Abstract

Esomeprazol magnesium trihidrat merupakan obat golongan proton pump inhibitor (PPI) yang dapat digunakan dalam pengobatan tukak lambung dengan menghambat sekresi asam lambung. Namun esomeprazol dikategorikan ke dalam BCS kelas 2 dengan kelarutan yang buruk dalam air sehingga dapat berdampak pada kemampuan disolusi dan bioavailabilitasnya. SNEDDS umum digunakan untuk meningkatkan kelarutan obat lipoflilik dimana SNEDDS merupakan campuran zat aktif, minyak, surfaktan, dan ko-surfaktan yang akan membentuk nanoemulsi minyak dalam air secara spontan ketika kontak dengan fase cair dengan agitasi yang ringan. Tujuan dari penelitian ini yaitu untuk mengetahui formula SNEDDS esomeprazol magnesium trihidrat yang paling baik serta mengetahui karakteristiknya. SNEDDS diformulasikan menggunakan fase minyak berupa VCO, surfaktan berupa tween 80, dan ko-surfaktan berupa PEG 400. Sediaan SNEDDS dilakukan evaluasi berupa persen transmitan, dispersibilitas, robustness, stabilitas termodinamika, indeks bias, ukuran globul, PDI, zeta potensial, dan uji disolusi. Hasil menunjukkan bahwa SNEDDS esomeprazol magnesium trihidrat dengan rasio minyak:Smix 1:6 dan perbandingan Smix 2:1 mampu membentuk nanoemulsi secara spontan dan stabil berdasarkan uji stabilitas termodinamika, dihasilkan rata-rata ukuran globul 78,03 nm, nilai PDI 0,667, nilai zeta potensial -14,57 mV, dan dapat meningkatkan kecepatan disolusi yang lebih baik dibandingkan bentuk murninya.

Abstract

Cinnamon bark oil is known to contain high levels of cinnamaldehyde (>50%). Cinnamaldehyde is an aromatic compound that is conjugated with a carbonyl group and potential to have photoprotective activity. The purpose of this study was to develop an emulgel preparation containing cinnamon bark oil and to evaluate the  emulgel in terms of its physical properties, stability, and sunscreen activity. Cinnamon bark oil was characterized based on SNI 06-3734-2006. Cinnamon bark oil at concentrations of 0.5 and 1 % was formulated into an emulgel using a combination of stearic acid and triethanolamine as an emulsifier and viscolam mac 10 as a gelling agent. The preparation was evaluated for its physical characteristics and stability. The sunscreen activity test was carried out in vitro by determining the Sun Protection Factor's values (SPF), % erythema, and % pigmentation.  The results showed that cinnamon oil met the SNI standards of cinnamaldehyde concentration (53.33 %). Cinnamon oil emulgel preparation has good physical properties in terms of organoleptic, pH, viscosity, rheology, spreadability, and stability based on centrifugation and freeze-thaw tests. The addition of 0.5 and 1% of cinnamon oil significantly increased the sunscreen activity of the emulgel based on the SPF, % erythema, and % pigmentation values with the extra protection category. An emulgel containing cinnamon bark oil has been successfully developed which has good physical properties and stability and has sunscreen activity based on the value of FPS, % erythema, and % pigmentation.

Abstract

Teh putih adalah salah satu jenis teh dengan kandungan polifenol yang tinggi. Senyawa polifenol sudah terbukti memiliki banyak khasiat untuk kesehatan, seperti sebagai agen sitotoksik. Polifenol teh putih bersifat hidrofil sehingga memiliki keterbatasan dalam menembus membran lipid. Sistem fitosom diketahui dapat membantu meningkatkan permeasi membran dari fitokonstituen yang bersifat hidrofilik. Tujuan dari penelitian ini adalah untuk mengembangkan formula fitosom yang optimum mengandung ekstrak teh putih serta mengetahui karakteristik fitosom yang dihasilkan. Teh putih diekstraksi dengan metode refluks menggunakan etanol 70%. Terhadap ekstrak dilakukan pengukuran kadar polifenol total dengan metode Follin-Ciocalteu. Ekstrak selanjutnya dikembangkan menjadi sistem fitosom menggunakan metode hidrasi lapis tipis, dengan variasi perbandingan ekstrak dan fosfatidilkolin. Hasil penelitian menunjukkan formula terbaik untuk fitosom teh putih diperoleh pada perbandingan ekstrak dan fosfatidilkolin (2:3), dengan nilai efisiensi penjerapan sebesar 91,31 % ± 0,29, ukuran partikel sebesar 423,67 ± 3,88 nm, indeks polidispersitas sebesar 0,50 ± 0,07, dan potensial zeta sebesar -25,20 ± 14,50 mV. Dari hasil penelitian dapat disimpulkan bahwa ekstrak teh putih telah berhasil dikembangkan menjadi sistem fitosom dengan karakteristik yang baik. Penelitian lanjutan terkait pengaruh pengembangan fitosom teh putih pada aktivitas sitotoksis perlu dilakukan.

