Abstract

Cancer is a non-communicable disease that causes morbidity and mortality in all regions of the world. Cancer treatment is important in reducing the high number of deaths (mortality) due to cancer sufferers. One of the cancer treatments is using chemotherapy drug cisplatin. However, cisplatin drug has side effects, namely nephrotoxicity, ototoxicity, neurotoxicity, hemotoxicity, cardiotoxicity, and hepatotoxicity. Therefore, the combination of cisplatin in albumin nanoparticles and modification of the bond using folic acid as an alternative solution minimizes the resulting toxic effects. This study aimed to determine the toxicity effect of the cisplatin drug formulation with a combination of bovine serum albumin (BSA) nanoparticles and folic acid through the Brine Shrimp Lethality Test (BSLT) method. The research methods used were BSA nanoparticle synthesis (NP-BSA), NP-BSA Synthesis with Cisplatin Drug (CP-NP-BSA), Folic Acid Synthesis with CP-NP-BSA Combination (As-CP-NP-BSA). Then, a toxicity test was carried out using the BSLT method to determine the toxicity level of BSA nanoparticles and the effect of adding folic acid in the cisplatin drug in the nanoparticles. The results showed that the absorbance of the 358 nm As-CP-NP-BSA UV-Vis spectrophotometer was 0.86. The toxicity level of the NP-BSA with the LC50 value was 69.23 ppm while the As-CP-NP-BSA LC50 toxicity level was 56.56 ppm. Nanoparticles consisting of a combination of bovine serum albumin, cisplatin and folic acid can be used as candidates for anticancer drugs

Abstract

Seborrheic dermatitis (SD) occurs due to excessive sebum production resulting in the growth of the fungus Mellasezia sp. on the skin becomes uncontrollable. Some of the receptors that play a role in SD regulation are Low Density Lipoprotein (LDL) protein, Lanasterol 14-α Demetylase enzyme, and Lipase enzyme. This study aims to determine the potential compounds of the endophytic fungi Trichoderma sp. as an antiseborrheic and knowing the interaction between ligands as inhibitors in SD regulation. The research was conducted with computer-based molecular docking. The results showed that there were two inhibitor candidate compounds based on the selection by binding affinity value, namely Chloromycetin and Benzyl benzoate. Based on the results of docking with VINA autodock, the potential compounds were Benzyl benzoate on LDL protein with binding affinity values of -7.3 kcal/mol, compounds were Chloromycethin on Lipase enzyme with binding affinity values of -7.3 kcal/mol and Lanasterol 14-α demethylase enzyme with a binding affinity value of -7.6 kcal/mol. These compounds form hydrogen bonds and hydrophobic bonds. Based on the prediction of Lipinski's rule of five, the two compounds are effective orally used as anti seborrheic dermatitis drugs

Abstract

Toraja arabica coffee is well known for its higher price and quality compared to robusta coffee, but its commercial is often added with other ingredients, including robusta coffee which has low economic value. Visual inspection is unreliable in roasted ground coffee due to the resemblance of its chemical content. The ATR-FTIR method tandem PCA was able to provide an overview of the typical chemical content of the coffee preparation. The purpose of this study was to evaluate the robusta coffee as adulterant in Toraja arabica coffee preparation by using ATR-FTIR. Toraja arabica coffee beans were obtained from three smallholder plantations around Toraja and Robusta coffee beans were obtained from Toraja, Lampung, and West Java coffee plantations. The coffee beans were roasted and then macerated using 96% ethanol for 3×24 hours and concentrated using a rotary evaporator until being thick. The IR spectrum of each extract was measured using the ATR-FTIR spectroscopy at a range of 4000-650 cm-1. The results show there is a similarity in the IR spectrum patterns and there is only a small difference in the transmittance of Toraja arabica coffee and robusta coffee. Furthermore, the IR spectrum is clustered by using PCA in R program. The projection of three commercial samples shows that samples 1 and 2 do not contain robusta coffee while sample 3 shows the presence of robusta coffee. In conclusion, the ATR-FTIR spectroscopic method tandem PCA was able to clustered the presence or absence of robusta coffee content in the Toraja arabica coffee.

Abstract

Depression is a psychiatric disorder characterized by decreased mood and affects daily activities. The development of herbal ingredients as complementary therapy becomes increasing in line with the number of patients who avoid side effects in conventional treatment. The purpose of this study is to investigate the potential effects of ethanolic extract of Ocimum sanctumL. as an antidepressant as well as evaluate the potential mechanism of action in the monoaminergic system in invivo study. For evaluation of mechanisms of action, mice were treated serotonin synthesis inhibitors (pCPA), non-selective dopamine antagonists (Haloperidol) and selective antagonists D2 dopamine (Sulpiride) as pre-treatment.The Tail Suspension Test(TST)method is used as a standardized test that has generally been used to evaluate the antidepressant effects of a drug compound. The results showed that the ethanolic extract of OcimumsanctumL. has antidepressant effect at the dose of 150 mg/kg.Ethanol extract of OcimumsanctumL. was able to decrease the immobility time in TST mice model significantly when given pre-treatmentwith pCPA (p<0.05) and haloperidol (p<0.05), while sulpiride (p<0.05) inhibits the antidepressant effects of ethanolic extract of OcimumsanctumL. significantly. These results showed that the monoaminergic system especially serotonergic and dopaminergic were involved in the antidepressant-like effects of ethanolic extract of OcimumsanctumL. by involving dopamine receptors and has a role in serotonin synthesis.
