Abstract

ABSTRACT Vesicular Arbuscular Mycorrhizae (VAM) Identification of Chilli (Capsicum annuum L.) and Tomato (Solanum lycopersicum L.) Rhizosphere and Its Spore Multiplication in Zeolite Media Various attempts have been made to increase the production of chilli and tomato plants by farmers, including by using  inorganic fertilizer application on and on going basis. Considering potential problems that may occured due to inorganic fertilizer use, aplication of  biological fertilizer which one of them is containing vesicular arbuscular mycorrhizae (VAM) could be expected to assist the growth of chilli and tomato in more naturally manner. This study was aimed to determine genus and species of VAM in chilli and tomato rhizosphere, its colonization in root tissue, and to examine zeolite media compatibility with corn as a symbiont. Based on the results of the study, it was found four species namely Acaulospora fofeata, A. colombiana, A. Laevis, and Glomus ambisporum in chilli and four species in tomato that were identified as A. fofeata, A. colombiana, Scutellospora calospora, and G. ambisporum. Colonization were found in roots of chilli, tomato, and corn indicated by symbiotic structures arbuscules, vesicles, and inner spores. Zeolite media with corn as symbiotic plant is considered suitable for VAM spore propagation. Keywords: rhizosphere, Acaulospora fofeata, zeolite, inner spore

Abstract

Application Some Types of Organic Fertilizer in a Pack To Suppress Clubroots and Improve Cabbage (Brassica oleracea L.) Plant Growth Cabbage is one type of leaf vegetables from subtropics area that has been known for a long time and cultivated in Indonesia. One of the hardest cabbage diseases is clubroots caused by the Plasmodiophora brassicae pathogen. This pathogen can cause high damage to cabbage plants. The used of synthetic pesticides is less success in the implementation, and caused environmental pollution, especially the soil, this study tried to control clubroots in the cabbage by using 8 types of organic fertilizer in a pack. This study used a Completely Randomized Design (CRD) with 9 treatments including controls with three replications. The results obtained four best treatments that were effective in suppressing clubroots while increasing the leaf height, leaf amount, leaf area, and maximum dry weight of the plants, that three best treatments were organic fertilizer PSBN, Multitonik, and Biotonik. The best treatment to suppress clubroots is found in organic fertilizer PSBN, Multitonik, and Biotonik which has the lowest percentage of ulcer attack by 33,33-55,56% compared to control (100%).

Abstract

Application of Biological Agent and Humus to Suppress Population Fusariumoxysporum f.sp. cubense the Cause of Banana Stem Rot. The study aimed to determine the interaction between P. fluorescens, Trichoderma spp. fungus, and humus in suppressing the population of Fusariumoxysporum f.sp. cubense in the soil. The design used was a randomized blockdesign (RBD) of 2 factors namely antagonistic microbes and humus. There were 18treatments with 3 replications so that 54 experimental units were obtained. Thevariables observed were the population of Foc, population of P. fluorescens,population of Trichoderma spp., and soil pH. The results showed that thecombination of biological agent and humus could reduce the population of Foc. Thehigher concentration of humus combined with biological agent showed thedecreasing population of Foc which causes rot of banana stems.

Abstract

ABSTRACT Effectiveness Test of Some Plant Seeds Extract to Inhibit the Growth of Spotting Leaf Bacteria (Xanthomonas campestris) on Tomato Plant. The X. campestris bacterium is very harmful to tomato plant because the bacteria able to attack the plant seedling up to full grown stage. This bacterium  not only attack the root, but also infect the leaves, fruits and stems. Utilization of botanical pesticides to inhibit the pathogen becomes one alternative option to be recommended. The control method has selective activity and safe for the ecosystem. The purpose of this study is to determine the ability of some plant seeds extract in inhibiting the growth of X. campestris the causal agent of leaf spot on tomato plants. Rose apple seed extract contains phyto-pesticide that can be used to inhibit the growth of leaf spot caused by X. campestris. The minimum inhibitory concentration of the extract was at a concentration of 0.312% with the diameter of inhibition zone of 8 mm. It was expected that rose apple seed extract may be used as an alternative subtance to inhibit the growth of bacterium X. campestris bacterium of tomato plants. Abstract

Tissue Culture of Anthurium Plants (Anthurium andraeanum var. tropical) in MS Media with Growth Regulating Substances BAP and NAA Anthurium (Anthurium andraeanum var. tropical) is an ornamental plant that has been cultivated on a wide scale in the floriculture industry. The advantages of anthurium ornamental plants has beautiful flowers that are suitable for ornamental plants and cut flowers, and the cut flowers have good opportunities in the domestic and international markets. So makes it suitable to be in-vitro developed to produce seedling that are disease resistant, quality and quantity. This experiment was to determine the optimal concentration of 6-Benzylaminopurine and Napthalene Acetic Acid to induce callus from leaves and shoots from anthurium seeds on Murashige and Skoog base media. Experiments on leaf explants using factorial completely randomized design with 25 treatment levels of growth regulator substances BAP and NAA. The results showed that BAP and NAA treatments could increase the growth of anthurium plants. The treatment that has the best result in anthurium leaf curvature is A3B2 (5 mg/l NAA + 3 mg/l BAP), with the fastest curvature time is 2 hst and the explant curves 100%. The treatment that has the best results in shoots induction from anthurium seeds is the treatment of A3B3 (5 mg/l NAA + 5 mg/l BAP) with the appearance of shoots at 7.33 days after planting, having 8.92 of shoots, having 3.33 of leaves, and having 5.85 of roots. The A3B3 treatment showed significantly different from the control treatment. The appearance of shoots on control treatment was 8.66 days after planting, having 3.66 of shoots, having 3.33 of leaves, and having 2.33 of roots. The combination of 5 mg/l BAP + 5 mg/l NAA (A3B3) was the best treatment for shoots growth and was suitable for the purpose of multiplying anthurium seedling.