Abstract

Abstract. Oral candidiasis is an infection of the oral mucosa caused by the fungus Candida albicans, characterized by the formation of complex biofilm lesions on the mucosal surface. One natural substance with antifungal activity is cinnamon bark (Cinnamomum burmanii), which contains cinnamaldehyde. This study aims to determine the characteristics of cinnamon bark oil and evaluate its antifungal activity against C. albicans by determining its minimum inhibitory concentration (MIC). Cinnamon oil was characterized according to the quality standards of SNI 06-3734-2006, including organoleptic testing (light yellow color and characteristic aroma), determination of specific gravity (1.030 g/mL), solubility testing in 70% ethanol (1:3), and determination of cinnamaldehyde content (62.63%). The antifungal activity of cinnamon bark oil was tested using the agar diffusion well method with test concentration variations of 0.025, 0.05, 0.1, 0.2, 0.4, and 0.8%. The antifungal activity was indicated by the presence of an inhibition zone, marked by a clear area around the well. Cinnamon bark oil produced a minimum inhibitory concentration (MIC) of 0.05%, which falls into the moderate inhibition response category. Abstrak. Kandidiasis oral adalah infeksi mukosa mulut akibat jamur Candida albicans, ditandai dengan terbentuknya lesi biofilm kompleks pada permukaan mukosa. Salah satu bahan alam yang memiliki aktivitas antijamur adalah kulit batang kayu manis (Cinnamomum burmanii), mengandung sinamaldehid. Penelitian ini bertujuan untuk mengetahui karakteristik minyak kulit kayu manis serta mengetahui aktivitas antijamur minyak kayu manis terhadap jamur C. albicans melalui penentuan konsentrasi hambat minimum (KHM). Minyak kayu manis di karakterisasi sesuai dengan standar mutu SNI 06-3734-2006 meliputi uji organoleptis (kuning muda dan bau khas), penetapan bobot jenis (1,030 g/mL), penentuan kelarutan dalam etanol 70% (1:3), dan penentuan kadar sinamaldehid (62,63%). Uji aktivitas antijamur minyak kulit kayu manis dengan metode difusi agar cara sumuran dilakukan pada variasi konsentrasi uji sebesar 0,025; 0,05; 0,1; 0,2; 0,4; dan 0,8%. Parameter pengujian aktivitas antijamur ditunjukkan dengan adanya zona hambat yang ditandai oleh zona bening disekitar sumuran.  Minyak kulit kayu manis menghasilkan konsentrasi hambat minimum (KHM) sebesar 0,05%, yang termasuk kategori respon hambatan moderat.

Abstract

Abstract. Almond oil is believed to be able to lower LDL levels in the blood so that it is beneficial for hyperlipidemia conditions, but its absorption is low due to its solubility. Therefore, an effective delivery system such as SNEDDS is needed to maximize the absorption of almond oil. This study aims to determine the effect of SNEDDS components (oil, surfactant and cosurfactant) on its characteristics using d-optimal mixture design where the independent variables are oil (X1), surfactant (X2), and cosurfactant (X3), and the response variables are emulsification time (Y1) and percent transmittance (Y2). The results show that each component of SNEDDS has an effect on the characteristics of SNEDDS, especially on emulsification time and percent transmittance, where the results show that the greater the concentration of surfactant and the smaller the concentration of oil and cosurfactant, the faster the emulsification time and the higher the percent transmittance. Abstrak. Minyak almond dipercaya mampu menurunkan kadar LDL dalam darah sehingga bermanfaat untuk kondisi hiperlipidemia, namun penyerapannya rendah karena sifat kelarutannya. Untuk itu, diperlukannya sistem penghantaran yang efektif seperti SNEDDS untuk memaksimalkan penyerapan minyak almond. Penelitian ini bertujuan untuk mengetahui pengaruh komponen SNEDDS (minyak, surfaktan dan kosurfaktan) terhadap karakteristiknya dengan menggunakan d-optimal mixture design dimana variabel bebas berupa minyak (X1), surfaktan (X2), dan kosurfaktan (X3), serta variabel respon berupa waktu emulsifikasi (Y1) dan persen transmitan (Y2). Hasilnya menunjukkan bahwa setiap komponen SNEDDS memiliki pengaruh terhadap karakteristik SNEDDS terutama terhadap waktu emulsifikasi dan persen transmitan, dimana diperoleh hasil bahwa semakin besar konsentrasi surfaktan dan semakin kecil konsentrasi minyak serta kosurfaktan maka akan semakin cepat waktu emulsifikasi dan semakin tinggi persen transmitannya.

