Abstract

The most common skin disorder is acne (Acne vulgaris) which is a chronic inflammatory disease that occurs in the pilosebaceous unit. Infection can be caused by Propionibacterium acnes and Staphylococcus aureus. Moringa leaves are known to inhibit acne growth. Piper crocatum leaves are also thought to have antibacterial activity. The study aims to determine the effect of the combination of the two extracts in inhibiting the growth of acne-causing bacteria, also tested the gel formula to meet physical and chemical parameters as well as stability to temperature and storage, also to test the acute dermal irritation against rabbit. Each extract with a concentration of 0.3%; 0.6%; 1.25%; 2.5%; 5%; 10%; 20%, incubated then measured the inhibition zone, then determined the minimum inhibition zone concentration of the two extracts to determine the lowest levels of the extracts which still gave antibacterial activity against the tested bacteria by the well diffusion method. The extract combination was formulated in a gel preparation with the excipients Carbopol 940, PG, Phenoxyethanol, TEA, CMC, and Aquadest. Antibacterial activity test for gel preparations used the well method with positive control Medi-Klin® gel. Then the formula evaluation includes physical, chemical, and microbiological evaluations. The results showed that the combination gel form had activity against P. acne and S. aureus at concentrations of 2.5% : 2.5%. The combination gel preparation of the two extracts also has synergistic effect in inhibiting acne bacteria. The formula can meet physical and chemical parameters and is stable to temperature and storage.

Abstract

Almost all diseases in the body cause pain. Currently the most widely used analgetic has not regardless from side effects. The development of new and effective pain medications that are natural is expected to have the benefits of treatment. 75% ethanol extract of dandang gendis leaf (Clinacanthus nutans (Burn F) Lindau) dosage 279,3 mg/kg BW and 96% ethanol fraction of bakung leaf (Crinum asiaticum) dosage 250 mg/kg BW were shown to have antinociceptive activity. The purpose of this study was to determine the antinociceptive effectiveness of a combination of 75% ethanol extract from dandang gendis leaves and 96% ethanol fraction of bakung leaf. In this study was used chemical method induction (Sigmund's method) in 25 male mice which were divided into 5 treatment groups, namely treatment group I as negative control was given CMC 0.5%, treatment group II as positive control was given acetosal 100 mg / kg BW, treatment groups III, IV and V were given combinationsextract. Each group was given treatment orally, thirty minutes then induced with 0.6% acetic acid intraperitonially, after which it was observed and counted for the amount of stretching for sixty minutes. Data were analyzed by one-way ANOVA to see differences in antinociceptive effects in each treatment group. The results of this study showed that all treatment groups had a significant difference (p <0.05), the combination of 75% ethanol extract dandang gendis leaf and 96% ethanol fraction of bakung leaf with a dose of 100 mg / kg BW had good effectiveness with a percent effectiveness value of 92.50%. The results of this study indicate that the combination of 75% ethanol extract from dandang gendis leaf (Clinacanthus nutans (Burn F) Lindau) and 96% ethanol fraction from bakung leaf (Crinum asiaticum) with the same concentration ratio, have relatively higher antinociceptiveof the activity of each extract, in other words both of them work together.

Abstract

Arbenan leaf is a medicinal plant that used in the healing of inflammation. The content of phenolic compounds, flavonoids, steroids and tannins contained in arbenan plants which play an important role in alternative medicine. Currently, scientific work of phytotherapy is an important alternative route in finding effective natural medicines with minimal side effects. The aim of this study was to determine the phytochemical properties and to determine the anti-inflammatory effect of arbenan leaf [Duchesnea indica (Jacks.) Focke] extract. In this study, nine male Wistar rats were tested in three treatment groups. Group I Na CMC, group II Na diclofenac and group III arbenan leaf extract. Inflammation in rats by inducing 1% carrageenan as much as 0.10 mL. The volume of edema per hour is known from the difference in the volume of the foot at certain hours with the volume of the normal foot. The AUC value of edema volume was calculated by the trapezoid method every one hour and the percen of anti-inflammatory power was calculated. The statistical analysis of the Least Significance Different (LSD) AUC value from the edema volume data showed that the two treatment groups, namely the Na diclofenac and the arbenan leaf extract group, were significantly different from the Na CMC group (P <0.05). So that both treatment groups have anti-inflammatory effects. The results showed that the ethanolik extract of arbenan leaves contained phenolic compounds, flavonoids and tannins. Then the ethanolic extract of arbenan leaves has anti-inflammatory activity of 0.31%.

