Abstract

The Crested black macaque (Macaca nigra) is an endemic species to the Sulawesi Island, limited distribution, and Critically Endangered on the IUCN red list. The population density in the Tangkoko Conservation Forest Management Unit (CFMU) has decreased significantly due to habitat destruction and hunting. Existing data are based on the study in the northern part of Tangkoko. Currently, data from the southern is urgently needed so that population data covers the entire area, including to identify the habitat quality. The aims of this study was to (1) analyze the population density level of Crested black macaque at the Tangkoko CFMU, (2) identify the habitat characterize of Crested black macaque at the Tangkoko CFMU, (3) analyze the correlation between Crested black macaque density and habitat quality at the Tangkoko CFMU. The study was conducted from January to March 2020 in the sothern of Tangkoko CFMU, North Sulawesi. The data collection on the density of Crested black macaque is done by animals directly inventorying through surveys in the field. Observations are carried out by walking at speeds of 1 - 1.5 km / h on each track and stopping at every 100 m to observe the surroundings. Data recorded is the number of individuals found using the line transect method. Vegetation data collection method used is habitat quality data collection carried out by making sample plots measuring 25 m x 25 m for tree level observations in lines with each distance between plots is 250 m. Data recorded for dominance of vegetation are species names, number of individuals, and diameter at breast height of trees. The results showed that (1) the population density level of Crested black macaque was 15 individuals/km2. The highest amount of density is found on the D4 was 72 individu/km2, the lowest on D1, D5, D6, and D7 with no macaques; (2) vegetation analyzed at 45 plots were important value index with the highest value of 86,95 is Ficus variegata on line D7 for tree level, and Theobroma cacao for pole level with highest value of 120,80 on line D4; (3) the correlation between density of Crested black macaque and habitat quality that shows a positive value based on the Pearson correlation test. Further research is needed to determine the relationship between population density, habitat disturbance, and hunting pressure.

Abstract

Payangka fish (Ophieleotris aporos (Bleeker)) is one of the most common fish species in Lake Tondano, North Sulawesi. Due to the availability of this fish throughout the year, it is one of the types of fish consumed by many people. However, the economic value tends to be below. This study diversified payangka fish into nuggets with clove spice (Syzygium aromaticum) and brown seaweed (Sargassum sp) supplementation. The research conducted aims to obtain a proximate profile and fatty acid content of payangka nugget. The research method used is an experimental laboratory method. The study consisted of preparing payangka fish, making payangka nuggets supplemented with clove and seaweed extracts, proximate analysis by gravimetric method and analysis of fatty acid content by gas chromatography method. The results showed that the supplemented payangka nugget had 67% water content, 0.80% ash content, 0.05% total fat, and 10.50% crude fiber. Omega 3 content is 4.75%, while Omega 6 content is 34.16%. Payangka fish nuggets that were supplemented had a higher nutritional content than those without supplementation. Payangka fish nuggets supplemented with clove extract and brown seaweed contain good dietary content.

Abstract

Mosquitoes often being the vector of some diseases, such as dengue hemorrhagic fever. To prevent these diseases vectored by mosquitoes, we could use mosquito repellent. But based on some research, these mosquito repellents on the market contain some dangerous chemicals, for example, Dichlorvos, Porpoxure, and synthetic pyrethroid. These kinds of chemicals can cause some side effects that are not good for health, ranging from headaches, nausea, to cancer and organ dysfunction. Therefore, it is necessary to have a mosquito repellent that is safer for health with minimal side effects. This research is an experimental type of research that aimed to determine the effectiveness of anti-mosquito in the form of an aromatherapy candle made of the beehive with the consistency of pala pulp extract and langsat seed extract. Pala pulp extract and langsat seed extract were obtained from the maceration method and then processed by Rotary Evaporator.  The concentrations used are Pala pulp extract: langsat seed extract. I: 0,5gr:0,25gr, II: 5gr:2,5gr. Both of the concentrations will be blend into 200gr of solid candle that will be melt. A test was made on some adult mosquitoes. Mosquitoes are being bred. The mosquitoes were placed on a cage that had been build by the researchers first. The measurement scale on this research is the mortality rate of the mosquitoes counted from the second we light up the anti-mosquito aromatherapy candle. There are two types of data analysis on this research, includes the anti-mosquito test and organoleptic test. The first concentration test's result shows that there is a very minimal effect on the mosquitoes. when the test was run, there were five adult mosquitoes inside the cage, an observation was done for 10 minutes counted from the time the candle was lit up. At the first 2 minutes 10 seconds, there is 1 mosquito that died, and the rest of the time for 10 minutes there was no other  mosquito died. The second concentration test's result shows a really big impact on the mortality rate of the mosquito. There were seven adult mosquitoes inside the cage when the test was run. And for 10 minutes, 6 mosquitos died. This means that the second concentration was the most effective amount of the concentration to apply on the candle. Hopefully, the result of this research could be useful for society in terms of environment-friendly anti- mosquito usage.

