Abstract

Human insulin potential has become an interest and is important in maintaining the success of therapyin patients with the availability of chemical-based analytical methods, however, only a few have beenusing immunoassays. This study aimed to produce IgG polyclonal antibodies from rabbits immunizedwith 1 mg/mL rhINS subcutaneously and validated by indirect ELISA. Antibody was precipitated andfractioned on a HiTrap® Protein A HP column before being quantified with a UV spectrophotometerat λ 280 nm. The characterization was conducted by Dot Blot test on a BCIP-NBT substrate, as well asSDS-PAGE and Western Blot with polyacrylamide gel concentrations of 7.5% and 17.5%. Validationwas performed using solutions containing glycerol and m-cresol as matrices spiked with rhINS. Thelinearity test in the rhINS concentration range of 80.11-200.28 μg/mL (r = 0.99) showed the linearresult. The accuracy and precision obtained an average of 99.11%±5.01 and 3.91%, while the LODand LOQ were 22.05 μg/mL and 73.51 μg/mL, respectively. Human insulin was stable at 2-8oC for 24hours (α: 0.05, ANOVA). In conclusion, in-house produced IgG polyclonal antibodies and goat anti-IgG peroxidase conjugate can be used for routine testing of human insulin.

Abstract

The development of analog curcumin compounds by modifying the structure of monocarbonyl into an analog indazole of curcumin (AIC) is recognized to have a great potential. Still, only a few reports have been available. Rarely occurring in nature, indazole molecules are typically created through chemical synthesis. Therefore, this study aimed to synthesize six new AIC compounds with a particular focus on testing in vitro antioxidant activity using the DPPH and FRAP methods, as well as anti-inflammatory activity using the protein denaturation method. The results showed that the compounds formed had high anti-inflammatory activity but low antioxidant activity. All synthesis products produced higher anti-inflammatory activity than standard diclofenac sodium and curcumin compounds. Specifically, compound 3a showed the highest anti-inflammatory activity with an IC50 = 0.548 ± 0.062 μM. Therefore, it was concluded that compound 3a has the potential to be further studied for anti-inflammatory activity.

Abstract

Indonesia has a diverse marine biota with bioactive substances that may provide a way to overcome COVID-19. In this research, we explored secondary metabolites isolated from marine invertebrates in Indonesia. A total of 137 compounds from different types of invertebrates were screened against ACE2 and SARS-CoV-2 Omicron spike protein. In relation, molecular docking and ADMET prediction were investigated to find the best compound. A molecular dynamics study was performed to determine the stability of the binding between the compound and ACE2 and RBD of the Omicron spike protein virus receptor. The results showed that acanthomanzamine E and cortistatin L have prominent molecular docking properties as ACE2 and SARS-CoV-2 Omicron spike protein blockers with binding energies of -12.87 and -9.96 kcal, respectively. The ADMET results also showed that both compounds have a promising drug-likeness, with only minor exceptions in partition coefficient (logP) and half-life. In conclusion, acanthomanzamine E and cortistatin L have shown significant potential as lead compounds for drug targeting ACE2 and SARS-CoV-2 blockers.

Abstract

Karbamat merupakan salah satu jenis pestisida yang banyak digunakan untuk membasmi hama buah dan sayur. Untuk menentukan bahwa residu karbamat dalam sayuran masih aman dikonsumsi manusia, telah dilakukan analisis beberapa residu karbamat seperti metomil, karbaril, karbofuran, dan propoksur. Sampel-sampel tomat, apel, selada air, kubis, dan sawi hijau dikumpulkan dari tiga supermarket dan satu pasar tradisional di Depok, Jawa Barat. Analisis dilakukan serempak untuk ke empat residu karbamat menggunakan kromatografi cair kinerja tinggi denganpereaksi o-ftalaldehida dan 2-merkaptoetanol dalam reaktor pascakolom dengan detektor fluoresensi. Dari sampel-sampel buah dan sayur yang dianalisis, hanya sawi hijau asal pasar tradisional yang positif mengandung propoksur dengan kadar 1,2 mg/25 gram berat basah (0,048 mg/g berat basah). Dengan Acceptable Daily Intake(ADI) propoksur 0,005 mg/kg berat badan/hari, konsumsi sawi hijau harian seberat 20 g/hari masih cukup aman dari gangguan kesehatan akibat pajanan kronik propoksur dengan margin of safety 298,7 (> 100 sebagai batas aman). Carbamat is a group of pesticides which is commonly used to control fruits and vegetables pests. To determine that carbamat residues in fruits and vegetables are safe for human consumption, carbamate residues such as methomyl, carbaryl, carbofuran, and propoxur in vegetables and fruits have been analyzed. Samples of tomato, apple, water lettuces, cabbage, and mustard greens were collected from three supermarkets and one traditional market in Depok, West Java. The analysis was carried out simultaneously for all four carbamate residues by high performance liquid chromatography using o-phtaladehyde and 2 mercaptoethanol reagents in post-column reactor with a fluorescence detector. Of fruits and vegetable samples analyzed, only mustard greens from traditional market positively containe propoxur at 1.2 mg/ 25 gram wet weight (0,048 mg/gram wet weight). With Acceptable Daily Intake (ADI) of 0.005 mg/kg body weight/day, mustard greens consumption of 20 g/day is safe from adverse health effect from chronic exposure to propoxur with Margin of Safety of 298.7 (> 100 as safe limit).

