Abstract

This study aimed to findthe best combination of glucose concentration with the type of extender to maintain progressive motility and viability of Native Rooster spermatozoa, which stored for 48 hours at 5oC. This study used a completely randomized design (CRD) factorial pattern of 4 x 3. The first treatment was a different type of extender: LE (lactated ringer egg yolk), LS (lactated ringer skim milk), CE (coconut water egg yolk) and CS (coconut water skim milk), while second treatment was glucose concentration (20mM, 50mM, and 80mM).  Native Rooster semen diluted with a ratio of 1:10, thensperm was stored in the refrigerator (5oC). Progressive motility and viability were observed at 0, 24, 48, and 72 hours. The results showed the type of extender treatment had a very significant effect (P 0.01) on the viability of spermatozoa, which were stored 24-hour. The treatment of extender type, glucose concentration, and interaction of extender type with glucose concentration had a very significant effect (P 0.01) on the quality of spermatozoa, which were stored 48 hours and 72 hours. The highest progressive motility of spermatozoa was maintained up to 46.67 ± 2.89% and spermatozoa viability up to 74.68 ± 4.51% in WE 80mM glucose extenders, so the conclusion of this study is the combination of egg yolk coconut water with 80mM glucose is the best extender to maintain progressive motility and viability of spermatozoa of Native Rooster which is stored for 72 hours at 5oC.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh pengaturan suhu dan kelembaban terhadap performa broiler di kandang closed house pada pemeliharaan usia 15-30 hari. Penelitian ini menggunakan 120 ekor broiler strain Cobb 500 berusia 15 hari, yang dipelihara selama 15 hari yang disusun berdasarkan rancangan Rancangan Acak Lengkap (RAL) dengan 3 taraf perlakuan dan 4 kali ulangan. Setiap unit percobaan terdiri dari 10 ekor broiler dengan perlakuan suhu dan kelembaban masing-masing 27 0C dan 88 %, 28 0C dan 87 % dan 29 0C dan 86 %. Parameter yang diamati yaitu performa broiler yang terdiri dari konsumsi pakan, pertambahan bobot badan, feed convertion ratio (FCR), dan mortalitas. Hasil penelitian ini menunjukkan bahwa pengaturan suhu dan kelembaban memberikan pengaruh sangat nyata (P<0.01) terhadap konsumsi pakan, namun tidak berpengaruh nyata (P>0.05) terhadap pertambahan bobot dan Feed Convertion Ratio (FCR), mortalitas broiler selama penelitian sebesar 0.83%.

Abstract

Semen preservation can be done by freezing or cold storage (chilling). The purpose of this study was to determine the effect of the type of semen extender on the quality of Bangkok chicken spermatozoa, and to determine the best type of extender used for Bangkok chicken spermatozoa. This study used a completely randomized design (CRD) with six treatment levels (P1, P2, P3, P4, P5, P6) with four replications. The treatment level used was: P1 = Coconut Water, P2 = Coconut water + Egg Yolk, P3 = Coconut Water + Egg Yolk + Glucose, P4 = Ringer Lactate, P5 = Ringer Lactate + Egg Yolk, P6 = Ringer Lactate + Egg Yolk + Glucose. Parameters observed were motility, viability, and abnormality. The results showed that the type of extender treatment had a very significant effect (P<0.01) on spermatozoa motility, spermatozoa viability, and spermatozoa abnormalities. The conclusion of this study is that the effect of the type of semen extender on the quality of spermatozoa in Bangkok chickens is very significant, especially with the addition of egg yolk components. The best type of extender to use is coconut water extender with the addition of egg yolk.

Abstract

Semen storage at a temperature of 50C affects the quality of spermatozoa. The purpose of this study was to determine the differences in motility and plasma membrane integrity of Kampung chicken spermatozoa variables that were stored directly at 5 °C with storage with a gradual decrease in temperature after the semen collection process. This study used a completely randomized design with 3 treatment levels and 9 replications. The treatment used was storage at a fixed temperature of 5 ⁰C for 75 minutes (P1), storage at a gradually lowered temperature from 41 ⁰C 30 minutes - 21 ⁰C 30 minutes - 5 ⁰C 15 minutes (P2), and storage at a gradually lowered temperature from temperature 41⁰C 15 minutes - 31 ⁰C 15 minutes - 21 ⁰C 15 minutes - 11 ⁰C 15 minutes - 5 ⁰C 15 minutes (P3). Research data that were normally distributed were analyzed using analysis of variance (ANOVA). The results showed that different temperature reduction treatments did not affect (P>0.05) the motility and plasma membrane integrity of spermatozoa variables during storage. The mean total motility, progressive motility and plasma membrane integrity were 25.36-41.22%, 67.30-77.08% and 92.35-96.29%, respectively. The conclusion of this study was that collected Kampung chicken spermatozoa could be directly stored at 5°C without any gradual reduction in temperature because no differences in plasma membrane integrity motility variables were found in the treatment.

