Abstract

This research aims to describe the ecological, socio-economic, institutional, and infrastructural conditions. Primary data collection was carried out through direct observation in the field, measuring the potential of mangrove forests, observing biota, and conducting direct interviews with local communities and relevant stakeholders. Secondary data collection was conducted by gathering documents from previous studies/research, legislation, and other supporting data. Four types of mangroves were found: Rhizophora apiculata, Rhizophora mucronata, Sonneratia alba, and Avicennia marina. The highest species density was Rhizophora apiculata with 6.56 individuals/m², the highest species frequency was 1 for Rhizophora apiculata, the highest species coverage value was Sonneratia alba at 34.02, and the highest Importance Value Index (IVI) was Rhizophora apiculata at point 3 with a value of 226.98. The mangrove diversity index (H') was 2.66, indicating a moderate category and the highest evenness index was at point 2, with a value of 0.92. The Mangrove Tourism Suitability Index (IKW) value was 2.36, indicating a Suitable category. The mangrove area in Pinasungkulan Village can accommodate a 350 square meter mangrove tracking area. The Area Carrying Capacity (DDK) is 56 people per day, with an operational time of 8 working hours per day. The study on community perceptions regarding the benefits of the mangrove ecosystem and its potential to be developed as an ecotourism destination is very positive, and it is expected that this can improve the community's welfare in Pinasungkulan Village. Keywords: ecotourism, mangroves, carrying capacity, suitability, Pinasungkulan Abstrak Penelitian ini bertujuan mendeskripsikan kondisi ekologi, sosial ekonomi, kelembagaan dan infrastruktur. Pengumpulan data primer dilakukan melalui pengamatan langsung (observasi) di lapangan, melalui pengukuran potensi hutan mangrove, pengamatan biota dan wawancara langsung dengan masyarakat lokal dan pihak terkait. Pengumpulan data sekunder dilakukan dengan cara mengumpulkan dokumen hasil studi/penelitian, peraturan perundang-undangan dan data pendukung lainnya. Terdapat 4 jenis mangrove yang ditemukan yaitu Rhizophora apiculata, Rhizophora mucronata, Sonneratia alba dan Avicennia marina. Nilai kerapatan jenis tertinggi adalah Rhizophora apiculata yaitu 6,56 individu/m², frekuensi jenis tertinggi adalah 1 pada jenis Rhizophora apiculata, nilai penutupan jenis tertinggi Sonneratia alba yaitu 34,02, Indeks Nilai Penting (INP) tertinggi jenis Rhizophora apiculata di titik 3 dengan nilai 226,98, indeks keanekaragaman mangrove H’= 2,66 dengan kategori sedang, indeks kemerataan tertinggi pada titik 2 yaitu 0,92. Nilai Indeks Kesesuaian Wisata (IKW) mangrove 2,36, menunjukkan kategori Sesuai. Kawasan mangrove Desa Pinasungkulan dapat dibangun tracking mangrove seluas 350 meter². Daya Dukung Kawasan (DDK) adalah 56 orang/hari dengan waktu operasional 8 jam kerja per hari. Kajian persepsi masyarakat tentang manfaat ekosistem mangrove dan potensinya untuk dikembangkan sebagai tujuan ekowisata sangat baik sehingga diharapkan hal tersebut dapat meningkatkan kesejahteraan masyarakat di Desa Pinasungkulan. Abstract

