Abstract

Penelitian bertujuan untuk mengetahui intensitas kalsium (Ca2+) intraseluler dan ultrastruktur pada beberapa bagian spermatozoa ayam setelah pembekuan. Analisis intensitas Ca2+ intraseluler menggunakan confocal laser scanning microscopy (CLSM) pada akrosom, kepala, bagian tengah dan ekor spermatozoa ayam sebelum dan setelah pembekuan, serta analisis Ca2+ juga dilakukan pada setiap lapisan spermatozoa. Analisis ultrastruktur spermatozoa menggunakan field emission-scanning electrom microscopy (FE-SEM) setelah pembekuan. Hasil penelitian menunjukkan bahwa intensitas Ca2+ intraseluler lebih tinggi pada bagian akrosom dan bagian tengah, sedangkan pada bagian kepala dan ekor lebih rendah. Proses pembekuan-thawing menyebabkan peningkatan intensitas Ca2+ intraseluler di semua bagian spermatozoa. Intensitas Ca2+ intraseluler pada lapisan bagian tengah lebih tinggi dibandingkan pada  bagian permukaan atas dan bawah spermatozoa. Pengamatan ultrastruktur menunjukkan adanya kerusakan pada bagian tengah, terlepasnya tudung akrosom, ekor melipat dan ujung ekor yang sedikit terbelah setelah pembekuan-thawing.

Abstract

Tujuan program pengabdian Masyarakat Pemula (PMP) ini adalah untuk meningkatkan pengetahuan Masyarkat mengenai optimasi green concentrate sebagai stimulan produksi susu sapi Bali pra melahirkan. Kegiatan pengabdian inidilaksanakan di desa Arabika, kecamatan Sinjai Barat, kabupaten Sinjai. Sasaran kegiatan dari pengabdian ini adalah anggota kelompok Tani Tabbangka. Pelaksanaan kegiatan dilakukan dengan pemaparan materi dan praktek secara langsung mulai proses penepungan green concentrate. Tingkat pengetahuan peserta dievaluasi dengan pembagian kuesioner preest dan posttest kemudian data hasil evaluasi dianalisis deskriktif. Hasil evaluasi menunjukkan bahwa terjadi peningkatan pengetahuan peserta kegiatan pengabdian dari skor 0 (pretest) menjadi skor rata-rata 60,82 (posttest). Skor tertinggi rata-rata pada pengetahuan pada proses pengolahan dan manfaat green concentrate. Tingkat Pendidikan dan umur berpengaruh terhadap daya serap ilmu peserta. Semakin tinggi Pendidikan kecenderungan skor semakin tinggi pula. Sedangkan peningkatan umur 40 hingga 80 tahun mengalami penurunan daya serap pengetahuan yang ditandai dengan skor yang semakin rendah. Kesimpulan yang diperoleh yaitu kegiatan pengabdian di desa Arabika kecamatan Sinjai Barat mampu meningkatkan pengetahuan Masyarakat tentang pemanfaatan green consentrate sebagai stimulant produksi susu sapi Bali pra melahirkan.

Abstract

This study aimed to evaluate the motility and kinematic parameters of Gaga chicken sperm at different ages using Computer-Assisted Sperm Analysis (CASA). Semen samples were collected seven times each from two roosters aged 6 months (young) and 12 months (mature) using the massage method. CASA evaluation assessed multiple parameters total motility, progressive motility, slow, static, distance average path (DAP), distance straight line(DSL), distance curvilinear (DCL), velocity average path (VAP), velocity straight line (VSL), velocity curvilinear (VCL), straightness (STR), linearity (LIN), amplitude of lateral head displacement (ALH), beat cross frequency (BCF), and wobble (WOB). Data were analyzed using an independent samples t-test. The results showed significant differences (P0.05) in progressive motility, DCL, VAP, and VCL between the two age groups, with the 12-month-old rooster showing higher values compared to the 6-month-old. In conclusion, the significantly higher sperm concentration and viability observed in mature roosters in this study underscore the importance of age-related reproductive maturity in optimizing semen quality for breeding purposes.

Abstract

This study aims to analyze the growth pattern of body weight of superior Sensi native chickens in an intensive maintenance system and to evaluate the effectiveness of the system in supporting production performance. This study was conducted for 75 days, covering three maintenance phases: starter (1–21 days), grower (22–42 days), and finisher (43–75 days), with a sample of 20 chickens from a population of 275 DOCs observed individually. The method used was a field experiment in a closed cage with ad libitum feeding. Data collected included initial body weight, weekly weight, daily weight gain (DWG), feed conversion (FCR), and mortality. The results showed a sigmoid growth pattern, where body weight increased consistently from 28.25 g in DOC to 1080 g on the 75th day. The highest DWG was recorded at 75 days of age at 14.02 g/head/day. The FCR value of 2.31 indicates good feed efficiency for native chickens, with a very low mortality rate of 0.88%. The conclusion of this study is that Sensi native chickens have high potential to be cultivated in an intensive system, both in terms of growth, feed efficiency, and resistance to mortality. These results can be a reference for breeders, researchers, and policy makers in developing superior native chicken cultivation strategies based on scientific data and production efficiency.

