Abstract

Blumea balsamifera extract has been shown to have biological activities such as anti-fungal, anti-tumor, radical scavenger, anti-obesity, hepatoprotective, antioxidant, anti-microbial, anti-inflammatory, antiplasmodial, and wound healing. The quality and efficacy of B. balsamifera extract are determined by its specific and nonspecific characteristics. This study explored the characteristics of B. balsamifera extract through a series of standardization processes. The dried leaves of B. balsamifera were powdered and macerated with 70% ethanol. After the solvent evaporation, a thick extract was obtained and stored in a refrigerator. The specific characteristics tested were organoleptic evaluation, total phenolic content (Folin-Ciocalteu method), total flavonoid content (aluminium chloride colorimetric method), and phytochemical screening using thin layer chromatography. The nonspecific characteristics tested were extraction yield, moisture content, total ash content, and acid-insoluble ash. The results of the study showed that the extraction process resulted in a yield of 13.4%. The extract had a thick and sticky appearance, a dark brown colour, a characteristic aroma, and a bitter taste. The moisture content, total ash content, acid-insoluble ash content, total phenolic content, and total flavonoid content of the extract were found to be 8.33±0.57 %ml/g, 11.63±0.86 %g/g, 2.26±0.12 %g/g, 43.86±0.89 mg GAE/g extract and 25.48±0.42 mg QE/g extract, respectively. Analysis using thin layer chromatography demonstrated that the extract included phenolics, flavonoids, terpenoids, and steroids.

Abstract

ABSTRAKKondisi ekosistem terumbu karang di perairan Ped, Nusa Penida berada dalam kategori sedang.  Kategori ini mengindikasikan pentingnya kegiatan restorasi. Kegiatan restorasi dengan metode transplantasi ini telah dilakukan pada bulan Agustus 2022 oleh NGO lokal yang bekerja sama dengan kelompok masyarakat dalam segi pemeliharaan berkelanjutan. Tujuan penelitian ini adalah untuk mengetahui kondisi pertumbuhan fragmen karang, rata-rata pertumbuhan dan faktor keterlibatan kelompok masyarakat yang berpengaruh terhadap pertumbuhan fragmen karang hasil kegiatan restorasi di perairan Ped, Nusa Penida. Metode penelitian ini adalah dengan menggunakan perangkat lunak bernama Coral Point Count with excel extensions (CPCe) untuk menghitung luas 80 fragmen karang pada periode 1 atau 1 bulan setelah transplantasi, periode 2 atau 3 bulan setelah transplantasi dan periode 3 atau 5 bulan setelah transplantasi, kemudian dilanjutkan dengan melakukan uji R2. Variabel independen yaitu faktor eksternal dan kondisi perairan serta variabel dependen yaitu pertumbuhan fragmen karang. Hasil penelitian ini adalah pertumbuhan fragmen karang meningkat relatif linear selama 5 bulan penelitian. Total fragmen yang hidup adalah 76 dari 80 fragmen setelah 5 bulan. Laju pertumbuhan fragmen maksimal adalah pada periode 2 yaitu 0,46 cm2/hari dan menurun pada periode 3 yaitu 0,42 cm2/hari, hasil laju pertumbuhan merupakan landasan pentingnya pemeliharaan pada 3 bulan pertama setelah dilakukannya transplantasi. Uji R2 menunjukan hasil 0,99 yang berarti faktor pertumbuhan fragmen adalah 99% waktu pengukuran dan 1% faktor lainnya. Pemeliharaan dapat dilakukan oleh kelompok masyarakat karena kemampuannya dalam memiliki pendanaan dalam pemeliharaan yang membantu kelompok dari segi operasional. Hasil penelitian pertumbuhan fragmen karang pada substrat reef star menyatakan bahwa pertumbuhan fragmen selama 5 bulan adalah relatif linear dengan tingkat hidup fragmen karang tinggi.Kata kunci: reef star, terumbu karang, restorasi, laju pertumbuhan.ABSTRACTThe condition of the coral reef ecosystem in Ped waters, Nusa Penida is in the medium category. This category indicates the importance of restoration activities. Restoration activities with the transplantation method have been carried out in August 2022 by local NGOs in collaboration with community groups regarding sustainable maintenance. The purpose of this study was to determine the condition of coral fragment growth, average growth, and community involvement factors that affect the growth of coral fragments resulting from restoration activities in the waters of Ped, Nusa Penida. The method of this study is to use a software called Coral Point Count with excel extensions (CPCe) to calculate the area of 80 coral fragments in period 1 or 1 month after transplantation, period 2 or 3 months after transplantation, and period 3 or 5 months after transplantation, then continued with R2 test. The independent variables are external factors and water conditions and the dependent variable is the growth of coral fragments. The results of this study were that the growth of coral fragments increased relatively linear over 5 months of the study. The total number of living fragments was 76 out of 80 after 5 months. The maximum fragment growth rate was in period 2 which was 0.46 cm2 / day and decreased in period 3 which was 0.42 cm2 / day, the results of the growth rate are the basis for the importance of maintenance in the first 3 months after transplantation. The R2 test showed a result of 0.99, which means that the fragment growth factor is 99% of the measurement time and 1% of other factors. Maintenance can be carried out by community groups because of their ability to have funding in maintenance that helps groups in terms of operations. The results of coral fragment growth research on reef star substrate stated that fragment growth for 5 months was relatively linear with a high survival rate of coral fragments.Keywords: reef star, coral reefs, restoration, growth rate.

