Abstract

Diabetes melitus (DM) merupakan penyakit dengan angka kejadiannya cukup besar dan merupakan salah satu masalah kesehatan yang cukup tinggi di belahan dunia. Angka prevalensi kejadian diabetes pada anak pada Januari 2023 meningkat 70 kali lipat. Kesehatan seorang anak sangat dipengaruhi oleh genetik orang tuanya, faktor ini juga termasuk pola makan orang tuanya. Pola makan orang tua juga mempengaruhi status genetik anak. Asupan nutrisi dapat mempengaruhi perubahan DNA yang dapat diturunkan kepada keturunannya. Kebiasana makan cenderung kurang sehat dapat memicu penyakit diabetes di kemudian hari. Pengabdian masyarakat ini bertujuan untuk meningkatkan kesadaran masyarakat terhadap pencegahan diabetes melitus pada anak. Sasaran kegiatan ini meliputi kader posyandu, ibu dan balita di Posyandu Rosela Kelurahan Lempake Samarinda. Tim pengabdian melakukan penyuluhan dan melakukan evaluasi terkait kegiatan ini. Peserta akan mendapatkan penyuluhan dan buku saku yang akan diberikan secara selesai kegiatan. Hasil evaluasi kepuasan  kegiatan dilakukan secara deskriptif menunjukan 50 % peserta merasa sangat puas sebanyak  karena dinilai memberikan kebermanfatan dalam meningkatkan pengetahuan dan pemahaman masyarakat terhadap pencegahan diabetes melitus pada anak.  Kata Kunci: Anak, Diabetes Melitus, Pencegahan, Pengetahuan, Penyuluhan Diabetes mellitus (DM) is a disease with a significant incidence rate and is one of the major health issues around the world. The prevalence rate of diabetes in children increased 70-fold in January 2023. A child's health is greatly influenced by their parents' genetics, which includes their eating habits. Parental eating patterns also affect the child's genetic status. Nutritional intake can influence DNA changes that can be passed on to offspring. Unhealthy eating habits can trigger diabetes in the future. This community service aims to raise public awareness about preventing diabetes mellitus in children. The target audience for this activity includes posyandu cadres, mothers, and toddlers at Posyandu Rosela, Lempake Village, and Samarinda. The service team conducts outreach and evaluates the activity. Participants will receive outreach education and a pocketbook provided at the end of the activity. The satisfaction evaluation results descriptively show that 50% of participants are very satisfied, as it is considered beneficial in increasing public knowledge and understanding of diabetes mellitus prevention in children.

Abstract

The community service activity themed "Preparation of Rhizome Simplisia and Encouragement for the Formation of Ukot Umot in the Farmer Group of Lempake Village, Samarinda" aimed to increase public knowledge about the preparation of high-quality rhizome simplisia and to encourage the establishment of local institution-based organizations (Ukot Umot). The activity involved farmer groups, women's PKK groups, and village officials. The materials presented included stages of washing, cutting, drying, and storing simplisia. Additionally, a participatory discussion on the importance of Ukot Umot institutions was conducted to strengthen collaboration and improve market access. Based on an evaluation through a questionnaire, 100% of participants stated that the activity was highly beneficial, with high satisfaction levels regarding the relevance of the material, the delivery method, and the benefits of the activity. The initial impacts of the activity included increased participant knowledge, a commitment to forming Ukot Umot, and motivation to process rhizomes independently. Despite challenges such as limited facilities, this activity successfully laid the foundation for the sustainable development of rhizome-based businesses.

Abstract

Skin wrinkles are caused by the cleavage of collagen fibers by the major collagenase enzyme matrix metalloproteinase-1 (MMP-1). Peptide-based matrix metalloproteinase-1 inhibitors present a potential new therapeutic strategy to prevent MMP-1-mediated collagenolysis, which is induced by UV-induced MMP-1 overexpression. This in silico study investigated the MMP-1 inhibitory capacity of carefully selected and well-designed hexapeptides. A molecular docking method was employed to screen 14 hexapeptides. The target ligands were observed to interact non-covalently. The LYYYGP peptide sequence was found to form three hydrogen bonds (H-bonds) with amino acid (AA) residues in the catalytic domain of MMP-1. Additionally, hydrophobic interactions between these peptides and the protein were observed at certain sites. The maximum binding affinity was observed for the LYYYGP sequence (binding free energy, ?G = -9.00 kcal/mol). These sequences exhibited binding energies comparable to those of the LSYYGP sequence (-8.48 kcal/mol) and YSGYYP sequence (-8.27 kcal/mol). The YSGYGY sequence demonstrated a lower binding affinity for the target receptor, with ?G = -5.59 kcal/mol. This study indicates that specific peptide sequence components are crucial for forming hydrogen bonds that link proteins to peptides. Further investigation, encompassing chemical synthesis and laboratory experimentation, is requisite to substantiate the efficacy of the two hexapeptides.

