30 0/00 – 32 0/00, the brightness of 12 m – 14 m, and pH of 8 – 10.*Keywords: Ascidian; diversity; ecological index; distribution patternAbstrakTujuan dari penelitian ini adalah untuk mengetahui distribusi dan keanekaragaman jenis ascidia di perairan Teluk Manado meliputi: komposisi jenis, kepadatan individu, keanekaragaman, dan dominansi. Serta mengetahui pola distribusi ascidia. Penelitian ini menggunakan metode transek kuadran. Pada penelitian ini ditemukan perbedaan jumlah spesies ascidia menurut kedalaman, 11 spesies dari 5 family pada 15 M dan 8 spesies dari 3 famili pada kedalaman 7 m. Nilai indeks keanekaragaman ascidia di kedalaman 15 m = 0.868 – 1.844 dan 7 m = 0.965 - 1.864, indeks keseragaman 15 m = 0.533 – 0.839 dan 7 m = 0.600 – 0.897, indeks Dominasi15 m = 0.254 – 0.745 dan 7 m = 0.254 – 0.708. Ascdia di perairan Teluk Manado memiliki dua pola distribusi yaitu pola distribusi seragam dan pola distribusi mengelopok. Abstract

This study aims to analyze the structure of the Polychaeta community on the soft substrate of the subtidal zone in Manado Bay. Samples were taken by grab at 3 stations namely ST1 located at a depth of 8 m with black mud substrate; ST2 is located at a depth of 26 m with blackish sand substrate, and ST3 are located at a depth of 18 m with blackish sand as a substrate. From the three sampling stations, 27 species of 253 Polychaeta individuals were identified. Station 1, which is located near the mouth of the Bailang River, has high individual abundance but low species diversity. Station 2, which is located near the Megamas area, has moderate individual abundance but high species richness. Station 3, which is located around the Faculty of Medicine, Unsrat Malalayang, has low individual abundance but high species richness. Substrate types and anthropogenic disturbances such as enrichment of organic matter are thought to be determinants of individual abundance, composition, and species richness of Polychaeta in Manado Bay.Keywords: Polychaeta; biodiversity; soft substrate; Manado BayAbstrak Penelitian ini bertujuan untuk menganalisis struktur komunitas Polychaeta pada substrat lunak zona subtidal di Teluk Manado. Sampel diambil dengan grab pada 3 stasiun yakni ST1 terletak pada kedalaman 8 m dengan substrat lumpur berwarna hitam; ST2 terletak pada kedalaman 26 m dengan substrat pasir berwarna kehitaman; dan ST3 terletak pada kedalaman 18 m dengan substrat pasir berwarna kehitaman. Dari tiga stasiun sampling tersebut berhasil diidentifikasi 27 spesies dari 253 individu Polychaeta. Stasiun 1 yang terletak dekat muara Sungai Bailang memiliki kelimpahan individu tinggi tetapi keanekaragaman spesies rendah. Stasiun 2 yang terletak dekat kawasan Megamas memiliki kelimpahan individu sedang tetapi kekayaan spesies tinggi. Stasiun 3 yang terletak di sekitar pemukiman belakang Fakultas Kedokteran Unsrat Malalayang memiliki kelimpahan individu rendah tetapi kekayaan spesies tinggi. Jenis substrat dan gangguan antropogenik seperti pengayaan bahan organik diduga merupakan faktor penentu kelimpahan individu, komposisi dan kekayaan spesies Polychaeta di Teluk Manado. Kata Kunci: Polychaeta; keanekaragaman hayati; substrat lunak; Teluk Manado

Abstract

This study aims to determine the biometric comparison of otoliths of Banggai cardinal fish based on gender and to determine the comparison of otolith biometrics at two locations in the Waters Front of TPI Dudepo Kab. South Bolaang Mongondow and in the Lembeh Strait, Bitung City. Data collection in the field using the roaming survey method. Fishing is done by snorkelling, catching a minimum of 50 individuals representing each size class using Chang net/Sibu fishing gear. The differences in otoliths in male and female sexes have seven different otolith characters in otolith length (OL), otolith width (OW), otolith area (OA) and otolith perimeter (OP), roundness (Rnd), ellipticity (Ell) and aspect ratio (Ar), and three characters that do not show significant differences, namely form factor (Ff), circularity (Cir), rectangularity (Rec). The differences in otoliths based on location in front of the Dudepo TPI and in the Lembeh Strait have seven different otolith characters in otolith length (OL), otolith width (OW), otolith area (OA) and otolith perimeter (OP), roundness (Rnd), ellipticity (Ell). and aspect ratio (Ar), and three characters that do not show significant differences, namely form factor (Ff), circularity (Cir), rectangularity (Rec). Bolaang Mongondow Selatan dan di Selat Lembeh Kota Bitung. Pengambilan data di lapangan menggunakan metode survei jelajah. Penangkapan ikan dilakukan dengan penyelaman snorkling, penangkapan minimal 50 individu yang mewakili setiap kelas ukuran dengan alat tangkap Chang net/Sibu. Perbedaan otolit pada jenis kelamin jantan dan jenis kelamin betina memiliki tujuh perbedaan karakter otolit pada panjang otolit (OL), lebar otolit (OW), area otolit (OA) dan perimeter otolit (OP), roundness (Rnd), ellipticity (Ell) dan aspect ratio (Ar), dan tiga karakter yang tidak menunjukkan perbedaan yang signifikan, yaitu form factor (Ff), circularity (Cir), rectangularity (Rec). Perbedaan otolit berdasarkan lokasi di depan TPI Dudepo dan di Selat Lembeh memiliki tujuh perbedaan karakter otolit pada panjang otolit (OL), lebar otolit (OW), area otolit (OA) dan perimeter otolit (OP), roundness (Rnd), ellipticity (Ell) dan aspect ratio (Ar), dan tiga karakter yang tidak menunjukkan perbedaan yang signifikan, yaitu form factor (Ff), circularity (Cir), rectangularity (Rec). Abstract

