Abstract

Research on the "effectiveness of the fungus Metarhizium anisopliae as a biological control agent against the larvae of the fly Musca domestica (L)” was done and focusingin determination of the effectiveness of this fungus as a biological control agent against Musca domestica larvae and to determine LC (Lethal Concentration) 50 and LC 90 on this biological control proceses. This study was designed in a completely randomized design consisting of 6 treatments and 3 replications. Arrangement of treatments i.e. the concentration of M. anisopliae in distilled water was; Po = distilled water without spore suspension, P1 = 5.1 × 1010 spores / mL, P2 = 2.0 × 1010 spores / mL, P3 = 1.0 × 1010 spores / mL, P4 = 5.1 × 109 spores / mL, and P5 = 2.0 × 109 spores / mL. Based on the observations, the mortality percentage rate was reached 93.33% as it in the treatment of spora concentration of 5.1 x1010 spores / mL.Probit analysis showed that LC50 was 8.3 x109 spores / mL with the range between the lower limit and the upper limit were 6.24 x109 spores / mL -11.43 x109 spores / mL. While the LC 90 was 5.0 x1010 spores / mL with the range of the lower limit until the upper limit were 2.8 x1010 spores / mL - 9.9 x1011 spores / mL. Key words: Metarhizium anisopliae, Musca domestica, effectiveness, mortality, biological control

Abstract

The study about “Screening of Aspergillus antagonistic to Phytopthora palmivora causing black pod disease of cocoa at Sulawesi Tengah” has been carried out during three months. The purpose of this study is how to getting the best of local isolats of Aspergillus sp. antagonistic that can be controlling of Phytopthora palmivora. The method of this study is exploring the local Aspergillus fungi as biological control of the P. palmivora causing black pod disease of cocoa in five Districts central of production kakao, such as Donggala District, Parigi Moutong, Poso, Morowali, and Buol. Parameter observed were antagonist test in vitro and pra in vivo on the cocoa. The result of this study was obtained fourteen isolats of Aspergillus sp namely are (1) Poso District: L-1, L-2, L-3 ; (2) Morowali District, L-4, L-5, L-6; (3) Donggala District, L-7, L-8, L-9; (4) Parimo District , L-10, L-11, L-12; (5) Buol District, L-13, L-14. All isolates of Aspergillus sp. were antagonistic in vitro on P. palmivora, but only six isolates, namely are L-1, L-10, L14, L-4, L-6, dan L-8 were have the best ability to controlling P. Palmivora on the cocoa pod. Key words: Aspergillus sp., antagonism, Phytophthora palmivora and Cocoa pod.

Abstract

The study about “Formulation on tablet of Aspergillus sp. bioantagonistic agent to Phytophthora palmivora Butler causing black pod disease of cocoa”, was done in the purpose or determining the best formula in Aspergillus sp. teblet formulation, and to determine it viability during 30 days of stronge. This experiment was conducted based on the Completely Randomized Design (CRD), with 8 tretmens and 3 replication. The treatments tested were P0 (100% of chaff), P1 (95 gr chaff + 5% bond supplement), P2 (90 gr chaff + 10% bond supplement), P3 (85 gr chaff + 15% bond supplement), P4 (80 gr chaff + 20% bond supplement), P5 (75 gr chaff + 25% bond supplement), P6 (70 gr chaff + 30% bond supplement), and P7(65 gr chaff + 35% bond supplement). The result showed that the viability of fungi, after 30 days stronge were 100%. The best formulation was P4 as determined based on brittleness test with score 6,73%, its higher than its other formula. Key words: Formulation, biological control agent, Aspergillus sp., and tablet.

Abstract

The study was held on effectiveness of extract lemongrass leaf (Cymbopogon nardus L.) as a anti septic of Candida albicans. The study conducted as long as 3 months, from march to may 2013. Extracts was obtained by maceration method. This study aims to determine the extract can be prevent the growing of the Candida albicans, and to determine the effective concentration to prevent the growing of the fungus Candida albicans. This study is an experimental research with a completely randomized design. Bes trew diffusion method by a concentration of 25%, 50%, 75%, 100% and metronidazol liquid is used to determine the zone diameter inhibition in the five treatment and five replications. Inhibition zone diameter data were analyzed statistically ANOVA and Duncen Multiple Range Test (DMRT). The concentration stratified of extract effect on the zone diameter growth of Candida albicans. Based on the study, concentration at 75% is the best to be inhibited the growth of Candida albicans in conclusion, the extract is anti septic to the growth of Candida albicans. Keywords: Cymbopogon nardus L., Maceration, Candida albicans

Abstract

Research entitling "Microbiological Analysis of Stik Potato Fry in Cafe Lesehan Talise Palu" have been executed in March until May 2013 as a mean to analyse the cemaran microbe of at stik potato fry in Cafe Lesehan Talise Palu and to know what stik potato fry the have fulfilled the Health Standard specified by BPOM by mikrobiologis. Result of research indicate that pursuant to microbiological analysis quantitatively is ALT And MPN of at sampel stik potato fry in Cafe Lesehan Talise Palu show 2 sampel which is not growed by colony, 6 sampel which is < 3,0 x 104 CFU/g, 1 incalculable sampel and 1 sampel with the result 3,1 x 104 CFU/g for the ALT of bacterium, while for the ALT of kapang obtained by 1 sampel which is not growed by the colony and 9 sampel which is < 3,0 x 104 CFU/g, and also for the ALT of khamir obtained by 3 sampel which is not growed by the colony, 6 sampel which is < 3,0 x 104 CFU/g and 1 sampel with the result 3,7 x 104 CFU/g. Most Probable Number (MPN) show 9 sampel which is 3,0 x 100 MPN/g (<3) and 1 sampel with the result 2,3 x 101 MPN/g namely Cafe C. From examination result 10 sampel stik potato fry knowable that 90% still fulfill the Health Standard specified by Director General BPOM of while 10% do not fulfill the Health Standard specified by Director General BPOM. Keywords: Microbiological Analysis, Stik Potato Fry, ALT and MPN