Abstract

Penelitian ini bertujuan untuk mengethui pengaruh pemberian pakan yang disuplementasi berbagai konsentrat hijau (lamtoro, gamal dan Indigofera) terhadap kandungan serat kasar feses sapi perah. Sapi perah Fries Holland (FH), sebanyak 12 ekor digunakan dalam penelitian ini. Penelitian menggunakan Rancangan Acak Lengkap (RAL) terdiri dari 3 perlakuan yaitu: pemberian pakan jerami padi fermentasi + lamtoro 30 % (P1), pemberian pakan jerami padi fermentasi + gamal 30 % (P2), pemberian pakan jerami padi fermentasi + indigofera 30 % (P3). Parameter yang diamati yaitu persentase serat kasar pada feses sapi berupa ADF (Acid Detergent Fiber), NDF (Neutral Detergent Fiber), selulosa, hemiselulosa dan lignin. Hasil penelitian menunjukkan bahwa persentase ADF berbeda nyata (P<0.05) antar perlakuan, perlakuan P2 menghasilkan ADF terendah (62.12%). Persentase selulosa berbeda nyata (P<0.05) antar perlakuan, P3 menghasilkan selulosa terendah (24.61%) dan P1 tertinggi (28.13%). Persentase NDF, hemiselulosa dan liginin tidak menghasilkan perbedaan dengan nilai masing-masing 75.90-76.82%, 11.92-13.2% dan17.55-19.59%. Kesimpulan ini adalah suplementasi Indigofera pada jerami fermentasi sebagai pakan mampu mereduksi kandungan selulosa dan meningkatkan ADF pada feses sapi perah jika dibandingkan suplementasi lamtoro dan gamal.

Abstract

This study aims to determine the morphometric comparison and identify abnormal sperm morphology in six types of Indonesian local chickens. Sperm observed came from six types of local chickens: red jungle fowl, Sentul chicken, Gaga’ chicken, Kampung Unggul Balitnak (KUB) chicken, Arab Silver chicken, and Bangkok chicken. Semen was collected by sequencing technique, then fresh semen was blotted on glass slides with eosin-nigrosine dye. Morphometrics and morphology of sperm were observed using a light microscope with a magnification of 100x16. ImageJ software was used for sperm morphometric observation on 60 sperm cells, while morphological observations were carried out by counting normal and abnormal sperm on 200 sperm cells. The results showed that there were variations in the size of sperm among the six types of chickens. Red jungle fowl had a higher total length and tail length than Sentul, Gaga', KUB, Arab, and Bangkok chickens. Observation of morphological characteristics showed that sperm with the highest abnormal morphology were in red jungle fowl and gaga' chickens. Tail bending and mid-piece bending is the most common abnormalities found in Indonesian local chickens.

Abstract

Penelitian ini bertujuan untuk mengetahui performa penetasan dan karakteristik tingkah laku mengeram pada ayam Kampung. Materi penelitian ini adalah ayam Kampung induk sebanyak 5 ekor, dan telur yang akan ditetasi sebanyak 11 butir per indukan. Parameter penelitian ini adalah berat badan induk, berat telur tetas, daya tunas, daya tetas, penyusutan telur, berat DOC, jumlah dan lama turun saat mengeram, suhu pengeraman dan reaksi ayam ketika didekati. Analisis data yang digunakan dalam penelitian ini adalah Analisis Deskriptif. Berdasarkan penelitian yang telah dilakukan maka diperoleh berat badan induk ayam yang mengeram 979.6-1350 g, berat telur tetas 41.9-47.4 g, daya tunas 81.63%, daya tetas 91.06%, penyusutan telur 11.97%, berat DOC 35.35 g, jumlah dan lama turun saat mengeram masing-masing 6.6 kali dan 42.06 menit, suhu pengeraman 37.97-38.19 °C dan reaksi ayam yang mengeram ketika didekati adalah selalu menyerang.

