Abstract

Fenol dan senyawa fenolik merupakan salah satu xenobiotik yang menjadi salah satu faktor stres lingkungan pada biota yang terpapar dan telah menjadi masalah lingkungan akibat dampak antropogenik lingkungan. Konsentrasi fenol yang melebihi batas ambang ke dalam ekosistem perairan dapat menjadi stresor kimia bagi organisme akuatik, termasuk kepiting bakau (Scylla serata). Teknik histologi juga merupakan salah satu metode umum yang dapat digunakan untuk mengetahui efek sublethal polutan. Sebelum menetapkan konsentrasi sublethal, terlebih dahulu dilakukan penentuan konsentrasi LC50 (96 jam). Berdasarkan data mortalitas dari hasil uji toksisitas akut LC50 (96 jam) fenol terhadap kepiting bakau (Scylla serata) yang dianalisis dengan analisis probit, diperoleh konsentrasi LC50 (96 jam) sebesar 26 mg.L-1. Dari hasil tersebut kemudian ditentukan perlakuan konsentrasi sublethal fenol (96 jam) yang dilakukan, yakni perlakuan A (1,62 mg.L-1), B (6,5 mg.L-1), C (13 mg.L-1) serta kontrol. Hasil pengamatan pada akhir perlakuan diketahui terdapat perbedaan yang signifikan terhadap histologi organ insang dan hepatopankreas pada tiap perlakuan dimana pada konsentrasi yang semakin tinggi menunjukkan kerusakan jaringan yang semakin terlihat jelas pada hepatopankreas juga terlihat perubahan yang signifikan yang terlihat pada tubulus hepatopankreatik kepiting bakau (S. serata) yaitu vakuola lebih terbentuk, epitel tampak tidak teratur, hilangnya bentuk bintang pada lumen. Abstract

Hadirnya fenol yang melebihi batas ambang ke dalam ekosistem perairan dapat menjadi stresor kimia bagi organisme akuatik, termasuk juga bagi ekosistem muara oleh karena muara (estuaria) merupakan daerah pertemuan antara air tawar dari perairan sungai dan air laut sehingga berpotensi mengandung bahan kimia antropogenik. Kepiting bakau (Scylla spp.) memiliki siklus hidup yang sebagian besar berada pada ekosistem mangrove dan umum digunakan dalam studi ekotoksisitas. Total Haemocyte Count (THC) dan histologi organ insang dapat menginformasikan perubahan histologi akibat stressor oleh karena paparan toksik, terutama fenol. Perlakuan sublethal fenol dengan konsentrasi yang berbeda  terhadap kepiting bakau (Scylla serata) pada hari ke-1, hari ke-3, hari ke-5 dan hari ke-8 tidak terdapat perbedaan yang signifikan (p> 0,05) (Lampiran) terhadap rata-rata Total Haemocyte Count (THC). Namun pada hari ke-7 menunjukkan penurunan jumlah THC pada tiap perlakuan bila dibandingkan dengan kontrol (tanpa perlakuan). Perubahan histologi yang terjadi meningkat sebanding dengan konsentrasi fenol yang diperlakukan terhadap kepiting bakau (S. serata). Perubahan histologi yang terjadi adalah kerusakan struktur dari lamela insang (l) dan bagian terluar sinus lamela atau outer lamellar sinuses (ols) yang meliputi infiltrasi hemosit, hiperplasia maupun nekrosis.Kata Kunci : fenol, hepatopankreas, kepiting bakau.

Abstract

Porphyridium cruentum microalgae have great potential to be developed as a source of active ingredients in various fields of health. It excretes secondary metabolites in the form of extracellular polysaccharides (EPS), potentially as antibacterial, antihyperglycemic, and immunostimulant components. This study aims to obtain the best culture density of P. cruentum, the active component of P. cruentum (EPS), and the chemical composition of EPS. The P. cruentum was cultivated on 15%, 20% and 25% culture stock, with an addition of silicate, Fe and vitamins in sterile seawater medium with salinity 35 ‰, pH 8, temperature 25-27°C, with continuous aeration and 2500 lux continuous 24 hours, for 14 days of culture period. The results of this study indicate differences in growth rates and abundance of microalgae to each culture stock. The 15%, 20% and 25% culture stocks each showed stationary phase at days 10th, 12th and 14th, with EPS concentrations of 10 ppt, 12 ppt, and 15 ppt, respectively. Variation of P. cruentum culture showed different EPS results, with a density of 25% capable of producing the highest EPS extract of 15,000 mg.L-1. The EPS is known to contain glucose and carboxylic acid compounds that can be utilized in the health and industrial fields. Keywords: culture, extracellular polysaccharides, Porphyridium cruentum.

