Abstract

Ikan gabus (Channa striata) mengandung fraksi protein albumin yang besar dan potensial digunakan sebagai biofarmasi serta bahan substitusi albumin manusia. Penelitian ini bertujuan untuk menentukan lama waktu pengukusan dan profil tepung ikan serta ekstrak daging dan ekstrak daging ikan dengan penyaringan dari ikan gabus sebagai sediaan imunomodulator. Ekstraksi albumin menggunakan metode pengukusan dengam lama pengukusan berbeda (10, 15, 20, dan 25 menit) dan albumin dianalisis menggunakan metode Bromocresol Green (BCG). Penelitian ini meliputi analisis proksimat dan logam berat ikan gabus, ekstraksi, analisis mineral, kadar albumin, berat molekul menggunakan SDS-PAGE, gugus fungsi menggunakan FTIR, ukuran partikel dan asam amino. Hasil penelitian menunjukan bahwa daging ikan gabus mengandung air 80,93%, lemak 1,37%, protein 16,76%, abu 0,65% dan karbohidrat 1,28%. Kandungan logam berat daging ikan gabus yaitu Hg 0,013-0,28 ppm dan Pb 0-0,637 ppm. Ekstraksi menghasilkan albumin daging segar 4,8 g/100 mL, ektrak daging 3,76 g/100mL, ekstrak daging dengan penyaringan 4,6g/100 mL dan tepung ikan 3,8 g/100 mL. Tepung ikan mengandung kadar protein 85,80%, air 9,26%, abu 2,67%, lemak 2,06% dan energi total 362,58 kkal/ 100 g. Ekstrak daging dengan penyaringan mengandung asam amino serina, lisina, asam aspartat, alanina, dan asam glutamat, serapan gelombang albumin 1.644,96 dengan berat molekul 63 hingga 70 kDa dan terdapat molekul IgG sebagai sediaan imunomodulator.

Abstract

Collagen is a connective tissue protein composed of the amino acids glycine, proline, alanine and hydroxyproline, which are found in the skin, bones, scales and swim bladder tissue of fish. Fish swim bladder as a by-product of the fishing industry have the potential to be converted into halal collagen. This study aims to evaluate the effectiveness of the pre-extraction process of the combination of NaOH and ultrasonication and the use of papain enzyme in the extraction process on the physicochemical characteristics of tuna swim bladder collagen. The research method was a combination of pre-extraction of NaOH and 150 minutes of ultrasonication and immersion for 2; 4; 6; 8; 10; 12; 14; 16; 18; 20; 22 and 24 hours, extraction using the papain enzyme with a concentration of 0; 3,500 and 5,000 U/g and extraction time of 36 and 48 hours. The results showed that the pre-extraction process using 150 minutes of ultrasonication with soaking for 22 hours was the best time to eliminate non-collagen proteins, and extraction using papain enzyme with a concentration of 5,000 U/g and soaking for 36 hours was the preferred treatment for the degree of collagen development. The yield of collagen obtained was 7.01±0.4%, the dominant amino acids were glycine, arginine and proline, tuna swim bladder collagen had a protein band of α1 chain of 117 kDa, α2 chain of 107 kDa and β chain of 244 kDa and zeta potential value of 0.496 mV.

Abstract

In recent years, shrimp farming have experienced a variety of problems, both technical and non-technical, causing the shrimp farm do not work properly (idle). In 2012 the Ministry of Maritime Affairs and fisheries formulate revitalization program which focused on the rehabilitation ponds channel through Demonstration Farm (Demfarm) program. The purpose of this study is to analyze the effectiveness of the implementation of the Demfarm program in the Blanakan District in Subang regency. The data has been analyze using qualitative analysis, quantitative analysis, SWOT analysis (Strengths, Weaknesses, Opportunities, and Threats), and AHP (Analytical Hierarchy Process). The result of the study shows that Demfarm implementation in Blanakan District, Subang Regency is quite effective. Blanakan District in Subang Regency is an ideal area for shrimp farm, the application of Demfarm technology can improve the productivity of farms  from 0-17  up to 7-75 tons / ha, the programs also help to establish the partnership system between farmers and the private sector. Shrimp farms have 0.148 IFE and 0.495 EFE, another most important thing for shrimp farming development in the Blanakan District is technology have 0.341. Shrimp farming worth to be developed, the value of Benefit Cost Ratio (B/C ratio) > 1, the Net Present Value (NPV) generated > 0 Break Event Point (BEP) the lowest production 7000 kg and the highest production is 7.500 kg, for BEP the lowest price is Rp60 000 and the highest price is Rp70.000

Abstract

Case study is conducted towards 12 SME milkfish processors in Bekasi, at which its raw materials are originated from Karawang Regency through a fresh milkfish supply chain. The potentcy of Karawang Regency as a milkfish producer does not guarantee continuous availability of the raw materials. The objectives of this study were: (1) To evaluate supply chain; and (2) To compile a priority strategy of fresh milkfish supply chain management in Bekasi, West Java. Data collection was conducted by purposive and snowball sampling techniques, starting from SME processors, retailers and collective traders in Bekasi, to milkfish farmers in Ciparage Jaya Village, Tempuran Sub-District, Karawang Regency. The study used descriptive analysis, evaluation by comparing the activities of the members of the supply chain with the existing standards, Strength, Weaknesses, Opportunities and Threats (SWOT) analysis and Analytic Hierarchy Process (AHP). The evaluation result showed there was gap between the activities of each supply chain member with the Indonesian National Standard (SNI) namely (1) implementation of GMP and SSOP by SME processors, (2) means of transport and storage of fresh fish by collective trader and retailer, (3) good aquaculture practices by farmer and (4) supply chain management by all members along the supply chain. Based on the SWOT and AHP matrix analyses, the development strategies of supply chain by actor was (1) the formation of raw material procurement network and marketing network by SME processor (0.326); (2) the quality improvement of human resource by the government (0.249); (3) the formation of organizations by farmer (0.230), and (4) maintaining partnership in business procurement cost by collective trader (0.195).

Abstract

Pangasius fillet industry in Indonesia that has grown in recent years will face the ASEAN regional competitiveness that began in late 2015. To face competitiveness from other countries, especially Vietnam as the largest Pangasius fillet producer, this study was carried out with regard to internal and external factors as a consideration in determining future strategies. This study aims to generate alternative competitiveness strategies of Pangasius fillet through descriptive analysis, SWOT, and AHP. The analysis result of internal forces obtains the total scores of 2.645 and external forces with the score of 2.505; this means that suggested strategy belongs to hold and maintain category. AHP analysis shows that alternative strategy with the highest score consecutively is the development of aquaculture technology for quality and efficient raw material, institutional strengthening of fish farmers and suppliers, diversification and product innovation, encouragement in investment on an integrated Pangasius fillet industry, branding of the Indonesian yellow fillet. The main objective of the strategy is focusing on the continuity of production and fulfillment of consumer preferences in order to create stability of supply and demand.Keywords: AHP, competitiveness, Pangasius fillet, SWOT

Abstract

By product that rich in protein such as fish roes are potential as raw material for protein concentrate. This research aimed to utilize skipjack roes to produce protein concentrate and to characterize its functional properties. The method used to extract protein was defatting method using isopropyl alcohol and ethanol with extraction times of 1, 2, and 3 hours.  The results showed that skipjack roes contained 19.81% of protein, 3.41% of fat, 71.32% of moisture, 2.04% of ash, and 1.53% of carbohydrate (by difference). Defatting method using isopropyl alcohol for 3 hours produced the best roe protein concentrate (RPC).  The product meets to the quality requirements of fish protein concentrate type B, contained protein and fat of 71.79% and 2.78%, respectively. This product also had functional properties as follows: water absorption capacity (1.57 ml/g), oil absorption capacity (1.82 g/g), emulsion capacity (81.65%), bulk density (0.51 g/ml), foaming capacity (1.90 ml), foaming stability (0.22 ml) and protein digestibility (95.86%).  Lysine and leucine became the major essential amino acid of RPC, with values were 70.76 and 64.91 mg/g protein, respectively. The composition of amino acids of RPC skipjack consisted of 8 essentials amino acids, 5 non-essentials amino acids and 2 semi-essentials amino acids. Abstract

ABSTRACT Spirulina contains complete nutrition and bioactive components as a source of antioxidants. The aims of this research were to determine a formula of troche from Spirulina platensis according to the physical characteristic standard of the Indonesian Health Ministry, and to determine the antioxidant activity of the selected troche from Spirulina platensis. This research was conducted in three stages. The first stage was troches formulation. The second stage was troches physical analyses, including weight uniformity, friability, hardnes, and disintegration time. The third stage was to determine the antioxidant activity of the selected formula of troche from Spirulina platensis. There were five formulas of troches i.e., FTS 1, FTS 2, FTS 3, FTS 4, and FTS 5. The troche consisted of Spirulina platensis powder, carrageenan, gelatin, corn flour, sugarleaf, and mint powder. Based on the weight uniformity of the troche, all formulas met the standard. However, according to friability aspect, the best formula was FTS 1 with rate of 1.7%. The hardness of five formula ranged from 0.65 to 2.3 Kp, and none met the standard. The best formula based on disintegration time measurement was FTS 1 (24:47 minutes). Based on the physical characteristics of the troches, the selected formula was FTS 1. Abstract

Quality deterioration of fish is caused by various factors, one of them is due to the activity of cathepsin enzyme. Cathepsin enzyme activity can be inhibited by enzyme inhibitors. Inhibitors of the enzyme can be obtained from fish meat tissue by separating the enzyme-inhibitor complex with heat treatment. The purpose of this research was to extract and apply the cathepsin inhibitors to inhibit deterioration of fish quality. This research was conducted in three  steps: (1) the determination of the optimum extraction temperature for extraction of cathepsin inhibitors, (2) the determination of the most potent cathepsin inhibitors with various concentrations, and (3) the application of cathepsin inhibitors in inhibiting the deterioration of fish quality. The results showed that the cathepsin inhibitors was optimally extracted at 80°C. Cathepsin inhibitors diluted using buffer with a ratio of 1: 1 had the same relative activity without dilution. The process of quality deterioration of milkfish  control in chilling temperatures occured after 504 hours (21 days) storage with a 24-hour observation time interval. The complete process during observation was pre-rigor (0 hour ), rigor mortis (96 hours), post-rigor (360 hours), and deterioration (504 hours). The process of milkfish quality deterioration treated with soaking in  cathepsin inhibitors occured after 624 hours (26 days) storage with the detailed process as follows: 0 hour (pre-rigor), 144 hours (rigor mortis), 408 hours (post-rigor), and 624 hours (deterioration). Thus fish marinated with cathepsin inhibitors achieve slower deterioration phase 5 days. Keywords: cathepsin inhibitor, deterioration, pre-rigor, rigor mortis, post-rigor, quality deterioration.

Abstract

Tuna Thunnus sp is a high-value fisheries products. The abundance of waste from tuna’s processing industry has not been used optimally. The objective of this study was to produce and characterize nanohidroxyapatite (Ca5(PO4)3((OH)0.8F0.2) of tuna’s Thunnus sp bone waste using high temperature sintering of 400-800OC continuing by nanomilling method. The results of the characterization showed that the sintering temperature of 800OC was the best treatment yielding of hidroxyapatite 57.73%, 89.27% white degree level. The result of statistic analysis showed that the sintering temperature variation gave a significant effect on the white degree at the test level (p <0.05). Proximate result after sintering showed moisture content of 0.15%, 99.83% ash, 0.05% fat, 0.73% protein and 0% crude fiber. Results of heavy metal analysis showed the content of lead (Pb) and mercury (Hg) was negative, and cadmium (Cd) 0.018 ppm. The mineral test consist of calcium, phosphor, kalium, magnesium, iron, zinc, manganese, and sodium with the dominant elements were 49.76% calcium and 9.02% phosphor. The result of statistic analysis showed that the sintering temperature variation gave significant effect to the three main minerals at the test level (p<0.05). The Particle Size Analyzer test showed that nanohidroxyapatite particles was measured at 800-900 nm. The X-Ray Diffractometer analysis showed the results of sample identification contains hydroxyapatite and fluorian, with syn 100.00%. The hidroxyapatite was a compound complex and crystals formed has a hexagonal shape with a percentage of 82.9% crystallinity. Keywords : bone, nanohidroxyapatite, particles, sintering, tuna

Abstract

By product Muarenesox talabon swim bladders can be used as a raw material for desperately needed in the food, biomedical, pharmaceutical, and cosmeceuticals industries. The aims of the research were to observed the histological and determine chemical characteristics of swim bladder including proximate and amino acids; extraction of acid soluble collagen and determine the characteristics of collagen including proximate, pH, heavy metals, microbial, amino acids, functional groups, molecular weights, and thermal stability. The morphology of cunang swim bladder consists of outer, middle, and inner layers containing collagen fibers; 33.67±0.71%wb and protein whichwere dominated by three amino acids that were glycine, proline, and alanine. Pretreatment by 0.1 M NaOH for 8 hours (K1T4) and acid extraction by 0.25 M acetic acid for 72 hours (M1T3) was the best treatment yielding 14.51± 0.43% of collagen; having 12.12±0.04% wb of moisture; 88.54 ± 0.08% wb of protein; 1.31±0.23% wb of fat; 0.17±0.03% wb of ash. Not detected any heavy metals (Pb, Hg, As, Cd). Acidity pH was 4.31 and negative of E. coli and Salmonella. The main amino acids detected were glycine 241.06 mg/g; proline 88.73mg/g; and alanine 86,98 mg/g; FTIR spectra were revealed the presence of triple helix structures; electrophoresis patterns consisted of 136 kDa of mol weight of α1 and 117 kDa of mol weight of α2 were characterisedto be type I collagen; which had Tmax of 195.59ºC and ΔH 7.8113 J/g. Keywords: acid extraction, swim bladder, collagens, thermal stability
