Abstract

This paper was the result of a feasibility analysis of Talengen Bay Waters for aquaculture of Kappaphycus alvarezii. Feasibility analysis of water was based on the environmental data and water quality parameters of Talengen Bay. One mistake in the development of aquaculture is environmentally unsuitable. In order to develop aquaculture properly, appropriate water conditions data were required. Suitable aquatic environments are required by organisms for their survival because they are related to the patterns and behavior of the biota. In connection with the problems of mariculture at Talengen Bay, several efforts were required to overcome them. One of the efforts was made through this study that was assessment of appropriate location for the cultivation of seaweed. Talengen Gulf was located in  in the territory of Kampung  Talengen and  Kampung Bunga Lawang, Central District of Tabukan, Sangihe Islands Regency, North Sulawesi. Laboratory analysis was conducted at the Laboratory of Industrial Baristand Manado. Determination of research stations done intentionally (purposive sampling). Based on the results obtained it could then be concluded that the condition of the aquatic environment and water quality parameters in the Talengen Bay were categorized as suitable for seaweed cultivation K. alvarezii. Phosphate, nitrate, Total Suspended Solid data were needed to be considered. Keywords: Environmental characteristics, seaweed, aquaculture; Kappaphycus alvarezii

Abstract

Title (Bahasa Indonesia): Pengaruh pupuk NPK [nitrogen, fosfor, kalium] terhadap pertumbuhan dan penanggulangan penyakit ‘white spot’ pada rumput laut Kappaphycus alvarezii This study aimed to analyze the effect of NPK fertilizer absorption on the growth of seaweed, Kappaphycus alvarezii, and the white spot disease prevention. This study could become a source of information for seaweed farmers to increase seaweed production through the use of NPK fertilizers. This study was conducted from January until March, 2014 in the waters of Toropot Village, the District of Bokan Islands, Banggai Laut. To know whether the different doses affect the white spot infection, ANOVA with Tukey's test was used. Results showed that all doses had the same potential to recover from white spot disease and heal faster than the control (no dose). The addition of nutrients N, P, and K at high dose could accelerate the recovery of the algae from white spot desease and increase the growth rate.

Abstract

Kata kunci: Tumbak; nilai manfaat langsung; terumbu karang; ekosistem   The issues of destructive fishing practices (using of bombs and poisons) and the use of corals as construction building materials have become issues in coastal management in Tumbak Village. The aims of this study are (1) to find out the value of the direct benefits of coral reefs as fishing ground to the communities in Tumbak village, (2) to find out the value of the direct benefits of coral reefs as building materials to the communities in Tumbak village; (3) to find out the total value of the use of coral reefs as a fishing ground and as a construction materials for to the communities in Tumbak village. The value of direct benefit of the coral reef for building foundation was Rp 6,177,600 per year and the value of direct benefits for fishing was Rp 4,860,000,000. Total value of direct benefits from both of these uses was Rp 4,866,177,600. Total value did not include the use of other forms of coral reef ecosystems. In conclusions, the use of coral reefs as a building materials were mainly due to the low cost and the proximity to residential areas. Such category unsustainable practice has led to the degradation of coral reefs down to damaged category (medium category). If this happened continuously, the ecological function as wave protectors and economic functions as a source of food could be eliminated. Abstract

The aimed of the study was to isolate and select the probiotic Lactobacillus from carp intestine potential for use in aquaculture. Carp as a source of probiotics with a size of about 200 g was  taken from the Center for Fish Hatchery and Disease Control (BP3I), Tateli. After aseptic surgery, the intestine was separated and cleaned, weighed as much as 1 g, put in a mortar and crushed until smooth. The scoured results were put into a tube and added 9 ml NaCl to get a ratio of 1:10. The tube that already contains intestinal scouring was then centrifuged at a speed of 1000 RPM for 10 minutes. The centrifugation process was carried out 2 times to get the supernatant that has been cleared of debris. The supernatant was removed and diluted to obtain a concentration of 10-2  to 10-3. The solution was then spread on MRS (De Man Rogosa and Sharpe Agar) media and incubated at 28oC in the Incubator for 24-48 hours. The bacteria that grew in culture fmedia was reisolated several times to get a single pure colony.. The pure colonies were propagated in the same media to be further identified through gram staining, biochemical, proteolytic, amylolytic, cellulolytic, antibiotic resistance, antagonistic, pathogenicity tests. The results of the isolation obtained the characteristics of a colony of milky white color, round and convex with long cells, gram-positive and confirmed based on biochemical tests as Lactobaccilus sp. The bacterium also had proteolytic, amylolytic, cellulolytic, antagonistic, antibiotic-sensitive activity that were characterized by the formation of clear zones in the area around probiotic discs, and did not cause death either as a disease or as a result of poisoning. Thus it can be concluded that Lactobaccilus sp. isolated from carp intestines can be used as probiotics for cultivation.
