Abstract

In this study, in vivo hypoglycemic activity of mahogany (Swietenia macrophylla) bark extracts was evaluated against alloxan-induced diabetic rats. The hypoglycemic effect was compared to that of standard glibenclamide. Oral administration of hot water and methanol extracts at a dose of 250 mg/kg body weight for thirteen days of daily treatment to diabetic rats was found to possess significant dose dependant hypoglycemic effect in diabetic rats. It less active than that of glibenclamide at dose of 3.22 mg/kg. However, the hot water extract showed significant hypoglycemic activity compared to that standard drug.  Phytochemical analysis of hot water and methanol extracts has shown posistive  test for the presence of  alkaloids, flavonoids, tannins, saponins, dan terpenoids. Histopathological studies of pancreas revealed its significant effect of β-cell count. Therefore, the hot water extract could serve as good adjuvant to other oral hypoglycemic agents and seems to be promising for the development of phytomedicines for diabetes mellitus.

Abstract

Kolagen memiliki aktivitas antioksidan dan mampu menghambat aktivitas tirosinase pada prosesmelanogenesis, salah satu biota laut yang dapat dijadikan sumber kolagen alternatif adalah teripang.Penelitian ini bertujuan menentukan karakteristik kolagen dan aktivitas penghambatan enzim tirosinaseoleh kolagen teripang emas (Stichopus hermanii). Penghilangan protein non kolagen dilakukan dengancara perendaman daging teripang menggunakan NaOH 0,1 M selama 48 jam. Isolasi kolagen dilakukanmenggunakan asam asetat 0.5 M selama 48 jam. Analisis gugus fungsi kolagen dilakukan menggunakanFTIR dan analisis aktivitas tirosinase dilakukan menggunakan spektrofotometer. Kadar protein nonkolagen yang diperoleh yaitu 0,090 mg/mL dan kolagen teripang emas memiliki rendemen 0,66%. Kolagenteripang memiliki nilai pH 6,91, gugus fungsi khas amida A (3379,29 cm-1), amida B (2924,09 cm-1), amidaI (1681,93 cm-1), amida II (1568,13 cm-1), dan amida III (1269,16 cm-1). Aktivitas penghambatan tirosinasemenunjukkan bahwa kolagen teripang emas memiliki nilai IC50 5610 ppm. Kolagen teripang emas belummampu menghambat aktivitas tirosinase.

Abstract

Waste of parang parang fish (Chirocentrus dorab) skin can be used as a source of collagen. Collagen isolation can be done chemically by the Acid Soluble Collagen (ASC) method. The objective of this research was to isolate collagen with ASC method and characterize their physicochemical. Collagen isolation consisted of pretreatment and hydrolysis with acids. The pretreatment used NaOH 0.1 M for 12 hours, while hydrolysis used acetic acid 0.5 M. Pretreatment results indicated that the concentration of non-collagen protein was 0.1243 mg/mL, while the yield collagen was 2.61%. The collagen had the viscosity of 6.50 cP, the denaturation temperature of 4°C, the transition temperature of 77.30°C, and the melting temperature of 153.90°C. The obtained collagen also had pH of 6.25. The fourier transform infrared (FTIR) spectra analysis showed the collagen contained amide A (3425.58), B (2924.09), I (1647.21), II (1543.05), and III (1246.02) (cm-1). The collagen also contained glycine (26.69%), proline (12.24%) and alanine (9.51%).

Abstract

Spirulina platensis and Herrmann's sea cucumber (Sticophus hermanii) are marine biota that known have many health benefits. One of the benefit is as an antioxidant. The antioxidant activity of S.hermanii dan S. platensis from another country has been studied, but the antioxidant activity of both materials from Indonesia and the mixture haven’t studied yet. The purpose of this study was to test the antioxidant activity and to identify bioactive compounds of methanol extraxt of S. hermanii from Flores, S. platensis from Jepara and the mixture using DPPH method. Measurement of antioxidant activity used DPPH radical as an oxidant. Identification of bioactive compounds used a qualitative phytochemical test. The results showed that methanol extract of S.hermanii, S. platensis and the mixtures have IC50 value of 2,696.09±329.28 ppm, 324.92±4.06 ppm and 2,445.80±164.18 ppm. The methanol extract of Herrmann's sea cucumber contains alkaloids, saponins and triterpenoid, meanwhile S. platensis methanol extract contains alkaloids, flavonoids and steroids. Based on tests, the methanol extract from Herrmann's sea cucumber, S. platensis and the mixtures had very weak antioxidant activity with IC50 values more than 200 ppm.

Abstract

Hiperpigmentasi merupakan kondisi ketika melanin diproduksi secara berlebih sehingga menimbulkan bercak-bercak pada kulit. Kondisi ini selain menyebabkan masalah estetika juga meningkatkan produksi reactive oxygen species pada kulit. Salah satu cara menangani hiperpigmentasi adalah melalui inhibisi enzim kunci sintesis melanin yaitu tirosinase. Spirulina platensis adalah mikroalga yang telah banyak digunakan sebagai suplemen maupun obat karena kandungan nutrisinya yang tinggi. Ekstrak S. platensis telah terbukti secara in vitro memiliki kemampuan menginhibisi enzim tirosinase. Penelitian ini bertujuan untuk mencari senyawa aktif pada S. platensis yang berpotensi sebagai inhibitor enzim tirosinase. Senyawa-senyawa aktif yang terdapat pada S. platensis ditelusuri melalui studi pustaka kemudian diseleksi berdasarkan afinitas, toksisitas, dan aturan Lipinski. Analisis penambatan molekuler dilakukan dengan senyawa yang terpilih terhadap enzim tirosinase dengan asam kojat sebagai ligan kontrol. Hasil penambatan molekuler menunjukkan bahwa kaempferol paling berpotensi sebagai inhibitor tirosinase karena memiliki ΔG paling negatif sehingga memiliki afinitas terbesar terhadap enzim tirosinase.

