Abstract

Avocado is one of the fruits favored by many people because of the avocado's taste and content. Avocados contain antioxidants and nutrients such as fat which is 9.8 g / 100 g of fruit flesh. But until now, the use of avocados is still focused on the fruit while other parts such as seeds are underutilized. Catechins are flavonoid compounds contained in avocado seeds. Extraction of catechins in avocado seeds was carried out by maceration method, namely immersion using three variations of solvents namely ethyl acetate, 95% ethanol, and acetone for 24 hours. The results of maceration were filtered and then remaserated for 24 hours to obtain more filtrate, then concentrated with an evaporator rotary vaccum to obtain thick extracts of avocado seeds which were then tested using the TLC-Densitometry method. The results obtained were the highest levels of catechins obtained in maceration using ethyl acetate solvents, with levels of 25.55% b / b. Keywords: avocado seeds, catechins, maceration

Abstract

Breast cancer can be initiated by either overexpression of HER-2 protein which can induce dimerization and autophosphorylation so that it triggers the activation of Focal Adhesion Kinase (FAK) resulting in migration and metastasis in breast cancer cells. Quercetin which has another name 3,5,7,3 ', 4'-pentahydroxyflavon with the molecular formula of (C15H10O7) is a flavonoid compound which is very widely found in nature. The purpose of this study was to determine the mechanism of inhibition of overexpression of HER-2 proteins by quercetin compounds by in silico molecular docking. In silico molecular docking was carried out in several stages namely method validation, optimization of 3D quercetin compound structure, docking between quercetin compounds optimized with HER-2 protein based on bond energy parameters the lower the bond energy the stronger and the more stable the bond is. The results of docking expressed by the binding energy of quercetin compounds with HER-2 protein are -8.24 kcal / mol, while the energy of the native ligand bond with HER-2 protein is -10.45 kcal / mol. The bonding energy shows that quercetin compounds have the potential as breast anticancer because they can modulate the overexpression of HER-2 proteins. Abstract

Increasing melanogenesis process causes excessive melanin synthesis resulting in darkening of the skin color. The melanogenesis process requires mealnogenesis enzymes, one of which is tyrosinase-related protein 1. One of the flavonoid compounds that has the potential as a skin lightening agent is quercetin. The antioxidant activity of quercetin plays a very important role in antimelanogenesis. This study aims to determine the affinity and molecular mechanism of quercetin on the target protein tyrosinase-related protein 1 using in silico molecular docking method. Molecular docking is carried out through stages including optimization of the structure of quercetin compounds, preparation of the target protein tyrosinase-related protein 1, validation of the molecular docking method, and docking of quercetin on the tyrosinase-related protein 1. Docking of quercetin with tyrosinase-related protein 1 produces binding energy values of -7.81 kcal/mol, while docking of native ligand with tyrosinase-related protein 1 produces binding energy values of -5.39 kcal/mol. Quercetin has a strong affinity for tyrosinase-related protein 1 which is indicated by the binding energy from the docking results. Quercetin has activity as a skin whitening agent with in silico test with molecular mechanisms through inhibition of the activity of tyrosinase-related protein 1 enzyme. Abstract

Rheumatoid arthritis is an autoimmune disease that occur by inflammation chronic which persist as a permanent symptom. That inflammatory process caused joint destruction. Production of pro-inflammatory sytokin such as Tumor Necrosis Factor Alpha (TNF-?) stimulate an autoimmunity. Active TNF-? plays a role in the occurrence of chronic inflammation, in which the formation of active TNF-? is regulated by TNF-? Converting Enzyme (TACE). Brazilin and brazilein are known to have anti-inflammatory activity and immunommodulator potentially as anti-rheumatoid arthritis. The purpose of this study were to determine the affinity and mechanisms of brazilin and brazilein against TACE proteins as anti-rheumatoid arthritis perfomed using molecular docking method. The study was conducted exploratively with several steps such as databases preparation of 3D structures brazilin, brazilein, TACE protein, optimization of brazilin and brazilein 3D structures, protein preparation, molecular docking method validation, and docking brazilin and brazilein in these proteins. The docking results are assessed from the binding energy and hydrogen bonds formed between brazilin and brazilein in proteins. The smaller value to the binding energy, will made the bond between brazilin and brazilein with proteins will be stronger and more stable. The results showed that brazilin and brazilein have activities as anti-rheumatoid arthritis because they are able to inhibit TACE proteins with respective bond energy values -7,24 for brazilin and – 7,59 kcal/mol for brazilein. These results show that brazilin and brazilein have the potential to inhibit inflammatory process and joint destruction in rheumatoid arthritis. Abstract

