Abstract

Background: Candida albicans, a fungal normal flora that causes Denture stomatitis. Sargassum polycystum is a brown algae species (Phaeophyceae) which grows almost along the coasts of Indonesia islands, known to have antifungal and antibacterial activities. Purpose: To determine the inhibition of brown algae extract (Sargassum polycystum)’s on the growth of Candida albicans and determine whether Sargassum polycystum extract works as denture cleanser material through organoleptic test. Methods: The sample was Candida albicans equal to 0,5 McFarland (108 CFU/ml). The experiment groups comprises negative control group (K-) with 1% DMSO, positive control group (K+) with 0,2% chlorhexidine gluconate, and treatment groups 1-4 (P1;P2;P3;P4) consecutively with 2,5%; 5%; 10%; 20% Sargassum polycystum ethanol extract macerated for 15 minutes in 5 (Ø 6mm) paper discs. SDA medium was incubated for 24 hours and the visible transparent zone was calculated. Organoleptic test was conducted by asking 10 panelists about the observation on color, smell, and homogeneity level of brown algae extract (Sargassum polycystum) with the concentrations of 2,5%; 5%; 10%; and 20%. The experiment result was analyzed with the Kruskal-Wallis test and the Mann-Whitney test. Results: Experiment material’s inhibitory effect was undetectable as Sargassum polycystum ethanol extract against 1% DMSO solvent’s solubility was not homogeneous. The organoleptic test result showed that color, smell, and homogeneity level of brown algae extract (Sargassum polycystum) was not compliant with antiseptic standards. Conclusion: No inhibitory effect in brown algae extract (Sargassum polycystum) as the organoleptic test proved the antiseptic standards were not fulfilled.

Abstract

Introduction: Gold sea cucumber (Stichopus hermanii) is a marine organism that contains omega-3 and glycosaminoglycan for enhancing collagen needed in the healing process. This study was aimed to determine the effectiveness of gold sea cucumber extracts in accelerating the healing process of oral traumatic ulcer in Wistar rats, in terms of the correlation between collagen type I expression, ulcer diameter, and body weight loss. Methods: Twenty Wistar rats (Rattus norvegicus) was divided into four groups and treated with gold sea cucumber extracts with the concentration of 80%, 40%, and 20% in the form of 0.1 ml gel daily. The negative control group received no treatment. Ulceration of the lower lip of labial mucosa was induced by a burnisher. The animals were observed for 7 days, during which they were weighed and the ulcers diameter was measured. The rats were then sacrificed after 7 days of treatment. Histometric analysis of collagen type I expression was also performed. Data obtained were analysed for differences between the group with one-way ANOVA test and the correlation was analysed with the Pearson test. Results: The significant differences (p < 0.05) between treatment groups was found in the ulcer diameters and the collagen type I expression. The most decreased ulcer diameter and the highest collagen type I expression detected in the group of treatment with a concentration of 80% — no significant differences found in the weight loss between-groups (p > 0.05). Strong correlation was also found between the expression of collagen type I and diameter of ulcer (Pearson correlation = 0.599, p = 0.05), however, no correlation found between weight loss with the expression of collagen type I (Pearson correlation = -0.102, p = 0.66), and the ulcer diameter (Pearson correlation = 0.022, p = 0.92). Conclusion: Gold sea cucumber had a potential benefit to be used as a traumatic ulcer healing medicament. Water extract of gold sea cucumber able to accelerate traumatic ulcer healing by enhancing the production of collagen type I and reduce the diameter of ulcer. There is a strong correlation between collagen type 1 expression and diameter of ulcer, and a weak correlation found between body weight loss and collagen type I expression, also with reducing ulcer diameter. Keywords: Gold sea cucumber, Stichopus hermanii, healing process, traumatic ulcer.

Abstract

Background: Calcium hydroxide is a material that is widely used in pulp capping treatment. However, the use of this material causes tunnel defects due to the large absorption of calcium. This can be manipulated by creating an encapsulation formula. Stichopus hermanii is a marine biota that contains lots of hyaluronic acid and can be used as an encapsulation material which also plays a role in cell regeneration. The characteristics of the microencapsulated calcium hydroxide preparation with Stichopus hermanii as pulp capping material through an in vitro study.Method: Stichopus hermanii was made freeze dry and dissolved with 1% acetic acid to obtain a gel concentration of 0.2%, 0.4%, and 0.8%. Next, calcium hydroxide powder is added with a ratio of 1: 6 and tripolyphospate (TPP) is added as a cross-linker agent and the mixture is stirred until homogeneous. The test material formed was subjected to SEM, PSA and cytotoxicity tests.Result: The cytotoxicity test showed that all of the test materials (0.2%; 0.4%; 0.8%) were not toxic, especially at a concentration of 0.2%. The PSA test results indicate that this encapsulation shows the micro particle size formed. The SEM test shows a rough and stringy surface shape, where at a concentration of 0.2%, a smoother morphological formation is found. Conclusion: The in vitro study showed that the microencapsulation of calcium hydroxide with Stichopus hermanii as a pulp capping material showed the characteristics of the non-toxic material, the micro size and the rough and fibrous surface shape.

Abstract

Background: Biphasic calcium phosphate (BCP) is graft material contained HA and TCP. Tegilarca granosa shell is a natural source that may converted into BCP. This study aims to determine the composition and cytotoxicity of BCP synthesized from Tegilarca granosa shell used various hydrothermal hours and to evaluate the mechanical properties of BCP scaffold. Methods: Tegilarca granosa shell was converted into BCP using hydrothermal method at 200˚C for 6h (Group 1); 9h (Group 2); and 12h (Group 3). The composition was determined by XRD and the cell viability were evaluated using MTT Assays. Each group was added with 20% gelatin ratio 50:50 (w/v) and freeze-dried to form scaffold. Scaffolds (Ø6mm x 4mm) were prepared for diametral tensile strength (DTS) test (n=6) and scaffolds (Ø7mm x 11mm) were used for compressive strength (CS) test (n=6). All data were analyzed using Kruskall-Wallis followed by Mann-Whitney test. Results: The composition of BCP (HA/ TCP) at Group 1, Group 2, and Group 3 were 81.80%/14,10%; 87%/6%; and 72%/21%. The cell viabilities were good for all groups. The DTS and CS test showed there was a significant difference between Group 1 and Group 3 scaffold, meanwhile there was no significant differences between Group 2 and Group 3 scaffold. Group 3 scaffold showed the highest DTS and CS, 6.921 MPa and 1,233 MPa. Conclusion: The BCP composition were depent on hydrothermal hours. Although all scaffold groups were non-toxic, but BCP scaffold synthesized from Tegilarca granosa shell using hydrothermal for 12 hours showed the highest mechanical properties.
