Abstract

Induced mutagenesis is one way to improve the quality of crops, especially to increase the resistance to pests and diseases. This research aimed to determine colchicine and ethyl methane sulfonate (EMS) mutagenesis on cowpea M3 purple and mung bean Vima1, in concatenation for developing resistant crops. This research consists of four packages: (1) first package (cowpea M3 purple seeds treated with colchicine solution, (2) second package (cowpea M3 purple seeds treated with EMS solution), (3) third package (mung bean Vima1 seeds treated with colchicine solution) and (4) fourth package (mung bean Vima1 seeds treated with EMS solution). The results of this research revealed that induced mutagenesis by colchicine solution treatment reduced the incidence of bean leaf beetles up to 19% on cowpea M3 purple, as well leaf miners by 5% to 9% and bean leaf beetles up to 5% on mung bean Vima1. Treatment of EMS solution decreased the incidence of bean leaf beetles by 17% on cowpea M3 purple and pink mealybug by 5% to 15% on mung bean Vima1. Induced mutagenesis using EMS solution significantly decreased vegetable leaf miner incidence by 33% to 93% or 71% on average. Colchicine and EMS solution treatment caused aphid attacks in cowpea M3 purple, particularly in pods. The attacks did not happen on leaves and aphid incidence in pods was slower than in control. Some promising mutant candidates were found from this research that will be used in further crop development studies. It is expected that the method and results of this research could inspire faster development of resistant crops.

Abstract

Curvularia lunata is a pathogenic fungi that attack various plant species including lettuce. Its control needs to be carried out in an environmentally friendly manner, namely using botanical fungicides. M. pruriens is a legume plant that is widespread in Indonesia that has a potential as a botanical fungicide, beside as a traditional medicine. The purpose of this study was to study the chemical content of M. pruriens leaves and its potency to inhibit the growth of C. lunata. The determination of the compounds was carried out by Gas Chromatography-Mass Spectrophotometry and the inhibition test was carried out using diffusion well, and agar dillution method. The result showed that the crude extract of M. pruriens leaves contained ethyl isothiocyanate that act as an antifungal with AUC of 20,78%. The MIC of the extract was 0,9%. The extract at 5% concentration was capable to inhibit C. lunata growth, mycelium mass, and conidia production respectively 38,33%; 67,14%; 99,88% compared to control.

Abstract

Lake Buyan in Buleleng, Bali, has various purposes, such as supplying water for crop and livestock farming and the domestic sector. Wastes generated by human activities in its immediate surroundings are increasing population pressures on the lake’s environment. According to Gubernatorial Regulation Number 16 of 2016, the lake’s water quality is in a poor state and is categorized as ‘polluted’. One of the ways proposed in this research to reduce the pollution level was to build a constructed wetland, which is a waste treatment system combining aquatic plants, soil or other growing media, and microbial assemblages. Results showed that 50 sets of vertical flow constructed wetlands with a single capacity of 40 m3 are needed around Lake Buyan. Abstract

Subclinical mastitis is a disease that quite often affects sows during lactation without any apparent clinical symptoms, one of which is caused by Klebsiella Spp. The purpose of this study was to determine the quality of pig milk and whether Klebsiella spp. was detected in the pig milk. Thirty-four samples were tested using the California Mastitis Test (CMT) method, and samples positive for subclinical mastitis were further tested using MacConkey media. Additional tests were conducted, including the TSIA biochemical test and microscopic examinations of the growing bacterial isolates, to confirm the bacterial identity. The results of this study showed that 29 out of 34 positive samples had subclinical mastitis with different levels of severity. According to the laboratory results, the presence of Klebsiella spp (85%) bacteria was identified. The results of this study indicate that subclinical mastitis caused by Klebsiella spp remains a significant threat in pig farms in Puhu Village, Payangan Subdistrict, Gianyar Regency, particularly when cage sanitation and management are not optimal. Early detection and improved husbandry systems are needed to prevent similar incidents.