Abstract

Non-alcoholic fatty liver disease (NAFLD) is a condition marked by excessive fat accumulation in the liver and poses a significant health challenge. The leaves of Blumea balsamifera and Sargassum aquifolium have been reported to have anti-atherogenic effects. This study aims to determine the effectiveness of B. balsamifera extract (BBLE) and S. aquifolium extract (SAE) in preventing and treating liver fat accumulation in Wistar rats induced by a high-cholesterol diet through the expression of the AMP-activated protein kinase (AMPK)/ Sirtuin 1 (SIRT1)/peroxisome proliferator-activated receptor γ (PPARγ) pathway, and the leptin receptor. The experimental design of this study is laboratory-based, involving, 20 Wistar rats were fed a high-cholesterol diet over a period of 21 days. The rats were divided into four groups for the evaluation of BBLE and SAE effect: negative control (P0): induced with a high-cholesterol diet + distilled water, positive control (P1): induced with a high-cholesterol diet + simvastatin, P2: induced with a high-cholesterol diet + 4 mg/kg/bw BBLE, and P3: induced with a high-cholesterol diet + 4 mg/kg/bw BBLE and 4 mg/kg/bw SAE. The treatment duration extended over three months. Immunohistochemical analyses were performed on liver tissues to measure AMPK, SIRT1, PPARγ, and leptin receptor expression. The results indicated that leptin expression was lower in the BBLE+SAE group compared to the simvastatin group, and differences were significant between the BBLE and BBLE+SAE groups. No significant differences were noted in AMPK, SIRT1, and PPARγ expression between the simvastatin and BBLE+SAE groups (p≥0.05). In conclusion, BBLE and SAE effectively reduce liver lipid accumulation and enhance fat metabolism in hypercholesterolemic rats.

Abstract

Hewan coba adalah hewan yang dipelihara dan dikembangbiakkan untuk keperluan eksperimen, penelitian, pendidikan, atau tujuan ilmiah lainnya. Sebelum diuji pada manusia, penggunaan hewan percobaan dalam penelitian obat-obatan perlu dilakukan terlebih dahulu. Penggunaan hewan percobaan dalam penelitian harus disesuaikan dengan tujuan yang ingin dicapai dan didasarkan pada prinsip 3R, yaitu pengurangan jumlah hewan (reduction), penyempurnaan metode (refinement), dan penggantian dengan alternatif lain (replacement). Tujuan dari kegiatan ini adalah untuk memberikan pengetahuan dan keterampilan dasar bagi siswa jurusan farmasi SMK Kesehatan Gana Husada dalam menangani hewan coba. Pelatihan dan pendampingan diawali dengan memberikan pretest kepada para siswa dilanjutkan dengan sosialisasi materi dan praktik pelatihan untuk meningkatkan kemampuan siswa dalam penanganan dasar hewan coba, kemudian diakhiri dengan pengisian posttest oleh para siswa. Setelah dilakukan penyampaian materi, diperoleh peningkatan hasil evaluasi kemampuan siswa terkait tentang materi cara penanganan dan teknik handling dasar untuk hewan coba. Nilai rata-rata pre-test yaitu 55, dengan nilai terendah 40 dan tertinggi 65 mengalami peningkatan dalam rata-rata post-test menjadi 90, dengan nilai terendah 80 dan tertinggi 95. Hasil dari kegiatan ini dapat dikatakan mampu meningkatkan kemampuan dan ketrampilan siswa dalam penanganan dasar hewan coba.

Abstract

Brown algae are considered to be a source of antioxidants that can contribute to improving health. The purpose of this study was to determine the best combination of brown algae and solvent types based on the parameters of antioxidant capacity, toxicity to Vero cells, and bacterial contamination that affects brown algae simplicia during 30 d of storage. Brown algae, Sargassum aquifolium and Padina australis, were collected from Sanur Beach at the lowest point in the intertidal zone. Extraction was performed using three solvents: ethanol, methanol, and distilled water. The antioxidant capacity of the brown algae extracts was measured using the DPPH method. The brown algae extract, which showed antioxidant capacity and IC50 in the strong category, was used for cytotoxicity testing. Bacterial contamination testing was performed based on the simple storage time of brown algae for 30 d. The results showed that the combination of S. aquifolium and P. australis ethanol extract (ratio 1:10) had a yield percentage of 37.2% with an antioxidant capacity of 0.935±0.003 mg/L GAEAC and an IC50 value of 89.03 μg/mL (strong category). The combination of S. aquifolium and P. australis ethanol extracts had an IC50 value of 382.30 μg/mL (the weak category). Total plate count (TPC), Enterobacteriaceae, and coliforms increased with the storage time of simplicia for 30 days. These findings indicate that the combination of ethanol extracts of S. aquifolium and P. australis is a potential source of natural antioxidants.

