Abstract

Doxorubicin chemotherapy has been a strong focus in breast cancer research. Side effects, toxicity and resistance have been extensively studied. One proposed solution to these issues that its combination with deoxyelephantopin. Deoxyelephantopin is known to be toxic in many cancer cells but safe in normal cells. IC50 of each compound were determined by using a MTT assay, and the MDR-1 gene mRNA expression were determined by using qPCR method, while the interaction of doxorubicin in combination with deoxyelephantonin on Pgp receptor were predicted by using an in silico approach. The IC50 of deoxyelephantopin was found to be 11.2 g/mL, while IC50 of doxorubicin was 448 nM IC50 values showed a deoxyelephantopin-doxorubicin combination was able to reduce MDR-1 expression by 19% compared to doxorubicin and IC50 values indicated that the combination formula reduced the expression by 15% over doxorubicin alone. The affinity of doxorubicin and deoxyelephantopin is -12.16 kcal/mol and -9.51 kcal/mol, respectively, while the affinity of doxorubicin after combine with deoxyelephantopin decreases from -12,16 kcal/mol to -11,25 kcal/mol due to the release of one Leu 221 hydrogen bond. The combination of doxorubicin with deoxyelephantopin is able to reduce expression and suppress the function of the Pgp protein.

Abstract

Personal hygiene is an action taken to maintain cleanliness and health for an individual’s physical and mental well-being. Good personal hygiene practices minimize the entry of microorganisms, thereby reducing the risk of individuals contracting diseases. ASUH, which stands for Safe, Healthy, Whole, and Halal, can be used as an indicator of food suitability for processing and human consumption. This community service activity aimed to supervise the effort to ensure the eligibility of sacrificial animals in Gampong Tungkop, Darussalam District, Aceh Besar Regency, through ante-mortem and post-mortem examinations. The animals examined included 24 cows and 6 goats ready for slaughter. The examinations consisted of ante-mortem examination, which covered the general condition, teeth, health status, eye mucosa, anus, absence of defects, sufficient age, not emaciated, and male gender. Post-mortem examination, which assessed the condition of the meat and visceral organs, including the liver (hepar), small intestine (intestinum tenue), large intestine (intestinum crassum), stomach (ventriculus), kidneys (ren), lungs (pulmo), spleen (lien), and heart (cor). Observations showed that the eye mucosa color of the examined cows and goats was generally pink, indicating normal conditions. All cows and goats examined were healthy and had perfect physical conditions (no defects). The post-mortem examination results showed that the meat, stomach, spleen, heart, lungs, and kidneys were in normal condition. However, endoparasites of the Fasciola hepatica type were found in the livers of five slaughtered cows. Health conditions of the supervised cows and goats met the requirements for sacrificial animal slaughter.

Abstract

AbstractThe activity of managing organic waste as a manifestation of achieving a Green Campus is one of the efforts to create a better environment that can benefit the community, especially those living near the campus area. Effective Microorganisms (EM-4) is a mixture of beneficial microorganisms used to improve soil quality, accelerate waste decomposition, reduce environmental pollution, and enhance plant growth. Many plants on campus have suboptimal growth, requiring fertilization. Therefore, it is necessary to engage the campus community, especially students, to produce organic fertilizer by utilizing waste from the campus environment, cafeteria, as well as organic leaf and pine litter waste, so it can be used as compost. More than 95,24% of respondents have never participated in training on compost production. Further training is needed to ensure the target audience better understands waste management.Kegiatan pengelolaan sampah organik merupakan perwujudan untuk menuju Green Campus merupakan salah satu kegiatan untuk mewujudkan terciptanya lingkungan yang baik dan dapat dimanfaatkan bagi masyarakat terutama masyarakat yang dekat dengan areal kampus.Effective Microorganisms (EM-4) adalah campuran mikroorganisme yang bermanfaat yang digunakan untuk meningkatkan kualitas tanah mempercepat penguraian sampah, mengurangi polusi lingkungan dan meningkatkan pertumbuhan tanaman Banyak tanaman yang ada dikampus yang kurang optimal pertumbuhan sehingga perlu pemupukan. Untuk itu perlu mengajak masyarakat kampus khususnya mahasiswa untuk membuat pupuk organik dengan memanfaatkan sampah yang ada di lingkungan kampus atau kantin, kotoran ternak dan sampah organik daun kayu dan seresah pinus sehingga dapat dimanfaatkan sebagai pupuk kompos. Lebih 95,24% responden yang tidak pernah mengikuti dalam pelatihan tentang pembuatan kompos. Perlu pelatihan lebih lanjut sehingga sasaran lebih paham terhadap pengelolaan sampah tersebut.

