Abstract

Chitosan is the result of deacetylation process from chitin which it can be found on Crustacean outer shell such as crabs. Chitosan can bind fat if it was consumed by human. The fat-binding ability of chitosan depends on the deacetylation degree. The research have been made into two phases. The first phase, chitosan was made from crab shell using NaOH. The deacetylation degree from chitosan that was made from the earlier process was analyzed with FTIR. The deacetylation degree result of the research was 59.39% using NaOH 50%. The second phase was the process of adsorbing triglycerides using chitosan in 10, 30, 45, and 60 minutes which was analyzed using spectrophotometer UV-Vis. The result of this research showed that the optimum triglycerides with 0.5, 1, 3 gram of chitosan was 2.99%, 3.14%, and 3.36%.

Abstract

AbstrakSusu kedelai dan tempe adalah pangan olahan yang digemari masyarakat Indonesia dan menjadi salah satu sumber protein nabati yang terbuat dari kedelai. Kedelai mengandung senyawa isoflavon glikosida yang bermanfaat untuk kesehatan sebagai pencegah penyakit kardiovaskuler, kanker, diabetes, hipertensi, osteoporosis, obesitas dan menopause. Isoflavon glikosida memiliki struktur yang lebih kompleks, sehingga perlu mengalami proses biokimiawi untuk menyederhanakan strukturnya agar mudah diserap oleh tubuh. Isoflavon glikosida akan disederhanakan menjadi bentuk aglikon selama proses fermentasi dan hidrolisis kimiawi. Genistein merupakan salah satu bentuk isoflavon aglikon yang banyak ditemui pada produk kacang-kacangan. Susu kedelai dan tempe merupakan produk olahan dari kedelai, perbedaannya adalah tempe telah mengalami proses fermentasi, sehingga diyakini memiliki kadar isoflavon aglikon yang tinggi. Penelitian ini bertujuan untuk menentukan kadar genistein pada ekstrak susu kedelai dan tempe dengan menggunakan spektrofotometri UV-Vis. Sampel diekstraksi dengan menggunakan etil asetat, sehingga diperoleh ekstrak kental, selanjutnya diukur kadar genistein dengan metode spektrofotometri UV-Vis, menggunakan baku standar. Hasil penelitian diperoleh kadar genistein pada ekstrak susu kedelai dan tempe yaitu masing-masing 0.613 dan 5.519% (b/b). Hal ini menunjukkan bahwa kadar isoflavon aglikon lebih tinggi pada tempe dibanding susu kedelai.Kata kunci: genistein, isoflavon aglikon, susu kedelai, tempe AbstractSoymilk and tempeh are a processed food that is popular with Indonesian society and becomes one of vegetable protein source made from soybean. Soy contains isoflavones glycosides are useful for health as preventions of cardiovascular disease, cancer, diabetes, hypertension, osteoporosis, obesity, and menopause. Isoflavones glycosides have more complex structures, so they need to undergo biochemical processes to simplify their structure and are easily absorbed by the body. Isoflavones glycosides will be simplified into aglycone forms during chemical fermentation and hydrolysis processes. Genistein is one form of isoflavone aglycone that is commonly found in nuts products. Soymilk and tempeh are processed products from soybeans, the difference is tempeh has undergone a fermentation process, so it is believed to have high levels of isoflavone aglycone. The purpose of this study was to determine the genistein content in the extract of soymilk and tempeh by using UV-Vis spectrophotometry. The sample was extracted by using ethyl acetate, so obtained by viscous extract, then measured its genistein level by UV-Vis spectrophotometry method, using standard reference. The result showed that genistein content of soymilk and tempeh extract were 0.613 and 5.519% (w/w), respectively. This indicates that isoflavone aglycone content is higher in tempeh than soymilk.Keywords: genistein, isoflavone aglycone, soymilk, tempeh

Abstract

Flavonoids are natural phenolic compounds found in almost all plants that have potential as antioxidants. Based on previous research, stating that black soybeans contain compounds of flavonoids. This analysis aims to establish the levels of total flavonoids on ethanol extract of black soybean hydrolyzed. Determination of total flavonoid levels of ethanol extract of black soybean hydrolyzed has been made with UV-Vis spectrophotometry method. Black soybean hydrolyzed using HCl and extracted using ethanol 70%. The extracts obtained were analyzed qualitative by the method of coloring reaction and quantitative analysis using UV-Vis spectrophotometer, where quercetin is used as a reference standard. Measurements were performed at maximum wavelength of 444.45 nm. The research showed the average level of total flavonoids is 11.83 mgQE / g extract or 1,163 %. In conclusion extract of black soybean hydrolyzed rich of total flavonoids, and it is related to the activity as antioxidant.

Abstract

Phenolic is a compound that having an aromatic ring with one or more hydroxyl groups. Phenolic compounds in foods can be grouped into simple phenols and folic acid. This study aimed to determine the total phenolic content of the leaf extract of nutmeg (Myristica fragrans Houtt). Leaves of nutmeg extracted using 70% ethanol, then the extract conducting qualitative analysis by reaction with FeCl3 as specific reagent. The result of this reaction is green solution, it means there are phenolic compound in the extract. The quantitative process of total phenolic was analyzed by UV-Vis spectrophotometry. A comparison used in this analysis is gallic acid; it is a standard reference in the determination of total phenolic content. Sample measured at a wavelength of 860.7 nm, which is the maximum wavelength of gallic acid. Total average content of phenolic is 183.56-mgGAE/g extract. The ethanol extract of nutmeg’s leaves have high total phenolic content, which means it is potential as antioxidant.

Abstract

The reasearch test effect anthypercholesterolemia Chytosan Windu Shrimp Shell with aimed to know the effect to decrease the total blood cholesterol degree on adult rabbit. These research include 3 group, namely 1 group of control and 2 groups of examined namely chytosan 5% and 10% w/v. The observation applied with measure of the total blood cholesterol degree on adult rabbit before induction of cholesterol, then inducted of cholesterol as long as 7 days, then observed  cholesterol degree, and then its gived chytosan 400 mg/kgBB (5%w/v) and 800 mg/kgBB (10%w/v) at the 8th and  measured again of the total blood cholesterol degree after examined Chytosan at concentration 400 mg/kgBB (5%w/v) dan 800 mg/kgBB (10%w/v), conduct decreasing of total blood cholesterol degree on adult rabbit. Key Word : Antihypercholestrolemia, Penaeus monodo, Oryctolagus cuniculus, Invivo

Abstract

Mungbean (Vigna radiate) and red bean (Phaseolus vulgaris) is a type of food containing isoflavone in the form of glycoside (genistin, daidzin, glycitin). Isoflavones are one type of phytoestrogen which have similar chemical structure with estradiol. Therefore, it can be used not only to inhibit but also to prevent many symptoms related to estrogen deficiency. Human body cannot adsorb isoflavone in the glycoside form. In order to make it absorbable, isoflavone should be hydrolyzed to defuse its glycoside chain to be aglycon (genistein, daidzein, glycitein). The aim of this study was to determine the influence of hydrolysis on chromatogram profile of mungbean and red bean. Hydrolysis was conducted by chemical method using hydrochloride acid (HCl). The chromatogram profile was determined by High Performance Liquid Chromatography method (HPLC) using C18 reverse phase column, the mobile phase was methanol : water (7:3), the sample injected automatically as much as 10 ml, the flow rate of 1 ml/min with a temperature of 28°C, at a wavelength of 254 nm. This study showed that chemical hydrolysis successes to remove the glycoside chain in both mungbean and red bean extract.

Abstract

Traditional medicine is very popular in Indonesia, so it is necessary to do this research to analyze the presence or absence of chemical substances contained in traditional medicine. This research aims to determine acetaminophen content on jamu pegal linu circulating in Makassar city by High Performance Liquid Chromatography (HPLC). The jamu were obtained from several drugs stores near Terong traditional market. Five brands of jamu pegal linu detected their acetaminophen content by HPLC method using a UV-Visible SPD 20A detector with a wavelength of 254 nm. The result showed that chromatogram on jamu pegal linu is the same as the acetaminophen at the retention time of 5.28 minutes. The whole of jamu pegal linu analyzed positively contain acetaminophen and prohibited for consumption in accordance with the regulations of National Agency of Drug and Food Control (POM) Republic Indonesia No.KH.00.01.43.2773/2008.

Abstract

Vitamin C is one of the nutrients that act as antioxidants derived from fruits and green vegetables, such as the broccoli (Brassica oleracea L.). This study aims to warming influence on levels of vitamin C in broccoli (Brassica oleracea L.) with qualitative and quantitative analysis. Qualitative analysis was performed using specific reagents that ammonium molybdate, Fehling Fehling's A and B which have indicated a positive sample containing vitamin C, and vitamin C were analyzed quantitatively using UV- Vis spectrophotometry at a wavelength of 570 nm. The results showed levels of vitamin C in raw broccoli 0.62915 mg/g and steamed broccoli 0.45449 mg/g.

Abstract

Polyphenols are a class of chemical compounds most commonly found in plants, these compounds have many health benefits associated with the character it has. The antioxidant activity of natural materials connected by polyphenol content, which can counteract free radicals. Objective: This study aims to determine the levels of total phenolic and flavonoid in crude extracts and purified extracts of the green tea leaves (Camellia sinensis). Methods: Determination of total phenolic content was using standard gallic acid by the Folin-Ciocalteau method. Total flavonoid assay using standard reference of quercetin by Chang method. Instrument used to measure the absorbance of the reaction is a UV-Visible spectrophotometer. Results: The results obtained after the measurement indicates that the sample of crude extract and purified extract of tea leaves containing total phenolic levels 40.5 and 43.9 mgGAE/g, respectively, while the levels of total flavonoids contained respectively by 2.77 and 3.14 mgQE/g. From the results obtained it can be concluded that the green tea leaves have a total phenolic content is greater than the total flavonoid content, thus it has great potential as an antioxidant

Abstract

Bitter leaf, Vernonia amygdalina (VA), is used in various alternative medicine in several countries,including Indonesia. This study aims to evaluate the antioxidant activity of VA leaves by three differentin vitro methods. The antioxidant activity was evaluated by radical scavenging of 1,1-diphenyl-2-picryl-hydrazyl (DPPH), ferric reducing antioxidant power (FRAP), and cupric reducing antioxidantcapacity (CUPRAC). The antioxidant capacity by FRAP and CUPRAC methods were expressed asmg of quercetin equivalents (QE) per g extract (mgQE/g) and mg of ascorbic acid equivalents (AAE)per g extract (mgAAE/g), respectively. All methods were measured by UV-Visible spectrophotometryat maximum wavelength. The results exhibited that VA leaves ethanolic extract contains flavonoidphenolic.The VA leaves ethanolic extract has an antioxidant activity by scavenging the DPPH methodwith the half inhibition concentration (IC50) was 8,22 μg/mL. The antioxidant capacity of VA leavesethanolic extract by FRAP and CUPRAC methods was 21,03 mgQE/g and 160,64 mgAAE/g extract,respectively. The VA leaves ethanolic extract has a powerful antioxidant potency which might correlateto the flavonoid-phenolic content in the extract.
