Abstract

This study was purposed to inquire the activities of benzopyran (chromones) and benzofuranones (coumaranones) derivatives against Steinernema feltiae (S. feltiae). The toxicity assay against S. feltiae showed that benzopyran derivatives 2, 3, 4, 6, and 9 have the highest activity on S. feltiae with viabilities percentage of 50%. The compound 9 demostrated the highest activity with LD 50 and LD values, 7.2 and 52.2 M, respectively. The activities of compound 7 and 10 showed the lowest toxicity. Interestingly, the activity of benzofuranone derivatives showed significant activities against S. feltiae. Compare to benzopyran derivatives, the benzofuranone derivatives has the highest toxicity, in particular compound 13 with LD 5.45 M. The nematicidal assay showed that benzofuranones (coumaranones) derivatives revealed higher activities than benzopyran (chromones) derivatives.Key words: chromones, benzopyran, coumaranones, benzofuranone, and Steinernema feltiae

Abstract

Penelitian ini bertujuan merancang kandidat probe dari urutan 529 bp (R529) yang terdapat 200-300 kopi dalam genom Toxoplasma gondii (T. gondii). Untuk menguji spesifitas rancangan probe digunakan perangkat lunak basic local alignment search tool (BLASTN) dari national center for biotechnology information (NCBI) dan Geniuous. Probe DNA sebesar 237 bp dilabel yang dengan digoksigenin disintesis dengan polymerase chain reaction (PCR). Kandidat probe yang dihasilkan berpotensi untuk digunakan lebih lanjut dalam deteksi keberadaan asam nukleat T. gondii pada berbagai teknik hibridisasi.

Abstract

This study aimed to analyze the muscle microstructure of longissimus dorsi Brahman cross (BX) cattle slaughtered at the slaughterhouse of North Aceh District. The meat samples for the study were taken from three male BX cattle aged 18-30 months with body weights ranging from 400 kg to 500 kg and the average body condition scores (BCS) of 3-4. The muscle used was the longissimus dorsi muscle. The mean (SE) diameter and cross-sectional area (CSA) of muscle fibers and the percentage of fast fibers in the longissimus dorsi BX muscle were 71.691.23 m, 4263.43305.69 m2 , and 48.178.81%, respectively. This study concluded that the longissimus dorsi muscle of the BX has relatively large muscle fibers, some of which in each fascicle are composed of fast muscle (fast fiber/type II).

Abstract

The aim of this study was to observe cortisol levels and liver histopathology of broiler chicken that were treated with black cumin oil (BCO) under heat stress. A total of 15 broiler chickens were used in this study and divided into 5 groups, K- (without treatment), K+ (given heat stress), P1 (given heat stress and 0.56mL BCO/400kg body weight, P2 (given heat stress and 1.11 mL BCO/400 g body weight), and P3 (given heat stress and 2.22 mL BCO/400 g body weight). Heat stress was given for 5 hours with temperature range of 34-35 C for 7 days. Cortisol was measured using the cortisol enzyme-linked immunosorbent assay (ELISA) kit. Liver histopathology was stained with hematoxylin eosin and observed with electron microscope. The data were analyzed using one way analysis of variance (ANOVA). This study found that application of heat stress to broiler chickens increased cortisol levels and induced histopathological changes in the liver. The BCO administration reduced cortisol level significantly (P0.05) in heat-stressed broilers. BCO administration also significantly reduced (P0.05) the degenerative changes in liver histopathology such as fat degeneration, hemorrhage and necrosis in broiler chickens under heat stress, but did not significantly influence the inflammatory cells infiltration. As conclusion, BCO administration to broiler chickens under heat stress can reduce cortisol levels and minimize histopathological changes in the liver.

Abstract

This study aimed to determine the ability of highly pathogenic avian influenza virus (HPAI) virus subtype H5N1 originated from Indonesia to induce apoptosis in Madin-Darby Canine Kidney (MDCK) cells. Three HPAI virus subtype H5N1 isolates with different genetic characteristic namely A/Bird/Bali1/2011, A/Chicken/East Java/BwiI2/2010 and A/Chicken/West Java/1074/2003, were cultured in MDCK cells. Apoptosis was identified by deoxyribonucleic acid (DNA) fragmentation of infected MDCK cells using Apoptotic DNA Ladder Kit. The results showed that all three HPAI virus isolates used in this study did not able to induce apoptosis in the MDCK cells within 5 to 72 hours post infection.

Abstract

This study aimed to determine the effect of black cumin (Nigella sativa) supplementation through drinking water on the histology of broiler chickens small intestine and large intestine. The research was conducted from April-September 2020 in a cage facility of the Integrated Field Laboratory, Faculty of Agriculture, University of Lampung. This research used a completely randomized design with four treatment groups and three replications (five broilers per replication) with a total of 60 male broilers. The treatments were drinking water without Black Cumin (P0, control); drinking water with Black Cumin 36 mg/kg BW/day (P1); 72 mg/kg BW/day (P2); and 144 mg/kg BW/day (P3). Three broilers from each group were randomly necropsied at 31 days old, and samples of the small intestine (duodenum, jejunum, and ileum) and large intestine were fixed with 10% formalin solution and sent to the Lampung Disease Investigation Center for histological preparations. The observation of preparations was carried out microscopically using the Leica DM500 Binocular Microscope to accurately calculate various parameter sizes. The results were analyzed statistically with one way Analysis of Variance at significant level 5% and if proven significant, then a Tukey test was conducted. The results of this study were that the supplementation of black cumin 72 mg/kg BW/day through drinking water could significantly increase (P0.05) 1) the average sizes of villi height and villi area of small intestine (duodenum, jejunum, ileum) and large intestine (colon) of broiler chickens; 2) the average sizes of basal villi width, villi apex width, and gland diameter of broiler duodenum; and 3) the average size of apex villi width of broiler colon. The conclusion of this study was the supplementation of black cumin at dose of 72 mg/kg BW/day through drinking water could increase the histological sizes of the small intestine and large intestine of broilers.

