Abstract

The aim of this study was to determine the protective effect of vitamin E on mice heart. Twentyfivemalemiceweighing40gattheageof3monthswereused.Themicewereadaptedforaweekandfedbasaldiet.Themicewerefednormaldietascontrol(group1),highfat+50IUVitaminE(groupII);highfat + 100 IU Vitamin E (group III) high fat + 200 IU Vitamin E (group IV); high fat + 400 IUVitamin E (Group V). Diet was given about 10% body weight, water was given ad libitum every day. After 3 months, all animal were then killed. The hearts were collected for routine histopathologicalexamination. The result indicate that lesions in the heart consist of swollen eosinophilic hyalinization ofcytoplasm, vacuolization, and necrosis myocardium (cardiomiopathy) in all mice receiving high fat dietwith less than 400 IU Vitamin E.Keywords: high fat diet, vitamin E, cardiovascular diseases

Abstract

Tujuan dari penelitian ini adalah mengidentifikasi keberadaan gen M, H5, dan N1 virus avian influenza (AI) melalui metode single step multiplex reverse transcriptase-polymerase chain reaction (RT-PCR), sebagai acuan untuk peneguhan diagnosis secara molekuler virus AI di Provinsi Aceh. Penelitian ini menggunakan 11 isolat virus AI asal Provinsi Aceh yang diperoleh dari Laboratorium Balai Penyidikan dan Pengujian Veteriner (BPPV) Regional I Medan di Sumatera Utara dari tahun 2006-2008. Penelitian dilakukan di Laboratorium Virologi BPPV Regional I di Medan, Sumatera Utara. Amplifikasi terhadap gen matriks (M) virus AI menggunakan metode simplex RT-PCR. Hasil simplex RT-PCR terhadap gen M diperoleh 10 isolat yang menunjukkan pita deoxyribonucleic acid (DNA) pada 276 bp dan satu isolat yang tidak muncul, kemudian dilanjutkan dengan metode single step multiplex RT-PCR menggunakan pasangan primer gen penyandi protein N1, H5, dan M. Produk PCR 131 bp (N1), 189 bp (H5), dan 276 bp (M) muncul sebagai hasil elektroforesis dari semua isolat virus AI. Semua virus AI yang mewabah dari tahun 2006-2008 di Provinsi Aceh termasuk ke dalam virus influenza A subtipe H5N1.

Abstract

This study was aimed at isolating and identifying Salmonella sp. and then conducting an antibiotics susceptibility test in order to detect resistant genes. One hundred and five chicken cloaca swab samples were used in this study. 30 samples were taken from a layer farm in Bogor, 45 from a broiler farm in Sukabumi and 30 from a broiler farm in Cianjur. In order to isolate and identify the bacteria, a tetrathionate broth was used, which was then cultured in a Salmonella-Shigella agar, and finally a Gram stain and biochemical test was conducted. To confirm the presence of Salmonella sp., a pair of primers were used for the polymerase chain reaction (PCR) method to determine the presence of the invA gene.. An antibiotics susceptibility test was used with the Kirby-Bauer disk diffusion method. Nine antibiotics were used in this study. Each primer pair was used for the detection of tetA, blaTEM, aac(3)-IV, gyrA and ermB genes, and for genes encoding antibiotic resistance a PCR test was used. Eight (7.6%) Salmonella sp. were isolated in this study. All isolates showed positive results with PCR confirmation. The results of the antibiotics susceptibility test showed that Salmonella sp. isolates were resistant to tetracycline (75%), oxytetracycline (75%), amphicillin (75%), gentamycin (12.5%), nalidixic acid (100%), ciprofloxacin (12.5%), enrofloxacin (0%), erythromycin (100%), and chloramphenicol (0%). The distribution of antibiotic resistance genes in Salmonella sp. were tetA (33.3%), blaTEM (100%), aac(3)-IV (0%), gyrA (100%) and ermB (0%) positive. In conclusion, Salmonella sp. was isolated. All isolates showed positive results in the PCR confirmation. Salmonella sp. isolates were resistant to tetracycline, oxytetracycline, amphicillin, gentamycin, nalidixic acid, ciprofloxacin, and erythromycin. Only the tetA, blaTEM, and gyrA genes were detected in Salmonella sp. isolates.