Abstract

Channa striata atau yang dikenal dengan ikan gabus adalah ikan air tawar yang memiliki banyak manfaat untuk kesehatan, khususnya kandungan albuminnya untuk penyembuhan luka. Jenis ikan air tawar lain yang diduga memiliki potensi albumin selain ikan gabus adalah ikan toman (Channa micropeltes) dan ikan betutu (Oxyeleotris marmorata). Penelitian ini bertujuan menentukan profil protein albumin pada ikan gabus, toman, dan betutu. Metode yang dilakukan yaitu ekstraksi protein, pengukuran kadar protein serta identifikasi profil protein dengan SDS-PAGE. Pengukuran kadar protein dilakukan pada ekstrak ikan segar, rebus, dan setelah pemurnian dengan presipitasi amonium sulfat. Kadar protein pada ekstrak ikan gabus segar 11,62±0,17 mg/g, rebus 6,28±0,57 mg/g dan murni 105,23±0,44 mg/g. Kadar protein ekstrak ikan toman segar 10,47±0,17 mg/g, rebus 8,19±0,33 mg/g dan murni 42,76±1,65 mg/g. Kadar protein ekstrak ikan betutu segar 4,47±0,33mg/g, rebus 2,95±0,17 mg/g dan murni 54,09±0,16 mg/g. Profil protein albumin pada ikan gabus, toman dan betutu memiliki bobot molekul 50-52 kDa.

Abstract

Söfö-söfö is a traditional medicinal plant from Nias Island that can cure fever, cough, diarrhea and fungal infections on the skin. However, the scientific basis of these plants is unknown. The aim of this research was to extract the Söfö-söfö leaf by maceration method using two solvents that were 70 % ethanol and ethyl acetate, to test the antimicrobial activity of the extract by Agar diffusion method against Staphylococcus aureus, Eschercia coli, and Candida albican, and analyze secondary metabolite compounds by fitochemistry test and to determine the components of bioactive compounds by GC-MS. The results showed that the best solvent for making Söfö-söfö extract as antimicrobial is ethyl acetate with a minimum inhibitory concentration of 4000 ppm to S. aureus (1.25 ± 0.35 mm), E. coli (1 mm) and C. albican 6000 ppm (1.5 mm). The secondary metabolite compounds of ethyl acetate extract were alkaloids, flavonoids and steroids. Bioactive compounds found in the Söfö-söfö ethyl acetate extract with potential antimicrobial activity were hexadecanoic acid, stigmasterol, neophytadiene, methyl ester, squalene and phytol.

Abstract

Curcuminoid, a natural compound isolated from Curcuma xanthorrhiza RoxB (Temulawak) has been reported to possess antioxidant, anti-inflammatory, anticancer, antimicrobial and anti-hyperglicemic properties. The curcuminoid is weakly soluble in water that restricts its bioavailability. This problem could be overcome by incorporating curcuminoid into solid lipid nanoparticles (SLN). Nanocurcuminoid was prepare by using homogenization-ultra sonication methods. Nanocurcuminoid obtained in this study was 523.5 nm in size and polydispersity index of 0.218 with entrapment efficiency of 24.2 %. Rats were made diabetic by induction of 50 mg / kg BW STZ, treated with the chosen formula (doses 5, 10, 20 mg/kg BW) orally for 15 days. Body weight and blood glucose levels were measured from day 0, 4th,7th,11th and 15th. The results showed that 15 days of daily treatment of 10 mg/kg BW nanocurcuminoid emulsion led to a reduction of blood glucose level by 30.93±14.90 % and body weight by 15.5±13.92 %. Curcuminoid formulated in solid lipid nanoparticles could suppress a decrease in body weight and lower blood glucose levels of rats.

Abstract

Piper crocatum is one of medicinal herbal plants with a large number of benefits. Usually herbal plants have activity as antibacterial agent. Therefore, the objectives of this research were to obtain information on antibacterial activities of the leaf extracts of Piper crocatum againts four types of bacteria, in that Staphylococcus, Bacillus substilis, Escherichia coli, and Pseudomonas aeruginosa and then to analyze the phytochemistry of the leaf extracts of Piper crocatum. The leaves of Piper crocatum were extracted by maceration and reflux using ethanol 30%. The assays of the antibacterial activities and phytochemistry on the extracts were carried out using the method of Maria Bintang. Results showed that the yield of the extraction using ethanol by maceration method was 20.8%. Meanwhile, using the reflux method, the yield was obtained about 26.25%. The phytochemistry analysis showed that the leaf extracts of Piper crocatum contained alkaloid, steroid and tanin. According to this study, it was found that the leaf extract of Piper crocatum can be used to inhibit the growth of B. subtilis and P. aeuruginosa, but can not inhibit the growth of E.coli and S. aureus.
