Abstract

The present study was undertaken with primary objective to characterize of Gembrong goat breeds. It is essential to characterize the germplasm for intragenetic variability, which will help in planning for conservation strategy as well as genetic improvement. DNA Genome was isolated from hairs. Nine microsatellite were amflified by PCR. PCR product were run on 6% bis-Acrylamide gel in automated DNA sequencer. Flourescent signals from the dye-labeled microsatellite were detecting using STRand software. The result show that all markers (INRA005, INRA063, INRA023, ILSTS19, ILSTS87, SRCRSP8, MAF65, OarFCB20, and McM547) successfully amplified in Gembrong goat microsatellite loci. The number of alleles per locus ranged from 1 (INRA005) to 4 (ILSTS87). All the microsatellites investigated were found to be highly polymorphic, except INRA063. In total, 23 alleles were observed for the 9 microsatellite loci. The allele sizes ranged from 99 bp (OarFCB20) to 240bp (SRCRSP8). The study can be extended to include large many microsatellites in different chromosome location to validate the results.

Abstract

The purpose of this study was to determine the effect of postmortem time on percentage of life epididymal sperm from postmortem canine caudae epididymides. A total of 9 dog were used and divided into three group. T0 was control group, T1, 3 hours postmortem and T2, 6 hours postmortem. This way, samples were obtained at different times postmortem. Sperm were extracted from the caudae epididymis by means of cuts. The result showed that the percentage of life sperm were 67,16 ± 5.67(T0), 46.33± 5.60 (T1) and 24.00 ± 4.35 respectively. We could appreciate that percentage of life was affected by postmortem time. There was significant decrease life sperm recovered from epididymides postmortem (P<0.01). In conclusion, epididymal sperm from dog undergo decrease of percentage of life, but it could stay acceptable within many hours postmortem. We intepreted these data to indicate that it may still be possible to obtain viable spermatozoa many hours later.

Abstract

This study was conducted to investigate the reproductive performance of bali cattle in breeding instalation bali cattle of Sobangan, Badung, Bali. Totaling 288 heifers were use to represents the benchmark reproductive performance of bali cattle. Data collected were service per conception (S/C), gestation length, birth weight, and postpartum estrus. The overall means for service per conception (S/C), gestation length, birth weight, and postpartum estrus were 1,3 times; 284,63±10,73 kg; 17,89±1,04 kg; dan 101,71±32,31 days.

Abstract

The Balinese believe that body size (the ratio of body length to snout length) in dogs is related to behavior, therefore body size is used to predict dog character. How to predict by measuring body size is a local wisdom that is still trusted for choosing a good dog. This belief has been running for a long time from their ancestors and has become a culture. Dog behavior according to local wisdom in Bali in general and in Sukawana Village, Kintamani District, Bangli Regency in particular, is categorized as: guna, jaya, forced, tek and kyul. Guna is a dog with an obedient character, useful and suitable as a hunting dog. Jaya is a dog with good character, attractive, energetic, tends to be hyperactive, this type of dog is also good for hunting. Force is a dog that is difficult to manage, requiring special handling, suitable for keeping the house. Tek is a dog with dirty behavior and difficult to train or manage and Kyul is a dog with a lazy character. A total of 66 Kintamani dogs were used as experimental animals. The research design used a cross-sectional study design. Body size observation data was then compared with clinical observations in the same dog. The results show that there is a relationship between behavior and body size (ratio of body length to snout length). It was concluded that body size (body length to snout length ratio) can be implemented to predict behavior.

Abstract

Tujuan penelitian ini adalah mendapatkan informasi dasar mengenai struktur genetika menggunakan marka melokuler DNA mikrosatelit gen bovine lymphocyte antigen (BoLA) dan hubungannya dengan kualitas semen sapi bali. Data diambil dari 8 ekor sapi jantan yang ada di Unit Pelaksana Teknis Daerah (UPTD) Inseminasi Buatan, Baturiti, Tabanan, Bali. Hasil pemeriksaan kualitas sperma menunjukkan bahwa volume semen sapi bali 3,0-6,5 ml, konsentrasi spermatozoa 800-11.000 juta sel/ml, dan persentase spermatozoa motil 70-75%. Hasil penelitian pada sapi bali menunjukkan bahwa ketiga lokus mikrosatelit yang digunakan teramplifikasi pada sapi bali dan jumlah alel pada lokus RM185 dan BM1815 adalah 2 sedangkan jumlah alel pada lokus DRB3 adalah 1. Heterozigositas per lokus berkisar 0-0,533. Nilai PIC per lokus antara 0-0,375. Dari hasil penelitian ini dapat disimpulkan bahwa ukuran alel pada ketiga mikrosatelit berpotensi sebagai marka molekuler yang berperan terhadap kualitas semen pada sapi bali.

Abstract

Aspergillus sp are fungi that commonly caused a systemic respiratory infection in Human and Dog. In Bali, most of the dog are free-roaming dogs, and has poor skin health condition. Some of them are suspected with fungal infection. Aspergillus sp has high risk to infect the dog, because they are available everywhere in environment. Therefore, this research is conducted for adding primary data of Aspergillus sp isolated from dog’s skin and their prevalence. The samples are 15 free-roaming Bali dogs with suspected fungal infection. Identification was carried out by using microscopic and macroscopic observation methods, skin swab was taken from the dog’s skin using the Mackenzie method, and the sterile toothbrush swab gently from the dog’s skin to the Sabouraud Dextrose Agar (SDA) media. The fungi were cultured on SDA media for macroscopic identification for approximately 3-4 days, mostly to observe how the fungal colony grew and its color. Microscopic identification are using adhesive tape, the tape was gently pressed into the fungal colony, the tape was stuck into the surface of object-glass with 3-4 drops of Methylene Blue staining, and the object glass was examined by using a light microscope, the findings of Aspergillus sp fungi are recorded and the data are descriptively presented. The result shows that most Aspergillus species that were found is Aspergillus fumigatus and Aspergillus niger according to their colonies growth and microscopic appearance, and the prevalence from 15 samples were 53.33%.