Abstract

ABSTRACT Identification of Endophytic Fungi from Rice Grain and It’s Inhibiting Ability by In Vitro Against Pyricularia oryzae Cav. Fungal endophyte is a fungi that lives in the tissues of plants without causing disease symptoms on host plants. During symbiotic, many endophyte could produce antibiotic compounds, those can make endophyte microbes become biological control agents against pests and plant diseases. The aims of this study is to determine the types and the number of endophytic fungi spesies that exist in rice seeds stadia, also growth inhibitory ability against Pyricularia oryzae Cav. in vitro. Based on microscopic identification and DNA analysis were found 14 species of endophytic fungi. Endophytic fungi were able to inhibit the develop of Pyricularia oryzae Cav. amount. The greatest inhibitor was Unidentified-2 isolates of 65.6% followed by fungi Phaeosphaeriopsis musae, Sarocladium oryzae, and Sordariomycetes sp., with inhibition of 63.3%, 61.1%, and 58.9% respectively. Abstract

ABSTRACT Inhibiting Ability of Some Fruit Rinds Extract Against the Growth of Ralstonia solanacearum on Chili Ralstonia solanacearum infect plant root through wound that occured indirectly. The symptoms of the disease are whole plant to wilt, the leaves turn yellow to blackish-brown, and eventually the plant dies. In the culture media contained  Triphenyltetrazolium Chloride (TZC) the bacterium tend to form white colony and irregular round shape. The method used to determine the extract activating was well diffusion method. The results of the study showed that application of coconut rind extract inhibited the bacterium. The minimum concentration was 1,25% with diameter of inhibition zone was 15 mm. The phenolic compounds contained on the coconut rind was possibly inhibit the growth of the bacterial colonies. Further research needs to be conducted in vivo to determine the effectiveness of antibacterial compounds contained in the extract of coconut rind. Abstract

The Effect of Pseudomonas spp. Indigenous Inoculation against Clubroot Disease and Cabbage (Brassica oleracea L.) Plant Growth Clubroot that caused by Plasmodiophora brassicae Wor. is a disease that can cause great losses to cabbage. Utilization of biological agents such as indigenous Pseudomonas spp. become an alternative option to be recommended to suppress the disease, because it is environmentally friendly and safe for the balance within ecosystem. The aim of this study was to obtain the most effective isolate of indigenous Pseudomonas spp. in suppressing clubroot disease and promoting growth of cabbage plants. The results showed there were three best effective isolates which able to suppress the clubroot disease as well increase the plant growth. The isolates were isolated from Brassica oleracea var. italica (Pf5), isolated from Brassica oleracea capitata var. rubra (Pf8), and isolated from Eruca sativa (Pf12).

Abstract

The effect of Pseudomonas fluorescens and Trichoderma spp. Applications against Clubroot Disease and Growth of Cabbage (Brassica oleracea L.) in Candikuning Village, Tabanan, Bali The research was aimed to find the effectiveness of P. fluorescens and Trichoderma spp. in suppressing clubroot disease caused by P. brassicae Wor. and increasing the growth of cabbage plants as well. The microbes treatments were set on randomized block design, consisting of 36 treatments and 3 replications. The variables observed were percentage of attack, number of clubroot, microbial density, plant height, leaf area, chlorophyll contents, and plant weight,. The results showed that the combination of P. fluorescens and Trichoderma spp. was able to suppress the percentage of pathogen attack and increase the growth of cabbage plants as well. The most effective treatment was combination between P. fluorescens-1 and Trichoderma-4, followed by combination treatment of P. fluorescens-1 and Trichoderma-3 and then P. fluorescens-1 and Trichoderma-5.

Abstract

The Effectiveness Test of Indigenous Endomycorrhiza on Maize Plant Growth (Zea mays L.). Efforts to increase crop productivity were hampered due to various obstacles such as genetic, environmental, and biotic factors, in order that alternative technologies such as endomycorrhizal biological agents were required. This study was aimed to determine the effectiveness of indigenous endomycorrhiza for the growth of maize. The experiments were carried out in the laboratory and greenhouse. The field experiment was designed in a nested randomized block design. The first factor was genus of indigenous endomycorrhiza (M) consisting of 3 lavels i.e genus Glomus (G), genus Gigaspora (F) and the mixed genus (C), the second factor was the spore dose (D) consisting of 3 levels i.e. D1 = dose of 50 spores, D2 = 100 spores and D3 = 150 spores. The genus Glomus was able to increase the weight of fresh and dry oven total stalk, the number of leaves and the height of the plants and those significantly higher than that of the genus of Gigaspora and the mixed genus. Spore doses (50, 100 and 150 spores) of the genus Glomus and the genus Gigaspora and mixture gave no significant different results on all observed variables.
