Abstract

Kebiasaan mengkonsumsi makanan yang mengandung lemak dan gula tinggi akan menyebabkan obesitas, yang pada akhirnya akan menimbulkan sindrom metabolik. Desain penelitian ini menggunakan rancangan penelitian post test only control group, dengan menggunakan sampel rattus norvegicus galur wistar berwarna putih, berjenis kelamin jantan, dewasa berusia 3-4 bulan, dengan berat badan 150-200 gram, dan dengan kondisi fisik sehat yang digunakan sebagai hewan model kolesterol tinggi dengan diberi pakan tinggi lemak, yang kemudian diberi bakteri probiotik Lactobacillus plantarum. Variabel bebas pada penelitian ini adalah Lactobacillus plantarum, sedangkan variabel tergantung adalah kadar kolesterol total dan kolesterol LDL. Pemeriksaan kadar kolesterol total dan kolesterol LDL dengan metode enzimatik kolorimetri dengan prinsip COD-PAP dilakukan pada akhir penelitian. Tujuan penelitian ini adalah untuk mengetahui dosis optimum probiotik yang dapat menurunkan kadar kolesterol tinggi dalam darah. Analisis data menggunakan one way Anova, dimana hasil yang didapat menunjukkan tidak adanya perbedaan yang signifikan pada pemberian bakteri Lactobacillus plantarum dengan konsentrasi 5x10⁸CFU dibandingkan konsentrasi 5x10⁹CFU. Oleh karena itu, pemberian Lactobacillus plantarum dengan konsentrasi sebesar 5x10⁸CFU merupakan dosis optimum yang dapat menurunkan kadar total kolesterol dan LDL kolesterol dalam darah.Kata kunci: Diet Tinggi Lemak, Tikus Putih, Kolesterol Tinggi, Lactobacillus plantarum; Kadar Kolesterol Total.

Abstract

This study aims to analyze the administration of mangosteen peel ethanol extract at different doses can reduce levels of malondialdehyde (MDA) and leukotriene B4 (LTB4) serum in male white rats (Rattus norvegicus) Wistar strains exposed to electric cigarette smoke. Electric cigarette induces mitochondrial ROS production and triggers oxidative stress. Antioxidants are needed by the body if there are many free radicals, for example mangosteen peel. This laboratory experimental study uses a randomized post test only control group design. Thirty male white rats were divided into 5 groups randomly (KK, KR, P1, P2, P3). KK given 0.5% Na-CMC, KR given smoke and 0.5% Na-CMC, P1, P2, P3 given smoke and mangosteen peel ethanol extract 100, 200, 300 mg / kg weight. Data were tested for normality and homogeneity tests. Data distribution was found not normal. Data were tested by Kruskal-Wallis Test and Mann-Whitney. Kruskal-Wallis Test results showed significant differences in MDA and LTB4 levels in each experimental group (KS, KR, P1, P2 and P3), p <0.05. Mann-Whitney test results showed a significant difference in the MDA levels of the KR against P3 and in the LTB4 level of the KR against P2 (p <0.05). The conclusion of the study was the administration of mangosteen peel ethanol extract can reduce MDA and LTB4 levels serum in male white rats (Rattus norvegicus) Wistar strains exposed to electric cigarette smoke.

Abstract

This study determined community knowledge, attitudes, and practices after COVID-19 socialization in Rambipuji and Suci Village, Jember District, East Java, Indonesia. This study used the analytic observational design study. As many as 40 people were given socialization about COVID-19 by gathering and online. The questionnaires were completed in two parts included before-socialization and after-socialization. The questionnaire data were analyzed descriptively by calculating frequency, percentage, and inferential statistics by t-test, Spearman correlation, and chi-square test. The data analysis used IBM SPSS version 23 software. The mean and standard deviation of percentage of knowledge pre-test, knowledge post-test, high attitude, moderate attitude, low attitude, very high practice, high practice, sufficient practice, and low practice were 58.33 ± 30.97, 77.70 ± 22.52, 62.5±21.7, 31±22.2, 6.5±17.1, 65±13.8, 32.3±13.8, 1.25±2.4, 1.25±1.8, respectively. Knowledge pre-test and post-test correlated 0.819 (p=0.001) and a t-test with p=0.003. Attitudes and practices correlated with p=0.001. Socialization of COVID-19 was useful to improve the community knowledge, attitudes, and practices in Rambipuji and Suci Village, Jember District, East Java, Indonesia that could prevent the transmission and inhibit the spread of the COVID-19 pandemic in Indonesia. Furthermore, continuous encouragement of COVID-19 socialization in wide areas was recommended.

Abstract

Diabetes mellitus (DM) is one of disease that its incidence increases every year worldwide. The condition of DM can cause various complications caused by oxidative stress. Stichopus hermanii (SH) or golden sea cucumber extract contains antioxidant compounds that have been proven to reduce oxidative stress conditions. The purpose of this study was to investigate the effect of Stichopus hermanii extract on condition of diabetes mellitus by looking at changes in fasting blood glucose, plasma insulin, and malondialdehyde levels in animal models of Wistar rats. This study was a laboratory experimental study using Randomized Control Trial Design with Post-test only control group design. Thirty-five male Wistar rats divided into five groups, i.e normal control group, positive control, negative control and two treatment groups with SH extract dose 8.5 and 17 mg / kgBW for 14 days once daily after induction of Streptozotocin at the Biochemistry Laboratory of the Faculty of Medicine, Airlangga University. Fasting blood glucose level was measured by a glucometer, plasma insulin measured by ELISA and MDA level was measured by a spectrophotometer. Data were analyzed statistically by using One Way ANOVA test and Kruskal Wallis. There were significant results of SH extract can reduce fasting blood glucose (Kruskal Wallis, p=0.030) and MDA (Kruskal Wallis, p=0.042) but not in plasma insulin (ANOVA, p=0.130). The lowest MDA level occurs in the K4 group that given SH extract dose 17 mg/kg BW than another experimental group. As the conclusion, this study showed SH extract can decrease fasting blood glucose and oxidative stress in diabetic-induced rats.