Abstract

Background: The incidence and mortality of cancer are rapidly growing worldwide. Modification on drug delivery systems based on nanotechnology was applied to improve the effectiveness and safety of treatment. Nanoencapsulation, a part of nanotechnology, was known can be involved in cytotoxic agents. Objective: This research was conducted to determine the type of polymers for nanoencapsulation of cytotoxic agents and analyze the effect of nanoencapsulation on the cytotoxic activity. Methods: The study was performed by systematic literature review using selected articles from reputable databases that meet the inclusion and exclusion criteria. Results: The results show that many cytotoxic agents have been developed in nanocapsules systems due to their low water solubility, chemical instability, and low bioavailability. The nanoencapsulation process was carried out using synthetic or natural polymers such as polylactic-co-glycolic acid (PLGA), PEGylated PLGA, polycaprolactone (PCL), chitosan-sodium tripolyphosphate, chitosan-sodium alginate, heparin-poly(l-lysine), and polymethyl methacrylate (PMMA). Those polymers are widely used for nanoencapsulation related to their biocompatible, biodegradable, non-toxic, and providing the desired coating properties. The nanoencapsulation on cytotoxic agents significantly increases the in vitro cytotoxicity, marked by the decrease of IC50 value in the range 1.4-15.4 folds compared to pure drugs. The increase in cytotoxicity could be caused by particle size reduction, modification of particle surface properties, and enhancement of drug stability. Conclusion: It can be concluded that nanoencapsulation can be applied for cytotoxic agents to increase their activity using the appropriate coating polymer.

Abstract

White tea and pomegranate seed oil are known to contain many bioactive compounds that make them potentially have antioxidant and photoprotective activities. This study aimed to examine the antioxidant and photoprotective activities of white tea leaf extract and develop it in an emulgel dosage form. White tea is extracted with 70% ethanol at 600C. The extracts were tested for antioxidant activity by the DPPH method and for photoprotective activity by the Mansur method. White tea extract was developed into an emulgel with pomegranate seed oil as the oil phase, stearic acid and TEA as emulsifiers, and viscolam mac 10 as a gelling agent. The results showed that the ethanol extract of white tea had a very strong antioxidant activity with an IC50 value of 10.80 ± 0.75 ppm. White tea ethanol extract also has photoprotective activity with an SPF value of 12.35 ± 0.63, %Te value of 2.13 ± 0.01 %, and %Tp value of 45.13 ± 0.08 % (ultra protection category). White tea extract has been successfully developed into an emulgel preparation with good characteristics and stability in terms of organoleptic, pH, viscosity, spreadability, and rheological behavior. White tea emulgel are stable based on centrifugation and freeze-thaw tests.

Abstract

Diabetic foot infection (DFI) is a common complication of diabetes, most frequently caused by Staphylococcus aureus. Green tea contains antibacterial compounds and may serve as a potential supportive therapy for DFI. This study aimed to develop a spray gel formulation containing green tea ethanol extract and evaluate its antibacterial activity against S. aureus. The extract was obtained using 96% ethanol through the reflux method and subsequently tested for antibacterial activity using the agar well diffusion method. The extract was incorporated into a spray gel formulation with Viscolam MAC 10 as the gelling agent. The resulting spray gels were evaluated for their physical properties and antibacterial activity. Green tea extract demonstrated antibacterial activity against S. aureus with a minimum inhibitory concentration (MIC) of 0.1%. Furthermore, the ethanol extract was successfully formulated into a spray gel using 5% Viscolam MAC 10, meeting quality requirements in terms of organoleptic properties, pH, viscosity, spray pattern, spray weight, drying time, and adhesion time. Formulations containing 0.75% and 1% GTE exhibited potent antibacterial activity against S. aureus. In conclusion, a green tea–based spray gel formulation with good physical characteristics and potent antibacterial activity has been successfully developed.

Abstract

Abstract. Skin is the outermost organ of the body that plays a role in the process of protection from UV radiation, which can cause the formation of free radicals and hyperpigmentation. Green tea leaves (Camellia sinensis (L) O. Kuntze) contain high polyphenol compounds, especially epigallocatechin gallate, which has potential as an antioxidant and tyrosinase inhibitor. This study aims to determine the antioxidant activity and tyrosinase inhibition in ethanol extract of green tea leaves. Ethanol extract of green tea leaves was obtained by reflux method using 96% ethanol solvent. Antioxidant activity test and tyrosinase inhibitor using DPPH method and dopachrome method based on enzymatic colorimetric testing. Antioxidant activity test results showed that green tea leaf extract has an IC50 value of 2.349 ppm, and has tyrosinase inhibitory activity with an IC50 value of 1000.422 ppm. Abstrak. Kulit merupakan organ tubuh terluar yang berperan dalam proses pelindungan dari radiasi sinar UV, yang dapat menyebabkan pembentukan radikal bebas dan hiperpigmentasi. Daun teh hijau (Camellia sinensis (L) O. Kuntze) mengandung senyawa polifenol yang tinggi khususnya epigallocatechin gallate yang berpotensi sebagai antioksidan dan inhibitor tirosinase. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan penghambatan tirosinase pada ekstrak etanol daun teh hijau. Ekstrak etanol daun teh hijau diperoleh dengan metode refluks menggunakan pelarut etanol 96%. Uji aktivitas antioksidan dan inhibitor tirosinase menggunakan metode DPPH dan metode dopakrom berbasis pengujian kolorimetri enzimatik. Hasil uji aktivitas antioksidan menunjukan bahwa ekstrak daun teh hijau memiliki nilai IC50 2,352 ppm, dan memiliki aktivitas penghambatan tirosinase dengan nilai IC50 1000,422 ppm.