Rumput laut atau alga laut (sea weed) dan garam merupakan salah satu komoditas perikanan yang telah dimanfaatkan sejak lama. Daerah Kutuh (Pantai Pandawa) merupakan salah satu wilayah penghasil rumput laut di Bali. Produksi rumput lautnya perbulan mencapai 130 sampai 225 per ton. Jenis rumput laut yang banyak dibudidayakan adalah jenis Euchema spinosum dan Euchema cottonii. Desa Kutuh memiliki potensi serta produktivitas yang tinggi untuk pengembangan budidaya rumput laut dan garamnya, namun dalam kenyataannya masih terdapat permasalahan yaitu strategi pengembangan usaha rumput laut masih kurang terencana. Dalam 3 tahun terakhir ini mengalami penurunan drastic dikarenakan para petani banyak yang beralih profesi saat ini menjadi pedagang dan penyedia usaha wisata di sekitar Pantai Pandawa. Menyusutnya jumlah petani tersebut semakin cepat seiring dijadikannya Pantai Pandawa sebagai obyek wisata. Untuk dapat tetap membudidayakan rumput laut yang menjadi salah satu keankeragaman hayati khas yang dimiliki oleh Bali maka perlu dilakukan suatu pengembangan usaha rumput laut dan dapat dikaitkan dengan pantai pandawa dijadikan sebagai obyek wisata. Salah satunya adalah pengembangan usaha mengolah rumput laut dan garam (komoditas terbesar di Pantai Pandawa) menjadi produk yang dapat dipasarkan didaerah wisata misalkan produk spa dan produk oleh-oleh. Salah satu yang dapat dilakukan adalah memberikan pengetahuan dan ketrampilan untuk mengolah rumput laut dan garam menjadi produk bernilai jual tinggi yakni sabun. Sabun diproduksi dengan cara mengkombinasikan rumput laut, garam, esensial oil, pewarna dan NaOH. Para petani perlu ditingkatkan pengetahuan dan ketrampilannya untuk memproduksi sabun yang berkualitas dengan cara menyelenggarakan workshop. Setelah diselenggarakan wokshop, para petani dapat mempraktekkan dengan baik cara membuat sabun dengan bahan kombinasi rumput laut dan garam.

Abstract

Examination of blood type has many benefits and shorten the identification time, in the interests of transfusion , right blood donor, and the identification in forensic cases such as the identification of criminal cases, in the other hand, blood type and rhesus checks also one of the prerequisites to attend junior high school from elementary school. Academic data of year 2014/2015 shows, none of the children in elementary school at Taro village, ever checked blood type or rhesus, so it is necessary to do this activity especially for 5 and 6 grade of students at elementary school. Examination includes blood type systems A, B , O and Rh. This examination followed by 139 students in grade 5 and 6 from SDN 1 , 2 , 3 , 4 of Taro, with the distribution age of the participants between 11 to 16 years. The test results showed 18.71 % of the participants have A blood type , 41.01 % with B blood type , 5.04 % AB blood type and 35.25 % with O blood type. All participants have a positive Rhesus ( Rh + ).

Abstract

Padatnya aktivitas yang dilakukan oleh setiap individu khususnya kaum perempuan saat ini, menuntutmereka untuk mendapatkan perawatan kecantikan yang cepat dan instan. Salah satu cara merawatkecantikannya adalah dengan menggunakan masker wajah. Masker wajah yang ada saat ini dibuat denganformulasi campuran bahan alami dan sintetis yang memudahkan konsumen memakainya, tetapi jarangterdapat masker yang memanfaatkan garam sebagai bahan utamanya. Garam sejak dahulu telah diketahuimemiliki banyak khasiat bagi tubuh dan kulit, tetapi pemanfaatannya sebatas digunakan sebagai garam mandiatau dioleskan diwajah dengan campuran madu. Oleh karena itu muncul inovasi untuk menjadikan garam danmadu sebagai bahan dasar dalam masker wajah dan memberikan pelatihan kepada para kelompok tani garammengenai pengolahan garam menjadi masker wajah alami. Kegiatan ini juga diharapkan mampumeningkatkan kondisi perekonomian dari petani garam di jimbaran sehingga tidak ada menjual garam sajatapi mampu mengolah garam tersebut menjadi produk dengan nilai jual yang lebih tinggi. Pelaksanaanpelatihan dilakukan dalam tiga tahap yaitu; kegiatan praproduksi yang mencakup tahap pengadaan alat danpenyiapan bahan baku, kegiatan pelatihan dan produksi yang mencakup tahap formulasi produk masker dantahap pengemasan produk, serta kegiatan pascaproduksi yang mencakup proses pemasaran, strategipemasaran produk. Pelatihan dilakukan pada tanggal 9 Juni 2015 di Jalan Setra Desa Jimbaran. Pelatihan inidisambut dengan sangat antusias dan berhasil mengolah garam menjadi produk masker wajah. Produkmasker wajah telah dilakukan pengujian secara mikrobiologi, fisika dan kimianya. Dari hasil uji diketahuiproduk memiliki kualitas yang baik.