Abstract

Objective: The purpose of this research was to evaluate and select the best architecture among native convolutional neural network (CNN), MobileNetV2, ResNet50V2, and EfficientNetB0 for predicting the endpoint of the high shear wet granulation process, with accuracy as the main evaluation metric. Methods: The dataset was captured from an industrial camera using static image analysis and was manually labeled as “NOT READY” and “READY” according to the traditional endpoint method based on the mixer’s ampere point in the granulator. The dataset contained a total of 180 images, which were split between training and validation sets. Native CNN and TensorFlow Keras application programming interface (API) were utilized with MobileNetV2, EfficientNetB0, and ResNet50V2 as base feature encoders. Hyperparameters, such as final Fully Connected (FC) layer width, dropout rate, and learning rate, were optimized for binary classification using Keras hyper tuning. Results: The best was the native CNN, it was also the fastest among the three other models, taking only 20-30 ms per step for inference during runtime, though it requires 9000 ms time for training, the longest time among the models. It achieved an accuracy of 98%, and a validation accuracy of 97%. Conclusion: The system was able to determine when a wet granulation process has reached its endpoint based on live images from a camera after being trained on previously labeled data. The native CNN was the best model, offering the fastest runtime performance and the highest accuracy.

Abstract

When playing sports, athletes are required to always maintain their health to provide maximum results in their performance. Supplements are products intended to complement nutritional needs, maintain, enhance and/or improve health functions and have nutritional value and/or physiological effects. L-Carnitine is a health supplement that is widely consumed by athletes. L-Carnitine has many effects on the body and the main effects are seen on blood pressure and VO2 max. The aim of this review is to look at the effects of L-Carnitine on blood pressure and VO2 max. It was found that L-Carnitine had more effect on a person's diastolic than systolic at doses of more than 2 grams/day. Meanwhile, the effect on VO2 max was seen in long-term use of 2 grams for 24 weeks and found a significant increase in VO2 max (p<0.05). L-Carnitine does influence blood pressure, VO2 max and is also correlated with endurance, but until now there is still a need for more research to show that the effects produced by L-Carnitine are significant both on blood pressure and VO2 max in athletes.

Abstract

Hypertension poses a significant global health burden, prompting exploration into alternative treatments. This study investigates the pharmacological network of Orthosiphon aristatus, aiming to uncover its therapeutic potential for hypertension. We compiled data on Orthosiphon aristatus chemical constituents and predicted potential targets for its key components. Through screening processes utilizing the Gencard database, active compounds and protein targets were identified. We examined protein target similarities between Orthosiphon aristatus and hypertension, constructing a network illustrating the relationships between active compounds and target genes. Additionally, we conducted GO function analysis and KEGG pathway enrichment to elucidate Orthosiphon aristatus role in hypertension. Notably, TP53 exhibited the highest degree of centrality, while Scutellarein and aurantiamide acetate displayed the highest affinities in molecular docking with TP53. These findings offer novel insights into Orthosiphon aristatus' potential as an adjunctive therapy for hypertension and contribute to the advancement of pharmacological interventions in this domain.

Abstract

Benzimidazole and vanillin are compounds which can act as anti-inflammatory. However, the potential of these two compounds is still low. The structure of benzimidazole and vanillin was combined with Mannich base substitution in order to increase potency and selectivity as an anti-inflammatory. In this study, benzimidazolvanillin (1) and its Mannich base derivatives were synthesized. The synthesized compounds (2a-d) show anti-inflammatory activity based on in vitro tests by using protein denaturation inhibition test. Furthermore, the IC50 obtained ranged from 210 -243 μM. This potency is lower than diclofenac (IC50: 2,99μM). In molecular docking, the results of the inhibition constant ratio between COX-1 / COX-1 as well as the interaction of the ligand and target protein show that benzimimidazolvanillin and its Mannich-base derivatives could be anti-inflammatory drug candidates that should be investigated further.

Abstract

This study systematically analyzed Indonesia's regulatory reforms for traditional medicines, health supplements, and cosmetics (TMHSC) issued between 2020 and 2024. The objective was to map the scope, distribution, and policy orientation of the 35 regulations enacted by the Indonesian Food and Drug Authority (BPOM) within the framework of the National Medium-Term Development Plan (RPJMN 2020–2024). A qualitative policy analysis was employed, combining document analysis, thematic coding, and comparative benchmarking with frameworks from the ASEAN, European Union (EU), and Organisation for Economic Co-operation and Development (OECD). The findings show that 25 regulations were directed toward strengthening regulatory services, nine targeted compliance and Good Manufacturing Practices (GMP), and one supported research and innovation through preclinical testing standards. This distribution reflects a policy trajectory that prioritizes service delivery and compliance assurance, while progressively integrating evidence-based approaches to research and innovation. A comparative analysis revealed a strong alignment with ASEAN harmonization initiatives and an incremental adoption of international benchmarks, such as ISO 22716 for cosmetics and the WHO GMP guidelines for herbal medicines. Overall, Indonesia's TMHSC regulatory transformation demonstrates a balanced and adaptive governance model that safeguards public health, promotes innovation, and enhances regional policy coherence. The results provide practical implications for policymakers, particularly BPOM and ASEAN member states, in developing regulatory frameworks that effectively balance consumer protection, innovation enablement, and market competitiveness within the TMHSC sectors.