The purpose of this study is to isolate and identify Vibrio spp. bacteria from vannamei shrimp (Litopenaeus vannamei) in PT. Pilar Persada Parigi Bajo Village, South Minahasa Regency. The research was carried out by starting with shrimp sampling from shrimp cultivation locations at PT. Persada Parigi Pillar in Bajo Village, Tatapaan, South Minahasa, North Sulawesi as many as 9 fish from 3 ponds (1 pond took 3 shrimp). The pond was recommended by the pond technician of PT. The Persada Parigi pillar is related to the condition of the pond which is suspected of having Vibrio spp. bacteria that need special attention. The shrimp samples were then taken to the Laboratory alive. During transportation, the samples were put in 3 separate containers equipped with battery aerators, then the identification of Vibrio spp. bacteria was carried out at the Laboratory of Pharmaceutical Microbiology, Faculty of Mathematics and Natural Sciences, Sam Ratulangi University and at the Manado Fishery Products Quality Control and Safety Quarantine Center. Isolation of Vibrio spp. bacteria using TCBS (Thiosulfate Citrate Bile Sucrose) agar media. The characterization of Vibrio spp. bacteria was carried out by a series of tests, namely gram staining tests (gram negative, positive and bacterial form), main tests (KOH 3%, H₂O₂ and oxidase), Oxidation/Fermentation (O/F), Glucose and Motility Indole Ornithin (MIO). The results of the isolation showed that there were 9 isolates of Vibrio spp. bacteria from 9 samples of vannamei shrimp (Litopenaeus vannamei) from 3 different ponds. The results of bacterial characteristics obtained negative grams, yellow and green colony morphology, bacil/stem cell shape, KOH 3% (-), catalase (+), oxidase (+), O/F (fermentative), indole (+), and motility (+), glucose (+) from the results identified by Vibrio spp. bacteria. Abstract

Tributiltin (TBT) is an organotin compound that is still widely used as a toxic biocidal additive in antifouling paints. These compounds are classified as pollutants that are very difficult to decompose in waters. The test of the concentration of TBT in seawater was carried out at the WLN Laboratory (Water Laboratory Nusantara) Manado. Nitrate, nitrite and ammonia concentration tests were carried out at the Laboratory of the Manado Industrial Service and Standardization Center (BSPJI). Measurement of the physical and chemical parameters of seawater used: Temperature, Brightness, pH, Dissolved Oxygen (DO), Salinity and Turbidity in situ. Analysis of the concentration of Tributiltin (TBT) compounds in seawater in the waters of the Lembeh Strait in the cultivation areas of Point 1 (Tanjung Rarandam), Point 2 (Kalapa Dua), Point 3 (Papusungan) in the Lembeh Strait showed a concentration of <0.01 μg/L, which is still in the safe range below the standard limit of seawater quality for biota (0.01 μg/L). Measuring water quality in the Lembeh Strait aquaculture area, the DO value ranges from 7.83-8.34 ppm. Salinity values ranged from 30.99-31.24 ppt. The pH value ranges from 9.37-9.42. The temperature value ranges from 27.87-27.69 °C. The brightness value ranges from 3.12 m-3.39 m. The turbidity value ranged from 0.63-1.22. The nitrate value of 1.98-2.19 mg/L, far exceeding the limit set (0.06 mg/L) for marine life, indicates a potential risk to the ecosystem. The concentration of Nitrite in the waters is still in accordance with the general water standards, ranging from <0.005 mg/L to 0.02 mg/L. The concentration of Ammonia is below the water quality standard threshold at all points of 0.3 mg/L, which is still safe for marine life and aquaculture activities in these waters in accordance with the seawater quality standards in Government Regulation No. 22 of 2021 Attachment VIII concerning the Implementation of Environmental Protection and Management.