Abstract

This research entitle "Detect the bacterium of Coliform and Escherchia coli at depot of drinking water refill the diKota Pasangkayu of Sub-Province of Mamuju of North of West Sulawesi" have been executed in March until May 2013, in Biological Laboratory of Biological Majors Base of Faculty MIPA Tadulako University Palu. Target of this research to detect the bacterium of Coliform and Escherchia coli at depot of drinking water refill in Town of Pasangkayu of Sub-Province Mamuju North West Sulawesi and also determine the quality by mikrobiologis pursuant to Standard of Indonesia National (SNI). Method used in this research is Most Probable Number (MPN). Result of Research indicate that at fifth of depot which have in test, that is at test of MPN Coliform of there is three depot which are positive namely depot B for the BI OF standard water with the value 4 MPN/100 mL sampel, for the depot of C namely standard CI water with the value 150 MPN/100 mL sampel, CII irrigate the process by niali 23 MPN/100 mL sampel, CIII irrigate the gallon with the value MPN 21 MPN/100 mL sampel, while for the depot of D namely IN standard water show the value MPN 43 MPN/100 mL sampel. Pursuant to examination of Coliform Fecal and Escherchia coli expressed by all sampel is negativity. Keywords: Coliform, Escherchia Coli, and Drinking water refill.

Abstract

Research entitle "Detect the Bacterium of Coliform and Escherichiacoli at Ices Beverage of Orange In Coastal Cafe Lesehan of Talise Palu" have been executed as a mean to detect and know the amount of bacterium Coiform and Escherichiacoli at ices beverage of orange in Coastal Cafe Lesehan of Talise Palu. This research have the character of descriptive, by using method of Standard Plate Count (SPC) And Most Probeble Number (MPN). Pursuant to research result from test SPC indicate that 50 % impure Cafe Lesehan because standard ineligibility of quality which is in specifying BPOM that is Cafe A of counted 1,1 x 104 CFU/ml, Cafe C of counted 4,3 x 104 CFU/ml, Cafe D of counted 1,5 x 104 CFU/ml, Cafe H of counted 1,8 x 104 CFU/ml. Examination of Method MPN from there are 30 % impure Cafe Lesehan by bekteri Coliform at ices beverage orange of because standard ineligibility of quality specified by BPOM that is at Cafe A of counted 143 x 101 MPN/ml, Cafe H of counted 2,33 x 101 MPN/ml, and Cafe J of counted 1,23 x 101 MPN/ml. Pursuant to examination of coliform fecal and test the Escherichia coli expressed by all sampel is negativity. Keywords: Coliform and Escherichia coli, ices beverage of orange, Cafe Lesehan.

Abstract

The research was about “Potential of Liquid Waste of Tempe Microbiologically as the alternative of Biogas Producer” had been conducted from July to September 2013 with the purpose of finding out of liquid waste of tempe microbiologically as the alternative of biogas producer and finding out the best concentration in media of liquid waste of tempe producer biogas and finding the best time for fermentation in producing biogas. The research method used is experimental which designed in the form of Complete Random Design (RAL), comprising 6 treatments and 3 times repetition. The research result shows that all treatment can produce biogas. The highest biogas volume is the treatment P5 (50% : 50%) which was as 128.10cm3. The best fermentation time occured on the third day (72 hours) up to the fourth day (96 hours) of fermentation. Keywords: Liquid Waste of Tempe, Microbiology and Biogas.

Abstract

This research was about “ The Usage of Liquid Waste of Tempe and Liquid Waste of Tofu by Using Different Inoculum to Produce Biogas”. The objective of the research was to use liquid waste of tempe and tofu by using different inoculum in producing biogas, finding out the interaction between the waste and microba inoculum in producing biogas, finding out the biggest amount of gas produced by liquid waste of tempe and tofu by using different inoculum in producing biogas and finding out the best time for fermentation in producing biogas. This research was designed by using Factorial Pattern of Complete Random Design (RAL) 2x2 comprising 4 treatments and 3 times repetition. The treatment examined was P1 )Liquid Waste of tempe + Rumen), P2 (Liquid Waste of tofu + Rumen), P3 (Liquid Waste of tempe + Rumen Contents), P4 (Liquid Waste of tofu + cow feces ) with the comparison of every treatment 50% :50%. Based on the research result, it shows that every treatment examined can produce biogas. The highest volume of examined treatment is P1 treatment (Liquid Waste of tempe + Rumen Contents) which was 57.15cm3. Abstract

The aim of this study is to characterize the growth rate of mycelium of oyster mushroom (Pleurotus ostreatus). It’s expected that the out come of this study is to find out the best medium which suitable for oyster mushroom propagation. The experiment was designedbased oncompletely randomized design(CRD) consistingof 6 treatmentsand 3 repetitions. The treatment were various ratio of rice bran and corn on the growth medium. The ratio used as treatment (rice : corn) were 100:0 (P1), 80:20 (P2), 60:40 (P3), 40:60 (P4),80:20 (P5), 0:100 (P6). Result of experiment showed that medium P3 was the bestin supporting the growth rate of mycelia which in average of 8,45 mm/day. It were higher than it in other media P4 (7,95 mm/ day), P6 (7,45 mm/ day), P5 (7,08 mm/ day), P1 (6,33 mm/ day) and P2 (5,5 mm/ day).
