Abstract

The research purpose to know ability of natural supplements in supporting the regeneration of banana cv. Barangan Musa acuminata Colla. Explants were cultured on MS basal medium (control) with natural supplements: coconut water (CW) 5; 10; 15 and 20%, green bean sprout extract (GBSE) and sweet corn extract (SCE) at 50; 100; 150 and 200 g/l. After 4 weeks of cultivation, maximum multiplication shoots (7.11 ± 0.19), shoot length (2.00 ± 0.22), and number of leaves (4.00 ± 0.38) was obtained in MS + 200 g/l bean sprout extract. The addition of natural supplements to the medium showed better results on all parameters of observation than control. This results indicates the potential of natural supplements for supporting of Barangan Musa acuminata Colla. propagation in vitro culture.

Abstract

Basal stem rot caused by Ganoderma boninense is one crucial disease in oil palm. The used of chitinolytic bacteria is one part of biological control to suppress G. boninense. This study aimed to isolate and characterize chitinolytic bacteria associated with roots of oil palm which have the potential as agents biocontrol against the pathogens of G. boninense, the caused of basal stem rot disease of oil palm. The study was conducted in North Luwu, East Luwu, and Makassar Districts from January to September 2017. The research was done in  four stages that consist of isolation of endophytic bacterial, testing chitinolytic activity, testing antagonistic and biochemical identification. The results showed that from 14 bacterial isolates obtained, two of them had chitinolytic activity with index of 2,35 and 3,37 respectively. Both of these bacteria can inhibit G. boninense on solid medium by 24,9% and 69,4% respectively and on the liquid medium by 47,5% and 68,5% respectively five days after inoculation. Based on biochemical characterization, these bacteria have similarities with Bacillus sp, and Serratia sp. To be concluded, Serratiasp. is a potential medium to be for controling basal stem rot disease on oil palm.ABSTRAKBusuk pangkal batang yang disebabkan oleh cendawan Ganoderma boninense merupakan salah satu penyakit penting pada kelapa sawit. Pemanfaatan bakteri kitinolitik merupakan salah satu bagian pengendalian hayati untuk mengontrol G. boninense. Penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi bakteri kitinolitik yang berasosiasi dengan akar tanaman kelapa sawit yang berpotensi sebagai agens biokontrol terhadap G. boninense, penyebab penyakit busuk pangkal batang kelapa sawit. Penelitian dilaksanakan di Kabupaten Luwu Utara, Luwu Timur, dan Makassar sejak bulan Januari sampai September 2017. Penelitian ini terdiri atas empat tahap, yaitu isolasi bakteri endofit, uji aktivitas kitinolitik, uji antagonis dan identifikasi secara biokimia. Hasil penelitian diperoleh 14 isolat bakteri dimana dua bakteri diantaranya memiliki aktivitas kitinolitik dengan indeks masing masing 2,35 dan 3,37. Kedua bakteri tersebut dapat menghambat G. boninense pada medium padat masing masing 24,9% dan 69,4% serta pada medium cair masing masing 47,5% dan 68,5% lima hari setelah inokulasi. Berdasarkan karakterisasi secara biokimia, bakteri tersebut memiliki kemiripan dengan Bacillus sp, dan Serratia sp. Hasil penelitian menunjukkan bahwa Serratia sp. berpotensi dalam mengendalikan penyakit busuk pangkal batang pada kelapa sawit.

Abstract

Rhizosphere is an ideal zone for microorganisms to grow and develop abundantly that influenced by plant roots exudates. Soil microorganisms have a symbiotic mutualism interaction with plants and their interaction influenced the availability of organic soil compound. Therefore, there is a need to investigate and analyze the diversity of fungi at the rhizosphere of economically important food crop like shutterstock taro potato (Satoimo). The study was conducted through several steps as follows: collection of soil samples, dilution of soil samples and isolation of fungi, identification and analyzing of variability of the isolates. The results showed that Satoimo rhizosphere had a diversity index 2.85 which was categorized as a modest index. This result indicated the stability of the fungi community at the Satoimo rhizosphere and its diversity as modest category. The diversity of the microorganisms for each soil sample indicate its organic compounds and as an important factor that determine quality and soil healthy status. Abstract

