Abstract

Diabetes Mellitus (DM) is an incurable disease that can be prevented clinically from worsening. This disease is based on classification related to pancreatic damage in total or partial so that the impact on insulin production. Glucose transporter is a protein that can glucose uptake from outside cell into cell and provides an important role in the body's glucose homeostasis. Glucose transporter-2 (GLUT-2) is found in the pancreas, liver, and kidney. Parameters used are the expression and amount of GLUT-2 density in pancreatic β cells and in hepatocyte cells in normal and diabetic Rat. Used Wistar strain male white rats. An immunohistochemistry (IHC) staining technique was performed to observe the expression of GLUT-2 and the macbiophotonics image J program to determine total density of GLUT-2. GLUT-2 is marked in brown in the cell nucleus surrounded by cytoplasm. The results showed an increased in GLUT-2 expression in hepatocyte cells with a total density of 25,447,052 and a decreased in GLUT-2 expression in pancreatic β cells with a total density of 5,074,544 by Streptozotocin 50 mg/kg BB-Nicotinamide 100 mg/kg BB (STZ-NA) Intraperitonial routes in induced DM rat. STZ-NA induced triggers damage in pancreatic β cells and significantly influences the expression of GLUT-2 protein. High blood glucose levels give different GLUT-2 activity in hepatocyte cells due to the effects of induced agent.

Abstract

Antibiotics will be beneficial and have the anticipated therapeutic effect if prescribed and administered as instructed. The purpose of this study was to evaluate community attitudes and knowledge levels regarding the use of antibiotics, as well as the relationship between these two variables. The sample size for this descriptive-analytic study was 95 respondents, and a cross-sectional design was used. The sampling technique utilized was purposeful sampling. Women made up the bulk of responses (65.3%), and their age range was 36 to 45 (44.2%). The latest educational levels indicated were high school (61.1%), antibiotic use in the past or present (100%), and housewife (38.1%). The respondent received scores of 62.1% for "excellent" knowledge, 3.5% for "adequate" learning, and 7.4% for "poor" expertise. In contrast, there were 0.0% of participants in a horrible disposition, 52.6% with an acceptable attitude, and 47.4% with an excellent attitude. The Chi-Square test findings showed a significant value of 0.097, or more significant than 0.05, between the variable amount of knowledge and attitudes against antibiotics. The p-value (> 0.05) indicates no correlation between the level of competence and perspectives regarding the use of antibiotics. Education, knowledge, and attitudes toward antibiotics were found to be correlated, with a p-value of (0.05). It was determined that the local population had a good and insightful understanding of antibiotics.

Abstract

Diabetes mellitus (DM) is a degenerative disease characterized by abnormalities in carbohydrate, lipid, and protein metabolisms. This research was aimed at the gel’s pharmacological activity, the extract’s potential in treating diabetic wounds in male rats of Melastoma malabathricum L leaves (ML) and Psidium guajava L leaves (PL), as well as microbiological activity. Viscous extracts of ML and PL were prepared in gel dosage form with concentrations of 4% and 6%. Clindamycin was used as a positive control. This study was conducted using an experimental laboratory method, and the study population included white male rats. The pharmacological activity was tested in the form of a gel dosage formula, and the rats were made diabetic using alloxan. The potential of the extract was observed in healing diabetic wounds in male rats. ML and PL leaf extract gels affect wound healing in diabetic patients. This can be seen from the change in diameter. Wound swab examination revealed the presence of bacteria Klebsiella pneumoniae and Staphylococcus aureus. Secondary metabolites of flavonoids, tannins, steroids, and saponins help stimulate the regeneration of epithelial cells and tissues. The results for wound healing of 4% and 6% gel formulations were derived from maturation data on day 10.
