Abstract

This study examined the differences in sperm quality and metabolite profiles among X and Y sperm in Friesian Holstein bulls. It also tried to find specific metabolites that can help improve the accuracy of sexed semen use in humid tropical dairy farms. Semen samples from five bulls were separated by sex and tested for movement, live sperm count, shape, and membrane and acrosome health. The tests used Computer-Assisted Sperm Analysis (CASA) and regular microscope checks. Metabolite analysis was performed using LC-HRMS, followed by various statistical tests, pathway checks, and ROC curve analysis. The outcomes showed that X sperm had better movement and acrosome health, while Y sperm had stronger membranes, fewer shape problems, and moved faster. The untargeted metabolite study showed clear differences between the two groups. X sperm had more identified metabolites and greater average levels. The crucial metabolites found more in X sperm were D-(−)-fructose, L-(+)-lactic acid, and sn-glycero-3-phosphocholine. Meanwhile, Y sperm had greater levels of acetyl-L-carnitine. The pathway analysis showed that X sperm primarily utilized carbohydrate and fat processing through glycolysis and phosphocholine pathways to maintain their movement and membrane integrity. On the other hand, Y sperm mainly use a carnitine-based energy pathway to support their faster and straighter movement. The identified metabolites can serve as reliable biomarkers to aid in laboratory quality control checks and enhance the quality of semen extenders. This outcome gives a good way to make sexed semen more stable and reliable in tropical dairy farming conditions.

Abstract

Semen quality is influenced by the frequency of ejaculation, which is significant to semen characteristics. The purpose of this study was to analyze the effect of ejaculation frequency on the quality of fresh Bali cattle semen, which includes volume, motility, and concentration of spermatozoa at the AI Center Pucak Maros. This study used secondary data from production records (recording) derived from 5 Bali cattle males, with a frequency of ejaculation of 2 times. The results showed that the frequency of ejaculation was statistically significantly different (P<0.05) in semen volume in males 11442 and 11434; the percentage of motility was significantly different (P<0.05) in male 11539; and the concentration of spermatozoa was significantly different (P<0.05) in males 11442, 11434, and 11539. The correlation coefficients for the three quality parameters showed negative correlations between semen volume and motility (R=-0.531) and concentration (R=-0.414), whereas motility and concentration were positively correlated (R=0.740). In conclusion, the second ejaculation has an impact on increasing semen volume but will reduce the percentage of motility and concentration of spermatozoa. Abstract

Tujuan pada penelitian ini yaitu untuk mengetahui pola kecerdasan emosional pada anak pasca pembelajaran daring. Penelitian ini merupakan jenis penelitian kualitatif naratif dengan pendekatan studi kasus yang dilaksanakan di Desa Sambung Kecamatan Gajah Kabupaten Demak. Teknik pengumpulan data dalam penelitian ini meliputi observasi, wawancara, dokumentasi, dan pencatatan. Teknik analisis data pada penelitian ini yaitu reduksi data, penyajian data, dan penarikan kesimpulan. Hasil penelitian menunjukkan terdapat lima pola kecerdasan emosional siswa yakni, kemampuan mengenali emosi diri, kemampuan mengelola emosi diri, kemampuan memotivasi diri, kemampuan mengenali emosi orang lain, dan kemampuan membina hubungan.