Abstract

This study aimed to determine whether the variant or biotype of Trypanosoma evansi can be seen from their polypeptide profiles using 12%sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE) stained with Brilliant Blue Commasie. The results generally showed thatthe molecular weight (MW) of polypeptides from nine isolates from East Java, Central Java, Banten, South Kalimantan, Central Kalimantan, andLampung provinces were in the range of 85.46 to 15.76 kD and each isolate has different polypeptide profile. Isolates A13 and A14 were isolatedfrom the same place but have different polypeptide profiles. Likewise, isolates S13 and S18 also have different polypeptide profiles despite beingisolated from the same place at the same time. On the other hand, isolate 372, 87, and 06 have different protein profiles but was classified in thesame biotype namely biotype I. Generally, the difference in protein profile actually more related to the biological diversity of the metabolism ofeach Trypanosoma evansi isolate from Indonesia.

Abstract

Penelitian ini bertujuan mengetahui pengaruh penambahan vitamin E dan mineral Zn terhadap kualitas semen kambing Boer. Rancanganyang digunakan adalah rancangan acak lengkap (RAL) dengan empat perlakuan, yakni P0= tanpa penambahan vitamin E maupun mineral Zn; P1= pemberian vitamin E 50 IU/ekor/hari; P2= pemberian mineral Zn 25 mg/ekor/hari; P3= pemberian vitamin E 50 IU/ekor/hari dan mineral Zn 25 mg/ekor/hari. Penelitian menggunakan 12 ekor kambing berumur 3 tahun, masing-masing perlakuan menggunakan 3 ekor kambing. Data yang diperoleh dianalisis menggunakan analisis ragam dan dilanjutkan dengan uji beda nyata terkecil (BNT). Hasil penelitian menunjukkan bahwa pemberian vitamin E dan mineral Zn serta kombinasi keduanya tidak memengaruhi volume semen kambing Boer, tetapi berpengaruh terhadap motilitas dan persentase spermatozoa abnormal (P0,05) dan terhadap persentase spermatozoa hidup (P0,01). Pemberian mineral Zn dan campurannya dengan vitamin E mampu meningkatkan kualitas spermatozoa kambing Boer.

Abstract

This study was aimed to assess the performance and suitability of commercial rapid test compared to the conventional test through verification process. This study used frozen meat from laboratory routine samples which divided into five groups, those were: natural group, low bacteria level group, medium bacteria level group, high bacteria level group, and control group, each sample test performed 9 replicates. All samples were tested for E. coli by conventional method (SNI 2897:2008) and commercial rapid test method. E. coli test result from both methods was calculated as precision (relative standard deviation), sensitivity, specificity, false negative, false positive, and kappa. The results showed that the commercial kit test had good precision with relative standard deviations score was 0.103. The sensitivity, specificity, false negative, false positive, and kappa score were 94.44%, 100%, 5.56%, 0%, and 0.872, respectively, indicates an equal performance with conventional method. The t student test showed that commercial rapid test method and conventional method had suitability on natural group, low bacteria levels group and medium bacteria level group.

Abstract

Penelitian ini bertujuan mengetahui pengaruh ekstrak etanol biji jintan hitam (EEBJH) terhadap aktivitas fagositosis dan sekresi reactive oxygen intermediates (ROI) makrofag peritoneal mencit Swiss yang diinfeksi Lysteria monocytogenes (L. monoytogenes). Dalam penelitian ini digunakan 72 ekor mencit jantan galur Swiss dengan berat antara 20-30 g. Mencit dibagi ke dalam enam kelompok, masing-masing terdiri atas 12 ekor. Kelompok 1 (kelompok kontrol negatif), diberi akuades secara per oral. Kelompok II (kelompok kontrol positif), hewan uji diberi imboost per oral. Kelompok III, IV, V, dan VI sebagai kelompok perlakuan, masing-masing diberi EEBJH dengan dosis 1, 5, 25, dan 125 mg/kg bobot badan/hari per oral selama 14 hari. Pada hari ke-15, semua mencit diinfeksi L. monocytogenes. Aktivitas fagositosis makrofag peritoneal diamati dengan metode lateks sedangkan aktivitas sekresi ROI diamati dengan metode nitro blue tetrazolium (NBT) reduction assay. Hasil penelitian menunjukkan bahwa pemberian EEBJH meningkatkan aktivitas fagositosis dan sekresi ROI makrofag peritoneal yang diinfeksi L. monocytogenes. Angka kematian hewan uji pada kelompok perlakuan lebih rendah dari kelompok negatif. Aktivitas fagositosis dan sekresi ROI makrofag tertinggi terdapat pada hari ke-14. Berdasarkan hasil penelitian maka dapat disimpulkan bahwa EEBJH memiliki efek meningkatkan aktivitas fagositosis dan sekresi ROI makrofag mencit Swiss yangdiinfeksi L. monocytogenes. Kelompok perlakuan dengan dosis 5 mg/kg bobot badan EEBJH memiliki aktivitas fagositosis dan sekresi ROI tertinggi.