Abstract

Hyperglycemia is a condition where blood sugar increases significantly which can lead to diabetes mellitus. Sambung Nyawa (Gynura Procumbens) leaves are a plant that is empirically believed to reduce hyperglycemia and has been scientifically proven based on several studies, however there has been no research that provides information about sembung nyawa leaves in infusion form. This study aims to determine the content of secondary metabolites in the infusion of the leaves of Gynura Procumbens and its antihyperglycemia activity in male males. This study consisted of phytochemical and antihyperglycemia screening using a tolerance method induced by glucose monohydrate at a dose of 2g/kgBW. The test group consisted of a negative control group (Na CMC 1%), a positif group (Glibenclamide), and 3 groups of sambung nyawa leaf infusion (5%, 10%, 15%). The results of the research showed that the infusion of sembung nyawa leaves contained secondary metabolites, namely tannins, phenolics, flavonoids and saponins. The effect of reducing blood sugar levels in test animals showed a significant reduction (0.00<0.05) at a 10% concentration infusion compared to the negative control and not significant to the positive control. The conclusion shows that the infusion of sambung nyawa leaves has an antihyperglycemic effect. Keywords:          Antihyperglycemic, Sambung Nyawa Leaves, Glucose Monohydrate   Abstrak Hiperglikemia merupakan kondisi peningkatan gula darah yang cukup signifikan yang akan berubah menjadi kondisi diabetes mellitus. Daun sambung nyawa (Gynura Procumbens) merupakan tumbuhan yang dipercaya secara empiris menurunkan hiperglikemia dan terbukti secara ilmiah berdasarkan beberapa penelitian dalam bentuk ekstrak. Penelitian ini bertujuan mengetahui kandungan metabolit sekunder pada infusa daun sambung nyawa dan aktivitas antihiperglikemia pada mencit jantan. Penelitian ini terdiri dari skrining fitokimia dan antihiperglikemia menggunakan metode toleransi yang dinduksi glukosa monohidrat dengan dosis 2g/kgBB. Kelompok uji terdiri dari kelompok kontrol negatif (Na CMC 1%), positif (Glibenklamid), dan 3 kelompok infusa daun sembung nyawa (5%, 10%, 15%). Hasil penelitian menunjukkan infusa daun sambung nyawa mengandung metabolit sekunder yaitu tanin, fenolik, flavonoid dan saponin. Efek penurunan kadar gula darah pada hewan uji menunjukkan penurunan yang signifikan (0,00<0,05) pada infusa konsentrasi 10% dibandingkan dengan kontrol negatif dan tidak signifikan terhadap kontrol positif. Kesimpulan menunjukkan bahwa infusa daun sambung nyawa mempunyai efek antihiperglikemia. Kata Kunci:         Antihiperglikemia, Daun Sambung Nyawa, Glukosa Monohidrat

Abstract

Kersen (Muntingia calabura Linn.) is a plant with a large population in Indonesia. Thus, the abundance of this number can be used as a source of raw materials for medicine. Kersen has many benefits, especially in the leaves as an antibacterial. The purpose of this study was to determine the extract yield, compound groups, and antibacterial activity against Salmonella typhi and Staphylococcus aureus. Antibacterial activity was measured using the well method and TLC Bioautography. The results showed an extract yield of 19.77%. The TLC profile identified flavonoid, alkaloid, tannin, and steroid compound groups. Kersen leaf extract showed antibacterial activity against Salmonella typhi and Staphylococcus aureus with moderate to strong antibacterial power at concentrations of 40%, 50%, and 60%. The groups of compounds suspected of playing a role in antibacterial activity are flavonoids, alkaloids, tannins, and steroids. This study shows that kersen leaf extract has the potential as an antibacterial agent against Salmonella typhi and Staphylococcus aureus bacteria.

Abstract

A workshop on making gummy candy based on stingless bee honey was held in Bangun Rejo Village, Kutai Kartanegara Regency, as an effort to empower the community through innovation in locally processed food products. The aim of this activity was to improve the skills of MSME actors and village residents in producing stingless honey gummy candy, and to elevate the added value of local honey. The methods used included a short theoretical session about the composition of stingless bee honey and the technical process of gummy candy production, a live demonstration, group practice among participants, as well as evaluation via pre-test and post-test on understanding and skills. The workshop was attended by 50 participants from various MSMEs and community groups. Results showed that after training there was an increase in participant understanding by approximately 75% compared to before the training. Furthermore, participants succeeded in making gummy candy with a chewy texture, the natural sweetness of stingless bee honey, and a distinctive aroma that was well accepted in organoleptic tests. This workshop made a tangible contribution to local product diversification, increasing production capacity, and the potential enhancement of village income through signature products based on stingless bee.

Abstract

Resistensi terhadap antibiotik konvensional memiliki tingkat kejadian yang cukup tinggi. Dibutuhkan pengembangan antibiotik baru salah satunya golongan senyawa peptida yang merupakan kandidat yang potensial sebagai agen antimikroba. Penelitian ini bertujuan untuk mensintesis peptida baru dan mengevaluasi aktivitas antimikrobanya. Peptida antimikroba dapat didesain menggunakan machine learning iAMPpred dengan menganalisis nilai tertinggi probabilitas antibakteri dan antijamur. Peptida disintesis secara kimia di laboratorium menggunakan metode sintesis peptida fase padat dan diuji aktivitas antimikrobanya dengan menggunakan metode dilusi untuk menghitung nilai Minimum Inhibitory concentration (MIC). Hasil penelitian menunjukkan desain heksapeptida linear Prolin-Leusin-Lisin-Leusin-Fenilalanin-Fenilalanin (PLKLFF) memiliki nilai probabilitas tertinggi dengan nilai probabilitas antibakteri 0,87 dan probabilitas antijamur 0,91. Heksapeptida linear ini berhasil disintesis menggunakan metode sintesis peptida fase padat dengan rendemen sebesar 93,32%. Keberhasilan sintesis dikonfirmasi menggunakan spektrofotometer infra merah dan spektroskopi massa. Senyawa heksapeptida linear PLKLFF memiliki aktivitas antijamur/fungistatik sedang terhadap jamur C. albicans ATCC 10231 dengan nilai MIC 312,5 µg/mL dan tidak aktif terhadap bakteri E. coli ATCC 11229 dan S. aureus ATCC 6538.