Abstract

Tujuan penelitian ini adalah untuk mengetahui pengaruh penambahan berbagai level tepung rebung terhadap kadar kolesterol dan kandungan nutrisi bakso daging sapi Bali. Rancangan yang digunakan pada penelitian ini adalah rancangan acak lengkap (RAL) pola faktorial dengan 3 kali ulangan. Faktor pertama yaitu level tepung rebung (0%, 10% dan 20%) dan faktor kedua yaitu lama penyimpanan bakso (0, 7 dan 14 hari). Parameter yang diamati yaitu kadar kolesterol, kalium, protein, lemak dan serat kasar. Hasil penelitian menunjukkan bahwa level tepung rebung berpengaruh nyata terhadap kadar kolesterol, kalium, protein, lemak dan serat kasar bakso. Sedangkan lama penyimpanan berpengaruh nyata terhadap kadar kalium, protein, lemak dan serat kasar bakso. Kadar kolesterol terendah (330.26 mg/100 g bakso) terdapat pada perlakuan 20% tepung rebung. Penambahan level tepung rebung meningkatkan kadar kalium, protein, serat kasar dan menurunkan kadar lemak pada bakso. Semakin lama penyimpanan menyebabkan peningkatan kadar kalium dan serat kasar serta  penurunan kadar protein.

Abstract

Tujuan penelitian ini adalah untuk mengetahui pengaruh bangsa sapi potong terhadap respon fisiologis di 3 desa Kecamatan Kajuara Kabupaten Bone. Sapi yang digunakan sebagai sampel penelitian yaitu 10 ekor sapi peranakan Limousin, 10 ekor sapi peranakan Simmental dan 10 ekor sapi Bali. Penelitian ini menggunakan rancangan acak lengkap dengan 3 perlakuan (bangsa sapi) dan 10 kali ulangan (individu sapi). Parameter yang diamati yaitu suhu rektal, frekuensi pernafasan, denyut nadi dan heat tolerance coefficient (HTC). Hasil penelitian menunjukkan bahwa frekuensi pernafasan paling rendah pada sapi Bali (17.1-19.6 kali/menit) dan paling tinggi pada sapi peranakan Limousin (34.7-37.6 kali/menit), frekuensi denyut nadi lebih rendah pada sapi Bali (61.4-62.4 kali/menit) jika dibandingkan peranakan Limousin dan Simental (67.2-68.7 kali/menit) sedangkan suhu rektal hampir tidak ada perbedaan pada ketiga jenis sapi kecuali di Desa Lemo dengan kisaran 37.7-37.93 °C. Nilai HTC pada sapi peranakan Simmental lebih tinggi (2.49-2.63) dibandingkan sapi Bali (1.71-1.83). Kesimpulan penelitian ini adalah jenis sapi potong berpengaruh terhadap frekuensi pernafasan, denyut nadi dan HTC namun relatif tidak berpengaruh terhadap suhu rektal kecuali di Desa Lemo.

Abstract

Indonesia is a tropical country that has two different seasons, namely the rainy season and the dry season. Embryo production can be influenced by several factors, one of which is the season factor. Seasonal factors can affect the quality and quantity of embryos produced. The purpose of this study was to determine the comparison of embryo production of Simmental cattle with Limousin cattle in the rainy season and the dry season. The material used was 40 donor cattle divided into 20 Simmental cattle and 20 Limousin cattle aged 3 to 6 years with a body weight of 450-700 kg. The research procedure starts from superovulation using reproductive hormone injection, artificial insemination, embryo collection using flushing technique and embryo evaluation. The research method used was a case study. Data processing and analysis in this study used the independent t-test with research variables of the number of embryos collected, the quality of embryos produced, the number of corpus luteum produced, the level of response, and the level of recovery. The results of this study were no significant difference (P> 0.05) in the production of Limousin and Simmental cattle embryos in the rainy season. In contrast to the dry season, the results were (P <0.05) which means there is a difference in the production of Limousin and Simmental cattle embryos in the dry season. On the other hand, Limousin cattle showed that Limousin cattle were slightly superior to all variables in the dry season.