Abstract

Daerah Kutuh (Pantai Pandawa) Badung Selatan merupakan salah satu wilayah penghasil rumput laut di Bali mencapai 130 sampai 225 per ton. Desa Kutuh memiliki potensi serta produktivitas yang tinggi untuk pengembangan budidaya rumput laut dan garamnya, namun kenyataannya masih terdapat permasalahan yaitu strategi pengembangan usaha rumput laut masih kurang terencana. Untuk dapat tetap membudidayakan rumput laut yang menjadi salah satu keanekaragaman hayati khas yang dimiliki oleh Bali maka perlu dilakukan suatu pengembangan usaha rumput laut dan dapat dikaitkan dengan pantai pandawa dijadikan sebagai obyek wisata. Salah satunya adalah mengolah rumput laut dan garam menjadi produk yang dapat dipasarkan didaerah wisata seperti lulur rumput laut sehingga perlu diadakan kegiatan pelatihan pada ibu-ibu PKK. Pelatihan ini bertujuan untuk melatih keterampilan Ibu-Ibu PKK Desa Kutuh Badung Selatan untuk menghasilkan produk bernilai jual. Kegiatan pelatihan pembuatan lulur telah dilaksanakan pada hari Senin, tanggal 27 Agustus 2018 bertempat di Aula Kantor Desa Kutuh Badung Selatan. Acara dibuka oleh bagian Hubungan Masyarakat Desa Kutuh Badung dilanjutkan dengan pemaparan materi tentang lulur dan pembuatannya serta pemanfaatan rumput laut dan garam sebagai bahan baku lulur. Kemudian Ibu-ibu PKK dibagi menjadi 5 kelompok untuk praktek pembuatan lulur rumput laut. Hasilnya kelompok PKK telah mampu menghasilkan lulur rumput laut dengan tingkat kehalusan yang berbeda-beda sesuai dengan selera. Lulur dikemas dalam plastik. Pelatihan pembuatan lulur rumput laut dan garam sangat bermanfaat dalam melatih keterampilan Ibu-ibu PKK dan meningkatkan nilai jual rumput laut. Abstract

Cisplatin (cisp) is the first line chemotherapeutic agent for several cancer diseases which can cause significant side effects and cellular resistance. Combination-chemotherapy treatment (co-chemotherapy) was reported to be able to reduce cisp effects. Therefore, this study was carried out to investigate the cytotoxic activity of ethyl acetate fraction of C. sappan (EFC) in combination with cisp by observing apoptosis induction and cell cycle profile. Cytotoxic activity was evaluated by MTT assay. Cell cycle and apoptosis analysis were performed using flow cytometry and p53 expression was analyzed using immunocytochemistry. EFC performed cytotoxic effect on HeLa cells by showing morphological changes such as cell shrinkage, rounding and decreasing of cells viability in concentration dependent manner, giving IC50 value of 65 μg/mL. Combination of EFC and cisp in low concentration decreased cell viability into 36.86%. Further assay indicated that this combination caused redistribution of cell cycle arrest in G1 and S phases through p53 stabilization in nucleus. However, that mechanism was not followed by apoptosis. These results provide evidence to support EFC development as the enhancer of cisp effect, by improving its cytotoxicity on HeLa cells. EFC increases HeLa cells sensitivity to cisp through G1 and S cells’ arrest depending on p53 expression. Key words: co-chemotherapy, EFC, cervix cancer HeLa cells, p53, G1 and S arrest.

Abstract

Brazilein is a compound obtained in a large amount from the dried heartwood of Secang (Caesalpinia sappan L.). Brazilein has strong cytotoxic effect in several cancer cell lines. This research was designed to evaluate the cytotoxic effect of brazilein and its combination with a chemotherapy agent, doxorubicin on MCF-7/DOX breast cancer cells. In the cytotoxicity assay, MCF-7/DOX cells were cultured in the presence of brazilein solely and in combination with doxorubicin for 24 hours and cell viability was evaluated by using MTT assay. MTT assay showed a dose-dependent inhibition of cell proliferation with IC50 value of 37 µM. Brazilein increased doxorubicin’s cytotoxic activity on MCF-7/DOX cells. Both of single treatment with different concentration of brazilein 12.5 and 25 mM or doxorubicin 0.8 and 1 mM gave cell viability percentage above 80%, but combination of them led to decrease the cell viability percentage significantly. Based on this research, it can be concluded that brazilein is potential to be developed as a co-chemotherapy agent on breast cancer cell that have been resistant to doxorubicin.
