Abstract

Telur ikan cakalang sebagai hasil samping pengolahan ikan asap potensial dimanfaatkan sebagai hidrolisat protein. Hidrolisis menggunakan fermentasi bakteri diharapkan menghasilkan peptida bioaktif yang bersifat antioksidan dan antibakteri pada hidrolisat protein telur ikan cakalang. Penelitian ini bertujuan menentukan aktivitas antioksidan dan antibakteri dari hidrolisat protein hasil fermentasi telur ikan cakalang. Penelitian dilakukan secara deskriptif. Pembuatan hidrolisat protein menggunakan 3 perlakuan fermentasi yaitu fermentasi spontan (FS), fermentasi menggunakan starter tunggal L. plantarum SK 5 (FL) dan fermentasi menggunakan bakteri endogenous serta ditambah bakteri L. plantarum SK 5 (FSL). Hasil pengamatan menunjukkan bahwa total BAL (bakteri asam laktat) perlakuan FS meningkat hingga jam ke-48 fermentasi lalu menurun hingga akhir fermentasi. Perubahan total BAL yang cenderung statis terjadi pada perlakuan FSL, sedangkan pada perlakuan FL terjadi penurunan hingga jam ke-48 lalu cenderung statis hingga akhir fermentasi. Total mikroba aerob pada perlakuan FS dan FSL mengalami penurunan hingga jam ke-48 fermentasi, sedangkan pada perlakuan FL total mikroba aerob dari awal hingga akhir fermentasi terhitung sama dengan total BAL. Nilai absorbansi pengujianasam amino bebas pada perlakuan FS meningkat paling tinggi selama fermentasi. Aktivitas antioksidan dari hidrolisat protein perlakuan FS dan FSL mengalami penurunan seiring bertambahnya waktu fermentasi, sedangkan aktivitas antioksidan pada perlakuan FL tidak mengalami peningkatan setelahfermentasi. Aktivitas antibakteri tertinggi terjadi pada hidrolisat protein perlakuan FL pada konsentrasi 0,5 mg/μL dengan lama waktu fermentasi 48 jam. Zona hambat yang terbentuk tergolong sedang.

Abstract

Gelatin komersial yang diekstraksi dari tulang dan kulit hewan (sapi dan babi) menimbulkan kekhawatiran masyarakat akan keamanan dan kehalalan produknya. Oleh karena itu, sumber gelatin, tulang dan kulit ikan dapat menjadi alternatif. Pengolahan kulit ikan kakap (Lutjanus sp) menjadi gelatin dapat dilakukan dengan penanganan asam atau basa. Tujuan penelitian ini adalah memanfaatkan kulit kakap merah sebagai sumber gelatin yang diekstraksi menggunakan asam (asam asetat 1-5%) dengan lama ekstraksi (12, 18, dan 24 jam). Produk gelatin terbaik diperoleh dari kulit ikan kakap merah yang direndam dalam asam asetat 3% selama 18 jam. Analisis fisika dan kimia ditujukan untuk mengkarakterisasi gelatin yang meliputi kadar air 10,19%; abu 0,40%; lemak 0,33%; dan protein 88,88%; kekuatan gel 312,5 bloom; viskositas 17,4 cP; pH 5,45; titik gel 10,15°C; titik leleh 27,26°C, titik isoelektrik 8; derajat putih 34,7%. Logam berat Pb dan Hg tidak terdeteksi.

Abstract

Tuna (Thunnus sp.) by-products from frozen loin and canning industry especially the eye is rich in proteins and in lipids consisting of polyunsaturated fatty acids (PUFA). That requires protective agent (antioxidant) to inhibit the oxidation naturally present and predicted to be protein peptides. Enzymatic hydrolysis of protein is an appropriate method to produce bioactive peptide with such nutraceutical/pharmaceutical function such as an antioxidant peptide. This study aimed to produce protein hydrolysate having a function as anwith an antioxidant activity from eye of tuna through enzymatic hydrolysis and determining the antioxidant activity by DPPH methods. Protein soluble content of tuna’s eye protein hydrolysate (TEPH) ranged from 59.98±0.130 to 94.90±0.002%. The degree of hydrolysis (DH) of TEPH was about 9.10±0.28 to 16.14±0.09%. The highest inhibition of DPPH radical scavenging activity was 93.57±0.05% (at 5 mg/mL) was obtained with a DH of 11.35±0.002% at the concentration 0.1% of papain for 6 hours hydrolysis. Abstract

Protein malnutrition in toddler is a problematic issue in Indonesia. Inclusion of roe protein concentrate (RPC) from skipjack in the breastfeeding complimentary foods of or infant foods can be an alternative to solve the protein malnutrition. This research was aimed to analyze the organoleptic score, nutritional facts and functional properties of the infant food supplemented with the RPC. Six different formulations (F1-F6) were prepared. The F2 formula was considered as the best formula. The infant food prepared using this formula contained 19.43% of protein and was higher than the control and the commercial product. The food also contained nutrition as required by the Indonesian National Standard (SNI) and Food and Agriculture Organization (FAO) standard. Furthermore, the functional properties of the infant food was comparable to that of the commercial and control product.

Abstract

Swim bladder is a marine byproduct and a potential source of collagen needed by food, cosmetical, biomedical and pharmaceutical industry. This study evaluated the effectiveness of alkaline pretreatment and papain enzyme on the characteristics of collagen from the catfish swim bladders. TThree concentrations of alkaline (0.05; 0.1 and 0.15 M) with were used to pretreat the swim bladders for 2; 4; 6; 8; 10 and 12 hours. The collagen was also treated by papain enzyme with concentrations of 0; 5,000; 10,000; 15,000 and 20,000 U/mg for 24 and 48 hours. The result showed that pretreatment using NaOH 0.05 M for 6 hours effectively reduced the noncollagen protein (p<0.05). Meanwhile, the treatment using papain enzyme with concentration of 5.000 U/mg for 48 hours improved the collagen solubility. The FTIR of collagen showed the existence of a triple helix structure. Meanwhile, the thermal analysis showed collagen gelation transition temperature (Tg) was at 88oC.

Abstract

Swim bladders of catfish contain high protein, therefore it is can be used as a raw material for collagen. The study aims to determine the chemical characteristics of swim bladders, the pretreatment of non-collagen, extraction of collagen dissolves acid and to evaluate the characteristics of collagen. The method of this study is KOH pretreatment with a concentration of 0,05; 0,1; and 0,15 for 12 hours. The extraction process is done by soaking the sample in a solution of acetic acid with a concentration of 0,25; 0,5 and 0,75 M (ratio 1:10; b/v) and extraction time for 24; 48; and 72 hours at 4oC. The experimental design used for alkaline and acetic acid pretreatment were factorial completely randomized design. The result showed that the protein content of swim bladder was 85,26% (db), the profile of amino acids were dominated by three amino acids namely glycine (56,85 mg/g), prolyne (31,03 mg/g), and alanyne (23,85 mg/g). Using 0,05 M KOH for 8 h was selected as the best pretreatment method for collagen extraction. Extraction method using 0,50 M acetic for 48 h resulted he best collagen which revealed the existence of a triple helix structure and had Tg 84oC.

Abstract

Spirulina is a microalga known as a superfood which contains high protein and is used in cosmetics, foods, and supplements industry. Collagen hydrolysate is the result of hydrolysis of collagen by protease. This study was aimed to determine the antioxidant activity and inhibition of tyrosynase from Spirulina, collagen hydrolysate and tablet formulated by combination of Spirulina and collagen hydrolysate. This research was conducted in three stages. The first stage was the extraction and hydrolysis of tilapia skin collagen, while the second stage was culturing and harvesting of Spirulina, and the third stage was determination of tablet formulation based on Spirulina and collagen hydrolysate followed by analysis of the antioxidant activity and inhibition of tyrosinase by selected tablet. Three formula with different ratio between Spirulina and collagen hydrolysates namely formula A (25%:53.75%), formula B (28.125%:50.625%), formula C (31.25%:47.5%) and additive such as vitamin C (12.5%), talk (1.25%), magnesium stearate (0.625%), aerosil (1.875%), avicel (5%) were developed. The result showed that formula A was the selected formula meeting the requirements of Ministry of Health based on the physical characteristics. The antioxidant activity of tablet A was 843 ppm and the tyrosinase inhibition was 163.56 ppm.

Abstract

Vitreous humor is a part of the tuna fish eye that plays a role in maintaining the brightness of the eye and contains hyalocytes having function to produce hyaluronan. Hyaluronan is thought as an antioxidant to prevent unsaturated fatty acids damage in tuna fish eye caused by free radicals. Hyaluronan from vitreous humor tuna fish eye has not been widely studied. This study was aimed to extract the hyaluronan found in vitreous humor of tuna fish eye and determine its characteristics. Extraction of hyaluronan consisted of presipitation, centrifugation, enzyme digestion, defatting, dialysis, and freeze drying. Antioxidant activity was determined by three methods comprising 1,1-diphenyl-2-picrylhydrazyl2,2-azinobis (DPPH), 3-ethylbenzo-thiozoline-6-sulfonic acid (ABTS), and ferric reducing antioxidant power (FRAP). Vitreous humor tuna fish eye had a proportion of 31% of eye, pH value 6.905±0.0083, viscosity 195.56±12.86 cP, water 98.09±0.49%, and glutamic acid 932.06±3.80 mg/L. The hyaluronan yield was about 0.005±0.001%. The IC50 antioxidant activity of hyaluronan against DPPH was 25.52±0.22 mg/mL while the inhibition capacity on ABTS was 1.92±0.08% and the ferric reduction capacity was 9.34±0.55 µM/g. These results indicates that the antioxidant activity of hyaluronan from vitreous humor tuna fish eye is low.

Abstract

Kulit ikan nila merupakan hasil samping proses pengolahan ikan yang dapat dimanfaatkan sebagai alternatif bahan baku kolagen dan hidrolisatnya. Hidrolisat kolagen diketahui memiliki aktivitas biologis yang potensial diantaranya sebagai antioksidan. Penelitian ini bertujuan untuk menentukan aktivitas antioksidan hidrolisat kolagen dari kulit ikan nila (Oreochromis niloticus) secara in vitro. Penelitian ini terbagi menjadi tiga tahap meliputi fase praperlakuan kolagen, ekstraksi kolagen dan hidrolisis kolagen. Pada proses hidrolisis digunakan enzim papain dengan konsentrasi yang berbeda (4.000 U/g, 6.000 U/g, 8.000 U/g sampel) dengan waktu hidrolisis selama 60, 120 dan 180 menit. Rerata nilai rendemen hidrolisat kolagen kulit ikan nila sebesar 15,17%. Waktu hidrolisis, konsentrasi enzim yang diberikan serta interaksi antar kedua faktor tersebut berpengaruh secara signifikan terhadap nilai derajat hidrolisis dan aktivitas antioksidan hidrolisat kolagen kulit ikan nila. Hidrolisat kolagen kulit ikan nila yang dihidrolisis selama 120 menit dengan pemberian konsentrasi enzim sebesar 8.000 U memiliki rerata persentase derajat hidrolisis sebesar 33,94%, serta rerata nilai IC50 terbaik yakni 93,32 µg/mL, termasuk pada kategori senyawa antioksidan kuat. Nilai zeta potensial hidrolisat kolagen kulit ikan nila adalah -10,9 mV.

Abstract

Tuna eye oil which is rich in omega-3 fatty acids eicosapentaenoic acid and docosahexaenoic acid can be obtained by low temperature extraction however the yield was not optimum yet. The yield value of tuna eye oil can be increased with enzymatic hydrolysis. Response Surface Methodology (RSM) can help analyze the effect of factors on the response to optimize the conditions of enzymatic hydrolysis. The purpose of this study was to maximization of oil yield, determine the optimum conditions for enzymatic extraction of tuna eye oil with different papain enzyme concentration and hydrolysis time using central composite design-face centered (CCDFC) modeling on RSM and determination of oil characteristics. The validation optimum of the enzymatic hydrolysis resulted in a yield value of 11.24±0.09% at the concentration of 1.02% papain enzyme for 3 hours. The EPA and DHA content of tuna eye oil at the optimum condition was classified as very high, 6.02% EPA and 30.30% DHA from 99.90% of the total identified fatty acids.