Abstract

Budidaya rumput laut Eucheuma cottonii di wilayah perairan Nusa lembongan sudah semakin berkembang sejak tahun 2020. Selain dikenal sebagai daerah pariwisata, Nusa Lembongan juga memiliki daya tarik lain yaitu sentra budidaya rumput laut. Rumput laut Eucheuma cottonii dikenal memiliki banyak manfaat dan kasiat salah satunya sebagai imostimulan alami. Selain itu keragenan yang dihasilkan rumput laut Eucheuma cottonii memiliki potensi besar digunakan dalam berbagai bidang baik itu untuk industri pangan dan non-pangan termasuk bidang kesehatan. Metode penelitian ini menggunakan metode deskriptif eksploratif dengan mengumpulkan informasi dari berbagai literatur penelitian terdahulu, baik dari jurnal nasional maupun internasional serta database online seperti Researchgate, Google Scholar, dan Direktori Jurnal Akses Terbuka. Hasil penelitian ini menunjukkan bahwa komponen aktif rumput laut Eucheuma cottonii mengandung alkaloid, saponin, steroid, flavonoid, dan tanin yang sangat bisa berperan dalam bidang kesehatan. Selain itu rumput laut Eucheuma cottonii juga memiliki aktivitas antioksidan dan kandungan karbohidrat yang tinggi dalam berat kering sehingga dapat dikembangkan dalam bidang pangan fungsional.

Abstract

Dalam penelitian ini dilakukan pengujian karakteristik dari bonggol pisang klutuk (Musa balbisiana Colla.) dan dilakukan juga uji ketahanan zat warnanya. Dalam penentuan karakteristik zat warna dilakukan uji rendemen, uji kadar abu, uji fitokimia kemudian dilakukan pengujian ketahanan zat warna dengan proses pencucian berulang menggunakan detergen dan dijemur pada terik matahari. Tujuan dari dilakukan penelitian ini adalah untuk mengetahui karakteristik dari sampel zat warna yang digunakan dan untuk mengetahui ketahanan warnanya. Hasil dari karakteristik dengan uji rendemen yaitu sebesar 2,19%, kadar abu 0,318%, kadar air 6,35% dan uji fitokimia mengandung metabolit sekunder flavonoid dan tanin. Hasil uji ketahanan warna dilakukan dengan pencelupan kemudian diberi perlakuan dan setelah itu dilakukan proses pencucian dengan menggunakan deterjen dan penjemuran langsung dibawah terik matahari dapat dikatakan bahwa warna yang diberikan mordan lebih tahan dibandingkan dengan warna yang tidak diberikan mordan.. Analisa kandungan boraks terlihat bahwa tidak terjadi perubahan warna sehingga disimpulkan bahwa keseluruhan sampel yang digunakan tidak mengandung boraks.

Abstract

Batu Lumbang Mangrove Ecotourism, located in Pemogan Village, Denpasar City, is one of the natural destinations located in a conservation area. Problems still faced by partners include not understanding the use of non-timber forest products (NTFPs) from mangroves as raw materials for food and traditional medicine and partners still need assistance to develop diversified tea bag products from mangrove leaves. Mentoring activities were carried out for 3 months at the Segara Guna Batu Lumbang Mangrove. The aim of this program is to provide education and training in making tea bags made from jeruju leaves (Acanthus ilicifolius) that are hygienic, in the right dosage, and increase partners' understanding of NTFPs through a pocket book. The method used in this community partnership program is a qualitative approach and observation. The target and output of this program is to increase partners' knowledge and skills in processing herbal drinks made from mangrove leaves and increase partners' understanding of the use of mangrove NTFPs. The results of the activity showed that there was an increase in partners' skills and knowledge of 88.87% after the program took place in processing and making herbal tea bags and utilizing mangrove pocket books.

Abstract

Problems related to landfills of plastic packaging waste are a source of environmental pollution. This research provides innovation in utilizing biodegradable plastic from cassava and leri peel starch with the addition of chitosan. This study aimed to analyze the results of the synthesis and characterization of chitosan membranes with a combination of cassava and leri peel starch and determine the biodegradation time of biodegradable plastic in EM-4 bioactivation. This study characterized biodegradable plastic membranes using test parameters, namely the SEM, FTIR, Swelling, and tensile strength tests. The biodegradation test was carried out by immersing biodegradable plastic membranes in an EM-4 bioactator for 7 days. Based on the results obtained from the characterization of biodegradable plastic membranes, namely the swelling test with concentrations of 5:3, 7:3, and 9:3, with yield values ​​of 62.9921%, 69.8902%, and 71.4286%. The SEM test obtained the best results at a concentration of 7:3. FTIR test on biodegradable plastic membranes containing hydroxyl (O-H) and carbonyl (C-O) groups. The tensile test obtained optimum results at the concentration of biodegradable plastic membranes, namely 9:3. As for the biodegradation test that has been carried out, the start time for degradation is 3 days.
