Abstract

Kappaphycus alvarezii seaweed is an important fishery commodity in Indonesia because it contains carrageenan, an important main ingredient in various industries. Research on the genetic characterization of seaweed in Indonesia is important as a source of information and genetic data needed to develop aquaculture practices such as breeding programs. Therefore, this study was conducted to determine the nucleotide composition of the rbcL gene sequence in Kappaphycus alvarezii as part of a genetic characterization study of Kappaphycus alvarezii seaweed in East Java. This study used five samples taken in Banyuwangi, Situbondo and Sumenep and compared them with the Kappaphycus alvarezii seaweed database at GenBank. The method used was the PCR method, and the primer pairs used were F-577 5'-GTATATGAAGGTCTAAAAGGTGG-3' and R-753 5'-GCTCTTTCATACATATCTTCC-3'. Based on research, primers F-577 and R-753 succeeded in amplifying the rbcL gene in Kappaphycus alvarezii samples in a partial manner with a length of 295 – 299 bp. The DNA sequence results of the sample have a similarity of 100% to the database in GeneBank. Nucleotide composition analysis shows the existence of genetic variations in the DNA sequences studied.

Moreover, there are other factors that cause the death of aquatic biota. Nowadays, the negative impact of plastic litter occurs on an ongoing basis through the food chain process which until now has not been well studied. Therefore, it is necessary to do further studies on the influence of the food chain as a vector of plastic litter distribution, particularly microplastic on aquatic biota from the first trophic level to high trophic levels.

Abstract

Highlight Research The endolithic bioeroders were identified. Cocconeis sp. was new for the science of euendolithic bioeroder of Scleractinian coral. SEM revealed Cocconeis sp. etch and bore of coral skeleton. Pennate diatom was present on coral cavities.   Abstract Laminar coral, Montipora, contributes to the coral reef ecosystem. The laminar life form is usually used by juvenile reef biota to shelter and prey. In an intertidal area, such as Pantai Kondang Merak, these corals are susceptible to erosion caused by mechanical and biological forces. Strong current or other anthropogenic activity may break coral colonies into pieces. Also, some grazers from reef fish and bioeroder potentially weaken coral structures. This study aimed to find the effect of biological agents from Bacillariophyceae, such as Cocconeis diatom, on the bioerosion process in laminar coral, e.g., Montipora. Ten montiporid corals from Pantai Kondang Merak were observed to find bio-eroding activities. Each coral colony was divided into 12 parts and photographed to record signs of bio-erosion on coral surface. While observing microborers, a 2x2 cm of the coral surface was observed using Scanning Electron Microscopy (SEM). SEM revealed that a frustule of Cocconeis sp. was found inside aragonite laminar coral from Pantai Kondang Merak. Cocconeis naturally grows on the coasts of tropical oceans as benthic organisms. They are reported as living attached to many substrates' surfaces and within the substrate as euendolith. This study revealed that Cocconeis sp. lived inside the coral skeleton as micro bioeroder by attaching their valve in coral aragonite. Further work needs more observations of another potential euendolith diatom living inside the coral and to build new information on their mechanism of bio-eroding process in more detail.

Abstract

Highlight Research S. platensis has been analyzed for its productivity. Analysis of the productivity of S. platensis on the use of fish culture wastewater media. S. platensis production can be increased by aquaculture wastewater. Aquaculture wastewater enhances the production of cell density, SGR, biomass, protein, and carotenoid of S. platensis. Abstract Increased productivity of Spirulina sp. in the form of high protein, carotenoids, and biomass content can be achieved by improving its nutrient supply. Inorganic fertilizers are nutrient sources, which are generally used in the culture of this organism on laboratory and industrial scale, but there are several drawbacks, including their high costs and limited availability. Several studies have also reported the use of zarrouk fertilizer as a standard culture medium for Spirulina platensis. Therefore, this study aims to determine the effect and the best concentration of fish culture wastewater treatment in Spirulina platensis culture using biomass, protein content, and carotenoid pigments as indicators. A two-factorial completely randomized designs (CRD) was used in this study, where the factors include the dose of organic waste and Zarrouk fertilizer. The microalgae samples, namely S. platensis were cultured using fresh water. This research consists of two factors. The first factor is the dose of organic waste, and the second factor is the dose of Zarrouk fertilizer. The wastewater treatment consisted of 0, 2, 4, and 6 ml/L, while Zarrouk dosages include 0, 0.5, and 1 ml/L. The best results were obtained from the sample treated with 6 ml/L aquaculture wastewater without the addition of Zarrouk. Furthermore, this treatment gave biomass production of 0.781 g/L, 50.441% protein, and 1.246 mg/L carotenoids. Based on the results, S. platensis culture can be carried out using fish culture wastewater without the addition of Zarrouk fertilizer.