Abstract

The online learning policy was implemented during the COVID-19 pandemic in various countries around the world. Problems arise in learning biology that requires laboratory experiment activities. This research provides an innovation in entomology learning with a project-based learning method using insect biodiversity in the local ecosystem, namely Lake Tondano. This study aims to determine the ability of science process skills and student responses using the project-based learning method. Project-based learning is one of the methods established for the independent learning process to learn the independent curriculum. The research was conducted with an exploratory, descriptive research method. Science process skills and student project products are measured using a rubric. Student learning outcomes are measured using quiz questions with the google form application.Meanwhile, student responses were measured using a questionnaire. The study results showed that the science process skills of students who were taught using the Project-based learning method were in the good and very good categories. The resulting project output is a practical guide based on the assessment rubric which is in good and very good grades. Student responses regarding the application of Project based learning in entomology courses are in the agree and strongly agree categories. Student learning outcomes showed an average in the good and very good categories. The application of project-based learning can stimulate the ability of science processes and student learning outcomes in entomology courses. Project-based learning is very appropriate to be applied to fields of science that require a lot of experimental activities. Because it is carried out in small research groups, the project-based learning method is appropriate for learning during the COVID-19 pandemic.

Abstract

Leilem (Clerodendrum minhassae Teism. & Binn.) is a plant endemic to Sulawesi. Leilem leaves are used as a typical food vegetable for the Minahasa tribe. However, leilem leaves were used as a medicinal plant for intestinal worms, abdominal pain and chest pain. There has been no research report on the use of leilem leaves as a source of antibacterial bioactivity. This study aims to obtain the best concentration of antibacterial Leilem leaf extract. Leilem leaves are obtained from North Minahasa. Extraction by maceration method using various solvents with a level of polarity. The test concentrations of the extract were 100 mg/L, 200 mg/L, 400 mg/L and 800 mg/L. As a positive control, clindamycin 400 mg/L was used. The results showed that the percentage of solvent yield was ethanol, which was 1.23% at 1:4 (w/v) maceration. The average diameter of bacterial growth is 12.6 mm. The results of the one way ANOVA analysis showed significant differences between test concentrations (p>0.5). Tukey's test showed that the three extracts significantly inhibited the growth of gram-positive bacteria Staphylococcus aureus strain ATCC 25923. The n-hexane extract showed the best antibacterial activity, followed by ethanol extract and ethyl acetate extract. Leilem leaves are potential to be developed as a source of antibacterial bioactive.

Abstract

One of the producers of honey bees in North Sulawesi comes from the Manembo Forest, South Minahasa. Today many ordinary people do not know the quality of good honey. This study aims to study the characteristics of bee honey from the South Minahasa Manembo Forest based on SNI. The research was conducted in April-July 2022. The research method used for testing the quality of honey was using qualitative research methods. The data obtained were then analyzed using descriptive data analysis. Determination of total flavonoid levels using quercetin as a comparison. The absorbance value was measured by UV-Vis spectro-photometer. The absorbance value is then entered into the linear regression equation. The results showed that the characteristics of Manembo forest honey, South Minahasa Regency based on SNI 2018 parameters met the standards and quality with the percentage of reducing sugar 68.53%, sucrose content 3.07%, insoluble solids 0.14%, diastase enzyme DN=4 and 0.03% ash content while those that do not meet the standard are water content with a percentage of 25%. The results of the analysis of the total flavonoid content showed a value of 1.6 mgQE/g. For further research, it is recommended to make an antidiabetic or anti-bacterial analysis in forest honey.