Penyakit busuk batang atau blendok merupakan salah satu penyakit utama yang disebabkan oleh cendawan Botryodiplodia theobrome yang menginfeksi batang tanaman dapat mengganggu proses metabolisme dan secara otomatis dapat mengakibatkan penurunan produktifitas tanaman. Pengendalian secara hayati dilakukan untuk mengurangi residu yang dihasilkan akibat menggunakan pestisida kimia yang berdampak pada kondisi tanah. Oleh sebab itu perlu dilakukan pengendalian ramah lingkungan melalui eksplorasi cendawan endofit yang dapat berperan sebagai antagonis dan mengendalikan penyakit busuk batang di pertanaman jeruk pamelo. Untuk memperoleh agensia hayati berupa cendawan endofit, telah dilakukan isolasi dari jaringan daun dan batang tanaman jeruk yang sehat. Terdapat 14 isolat yang berhasil diisolasi dengan karakteristik yang berbeda-beda. Setelah proses pemurnian isolat dan karakterisasi secara morfologis dan mikroskopis. Delapan diantara isolat-isolat tersebut teridentifikasi dalam Genus Trichoderma, Penicillium, Aspergillus dan 4 diantaranya tidak teridentifikasi karena tidak menghasilkan konidia sehingga sulit untuk diketahui identitasnya. Hasil uji dual kultur memperlihatkan bahwa terdapat variasi efektivitas dari isolat-isolat yang diuji. Isolat PK6, PK 13, PK 14, PK 11 dan PK 1 memperlihatkan persentase penghambatan yang terbaik mencapai nilai diatas 70%, bahkan PK 6, PK 13 dan PK 14 mampu menghambat hingga 100 % terhadap tiga isolat B. theobromae yang diuji. Terdapat satu isolat dengan daya hambat terendah yaitu PK 9 yaitu 18.91 %, 24.81% dan 19.81%. Mekanisme antagonistik yang diamati berupa kompetisi, dan antibiosis.

Abstract

The nickel (Ni) content in a post-mining soil of Pomalaa mines reached 14,200 mg.kg-1 and became a limiting factor in the plant growth process. A Ni reduction in the soil by using phyto-accumulator such as Jack bean (Canavalia ensiformis L.) can be improved by combining it with arbuscular mycorrhizal (AM) fungi. The purpose of this study was to determine the effect of the mycorrhizal fungus Acaulospora sp. on the efficiency of Ni reduction by C. ensiformis. This experiment was carried out by using a randomized block design with three different treatments, include: 1) C. ensiformis without Acaulospora sp. inoculation (negative control), 2) C. ensiformis inoculated with indigenous Acaulospora sp. and 3) C. ensiformis inoculated with non-indigenous Acaulospora sp. The study was conducted in the nursery that belongs to PT. Vale Indonesia Tbk., Sorowako, South Sulawesi, Indonesia. The results showed that highest nickel accumulation was found in the root inoculated with indigenous Acaulospora sp. (9500 mg.kg-1), followed by stem (1400 mg.kg-1), leaf and pod (1300 mg.kg-1), seed (1200 mg.kg-1), and flower (1100 mg.kg-1). This study indicates that application of the indigenous Acaulospora sp. can improve C. ensiformis efficiency to reduce Ni content at Sorowako post-mining area.

Abstract

Percepatan pengelolaan rehabilitasi lahan pasca tambang nikel di Sorowako merupakan hal yang sangat mendapat perhatian PT. Vale Indonesia, masyarakat Sorowako, dan Pemerindah Daerah Luwu Timur. Percepatan ini dapat dilakukan dengan memanfaatkan sumber daya alam khususnya mikroorganisme endomikoriza indigenous. Endomikoriza yang juga disebut mikoriza arbuscula (MA) mendapat banyak perhatian karena kemampuannya membentuk simbiosis mutualistik dengan 80% – 96% species tumbuhan. Penelitian ini bertujuan mengetahui genus spora endomokoriza indigenous yang dominan dan mempunyai potensi untuk percepatan pengelolaan rehabilitasi lahan pasca tambang nikel. Penelitian ini dilaksanakan dalam tiga tahap yaitu pengambilan sampel rhizosfer, trapping spora, isolasi dan identifikasi jenis spora MA. Hasil penelitian menunjukkan bahwa genus endomikoriza indigenous yang dominan dan berpotensi dalam percepatan pengelolaan rehabilitasi lahan pasca tambang nikel di Sorowako adalah genus Acalauspora sp (75, 06 %).

Abstract

Morphological characterization of Phytophthora isolates commonly was done on the basis of the colony and hyphal swelling characteristics, form, and shape of chlamydospore, sporangia as well as oogonia and antheridia. Molecular characterization was conducted using specific primer pairs Pal1 s and Pal2a for Phytophthora palmivora and universal primer ITS4 and ITS5 followed by sequencing of isolates. The virulence of Phytophthora palmivora isolates was tested on a cocoa pod of the Sulawesi2 clone. A total of 21 Phytophthora isolates were collected from Palopo, Gowa, and Pinrang Districts. Based on colony morphology and microscopic features i.e. chlamydospore, sporangium, and its papilla form, all isolates indicated Phytophthora palmivora. Molecular identification using a specific primer showed that all ten representative group isolates had the same band in size of 650 bp, whereas using universal ITS4 and ITS5 primers, all isolates generated one band in the size of 900bp. The sequencing result showed, that all isolates were placed in the group P. palmivora. The virulence of P. palmivora isolates on the Sulawesi2 clone revealed a varying degree of virulence, from less virulent to highly virulent. All isolates from Pinrang were classified as highly virulent because induced necrotic in more than 50% of the tested cocoa pod.