Abstract

Human activities such as agriculture, swimming, livestock, public washing, and the use of latrines around the river are considered as major causes of damage to the river ecosystem, affecting the growth of Macroinvertebrate, and thus assessing the condition of the river. This study aimed to study the composition of Macroinvertebrate and the quality of water health in Sumber Maron River, in the village of Gondanglegi. This research was conducted from March to April 2018. The method used in this research was the quantitative method using primary data. Research stations were identified by considering land-use in nine regions. The sampling technique of total macroinvertebrates was the kicking technique using the manual mesh with a 500 µm applied to the submerged solid substrates. The data were analyzed using a BMWP Index and interpreted by calculating the Average Scores Per Taxon (ASPT). The result of this research represented that there were 20 taxa of macroinvertebrates, which consisted of It belongs to 8 orders (Trichoptera, Coleoptera, Diptera, Tricladida, Decapoda, Odonata, Ephemeroptera, Plecoptera) and four class (Oligochaeta, Hirudinea, Gastropoda, Crustacea). Generally, the lowest taxa groups found in the station A1 (3 taxa), which were the agriculture and toilet (recreation area). Meanwhile, the highest taxa groups were found in station B3, which was the recreation area, by a total of 14 taxa. Macroinvertebrate with the lowest abundance was Gerridae at station A3, which respectively had the number of 1 ind.5m-2. Macroinvertebrate with the highest abundance was Baetidae at the station B3, which is the recreation area, by the number of 549 ind.5m-2. The result of modified BMWP - ASPT analysis showed a value of 3.3 - 5.75, which indicated that the condition of Sumber Maron River was ranging from bad to excellent category. The bad water condition was found in the agriculture and toilet (recreation area), whereas the excellent water condition was found in the swimming pool (recreation area). Canonical Correspondence Analysis (CCA) revealed that environmental variables were significant for an explanation of the variance in the family. Keywords: BMWP-ASPT, Macroinvertebrate, River.

Abstract

Phytoplankton plays an important role in Litopenaeus vannamei cultivation. It plays a role as natural feed, water quality control, and indicator of shrimp cultivation success. The community structure of phytoplankton can be influenced by organic matter concentration in ponds. On the other hand, water quality and phytoplankton community structure also influence the productivity of shrimp cultivation. The research aimed to analyze the phytoplankton structure community, cultivation performance and the water quality in one of vananmei shrimp cultivation located in Muncar District. The research used the descriptive method. The research was conducted in two shrimp intensive system ponds in Tembokrejo Village, Muncar District, Banyuwangi, on February-March 2020. Parameters observed were composition, diversity, and density of phytoplankton, water quality parameters, and production performance in each pond. Based on the results, five phytoplankton classes was found: Bacillariophyceae (12 genera), Chlorophyceae (4 genera), Cyanophyceae (8 genera), Dinophyceae (2 genera), and Euglenophyceae (1 genus). Based on the density, Cyclotella and Chlorella were dominated in both ponds. Diversity index values on ponds 1 and ponds 2 were 1.64 and 1.71, respectively. The productivity of both ponds was 10.794 kg.ha-1 and 11.698 kg.ha-1, FCR (Feed Conversion Ratio) were 0.99 and 1.18, and ADG (Average Daily Growth) on both ponds were 0.16 g.day-1. Water quality parameters in both ponds showed an optimal range for vannamei cultivation, except phosphate, which is quite high. Overall, the cultivation performance of both ponds in our research showed good results. However, cultivation performance obtained in this research was not on its best performance yet due to Infectious Myonecrosis (IMNV) infection.Keywords: Cultivation performance, Litopenaeus vannamei, muncar, phytoplankton.
