Abstract

Objective: To determine the expression of Phosphatase Regenerating Liver-3 (PRL-3) and E-Cadherin in the epithelial ovarian cancer on various stages and differentiation grades. Method: This was a cross-sectional study design conducted at Obstetrics and Gynecology Department of several teaching hospitals, Faculty of Medicine Universitas Hasanuddin from January to June 2015. The expression of PRL-3 and E-cadherin was assessed immunohistochemically in 40 patients with epithelial ovarian cancer including 15 patients in early stage and 25 patients in advanced stage. We used the Fisher’s exact test with the significance of p0.05). The significant difference was found in the expression of E-cadherin whereas the high expression was shown at early stage than advanced stage (p0.05). This study also pointed out no correlation between the expression of PRL-3 and E-cadherin in epithelial ovarian cancer (p>0.05). Conclusion: PRL-3 overexpression does not decrease E-cadherin expression in epithelial ovarian cancer. Keywords: E-cadherin, epithelial ovarian cancer, PRL-3

Abstract

ABSTRAK Latar belakang Tumor ganas tiroid merupakan salah satu dari beberapa jenis keganasan yang masih menjadi dilema bagi para klinisi dalam mendiagnosa. Fine needle aspiration biopsy (FNAB) adalah salah satu metoda diagnostik yang penting dalam menilai nodul pada kelenjar tiroid. Namun pada sebagian besar kasus, diagnosa tumor ganas tiroid masih belum dapat ditegakkan secara pasti. Galectin-3 adalah molekul protein 31-kDa yang tergolong dalam -galactoside binding family of lectins. Galectin-3 terlibat dalam berbagai proses biologik. seperti adhesi antar sel, proliferasi, transformasi maligna, dan metastasis. Tujuan penelitian ini untuk menilai peranan ekspresi galectin-3 dalam membedakan lesi jinak dan ganas pada kelenjar tiroid. Metode Dilakukan pemeriksaan immunohistokimia untuk menilai ekspresi galectin-3 pada 80 sediaan lesi kelenjar tiroid (20 nodular goiter, 20 adenoma folikuler, 20 karsinoma folikuler, dan 20 karsinoma papiler). Hasil Seluruh sediaan karsinoma folikuler dan papiler memberikan hasil positif untuk galectin-3 (100%). Sementara hanya 1/20 (5%) sediaan nodular goiter dan 3/20 (15%) sediaan adenoma folikuler yang memberikan hasil positif pada galectin-3. Didapatkan perbedaan bermakna antara lesi jinak dan ganas (p=0,0001). Sensitivitas galectin-3 untuk diagnosa lesi maligna adalah 100% dengan spesifitas 90%. Nilai prediksi positif dan negatif adalah 90,9% dan 100%. Kesimpulan Ekspresi galectin-3 memiliki sensitivitas yang tinggi pada lesi maligna kelenjar tiroid. Oleh karena itu, ekspresi immunohistokimia galectin-3 dapat digunakan sebagai petanda tambahan dalam membedakan lesi jinak dan lesi ganas kelenjar tiroid. ABSTRACT Background Thyroid cancer represents one of the few cancer types that remains a diagnostic dilemma for the clinician. Fine needle aspiration biopsy (FNAB) represents the critical initial diagnostic test used for evaluation of thyroid nodules. However, diagnosis of thyroid cancer still remains uncertain in a large number of cases. Galectin-3 is a 31-kDa protein that is a member of the endogenous –galactoside binding family of lectins. It appears to be involved with the several cell functions, including cell adhesion, proliferation, malignant transformation, and metastasis. The aim of this study is to evaluate galectin-3 expression in distinguishing between benign and malignant lesions of thyroid. Methods Expression of galectin-3 in a series of 80 thyroid lesion specimens (20 nodular goiters, 20 follicular adenomas, 20 follicular carcinomas and 20 papillary carcinomas) were investigated using immunohistochemically in order to differentiate benign lesions and malignant lesions. Results All specimens of follicular carcinoma and papillary carcinoma were positive for galectin-3 (100%). Meanwhile, only 1/20 (5%) of nodular goiter and 3/20 (15%) of follicular adenoma specimens were positive for galectin-3. Significant difference was found between the benign and malignant lesions (p= 0.0001). The sensitivity, specificity, positive and negative predictive value of galectin-3 staining were 100%, 90%, 90.9% and 100%, respectively. Significant differences were also found between the subgroups of benign and malignant lesions (p0.05) Conclusion Galectin-3 staining is highly sensitive for malignancy thyroid lesions. Galectin-3 immunostaining could be useful as an adjunct to distinguish benign from malignant thyroid lesions. Abstract