Abstract

This study was conducted to determine the effect of water salinity changes on methionine content in tiger grouper juvenile (Epinephelus fuscoguttatus). A total of 2,560 tiger grouper juveniles were used and divided into five groups consist of 1 control group (without exposed to salinity changes) and 4 treatment groups. The salinity was changed every 2, 3, 4, and 6 hours in treatment A, B, C, and D, respectively. Salinity levels were changed during 24 hours by lowering salinity level from 32 psu to 22 psu. Twenty five of fish were collected from each treatment for methionine content analysis. Data were analysed using one way analysis of variance (ANOVA). The results showed that there was a decrease in methionine content in all treatments. A significant decrease (P0.05) of methionine content in treatment A, B, and C were observed after 20 hours (1.15%), 18 hours (1.27%), and 16 hours (1.24%), respectively. While at 0 hours (control), the methionine content was 2.02%. Methionine level in treatment D was not significantly different (P0.05) compared to control group. As conclusion, rearing the tiger grouper juvenile with salinity fluctuations every 6 hours did not lead to methionine deficiency.

Abstract

Penelitian ini bertujuan menentukan hubungan clonal methicillin resistance Staphylococcus aureus (MRSA) antara isolat sapi dan manusia.Staphylococcus aureus yang digunakan dalam penelitian ini berasal dari infeksi kulit manusia di Yogyakarta (10 isolat) dan dari susu sapi perah (11 isolat) yang berasal dari Yogyakarta, Solo, dan Boyolali. Identifikasi bakteri dan uji resistensi S. aureus terhadap methicillin telah dilakukan pada penelitian sebelumnya. Hubungan genetika S. aureus antar isolat sapi dan manusia ditentukan menggunakan teknik single enzyme amplified fragment length polymorphism (AFLP). Hasil penelitian diketahui bahwa berdasar analisis AFLP S. aureus memperlihatkan 15 pola genetika (A sampai O) dan dapat dikelompokkan ke dalam 7 klas (I sampai VII). Staphylococcus aureus isolat asal sapi dari daerah yang berdekatan (Boyolali dan Solo), dapat dikelompokkan dalam 1 klas (kecuali 1 isolat sapi dari Yogyakarta), isolat sapi dan manusia dari Yogyakarta dapat dikelompokkan dalam beberapa klas. Masing-masing klas terdapat isolat S. aureus yang telah resisten terhadap MRSA. Hubungan genotipe S. aureus dalam penelitian ini dapat digunakan untuk mengetahui distribusi clonal antara isolat sapi dan manusia dan dapat digunakan sebagai kontrol adanya infeksi MRSA di Indonesia.

Abstract

ABSTRACT The purpose of this research was to examine the effect of various sugars addition in Tris extender on quality of frozen thawed epididymal spermatozoa of ram. Collected spermatozoa was divided into five tubes and centrifuged at 3,000 rpm for 30 minutes. Supernatants were removed and pellets were diluted with Tris extender (control), Tris extender + 0.4% dextrose (dextrose), Tris extender + 0.4% maltose (maltose), Tris extender + 0.4% lactose (lactose), and Tris extender + 0.4% sucrose (sucrose), respectively. There were no significantly difference among treatment on mean percentages of motile (MS), live (LS) and intact plasma membrane (IPM) after equilibration. Mean percentages of post-thawed MS, LS, and IPM for dextrose (43.0, 53.8, and 53.4%), maltose (44.0, 54.8, and 52.8%), lactose (44.0, 52.8, and 52.8%), and sucrose (44.0, 55.0, and 53.4%) were significantly (P0.05) higher than control (37.0, 44.0, and 40.8%). In conclusion, addition of 0.4% dextrose or 0.4% maltose or 0.4% lactose or 0.4% sucrose in Tris extender increased quality of frozen thawed cauda epididymal spermatozoa of ram.