The specific objectives of this study are: Identifying and carefully explaining the standard of living of fishing families based on women's empowerment/housewives in Tateli Dua Village, Mandolang District, Minahasa Regency. 1. The method used in this study is the census method. 2 Data Collection Techniques Data collected through two sources, namely primary data and secondary data. Primary data is data that is directly obtained through direct interviews at the research location, filling out questionnaires. 3 Data Analysis Methods Analysis of research data results is divided into two types, namely quantitative analysis and qualitative analysis. Engel Index Analysis, because the Engel index is one way to reflect the standard of living of a person or group of people.  The results of the analysis using the Engel Index obtained a figure of 28.52%, this means that the total income of fishermen who own fishing rods is not much, even less than half of their income is used for food needs only. It can be concluded that the level of welfare of fishermen who own fishing rods is classified as good / high because more than half of their income for food needs, namely 71.47%, is used for non-food needs. This means that their lives in the house already have other facilities, such as having a television in addition to children who are well schooled. This is greatly supported by the role of fishermen's wives in working and being able to provide economic support for the family. Keywords: Standard of Living, Women's Empowerment, Hand Fishing Abstrak Tujuan khusus  dalam penelitian ini, yaitu : Mengidentifikasikan dan menjelaskan dengan cermat  taraf hidup keluarga nelayan pancing berbasis pemberdayaan wanita/ibu rumah tangga di Desa Tateli Dua Kecamatan Mandolang Kabupaten Minahasa. 1. Metode dalam penelitian ini dipakai metode sensus. 2 Teknik Pengumpulan Data Data yang dikumpulkan melalui dua sumber yaitu data primer dan data sekunder. Data primer adalah data yang langsung diperoleh melalui wawancara langsung di loakasi penelitian, pengisian kuisioner. 3 Metode Analisis Data Analisis data hasil penelitian dibedakan dalam dua macam, yaitu analisis kuantitatif dan analisis kualitatif. Analisis Indeks Engel, karena indeks Engel merupakan salah satu cara untuk mencerminkan taraf hidup seseorang atau sekelompok orang. Hasil analisis dengan menggunakan Indeks Engel diperoleh angka 28,52 %, ini berarti bahwa total pendapatan nelayan pemilik pancing tidak banyak bahkan tidak sampai separuh  pendapatannya yang digunakan kebutuhan makanan saja.  Hal ini dapat disimpulkan bahwa tingkat kesejahteraan nelayan pemilik pancing  tergolong baik/tinggi  karena lebih dari separuh pendapatannya kebutuhan pangan saja yaitu sebesar 71,47% digunakan untuk kebutuhan non pangan. Ini berarti kehidupan mereka di dalam rumah sudah ada fasilitas yang lain misalnya memiliki televisi disamping anak-anak yang disekolahkan dengan baik. Hal ini sangat ditunjang oleh peranan istri nelayan dalam ikut bekerja dan bisa memberikan dukungan ekonomi bagi keluarga.  Kata kunci : Taraf Hidup, Pemberdayaan Wanita, Pancing Ulur.