Abstract

Burkholderia glumae and Xanthomonas oryzae pv. oryzae are the primary causes of bacterial disease in rice, capable of reducing production and posing a threat to food availability. This research aims to assess the efficacy of a consortium of five bacteria contained in Mikrobat as bioactivators. The goal is to enhance rice yields, mitigate the severity, and suppress grain rot disease and bacterial leaf blight in rice. The research employed a split-plot design encompassing 12 treatment combinations replicated three times. The experimental setup involved two factors: the main plot consisted of rice varieties-Ciherang, Inpari 4, and Trisakti-while the subplots represented types of bioactivators-Mikrobat, EM4, Trichoderma asperellum, and a control. Results demonstrated Mikrobat’s positive impact compared to the control, with an average of 17.37 productive tillers and higher yields than other bioactivator treatments. Notably, Mikrobat exhibited 50.04% spikelets per panicle and a 1000-grain weight of 19.77 g. The severity of disease with Mikrobat treatment was notably lower than with EM4 and T. asperellum, featuring 4.20% for grain rot disease and 4.41% for bacterial leaf blight. This was significantly different from the control, which showed severity rates of 9.94% for grain rot disease and 7.75% for bacterial leaf blight. Among the three tested varieties, Ciherang demonstrated the best response, displaying higher resistance to both tested diseases. The varietal treatments did not significantly differ from each other. Although Mikrobat’s effects were not significantly distinct from EM4 and T. asperellum, its usage still reduced the severity and suppression of grain rot and leaf blight diseases

Abstract

Cendawan Beauveria bassiana merupakan cendawan entomopatogen yang dapat menginfeksi hampir seluruh jenis serangga melalui interaksi endofit dengan tanaman, sehingga cukup prospektif digunakan sebagai alternatif pengganti insektisida kimia. Penelitian ini bertujuan mengetahui pengaruh aplikasi cendawan B. bassiana terhadap benih jagung sebagai bioinsektisida yang efektif dan efisien dalam pengendalian Spodoptera frugiperda, Ostrinia furnacalis dan Helicoverpa armigera di sentra pertanaman jagung . Penelitian ini dilaksanakan di Laboratorium Penyakit, Departemen Hama dan Penyakit Tumbuhan, Fakultas Pertanian, dan penelitian lapangan di laksanakan di pertanaman jagung GOR Unhas, Kecamatan Tamalanrea, Kota Makassar. Penelitian ini berlangsung pada Juli sampai Desember 2023. Penelitian ini menggunakan Uji T independen dengan ?= 0,05 untuk membandingkan mean antara kedua perlakuan yaitu benih dengan perendaman B. bassiana dan benih tanpa perendaman B. bassiana. Berdasarkan hasil penelitian menunjukkan rata-rata tinggi tanaman pada perlakuan benih dengan perendaman B. bassiana lebih tinggi dibandingkan benih tanpa perendaman B. bassiana. Rata-rata tingkat populasi larva Spodoptera frugiperda pada benih tanpa perendaman B. bassiana lebih tinggi daripada benih dengan perendaman B. bassiana. Begitupun dengan tingkat kerusakan tanaman jagung, menunjukkan rata-rata tingkat kerusakan tertinggi pada benih tanpa perendaman B. bassiana, hal ini diakibatkan populasi larva S. frugiperda lebih banyak ditemukan pada tanaman jagung dengan perlakuan benih tanpa perendaman. Kategori serangan yang dihasilkan hama S. frugiperda menunjukkan bahwa serangan sedang sampai berat.

Abstract

Efficiency and effectiveness in controlling crossbreeding are pivotal in yielding superior hybrid varieties with desired traits such as genetic purity and high productivity. Cytoplasmic Male Sterile (CMS) is a crucial component in hybrid variety assembly. This study aims to identify the genotypes that can induce male sterile lines based on genetic diversity and the level of pollen sterility in the prospective male sterile genotypes tested, consisting of seventeen genotypes. The research findings revealed that the genetic diversity among the seventeen observed male-sterile candidate genotypes tends to vary in quantitative traits. Among the quantitative traits, JHD14 exhibited indications of being a male sterility inducer compared to the other genotypes observed, demonstrating a value of 0.00 g in terms of pollen weight. This is further substantiated by the microscopic examination of JHD14 pollen grains, which displayed 100% sterility.
