Abstract

The study purple sweet potato is known to contain flavonoids, a class of natural polyphenols with the capability to mitigate skin pigmentation. The research aims to assess the antioxidant and tyrosinase inhibitory activities of purple sweet potato extracts obtained through hexane, ethyl acetate, ethanol, and water solvents. The results of phytochemical identification show that the extract contains various secondary metabolites which have the potential to act as antioxidants. The DPPH method IC50 values of 6948.12ppm for n-hexane, 3015.19ppm for ethyl acetate, 128.05ppm for ethanol, and 791.77ppm for water extract. The MDA inhibitor test IC50 values of 2067.02ppm for n-hexane, 1968.13ppm for ethyl acetate, 116.14ppm for ethanol, and 921.14ppm for water extract. In the tyrosinase inhibitor assay, IC50 values were 1328.29ppm for n-hexane, 1245.13ppm for ethyl acetate, 217.35ppm for ethanol, and 391.21ppm for water extract. Tuckey test statistics, ethanol extract was not significantly different from the positive control in the DPPH test, MDA inhibitor and tyrosinase test. These findings suggest that purple sweet potato extracts, particularly the ethanol extract, hold promise as natural ingredients with antioxidant and tyrosinase inhibitory properties, making them potential candidates for safe and effective skin brightening formulations.

Abstract

Alpha-glucosidase enzyme activity needs to be inhibited to avoid increasing blood sugar levels in diabetics. Red betel leaf extract is reported to have active compounds such as phenolics and flavonoids that have alpha-glucosidase enzyme inhibition activity. Previous research showed that 70% betel leaf extract has an ɑ-glucosidase enzyme inhibition activity of 69.84%, but it is not yet known how the effect of extracts with different levels of polarity on the inhibition activity of alpha-glucosidase enzyme. This study aimed to determine extracts that have optimal levels of phenolic and flavonoid content and the highest suppression of α-glucosidase activity. The multistage maceration method was used to extract red betel leaf simplicia using solvents with different levels of polarity (n-hexane, ethyl acetate, methanol). Tests were conducted on overall phenolic concentration, flavonoids, and ɑ-glucosidase inhibition ability. Data were analyzed using one-way ANOVA test with Tukey's further test (p<0.05). The results of the assessment of phenolic concentration, flavonoids, and α-glucosidase inhibitory activity as a whole showed statistically significant differences (p<0.05) in all extracts with the highest total phenolic and flavonoid content found in the n-hexane extract of 25.68 ± 0.29 mg GAE/g and 65.84 ± 0.96 mg QE/g respectively. Meanwhile, methanol extract of red betel leaf became the best extract in this study with the greatest inhibition among all extracts with a percent α-glucosidase inhibition value of 43.87 ± 1.83%.

Abstract

This study aims to determine the stability of total phenolic content, antioxidant activity, α-glucosidase inhibitory activity, physical properties (total dissolved solid, pH, color), and total microorganism of antidiabetic functional beverage at certain storage period. The functional beverage made from red betel leaf, cinnamon, and red ginger by using infuse method. Parameters were analyzed for 28 days on 10°C and 30°C of storage temperature. Antioxidant activity was measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, α-glucosidase inhibitory activity was determined in vitro, and total microorganism was measured by total plate count (TPC) method. The results showed that there were increasement in total dissolved solid of 1.92% (10°C) and 3.85% (30°C), and pH values of 15.30% (10°C) and 13.57% (30°C). In contrary, there were decreasement in total phenolic content of 61.63% (10°C) and 79.73% (30°C), antioxidant activity of 46.80% (10°C) and 53.19% (30°C), and α-glucosidase inhibitory activity of 5.43% (10°C) and 0.39% (30°C). Total microorganism at 10oC was <1.0×100 CFU/mL until day 28, while at 30oC was 2.0×100 CFU/mL on day 28. Based on the results, the functional beverage was good in physical and microbiological quality and still have biological activity as an antidiabetic beverage during storage.

Abstract

Toona sureni (Blume) Merr., known as Indonesian mahogany, is valued for its wood, while its leaves are used as vegetables and animal feed. However, its medicinal potential remains underexplored. This study evaluated the antibacterial and tyrosinase inhibitory activities of T. sureni leaf extracts from 16-, 27-, and 34-year-old trees. Extraction was performed by maceration using methanol, ethyl acetate, and n-hexane for antibacterial testing; and 30, 70, and 96% ethanol for tyrosinase inhibition. Total alkaloid content was determined using the bromocresol green method, while antibacterial activity was assessed by disc diffusion and MIC, and tyrosinase inhibition by spectrophotometry using kojic acid as a control. It revealed a 3.83% alkaloid content in the 70% ethanolic leaves extract of the 34-year-old tree. The 96% ethanolic extract from 16-year-old leaves exhibited the highest tyrosinase inhibitory activity at 86.66% with an IC50 value of 53.71±1.04 μg/mL. Antibacterial tests revealed that the methanol extract of 16-year-old trees inhibited Staphylococcus aureus (17.73 mm) and Propionibacterium acnes (15.80 mm) at 10,000 μg/mL, with MIC of 1000 and 500 μg/mL, respectively. These results suggest that the extract of 16-year-old tree has both antibacterial and tyrosinase inhibitory effects, with greater potential for cosmetic use, particularly in anti-acne and skin-brightening products.

Abstract

Stres oksidatif menghasilkan malondialdehida (MDA) sebagai produk peroksidasi lipid yang sering digunakan sebagai biomarker kerusakan oksidatif. Penelitian ini bertujuan untuk membandingkan pengukuran kadar MDA pada ekstrak teh hijau (Camellia sinensis) menggunakan spektrofotometer konvensional (kuvet) dan nano spektrofotometer (mikroplat 96 sumur). Ekstrak diperoleh melalui maserasi etanol 70% dengan rendemen rata-rata 36,14%. Penentuan MDA dilakukan dengan metode asam tiobarbiturat (TBA), dengan panjang gelombang maksimum 532 nm pada kedua instrumen. Hasil menunjukkan bahwa kadar MDA absolut berbeda signifikan antara kedua instrumen, di mana nilai absorbansi dan konsentrasi MDA pada nano spektrofotometer lebih rendah. Namun, analisis persen inhibisi dan IC50 tidak menunjukkan perbedaan signifikan (IC50 kuvet 83,997 ppm; nano spektrofotometer 83,065 ppm; p > 0,05), menandakan konsistensi dalam menilai aktivitas antioksidan relatif. Dari aspek efisiensi, nano spektrofotometer memerlukan volume sampel dan reagen lebih kecil (100 µL dan 200 µL), mengurangi limbah kimia, serta menurunkan biaya analisis hingga 71% dibandingkan kuvet. Selain itu, kapasitas analisis meningkat hingga 96 sampel per batch dengan waktu lebih singkat. Kesimpulannya, nano spektrofotometer memberikan hasil komparatif yang valid dan lebih efisien, sehingga layak direkomendasikan untuk analisis MDA di laboratorium dengan keterbatasan sumber daya.
