Abstract

Ethanol production by Saccharomyces cerevisiae is essential for the biofuel industry, but microbial contamination by Lactic Acid Bacteria (LAB) can significantly reduce yield and fermentation efficiency. This research aimed to identify Lactic Acid Bacteria (LAB) causing significant inhibition of Saccharomyces cerevisiae growth. LAB isolates were screened and identification was carried out by carbohydrate fermentation using the 50 CHL Analytical Profile Index (API) Kit, and the LAB isolates (MC2K1, MP2K1, and MP2K2) were identified as Lactobacillus rhamnosus and isolate MC2K2 as L. pentosus. The results demonstrated that bacteriocins did not significantly inhibit S. cerevisiae following heat treatment, indicating that metabolic byproducts, particularly lactic acid, were primarily responsible for yeast inhibition. Co-culture of LAB and S. cerevisiae supernatants was used to determine inhibition by metabolites produced by LAB. A higher inhibitory effect occurred in the supernatant from 48 hours of incubation with lower CFU results. The highest inhibition rate reached 42% in isolate MC2K1 and the incubation time of 6 hours with all isolates significantly inhibited the growth of S. cerevisiae. These findings challenge the prevailing assumption that bacteriocins are the primary inhibitors of yeast and highlight the importance of metabolic byproducts as dominant inhibitory factors. This insight contributes to developing targeted contamination control strategies, ultimately enhancing ethanol production efficiency.

Abstract

As part of implementing the Tri Dharma of Higher Education, the lecturer team in STAB Negeri Raden Wijaya Wonogiri, Central Java conducted community service activities in the form of "Training and Assistance for Buddhist Sunday School Management in Wonogiri Regency" (Efforts of Service Lecturers to Improve the Quality of SMB Management in Wonogiri Regency). The locations used as places to carry out community service activities are SMB Maitri Ratna in Giriwoyo District and SMB Dhamma Sasana in Slogohimo District. Based on observations in the field, it was found that Sunday school management had not been managed well and learning activities used monotonous material because Sunday school teachers had minimal training in both teaching and organizational management. This service activity is carried out by providing training and assistance to Sunday school coaches and teachers through 3 stages, namely observation, implementation, and ending with evaluation. The results of ongoing community service activities provide benefits, namely the preparation of Culla Sekha Sunday school learning plans, SMB teachers being able to use tangram media and interactive media in Sunday school activities, and the formation of the Maitri Ratna SMB management structure.

Abstract

Polyurethane is a class of polymers characterized by their substantial tensile strength and high melting points which enables them to be extremely durable. Despite its xenobiotic origin, polyure-thane has been found susceptible to biodegradation by microorgan-isms. The main object of this research was to investigate polyurethane degradation by the bacterium isolated from decayed teak wood (Tec-tona grandis Linn. f.). Polyurethane degrading bacteria were isolated using selective medium in the form of LB (Luria Bertani) with the ad-dition of 0.3% Impranil. Bacterial biodegradation activities shown by the changes of polyurethane structure analyzed by FT-IR spectrosco-py. The bacterial identification was carried out based on observations on the morphological characteristics of the colonies, cell morphology and sequences of 16S rRNA encoding genes. One of the potential iso-late that successfully isolated was K9, which demonstrates the disap-pearance of the 1735/cm peak of the characteristic function urethane in the FT-IR analysis. Analysis of the 16S rRNA encoding gene showed that the potential isolate having 98% similarity index to Bacillus safen-sis strain FO-36b. The activity shown by the isolate suggests that the bacteria could be a promising agent for polyurethane degradation.Â