Abstract

Honeycomb can be used as a source of antibacterial, this is due to the content of secondary metabolites in the form of flavonoids in the honeycomb which serves as a protective determinant of honey quality. This study aims to isolate, identify and test antibacterial isolates of Indigenous bacteria from Apis mellifera nests against gram-positive bacteria S. aureus and gram-negative bacteria E. coli. This research used descriptive method and the research data were obtained through laboratory experiments. The results of the isolation stage of indigenous bacteria obtained seven (7) isolates AM1, AM2, AM3, AM4, AM5, AM6, AM7 Indigenous bacteria from Apis mellifera bee hives. AM1 and AM4 isolates were similar to the Enterobacter genus, AM2 isolates to Corynebacterium genus, AM3 and AM6 isolates to Paracoccus genus, and AM5 and AM7 isolates to Azotobacter Sp. The seven isolates of Indigenous bacteria have potential as antibacterial and the diameter of the inhibition zone against E. coli bacteria is AM1 (8.88 mm), AM2 (8.65 mm), AM3 (8.03 mm), AM4 (6.41 mm) , AM5 (9.07 mm), AM6 (9.53 mm) and AM7 (9.44 mm) while against S. aureus AM1 (9.08 mm), AM2 (9.23 mm), AM3 (9.15 mm), AM4 (8.70 mm), AM5 (10.44 mm), AM6 (11.56 mm) and AM7 (9.0 mm).

Abstract

Nutmeg leaf, lemongrass, and bee nests contain bioactive compounds that have potential as mosquito repellents. The purpose of this study was to learn about the bioactivity of a combination extract of nutmeg leaf, lemongrass, and bee nests, as well as to determine the efficient mosquito repellent lotion recipe from a combination of nutmeg leaf, lemongrass, and bee nests extract. The maceration process was used for extraction. Total flavonoid content was estimated by comparing it to the flavonoid quercetin. The absorbance value was measured with a UV-Vis spectrophotometer. The concentration of the extract combination in the lotion used was 0%, 2.5%, 5% and 10%. The total flavonoid concentration was found to be 16.67 mgQE/g. The 2.5% formula lotion has a protective power of 22%, the 5% formula lotion has a power of 62.96%, and the 10% formula lotion has a power of 82.41%. From these results, it can be seen that a formula with a concentration of nutmeg leaf, lemongrass, and bee nests as much as 10% can be the most effective mosquito repellent. The outcomes of this study are expected to help the community select environmentally friendly and safe insect repellents.

Abstract

The Ranowangko River passes through Tomohon City and Minahasa Regency, ending in the Sulawesi Sea. Tombariri District, is the Bunaken National Park area. Ranowangko River Water Monitoring in 2022 by the Regional Environmental Service of North Sulawesi Province shows that Fecal Coli and Total Coli parameters in the Ranowangko River at Station 1 do not meet the requirements according to Government Regulation Number 22 of 2021 concerning Class 2 National Water Quality Standards (BMA). Research to assess water quality from biological, physical and chemical aspects as well as determine water quality status. Monitoring was carried out 4 times in 2 consecutive years at 3 observation stations. Quantitative data and qualitative data include water quality analysis through determination of 14 parameters compared according to BMA Class 2. The water quality data was analyzed using the Storet Method to determine water quality status in accordance with Minister of Environment Decree Number 115 of 2003. The results showed Fecal Coli and Total Coli (Biological Aspects), TSS (Physical Aspects), Total Phosphate (Chemical Aspects) exceeded the BMA. Score calculation results using the Storet method at Station 1: -68, Station 2: -72 and Station 3: -76. Based on the value system According to Minister of Environment Decree 115 of 2003, the score above shows that the Water Quality Status of the Ranowangko River is heavily polluted. Recommendations to the regional governments of Tomohon City and Minahasa Regency to carry out further monitoring of the water quality of the Ranowangko River.