Abstract

Nyirih is a plant from the Meliaceae family with the Latin name Xylocarpus granatum J.Koeing, this nyirih spreads in tropical waters and does not cluster in certain areas. This study aims to determine the total phenolic content of the stem bark extract of nyirih (Xylocarpus granatum J.Koeing) using the UV-Vis spectrophotometric method. The stem bark extract of nyirih (Xylocarpus granatum J. Koeing) was obtained with maceration method using 96% ethanol solvent. Determination of total phenolic content based on the Folin-Ciocalteau method, where the addition of 7% sodium carbonate provides an alkaline atmosphere that will produce phenolic ions. Furthermore, it will reduce Follin to form a blue complex. At a maximum wavelength of 755 nm, the results showed that the average total phenolic content of the stem bark extract of nyirih (Xylocarpus granatum J. Koeing) was 6.163 mg GAE / g extract.

Abstract

Bawang Dayak (Eleutherine palmifolia L. Merr) dalam bentuk segar maupun ekstrak telah dipergunakan secara tradisional oleh masyarakat untuk pengobatan berbagai penyakit. Saat ini sediaan ekstrak etanol bawang Dayak dalam bentuk tablet effervescent belum tersedia. Berdasarkan hal tersebut maka tujuan penelitian adalah formulasi dan karakterisasi tablet effervescent dari ekstrak etanol bawang Dayak. Ekstraksi bawang Dayak dilakukan dengan metode maserasi menggunakan etanol 96%. Ekstrak etanol yang dihasilkan diuji toksisitas dengan metode Brine Shrimp Lethality Test dan karakterisasi fitokimia. Pembuatan tablet effervescent menggunakan metode granulasi basah dengan variasi konsentrasi polivinilpirolidon (0%, 1%, 2%, 3%, dan 4%). Karakterisasi tablet effervescent meliputi penampilan, keseragaman ukuran, keseragaman bobot, kekerasan, dan waktu melarut. Hasil penelitian menunjukan bahwa rendemen estrak etanol yang dihasilkan adalah 8,45%. Ekstrak etanol bawang Dayak tidak bersifat toksik dengan LC50 sebesar 1.241 ppm. Uji fitokimia dari esktrak etanol memberikan hasil positif pada flavonoid, alkaloid, polifenol, dan tannin. Hasil uji keseragaman ukuran dan uji keseragaman bobot tablet effervescent tidak berbeda nyata. Hasil uji kekerasan dan uji waktu melarut tablet effervescent menunjukkan bahwa variasi konsentrasi PVP berpengaruh terhadap kekerasan dan waktu melarut tablet. Tablet effervescent dengan waktu melarut paling cepat diperoleh dengan menggunakan 1% PVP.

Abstract

Epidermal growth factor receptor (EGFR) is one of tyrosine kinase that overexpressed in many types of cancer. The ihbiition of EGFR activity has known as a rational target for cancer therapy. However,  several EGFR inhibitors have limitations related to resistance problems or reduced tumor response. Therefore, the effort to develop new inhibitors with greater efficacy is very important to overcome the problems. In the present study, we designed a computational method by combining ligand based pharmacophore modeling and molecular docking for screening the potential natural compounds as EGFR inhibitors. Validation result of ligand based pharmacophore modeling and docking molecular by ROC curves showed that both of these protocols had AUC ≥ 0.7. The Redocking erlotinib to EGFR in our docking protocols gave the RMSD < 2 Å. Therefore, our computational method could be applied for virtual screening. Initial screening based on pharmacophore modeling obtained 86 out of 179.816 compounds that could be mapped with the pharmacophore features.  Further screening through molecular docking showed two compounds had a lower docking score than erlotinib, i.e ZINC00941342 and ZINC72325782. However, by careful analysis of their interaction profiles, only ZINC00941342 which formed hydrogen bonds with Met769, which is known as an EGFR key residue. Additionally, it was also found to form hydrogen with Cys773. Therefore, we recommend ZINC00941342 as a potential EGFR inhibitor.

Abstract

Sembukan leaf is a medicinal plant that is widely used by the community, one of which is as an anti-inflammatory. This study aims to determine the effect of giving sembukan leaves (Paederia foetida L.) to decrease uric acid levels in male white rats (Rattus norvegicus). The study was carried out by testing 5 groups of white rats, namely positive control (Allopurinol 100 mg), negative control (Na.CMC suspension 0.5%), the dose of ethanol extract of sembukan leaves 10 mg/KgBB, 20 mg/KgBB, and 40 mg/KgBB. Each treatment group was measured initial uric acid levels (before induction), after induction of chicken liver juice and after giving treatment or test samples using test strips. The results showed that uric acid levels in all groups decreased with the percentage decrease in uric acid levels at a dose of 10 mg/Kg BB, 20 mg/Kg BB and 40 mg/Kg BB, respectively by 13.2%, 24.5% and 54.3%. The results of the One Way Anova statistical test showed a significant difference with P value = 0.010 (α <0.05). The concentration of the extract that gave the greatest reducing effect was the extract dose of 40mg/kgBB.

Abstract

 Inhibition of the α-glucosidase can be used as antidiabetic therapy because it can reduce postprandial blood sugar levels. The stem bark of mahogany (Swietenia macrophylla King.) have properties as inhibitors of enzyme α-glucosidase. The purpose of this study was to determine the active compound of mahogany stem bark against α-glucosidase enzyme in vitro and in silico. 96% ethanol extract of mahogany stem bark was partitioned with n-hexane, ethyl acetate and water. Testing of inhibition of α-glucosidase enzyme work done with substrate p-nitrophenyl-α-D-glucopyranoside. The water fraction was separated on silica gel column chromatography (SiO2 : dichloromethane-methanol = 10 : 1 ~ 1 : 1 and methanol). Molecular docking using AutoDock 4.2.6 was performed to predict the binding modes of α-glucosidase enzyme with chemical compound from water subfraction of stem bark mahogany. The results showed that water fraction of stem bark mahogany have antidiabetic activity with IC50 value of 1.51 µg/mL. Fraction Fr-1 gave an inhibition of 83.61 ± 1.11 %. Fr-1 contains chemical compound catechin, evodionol, swietenitin E, 11-deoxyswienialide D, swietenitin J and H. In silico study showed that catechin and evodionol free energy value (ΔG) -6.98 and -6.25 Kcal/mol and inhibition constanta (Ki) 7.61 and 26.18 µM.

Abstract

This study aims to determine and compare the phenolic and flavonoid content of methanol extract in Acacia rugata (Lam.) Fawc. Rendle leaves using UV-Vis spechtrophotometry and TLC-densitometry. The Acacia rugata (Lam.) Fawc. Rendle extracted by using maceration applied sonicator. The quantitative assay measured by spectrophotometer UV-Vis at wavelength 430 nm. The TLC–densitometry performed at wavelength of 254 nm and quercetin was as a standard. The results showed that the phenolic total showed 0.506 gGAE/g extract for spectrophotometer Uv-Vis and 0.879 gGAE/g extract for TLC-Densitometric. The flavonoid content reported 0.44 gQE/g extract and 0.73 gQE/mg extract for spectrophotometer UV-Vis and TLC Densitometric methods, respectively.

Abstract

Kersen parasite leaves (Dendropthoe pentandra L. Miq) is a type of parasite that is hemiparasitic and contains active compounds, namely, flavonoids, terpenoids and tannins. This compound has benefits as an antihyperglicemic. This study aims to determine the effect of lowering blood glucose levels from the infusion of Kersen parasite leaves (Dendrophthoe pentandra L. Miq) in male rats induced by alloxan. This research is a laboratory experimental study with a randomized preposttest one group design. The study used alloxan-induced male rats which were divided into 3 dosage groups (38 mg / kg, 77 mg / kg, and 154 mg / kg). In the results of the mean blood glucose levels of rats, there was a decrease in blood glucose levels on the 0th day of treatment, on the 7th day after the treatment there was an increase in blood glucose levels and on the 14th day after the treatment there was a decrease in blood glucose levels except in the dose II group and III. In this study, Kersen parasite leaf infusion had no effect on reducing blood glucose levels of alloxan-induced male white rats.