Abstract

We used the Nansen Bottle Sampler to collect water samples in the deepsea area, ranging from 150 meters to 175 meters in six water points around Manado Bay, to test the quality of e-DNA water samples to detect target species in the fishing area. Therefore, the basis of the case study method with a sampling technique was carried out on July 29 2023 using Power Water Sterivex Kits, water samples were then stored at -250C and were then taken to TBRC, University of the Ryukyus for further laboratory works, such as; eDNA extract, eDNA quality testing, 1st and 2nd PCR and Electrophoresis eDNA analysis processes using MiFish-U primers with a target of 163–185bp and 375 bp, following the MiFish protocol.  Based on the results of the eDNA extract solution, it is known that the quality of eDNA from the 6 sampling sites locations ranged between 2.8 µg/mL – 4.4 µ g/mL, which means a good quality of eDNA. Moreover, it showed that the presence of DNA fragments at Kappa 60◦C Gelelectrophoresis 1st-PCR, 12S rRNA gene product (163–185bp), and Kappa 60◦C and 65◦C Geklelectrophoresis 2nd-PCR Products according to the target amplicon 375 bp. This means we can conduct the next step, the PCR sequence analysis.  Then, eDNA quality testing, 1st and 2nd PCR, and Electrophoresis of e-DNA analysis process were using MiFish-U F/R primers with a target of 375 bp, it is known that the concentration of Nanodrop from the 6 sampling locations ranges between 2.8 µg/mL – 4.4 µg/mL while the core or quality eDNA ranged from 1.56 µg/mL – 2.50 µg/mL. Based on identification results, five types of species were detected; Myctophum lychnobium, Selar crumenophthalmus, Photonectes sp., Oreochromis sp. Thunnus obesus and Homo sapiens were generated using eDNA metabarcoding on the mitochondria genome database MitoFish. Keywords: e-DNA, eDNA quality, species target, fishing area, Manado Bay   Abstrak Kami menggunakan Nansen Bottle Sampler untuk mengambil sampel air pada laut dalam berkisar 150meter sampai 175 meter di enam titik perairan Sekitar Teluk Manado, untuk menguji Kualitas e-DNA sample air yang digunakan untuk mendeteksi target spesies pada daerah penangkapan. Selanjutnya dasar metode studi kasus dengan teknik pengambilan sampel secara sampling dilakukan pada tanggal 29 Juli 2023 menggunakan Power Water Sterivex Kits, sapel air disimpan pada -250C yang selanjutnya dibawa ke TBRC, University of the Ryukyus untuk pnelitian laboratorium lanjutan seperti ekstrak eDNA, Pengujian kualitas eDNA, 1st and 2nd PCR dan Elektrophoresis proses analisis eDNA menggunakan primer MiFish-U dengan target 375 bp, mengikuti MiFish protokol. Berdasarkan hasil pengujian larutan ekstrak eDNA diketahui bahwa kualitas eDNA dari 6 titik lokasi sampling berkisar antara 2.8 ng/mL – 4.4 ng/mL dan menunjukkan adanya fragment DNA pada Kappa 60◦C Geklelectrophoresis 1st-PCR Produk 12S rRNA gene (163–185bp, dan Kappa 60◦C dan 65◦C Geklelectrophoresis 2nd-PCR Produk sesuai amplikon target 375 bp. Hal ini berarti dapat dilanjutkan pada tahap analisis sekuens PCR. Pengujian kualitas eDNA, 1st and 2nd PCR dan Elektrophoresis proses analisis eDNA menggunakan primer MiFish-U F/R dengan target 375 bp, diketahui bahwa konsentrasi Nanodrop dari 6 titik lokasi sampling berkisar antara 2.8 µg/mL – 4.4 µg/mL sedangkan kemurnian atau kualitas eDNA berkisar antara 1.56 µg/mL – 2.50 µg/mL. Hasil identifikasi menyatakan lima jenis spesies terdeteksi; Myctophum lychnobium, Selar crumenophthalmus, Photonectes sp., Oreochromis sp. Thunnus obesus, Homo sapiens dihasilkan dengan menggunakan eDNA metabarcoding pada MitoFish database genom mitokondria. Abstract

Coastal reclamation in Sario District since 1995 has affected the socioeconomic conditions of coastal communities, particularly handline fishermen. This study aims to identify the socioeconomic conditions of fishermen, analyze internal and external factors using PESTEL and SWOT analyses, and determine appropriate adaptation strategies. The research was conducted from August to November 2025 in three villages within Sario District using a descriptive qualitative and quantitative approach. Results show that fishermen are mostly of productive age with low education levels. Declining income due to distant fishing grounds has driven diversification into side jobs. The PESTEL analysis revealed impacts of reclamation, rising costs, and weak marketing, while SWOT placed fishermen in an aggressive strategy position with strong internal capacity but serious external threats. Adaptive strategies involve pursuing side jobs while preserving traditional fishing livelihoods. Keywords: adaptation strategy; traditional fishermen; coastal reclamation; handline fishing; PESTEL analysis; SWOT analysis Abstrak Reklamasi pantai di Kecamatan Sario sejak 1995 berdampak pada kondisi sosial ekonomi masyarakat pesisir, terutama nelayan pancing ulur. Penelitian ini bertujuan mengidentifikasi kondisi sosial ekonomi nelayan, menganalisis faktor internal dan eksternal menggunakan PESTEL dan SWOT, serta menentukan strategi adaptasi yang tepat. Penelitian dilakukan selama Agustus–November 2025 di tiga kelurahan di Kecamatan Sario dengan pendekatan deskriptif kualitatif dan kuantitatif. Hasil menunjukkan nelayan didominasi usia produktif dan berpendidikan rendah. Penurunan pendapatan akibat jauhnya area tangkap mendorong diversifikasi pekerjaan. Analisis PESTEL mengungkap dampak reklamasi, biaya tinggi, dan lemahnya pemasaran, sementara SWOT menempatkan nelayan pada strategi agresif dengan kekuatan internal baik namun menghadapi ancaman eksternal. Strategi adaptasi dilakukan melalui pekerjaan sampingan tanpa meninggalkan profesi tradisional.