Abstract

Penelitian ini bertujuan mempelajari deformasi sendi tarsus yang mengalami tarsitis kronis. Penelitian ini dilakukan dengan mengamati perubahan ukuran sendi tarsus pada preparat kaki belakang kuda delman dan deformasi skelet sendi tarsus pada kuda delman. Dari hasil penelitian ditemukan adanya pembesaran di bagian medial tarsus kuda dan pembesaran pada lingkar tarsus kuda 3-7 cm dari bentuk normal dengan rata-rata lingkar tarsus kuda delman 23,18 cm dan rata-rata lingkar tarsus pada kuda yang mengalami tarsitis 27,83 cm. Kelainan berupa perlekatan tulang (ankylosis) dan pertumbuhan tulang baru (exostosis) pada permukaan tulang-tulang sendi tarsus kuda. Pada gambaran radiografiditemukan adanya penyempitan ruang sendi, kerusakan permukaan tulang dan pertumbuhan tulang baru yang abnormal. Deformasi yang terjadipada sendi tarsus preparat kaki belakang kuda delman diduga disebabkan oleh aktivitas yang berat semasa hidupnya.

Abstract

Hookworms infection remains as an important parasitic infections affecting animal and human health worldwide. However, information on the prevalence and the risk factors of hookworm infection on dogs in Indonesia is very limited. A cross sectional study to estimate the prevalence and to identify risk factors of hookworms infection on dogs was conducted in rural area with different topography, Sukabumi Regency, Indonesia. A Total of 204 dog stool specimens were examined for hookworm eggs using simple flotation methods. The data of related risk factors namely type of topography area (high and low land), purpose of having dog, dog demography, dog reared management and deworming were collected through dog owners interviews with completed a questionnaire. Those data were analyzed using the chi-square test (2) and logistic regression. The prevalence of hookworm on dogs in Sukabumi was 24.5 % (95% Confidence Interval [CI]=19.1-30.8). This prevalence was associated with topography of the area, the age of dog, and type of dog keeping methods. The prevalence of hookworms was higher in the high land area than low land area (OR=5.935, 95% CI=2.764-12.744 ). Logistic regression identified pupies as a high risk group to hookworms infection (OR= 2.041, 95%CI=1.035-4.055). The dog which kept in the cage/tied had higher risk than free roaming dog (OR=3.66, 95%CI=1.479-9.091).Key words: Dog, hookworms, prevalence, risk factors, Sukabumi.

Abstract

The research has been done to know prevalence of toxoplasmosis in livestock, to search the source of infection that have potency to cause toxoplasmosis in human and to make a map of toxoplasmosis infection area in Banda Aceh. The method used in this research was serologic test, Card Aglutination Test (CATT). Data of fertile women in this study are collected through questioner. The result of this research showed that the toxoplasmosis prevalency number of society in Banda Aceh was 3.15%, while at livestock were varied respectively: goats 40%, chickens 25%, cattles 23%, duck 20%, cats 16%, buffalos 15%, and sheeps 10%. The potencial sources of to coures toxoplasmosis in human are: goat, chicken, cattle, duck, cat, buffalo, and sheep. The area found toxoplasmosis in humans and animal were: Baiturrahman, Kuta Raja, Ulee Kareng, Kuta Alam, and Syiah Kuala. Abstract

Penelitian ini bertujuan mengetahui pengaruh dosis dan lama inkubasi anti MPS ecto-CIK dalam menghambat viabilitas spermatozoa kambing dan sapi. Rancangan penelitian yang digunakan adalah rancangan acak lengkap (RAL) dengan pola faktorial yang terdiri atas 2 faktor yakni dosis pengenceran (0, 5, 10, dan 15 l) dan lama inkubasi (5, 30, 60, dan 120 menit) masing-masing 6 kali ulangan. Data yang diperoleh dianalisis dengan analisis varian dua arah, yang dilanjutkan dengan uji jarak Duncan dengan taraf signifikansi 5%. Pemberian anti MPS ecto-CIK membran spermatozoa kambing dengan konsentrasi 0, 5, 10, dan 15 l dan lama inkubasi 5, 30, 60, dan 120 menit berpengaruh signifikan terhadap viabilitas spermatozoa kambing dan sapi (P0,05). Perlakuan anti MPS ecto-CIK pada dosis 15 l dan lama inkubasi 120 menit terhadap spermatozoa kambing dan sapi merupakan perlakuan yang paling optimal dalam menghambat viabilitas spermatozoa kambing (45,5011,16 dan 44,879,40%) dan sapi (39,0814,40 dan 36,6711,93%).