Abstract

Collagen is one of the determinant factors of beef tenderness. This study analyzed the expression and content of type III collagen connective tissue in two muscle types of Aceh cattle through immunohistochemical staining. Meat samples were obtained from the gluteobiceps and psoas major muscles of three Aceh cattle bulls weighing 250-300 kg, with a body condition score of three. Histology preparations were created from samples and subjected to immunohistochemical staining utilizing specific antibodies targeting type III collagen. Qualitative data were analyzed descriptively, while quantitative data were analyzed using IMAGE J software. The results showed that the expression of type III collagen connective tissue was found in the endomysium and perimysium layers of both muscles. The expression of type III collagen connective tissue varied significantly among the muscles (P<0.05). The gluteobiceps muscle had an average percentage of the area containing type III collagen connective tissue of 4.47±2.41%, while the psoas major muscle was lower at 2.51±1.69%. This study concludes that the gluteobiceps muscle of Aceh cattle has a higher amount of type III collagen connective tissue than the psoas major muscle.

Abstract

This study aimed to analyze the muscle microstructure of longissimus dorsi Brahman cross (BX) cattle slaughtered at the slaughterhouse of North Aceh District. The meat samples for the study were taken from three male BX cattle aged 18-30 months with body weights ranging from 400 kg to 500 kg and the average body condition scores (BCS) of 3-4. The muscle used was the longissimus dorsi muscle. The mean (SE) diameter and cross-sectional area (CSA) of muscle fibers and the percentage of fast fibers in the longissimus dorsi BX muscle were 71.691.23 m, 4263.43305.69 m2 , and 48.178.81%, respectively. This study concluded that the longissimus dorsi muscle of the BX has relatively large muscle fibers, some of which in each fascicle are composed of fast muscle (fast fiber/type II).

Abstract

The presence of glycoconjugates in caput, corpus, and cauda epididymis of the Javan muntjacs was performed using lectin histochemical method. The epididymis organs were obtained from two adult male Javan muntjacs in hard antler stage and then immediately fixed in Bouins solution and processed in histological manner. Six types of biotinylated lectins namely Peanut agglutinin (PNA), Soybean agglutinin (SBA), Wheat germ agglutinin (WGA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), and Ulex europaeus agglutinin I (UEA I) were applied to detect the specific glycoconjugates in the epididymal tissue of muntjacs. The result showed the presence of glycoconjugates were detected by five types of lectins except UEA I in caput, corpus, and cauda epididymis with specific distribution and also the intensity of lectins binding. Glycoconjugates -galactose, -glucose, mannose, N-acetyl-galactosamine, N-acetyl-glucosamine, and sialic acid were stained with various intensity in epithelial cells, basal cells, stereocilia, spermatozoa, and stroma of caput, corpus, and cauda epididymis. The presence of sialic acid on the surface of spermatozoa detected by WGA are thought to be play a role on sperm protection during their maturation and transit along epididymal lumen. In conclusion, the difference of glycoconjugates distribution pattern and lectin binding in the epididymal caput, corpus, and cauda of the Javan muntjac indicates the regional differences in epididymal function in spermatozoa maturation.

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 g/ml of concentration for PNA lectins and 15g/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates -galactose, -glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis.Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak
