Abstract

Urban farming activities with hydroponics techniques is one of the positive activities that can improve the creativity of people in urban areas. Urban farming activities can help improve the quality of people's lives, the environment, and play a role in maintaining food security. The study consisted of independent variables consisting of several groups, namely control (AB-Mix), 0.5 ppm of IAA (Indole Acetic Acid), BAP (Benzyl Adenine Purine) 0.5 and 1.0 ppm, and combination of IAA / BAP 0.5 /1.0 ppm. The purpose of this study was to study the effect of these treatments on water spinach plant yields that include leaves number, plant height, antioxidant content, and water content. The results of morphological growth showed that treatment with IAA 0.5 ppm was the most optimal while treatment with 1.0 ppm BAP had the average of the shortest plant height, the average number of leaves at least, and the shortest root length average. While the chlorophyll test showed that the system unit with the treatment of 0.5 ppm IAA had the highest total chlorophyll content with an average of 9.55 μg/ml. From the overall result, it can be assumed that the IAA of 0.5 ppm has the most positive effect on water spinach among other treatments.

Abstract

Abstract. Barus T, Halim R, Hartanti AT, Saputra PK. 2019. Genetic diversity of Rhizopus microsporus from traditional inoculum of tempeh in Indonesia based on ITS sequences and RAPD marker. Biodiversitas 20: 847-852. The main microorganism for tempeh fermentation is Rhizopus microsporus. These days, many tempeh producers use commercial inoculum, such as ‘Raprima’ as resource of R. microsporus. As a result, the genetic diversity of R. microsporus that had been reported in Indonesia has diminished. Information about genetic diversity is needed as a basis to select R. microsporus as tempeh inoculum. This research aims to investigate the genetic diversity of R. microsporus from waru leaves based on Internal Transcribed Spacer (ITS) Sequence and Random Amplified Polymorphic DNA (RAPD) markers. A total of 25 R. microsporus were isolated from traditional inoculum waru leaves (Inoculum 1) and traditional inoculum other than waru leaves (Inoculum 2). Amplification of ITS sequence was done using universal primer pairs of ITS-4 and ITS-5. Amplification of RAPD markers was done using primers OPC-08, OPC-19, OPQ-6, R-108, OPA-09 and OPJ-20. ITS sequence was not sufficient to compare the similarities among R. microsporus. On the other hand, RAPD markers successfully compared the similarities among 25 R. microsporus. A total of 25 R. microsporus were divided into 9 clusters. R. microsporus from Inoculum 1 grouped into Cluster 1, Cluster 3 and Cluster 4-8. Inoculum 2 grouped into Cluster 2 and Cluster 9. R. microsporus from tempeh grouped into Cluster 4 and was different from Inoculum 1 and Inoculum 2, except for TB3.

Abstract

Abstract. Hartanti AT, Susanti FN, Prasasty VD, Radiastuti N. 2021. Culturable endophytic fungal diversity in cassava tubers of Indonesia. Biodiversitas 22: 1250-1260. Fungal endophytes are found in plant tissue and do not harm the host plant. Cassava (Manihot esculenta Crantz) is the third staple food besides rice and corn. This study aimed to identify the diversity of endophytic fungi in the cassava from several regions in Indonesia, including Bogor, Lembang, Makasar, Serang, Singaraja (Buleleng), Subang, Wonogiri, and Wonosobo. The endophytic fungi were isolated and were molecularly characterized using the internal transcribed spacer (ITS) rDNA sequence (ITS4 and ITS5). Phylogenetic analysis was performed by using the MEGA 7 with a bootstrap value of 1000 repetitions. All 14 isolates of endophytic fungi were isolated from different regions. Of these six isolates belonged to genus Fusarium (F. solani FS5, F. lichenicola FS2, F. lichenicola FS4, F oxysporum FS1, F. falciforme FS3, F. falciforme FS6), and four to the genus Aspergillus (Aspergillus sp. FS7, Aspergillus sp. FS8, Aspergillus sp. FS9 and A. fumigatus FS10). In addition, Nectria pseudotrichia, Lasiodiplodia sp., Penicillium citrinum, and Schizophyllum commune were also isolated from cassava tubers. In conclusion, it suggested that diverse endophytic fungi colonize tubers of cassava growing in Indonesia.

Abstract

Abstract. Hartanti AT, Kurniawan BC, Gunawan AW. 2024. Potential of Rhizopus delemar and Rhizopus microsporus as tempeh starters. Biodiversitas 25: 4208-4214. The biodiversity of tempeh-associated Rhizopus in Indonesia has decreased due to the widespread use of commercial starters. This study aimed to conserve three pure Rhizopus strains (Rhizopus delemar ATH 53, R. microsporus ATH 1 and ATH 24) of tempeh starters and optimize their use in tempeh production. Using rice flour and Rhizopus spores at 106/mL concentration, tempeh starters were made and stored for up to 12 weeks, maintaining spore viability at 106/g with 7-14% water content. Tempeh production experiments utilized three starter doses (0.1, 0.2, 0.3 g per 100 g soybeans) and three different incubation temperatures (27, 30, and 35°C), all of which successfully yielded tempeh. Sensory evaluations revealed that ATH 1 and ATH 53 starters were superior over ATH 24, which produced a distinct yellow tempeh that sporulated easily. It was also noted that 0.1 g of starter per 100 g of soybeans was sufficient for producing high-quality tempeh, offering an efficient use of starter. By successfully conserving these Rhizopus strains as viable tempeh starters for at least 12 weeks, results of present study demonstrate a promising approach to maintaining biodiversity in Indonesian tempeh production, paving the way for further exploration of traditional Rhizopus strains in tempeh making.