BACKGROUND: Nasopharyngeal cancer (NPC) is known to release a specific exosome. NPC-derived exosome (NPC-Exo) could carry p53. However, information regarding the type of p53 carrier on NPC-Exo remains limited. This study aims to introduce our important findings regarding the type of p53 NPC-Exo cargo.METHODS: Serum from patients with NPC were prepared for exosome isolation with Seramir Exoquick by following the manual instructions. RT-PCR was conducted to determine the expression levels of latent membrane protein 1 (LMP-1) and p53 in the exosome isolate. Partial sequencing of p53 amplicon was conducted to determine mutation type of p53.RESULTS: There were 8 patients enrolled in this study. According to RT-PCR results, the expression levels of LMP-1 and p53 varied in the NPC-Exo isolate. Based on sequencing analysis, 1 case of p53 mutation was noticeable.CONCLUSION: According to current results, the NPC-derived exosome potentially carries not only wild type but also mutant type p53. Further research is needed to explore deeper the effect of the mutant type p53 as an exosome carrier in the clinical application.KEYWORDS: Nasopharyngeal cancer, exosome, p53, mutation

Abstract

BACKGROUND: Some of colorectal cancers (CRCs) are familial, however, heterozygote relatives have approximately 80% lifetime risk of cancer. Risk assessment of CRC’s family could be calculated by direct measurement of mRNA gene expression and Bayesian theorem which is modifying initial background of pedigree risk with additional conditional information. This application has not been reported.METHODS: The cross-sectional translational sequential studies were performed: (1) adenomatous polyposis coli (APC) and MutS homolog (MSH)2 mRNA quantitative RT PCR gene expressions in tissue and whole blood CRC patients; (2) gene expression was determined in matched controls; and (3) pedigree and Bayesian analysis was calculated in the patient’s family of Proband.RESULTS: Fourty CRC and 31 control subjects were enrolled. The mean blood APC level control’s group was 13,261±670 fold-change (fc) and blood MSH2 level was 12,219±756 fc. The cut-off points for hereditary APC was 12,195 fc and MSH2 was 11,059 fc. The mean APC blood level in CRC subject was 11,578±2,638 fc and MSH2 blood level was 11,411±2,912 fc. There were significant differences APC and MSH level between tissue and blood level in CRC. Eight of 40 CRC subjects had a history of familial CRC. Four patients and 10 Probands were available for recurrence risk evaluation of pedigree analysis, RNA PCR quantitative and Bayesian calculation.CONCLUSION: There was determined a cut-off point of hereditary mRNA quantitative expression. The APC and MSH2 levels in CRC subjects were significantly lower than controls. Bayesian analysis allowed for the calculation of relative risk in CRC family members and considered in clinical practice.KEYWORDS: hereditary CRC, APC gene, MSH gene, Bayesian analysis

Abstract

ABSTRACT Background: Hydatidiform Mole (complete and partial) is one of Gestational Trophoblastic Diseases (GTD) characterized by the occurrences of trophoblastic cell proliferation and abnormal placenta flakes. It can develop into malignancy, metastasis, and cause death. Therefore, characteristic data of the patients in applying preventive program and optimal therapy are required. This research evaluates the frequency and distribution of the hydatidosa mole patients and its type in the annual period from 2012 to 2015 and several responsible risk factors, such as age and obstetric history of the patients. Methods: reviews of histopatology specimen of hydatidiform mole were conducted as many as 255 cases which met the inclusion criteria in the period of January 2012 to September 2015. The review particularly determines the histopathology diagnosis of hydatidiform mole and its type. Results: The number of hydatidiform mole patients in the period of January 2012 to September 2015 is 255 patients. Approximately 70.65% cases of those are categorized as completed hydatidiform mole. The highest frequency of mole occurs in the range of ages from 21 to 25 years old (24.19%). About 28.57% cases occur on the first pregnancy and 7 out of 255 are recurrent case of mole pregnancy. Conclusions: The occurrences of hydatidiform mole found in Pathology Anatomy Laboratory of Medical Faculty of Hasanuddin University are quite high, and the cases of completed hydatidiform mole are in the highest number. The numerous patients of hydatidiform mole are commonly found in the youngster group and low gravidity.  Keywords: gestational trophoblastic disease, hydatidiform mole

Abstract

ABSTRACT Introduction: Ovarian carcinoma is the fifth most caused of gynecological death and its incidence has been shaply increased in the last five years. Phosphatase Regenerating Liver-3 (PRL-3) is a protein that play an important role in malignant cell proliferation, differentiation, and motilily. Methods: This study analized PRL-3 protein expression in paraffin block of ovarian carcinoma tissue sections by immunohistochemical analysis using PRl-3 monoclonal antibody. Results: We found that PRL-3 was expressed in all tissues analized. However, statistical testing did not found any relation between PRL-3 expression and clinical stages, also no relation found between PRL-3 expression and histological differentiation degree of ovarian carcinoma. Conclusion: No relation found between PRL-3 expression in early stage and advance stage of ovarian carcinoma, as well as among various degree of histological differentiation. Study using more variable obviously needed to gain the knowledge about the role of PRL-3 in ovarian carcinoma pathogenesis. Keywords: Ovarian carcinoma, Phosphatase Regenerating Liver-3

Abstract

Objective: To determine the role of TLR-7 expression on intrauterine vertical transmission in pregnancy through identification of serum hepatitis B markers in both maternal and umbilical cord blood.Method: Analysis of TLR expression was performed on 38 paraffin block samples of placental tissue acquired from mothers with HBV using TLR immunohistochemical staining.Result: 16 of 38 samples were acquired from mothers aged 26-30 years old. Most of the samples were from primiparous mothers (52.6%). This study found no significant association between TLR-7 expression and HBV DNA in the placenta and cord blood (p = 1.000). However, we found a significant association between placental TLR-7 expression and maternal HBV DNA (p = 0.034). Meanwhile, placental HBeAg and HBV DNA were not associated with placental TLR-7 expression (p = 0.082; p = 1.000).Conclusion: There was no significant association between TLR-7 expression and HBV DNA in the placenta and cord blood, but we found a significant association between TLR-7 expression and maternal HBV DNA.  Hubungan Ekspresi TLR-7 Plasenta dengan HBV DNA Tali Pusat dan HBV DNA Plasenta AbstrakTujuan: untuk melihat peran ekspresi TLR-7 terhadap transmisi vertikal intrauterina pada kehamilan melalui identifikasi marker serum hepatitis B pada darah ibu dan talipusar.Metode: Analisis ekspresi TLR dilakukan pada 38 sampel blok paraffin jaringan plasenta ibu yang menderita HBV dengan memakai pewarnaan imuhohistokimia TLR.Hasil: 16 dari 38 sampel berusia 26-30 tahun. Sebagian besar sampel merupakan kelompok primipara (52.6%). Penelitian ini tidak menemukan hubungan yang signifikan antara ekspresi TLR-7 di plasenta dan HBV DNA darah tali pusat (p = 1.000). Tapi, kami menemukan hubungan yang signifikan antara ekspresi TLR-7 plasenta dan HBV DNA ibu (p = 0,034). Sedangkan HBeAg dan HBV DNA plasenta tidak berhubungan dengan ekspresi TLR-7 plasenta (p = 0,082; p = 1.000).Kesimpulan: Tidak ada hubungan yang signifikan antara ekspresi TLR-7 dan DNA HBV di plasenta dan tali pusat, tetapi kami menemukan hubungan yang signifikan antara ekspresi TLR-7 dan DNA HBV ibu.  Kata kunci: toll-like receptor (TLR) 7, HBV DNA, tali pusat, plasenta, Hepatitis B, infeksi intrauterina