Abstract

This study aims to evaluate the effect of adding varying concentrations of sodium hyaluronate (SH) to a Ringer lactate-egg yolk diluent on the quality of native chicken spermatozoa during 48 hours of storage. A completely randomized design (CRD) was used with four treatment groups: control (diluent without SH), diluent with 63 M SH, 103 M SH, and 143 M SH. The study involved six replications with pooled semen from three native chickens aged approximately 10 months. Semen was collected, diluted, and stored at 5C for 48 hours. Parameters observed after storage included motility, viability, plasma membrane integrity, acrosome integrity, DNA damage, and mitochondrial activity. Data were analyzed using analysis of variance (ANOVA). The results indicated that the treatments had no significant effect (P0.05) on any of the spermatozoa quality parameters. The observed values for motility, viability, plasma membrane integrity, acrosome integrity, DNA damage, and mitochondrial activity were 46.67-50%, 95.11-96.55%, 92.77-95.55%, 4.77-5.44%, and 71.05-75.37%, respectively. In conclusion, the addition of sodium hyaluronate at concentrations of 63-143 M in a Ringer lactate-egg yolk diluent does not significantly affect the quality of native chicken spermatozoa during 48 hours of storage at 5C.

Abstract

The aim ofthis study was to examine the preservability of local chicken semen in extender added with various level extra virgin olive oil (EVOO).Semen was collected from three roosters using massage method. Immediately after collection the semen evaluated macro and microscopically. Semen demonstrated   motility, individually divided into five tubes. Each of them diluted with ringer fructose that supplemented with 0%, 4%, 6%, 8% or 10% EVOO. Semen was stored in  refrigerator (5 oC) for sixty hours and evaluated every twelve hours for spermatozoa motility and viability. Results revealed that supplementing the diluent of roosters semen with EVOO and then storing it for different storage periods (, , ,  or  hours) resulted in significant (p .) improvement in spermatooa motility and viabilityin comparison with the control group.Moreover, used of 8% EVOO surpassed other treatments afterin vitro storage for up to 60 hoursand resulted fertilizing ability up to 73.81%.This research conclude that 8% EVOO can be used as antioxidant source for roosters semen during preservation.Keywords : extender, local chicken, extract virgin olive oil, sperma

Abstract

This study aimed to examine the effect of adding different concentrations of sorbitol to the Ringer’s lactate-egg yolk (RLEY) diluent on the quality of Gaga roosters’ semen during cold storage. A completely randomized design was used with 10 replicates and 4 levels of diluent treatment, including control, 1%, 2%, and 3% sorbitol. Semen was collected using a massage method, followed by fresh semen evaluation and dilution with a ratio of 1:5. Semen was stored at a cold temperature (5 ºC) and observed after 0, 24, 48, and 72 hours. The variables observed were semen pH, motility, kinematics, viability, plasma membrane integrity, acrosome integrity, malondialdehyde (MDA), and intracellular calcium ion (Ca2+) concentrations of sperm. All data were analyzed using analysis of variance (ANOVA). Subsequently, the evaluation of sperm mitochondrial activity and DNA damage was carried out, and the data were analyzed using the T-test. The results showed that treatment with sorbitol in the diluent had no effect on semen pH, kinematics (except straightness), acrosomal cap integrity, DNA damage, mitochondrial activity, and MDA concentration. However, sorbitol concentrations significantly affect (p<0.05) total motility, progressive motility, static motility, STR, viability, plasma membrane integrity, and intracellular Ca2+. In conclusion, the addition of 1-3% sorbitol preserves the quality of Gaga roosters’ semen during 72 hours of cold storage.

Abstract

Tujuan kegiatan pengabdian kepada masyarakat ini adalah untuk memberikan pengetahuan dalam mengolah limbah perikanan dan limbah pertanian yang ada di sekitar Pulau Kodingare menjadi pakan ternak dengan teknik fermentasi dan memberikan vitamin pada ternak kambing. Kegiatan dilakukan di Desa Padaelo Pulau Kodingare Kecamatan Pulau Sembilan Kabupaten Sinjai. Pelaksana kegiatan adalah tim Dosen Fakultas Pertanian Universitas Muhammadiyah Sinjai. Kegiatan ini diawali dengan sesi penyuluhan dari narasumber dan dilanjutkan dengan pemberian (injeksi) vitamin B kompleks pada ternak kambing dan pembuatan pakan fermentasi jerami padi. Kegiatan ini menstimulasi dan memberikan semangat kepada masyarakat untuk penerapan dan meningkatkan kesadaran tentang menjaga kesehatan ternak kambing yang dimiliki. Pelatihan pembuatan pakan jerami padi fermentasi diharapkan dapat membuat masyarakat desa Padaelo mampu memanfaatkan limbah berupa jerami padi yang ada di kota Sinjai menjadi pakan ternak. Kegiatan ini juga berdampak pada aspek sosial yaitu seperti terjalinnya rasa partisipasasi, rasa kebersamaan, dan rasa kegontong-royongan dalam penyelesaian masalah yang terkait. Kesimpulan kegiatan ini adalah pengabdian kepada Masyarakat mampu meningkatkan pengetahuan, motivasi dan berdampak baik pada aspek social di Masyarakat Pulau Kodingare, Kabupaten Sinjai.
