Abstract

ABSTRACTThis study aimed to obtain the pathogenicity of isolated bacteria from maribund barramundi Lates calcarifer with black body syndrome (BBS). Moribund barramundi was collected from the Center for Mariculture Development (BBPBL), Lampung Province. Five of pathogenic bacteria were found, i.e. Pseudomonas stutzeri L01, Vibrioharveyi, Bacillus cereus, Salinococcus roseus, and Pseudomonas stutzeri L02. The bacteria were tested for LD50 to obtain two types of high virulent bacteria to be used for the pathogenicity test. P. stutzeri L01 and V. harveyi were the most virulent bacteria with a bacterial density of 107 CFU/mL. Both bacteria were used for pathogenicity test with three treatments: injection of P. stutzeri L01, V. harveyi, mix P. stutzeri L01 and V. harveyi, and phosphatebuffered saline as control. Clinical symptoms showed a blackened body, bleeding under the belly, and inactive. Mortality of fish injected with P. stutzeri L01, V. harveyi and mix P. stutzeri L01 and V. harveyi was 53.33%, 55.00%, and 58.33%, respectively. Erythrocyte and hemoglobin in all treatments were not significantly different (P>0.05). However, there was significantly different in the total leukocytes of mixed P. stutzeri and V. harveyitreatment (P<0.05). In conclusion, P. stutzeri L01, V. harveyi and mix of them resulted in black body syndrome (BBS) disease. Coinfection of P. stutzeri L01 and V. harveyi produce higher mortality than single infection.Keywords: barramundi, black body syndrome, histopathology, pathogenicityABSTRAKPenelitian ini bertujuan mendapatkan patogenisitas bakteri dari ikan kakap putih L. calcarifer yang memiliki gejala black body syndrome (BBS). Ikan diambil dari Balai Besar Pengembangan Budidaya Laut (BBPBL), Lampung. Lima jenis bakteri patogen ditemukan, yaitu Pseudomonas stutzeri L01, Vibrio harveyi, Bacillus cereus, Salinococcus roseus, dan Pseudomonas stutzeri L02. Bakteri diuji LD50 untuk mendapatkan dua jenis bakteri virulen tinggi yang akan digunakan untuk uji patogenisitas. P. stutzeri L01 dan V. harveyi adalah bakteri paling virulen dengan kepadatan bakteri 107 CFU/mL. Uji patogenisitas dengan tiga perlakuan: injeksi P. stutzeri L01, V. harveyi, campuran P. stutzeri L01 dan V. harveyi, serta phosphate-buffered saline sebagai kontrol. Gejala klinis menunjukkan tubuh menghitam, perdarahan di bawah perut, dan pergerakan tidak aktif. Mortalitas ikan yang disuntik dengan P. stutzeri L01, V. harveyi dan campuran P. stutzeri L01 dan V. harveyi masing-masing sebesar 53,33%, 55,00%, dan 58,33%. Eritrosit dan hemoglobin pada semua perlakuan tidak berbeda nyata (P>0,05). Namun, ada perbedaan yang signifikan dalam total leukosit perlakuan campuran P. stutzeri L01 dan V. harveyi(P<0,05). P. stutzeri L01, V. harveyi dan campuran P. stutzeri L01, V. harveyi dapat menyebabkan black bodysyndrome (BBS). Koinfeksi P. stutzeri L01 dan V. harveyi menghasilkan mortalitas lebih tinggi daripada infeksi tunggal.Kata kunci: barramundi, black body syndrome, histopatologi, patogenisitas

Abstract

ABSTRACT Cultivation of Panulirus homarus lobster is now carried out with sub-mersible net cage system at a certain depth in order to obtain optimal temperature, light and water pressure. The purpose of this study was to evaluate the health status of the sand lobster P. homarus which was kept in sub-mersible net cage system measuring 250 cm × 272 cm × 135 cm with a depth of 6 m and 8 m in the waters of Semak Daun Island, Seribu Islands, DKI Jakarta. The average size of lobster seeds used was 93.23 ± 0.99 g/head with a density of 4 lobsters/m2. Lobsters were fed trash fish, molluscs and crustaceans, with a frequency of twice a day at 07.00 WIB 30% and 17.00 WIB 70% of the lobster biomass weight. This study used a completely randomized design with the two depth treatments mentioned above and three replications. Observations of total haemocyte count, differential haemocyte count, phenoloxidase activity, respiratory burst phagocytic activity and histology of lobster hepatopancreas were performed twice every 14 days. Based on the above observations, the depth does not affect the immune response, there is no visible damage to the cells and tissues of the lobster hepatopancreas. Abstract