Abstract

Pabrik daur ulang plastik mengolah sampah-sampah plastik yang sudah tidak bernilai menjadi produk baru. Namun, industri daur ulang plastik ini menjadi false solution karena dapat menjadi sumber potensial pencemaran plastik dari limbah yang dihasilkan berupa mikroplastik. Penelitian ini bertujuan mengidentifikasi mikroplastik pada sedimen di saluran irigasi dan Kali Pelayaran tempat pembuangan limbah pabrik daur ulang plastik. Jenis penelitian yang digunakan adalah deskriptif kuantitatif diambil dari data kelimpahan mikroplastik dan deskriptif kualitatif diambil dari data tipe mikroplastik. Metode pengambilan sampel dilakukan secara purposive sampling pada dua stasiun lokasi pabrik daur ulang plastik. Sampel diambil pada tiga titik di setiap stasiun. Kelimpahan mikroplastik yang mendominasi didapatkan pada titik outlet pembuangan limbah pabrik daur ulang plastik Kabupaten Sidoarjo sebesar 3,96 partikel/50 gram dan 1,9 partikel/50 gram pada pabrik daur ulang plastik Kabupaten Gresik. Tipe mikroplastik yang ditemukan dari jumlah yang terbanyak ke terkecil adalah filament, fibre, fragment, foam, granule.

K.alvarezii is cultivated in several areas in Indonesia, including on the island of Lombok. The production of K.alvarezii is related to its growth and it is related to its phytohormones. K.alvarezii cultivated in Lombok is currently not known for its composition and amount of phytohormones. The content of phytohormones at different planting ages is of course different. The purpose of this study was to determine the number of phytohormones in seaweed K. alvarezii at different planting ages. This study was conducted in Ekas Bay, Lombok, Nusa Tenggara Barat from June to December 2020. The study used experimental methods and descriptive data analysis. The results of this study explain that currently cultivated K. alvarezii contains cytokinins, auxins, and gibberellins. Cytokinins obtained on the 9th day of planting were 11.62 ppm and increased on the 18th day to 12.34 ppm. At the next planting age, it continued to decrease on the 27th day to 11.43 ppm and at harvest time, the 45th day of 10.69 ppm. Auxin detected in the seeds used was 644.93 ppm, on the 27th day of 14.27 ppm, on the 36th day of 1078.25 ppm. Giberilin was detected on the 9th day at 0.49 ppm. This study concluded that K. alvarezii at different planting ages had different phytohormones content.

Abstract

Udang vaname merupakan biota budidaya yang sangat rentan terhadap lingkungan sekitarnya. Kondisi kualitas air yang tidak ideal dapat mengakibatkan udang mengalami stress dan menghambat pertumbuhannya. Tujuan dari penelitian ini adalah untuk menentukan dosis yang tepat dari pemberian fermentasi bekatul dan EM4 sebagai probiotik berdasarkan dampaknya terhadap perbaikan kualitas air kolam budidaya udang vaname. Penelitian ini dilakukan dengan mengaplikasikan 3 perlakuan berbeda terdiri dari kontrol, perlakuan 1 dengan dosis fermentasi bekatul 25 g dan EM4 10 ml dan perlakuan 2 dengan dosis fermentasi bekatul 50 g dan EM4 20 ml. Parameter kualitas air yang diukur sebanyak tujuh yaitu suhu, oksigen terlarut, pH, nitrat, nitrit, ammonia, dan fosfat. Hasil penelitian menunjukkan bahwa kondisi kualitas air kolam udang cukup ideal dan perlakuan 1 dengan dosis fermentasi bekatul 25 g dan EM4 10 ml secara konsisten lebih unggul dalam memperbaiki kondisi kualitas air dibandingkan dengan perlakuan lainnya.   Vannamei shrimp are cultured biota that are very vulnerable to the surrounding environment. Water quality conditions that are not ideal can cause shrimp to experience stress and inhibit their growth. This study aimed to determine the appropriate dose of fermented rice bran and EM4 as probiotics based on their impact on improving the water quality of vannamei shrimp culture ponds. This study was conducted by applying 3 different treatments consisting of control, treatment 1 with fermented rice bran dose of 25 g and EM4 10 ml and treatment 2 with fermented rice bran dose of 50 g and EM4 20 ml. Seven water quality parameters were measured, temperature, dissolved oxygen, pH, nitrate, nitrite, ammonia, and phosphate. The results showed that the water quality conditions of shrimp ponds were quite ideal and treatment 1 with a fermentation dose of 25 g rice bran and 10 ml EM4 was consistently superior in improving water quality conditions compared to other treatments.
