Abstract

Abstract

Atherosclerosis is a condition characterized by inflammation in the arteries, which is linked with the accumulation of lipids and alterations in metabolism. Considering that atherosclerosis is the main factor causing death in the world, it is necessary to carry out immediate prevention and treatment to reduce the risk of developing clinical severity. The creation of foam cells, which originate from macrophages, is considered a key element in cardiovascular ailments, particularly in the advancement of atherosclerosis. Two types of the nuclear receptors known as Peroxisome proliferator-activated receptor γ (PPARγ) and liver X receptor α (LXRα), which serve as a primary regulator of cholesterol, intracellular lipid homeostasis and they are instrumental in the process of reverse cholesterol transport (RCT). Activation of these receptors could potentially decrease foam cell formation, consequently lowering the risk of atherogenesis and reducing cardiovascular disease risk. This research aims to determine sterol derivative compounds in Tagetes erecta Linn. which have the best interaction and potential as anti-atherosclerosis through peroxisome proliferator-activated receptor γ (PPARγ) and liver X receptor α (LXRα) activation. The analysis of this study is using docking molecular analysis. The parameters observed in this study were docking score, visualization results, absorption, distribution, metabolism, excretion profile, and toxicity value. The molecular docking outcomes indicate that β-sitosterol and 7β-hydroxysitosterol possess the most favorable binding energy values. They exhibit a positive pharmacokinetic profile, with the exception of gastrointestinal absorption and respiratory toxicity.

Abstract

The cocoa seeds of the Gumbrih-Bali region are a natural commodity with great potential for development or processing by local communities. The purpose of this research is to provide information about the antioxidant potential contained in cocoa seeds. Testing the antioxidant capacity in this study uses the Uv-Vis spectrophotometry method at 517 nm wavelengths. Free radical scavanger is measured with control absorption and samples was analyzed with regression curve. The research results proved that the antioxidant (AAI) in the n-butanol fraction show result very strong ability of 4.64 compared to the 0.42 ethyl acetate fraction and 0.46 n-hexane fraction. These results prove that the potential chemical content of cocoa seeds is high in polar solvents (n-butanol fractions) such as flavonoid compounds. Furthermore, this research will be used as a basis for developing natural herbal ingredients such as cocoa which can be consumed in the community to prevent degenerative diseases.

Abstract

Indonesia's high UV-A exposure significantly contributes to skin hyperpigmentation conditions such as melasma. This study investigated the effectiveness of Jackfruit (Artocarpus heterophyllus Lamk.) leaf serum in reducing dark spots in a guinea pig model of melasma induced by UV-A radiation. Crude extracts were prepared and purified from Jackfruit (Artocarpus heterophyllus Lamk.) leaves, then formulated into serums at 4% and 5% concentrations. These formulations underwent physical quality evaluations, including organoleptic properties, pH, homogeneity, spread ability, adhesion, and viscosity. Anti-hyperpigmentation effects were assessed using Masson Fontana staining on UV-A-exposed guinea pigs treated for 14 days. The results demonstrated a reduction in melanin content, with mean values of 17.97±1.68 (negative control), 1.17±0.28 (positive control), 3.56±2.77 (4% serum), and 2.56±1.25 (5% serum). Histological analysis showed that fibroblast proliferation and epidermal hyperplasia were significantly reduced with serum treatment. These effects are attributed to the flavonoid content in Jackfruit (Artocarpus heterophyllus Lamk.) leaves, which exhibit antioxidant and anti-melanogenesis properties. In conclusion, Jackfruit (Artocarpus heterophyllus Lamk.) leaf serum, particularly at 5% concentration, is effective in reducing melasma-related hyperpigmentation and may offer a safer alternative to synthetic treatments.

Abstract

Background: Excessive ultraviolet (UV) exposure is a major contributor to skin photoaging through oxidative stress, inflammation, and collagen degradation. Mangrove leaf extract (Xylocarpus granatum J. Koenig) contains bioactive compounds with potential photoprotective properties. In this study, the extract was formulated into an oil-in-water (O/W) cream suitable for cosmetic applications. Objective: This study aimed to determine the in vitro sun protection factor (SPF), percentage of erythema transmission (%Te), and percentage of pigmentation transmission (%Tp) of the mangrove leaf extract cream, and to evaluate its effectiveness on UVB-exposed guinea pig skin using collagen density as an anti-aging biomarker. Methods: In vitro assessments of SPF, %Te, and %Tp were performed using UV–Vis spectrophotometry. In vivo evaluation was conducted using 20 guinea pigs to examine the effect of the cream on collagen density through histopathological analysis of UVB-exposed skin. Results: The mangrove leaf extract cream exhibited strong photoprotective activity, with SPF values ranging from 20.68 ± 0.51 to 33.70 ± 0.67, %Te values from 3.13 ± 0.08 to 1.75 ± 0.03, and %Tp values from 4.40 ± 0.12 to 3.31 ± 0.03. Furthermore, the cream significantly increased collagen density (71.77 ± 5.27–81.02 ± 0.96) compared with the positive control (52.16 ± 2.32) (p < 0.05).Conclusions: The mangrove leaf extract cream provides effective protection against UVB radiation, characterized by high-protection SPF, extra-protection %Te, and sunblock-level %Tp. The 5% formulation significantly enhanced collagen density, supporting its potential as a natural anti-aging active ingredient for photoaging prevention through improved UV protection and dermal structure.