This study aimed to evaluate the dietary supplementation of α-lipoic acid (ALA) on the growth performance, antioxidative capacity, and protein-sparring effect of striped catfish (Pangasianodon hypophthalmus) fingerling. The striped catfish (individual weight of 1.4 ± 0.0 g) were stocked in 25 cages at 2x1x1.5 m size in a 200-m pond at 50 fish. Fish were fed formulated diets with ALA supplementation, namely: diet A (27% protein: 390 kcal), diet B (27% protein: 390 kcal +ALA 8 g/kg), diet C (27% protein: 400 kcal), diet D (27% protein: 400 kcal +ALA 8 g/kg), and diet E (30% protein: 390 kcal). Feeding was performed until apparent satiation for 60 days. The ALA supplementation significantly produced higher final individual weight, specific growth rate, and protein efficiency ratio, followed by a lower feed conversion ratio on the 27:390+ALA diet treatment (P<0.05) than other diet treatments. The highest liver performance based on superoxide dismutase enzyme activity and glycogen contents was obtained from the 27:390+ALA diet treatment. The malondialdehyde, triglycerides, and lipid accumulation in the liver on the 27:390+ALA diet treatment were lower than other diet treatments (P<0.05). Therefore, the best growth performance and physiological status in striped catfish was obtained from 27:390+ALA diet treatment.

Abstract

This study aimed to identify the fish health status, based on the total erythrocytes (cells/mm3), total leukocytes (cells/mm3), hemoglobin level (g %), hematocrit, and histopathological profiles. This study was performed on February, 2021 – January, 2022. The study was located in floating-net cage of Kelapa Dua Island, Seribu Islands, Jakarta, Indonesia. Sampling was performed by a purposive-sampling method, environmental parameter measurements, containing temperature and salinity, clinical symptom observation in fish, total erythrocytes, leukocytes, hemoglobin, hematocrit, histopathological profiles of hybrid grouper liver and kidney, and existence of VNN virus. The observation results present the behavior changes in sick fish, namely whirling on the net cage surface, dead-sleeping or death-like condition below the net cage. Hybrid grouper has total erythrocytes of 1.36 x 106 – 5.13 x 106 cells/mm3, total leukocytes of 2.9 x 104 – 7.9 x 104 cells/mm3, hemoglobin level of 5.56 - 10.94 g %, and hematocrit of 13.31 - 24.78 %. From the histopathological observation, hepatocyte, necrosis, vacuolysis was occurred in liver and tubular, leukocyte infiltasi, nicrosis was occurred in kidney and with the highest prevalence of VNN at 20 – 80 %. Therefore, hybrid grouper is sick due to increased number of leukocytes at 5.13 x 106 cells/mm3 above the normal range, followed by decreased hemoglobin and hematocrit levels at 5.56 - 10.94 g % and 13.31 - 24.78 %, respectively, due to organ damage. Keyword: must be written in not more than 5 words and alphabeticaly listed ABSTRAK Penelitian ini bertujuan untuk mengetahui status kesehatan ikan meliputi total eritrosit (sel/mm3), total leukosit (sel/mm3), kadar hemoglobin (gram %) dan hematokrit (%) dan gambaran histopatolgi. Penelitian ini dilaksanakan pada periode bulan Februari 2021 hingga Januari 2022. Lokasi penelitian pada keramba jaring apung (KJA) di Pulau Kelapa Dua Kepulauan Seribu. Metode pengambilan sampel ikan dilakukan secara purposive sampling, pengukuran parameter lingkungan perairan yaitu suhu dan salinitas, pengamatan gajala klinis ikan, perhitungan jumlah eritrosit, leukosit, kadar hemoglobin dan hematokrit serta gambaran histopatologi pada organ hati dan ginjal ikan kerapu cantang dan keberadaan virus VNN. Hasil pengamatan menunjukkan perubahan tingkah laku ikan sakit yaitu berputar ke atas permukaan KJA, terjadi sleeping dead atau ikan berada di dasar seperti mati. Ikan kerapu cantang memiliki jumlah eritrosit 1.36 x 106 – 5.13 x 106 sel/mm3, leukosit 2.9 x 104 – 7.9 x 104 sel/mm3, hemoglobin 5.56 - 10.94 gr %, dan hematokrit 13.31 - 24.78 %, hasil histopalogi ikan mengalami perubahan sel pada hati yaitu hepatosit, vakuolisasi, nekrosis dan ginjal mengalami perubahan infiltrasi leukosit, nicrosis, tubulus dan kehadiran virus VNN tertinggi berkisar 20 % - 80 %. Berdasarkan hasil penelitian tersebut maka dapat disimpulkan bahwa ikan kerapu cantang yang sakit karena jumlah leukosit meningkat 5.13 x 106 sel/mm3 melebihi kisaran normal, kadar hemoglobin menurun 5.56 - 10.94 % dan kadar hematokrit 13.31 - 24.78 % rendah dan terjadi kerusakan jaringan pada organ dalam ikan.
