Abstract

Penelitian ini bertujuan mengetahui tingkat maturasi oosit sapi Simmental Peranakan Ongole (SimPO) dan Limousin Peranakan Ongole (LimPO) secara in vitro. Oosit dari Ovarium sapi yang berasal dari rumah potong hewan (RPH), dikelompokkan berdasarkan jenis sapi PO (kontrol), SimPO dan LimPO. Oosit diaspirasi dari ovarium dengan syringe 5 ml dan jarum 18 G. Oosit dengan kualitas A dan B yang digunakan dalam penelitian ini. Oosit dikultur dalam media maturasi TCM 199 100 l drop dan dilapisi minyak mineral, diinkubasi pada suhu 38,5 C, CO2 5% dan kelembaban 95% selama 24 jam. Tingkat maturasi in vitro ditentukan dengan pewarnaan aceto orcein 1% untuk melihat tahapan maturasi dengan adanya perubahan konfigurasi kromosom dan membran inti berupa germinal vesicle, germinal vesicle breakdown, metafase I, anafase/telofase I, dan metafase II. Hasil penelitian menunjukkan tingkat maturasi in vitro oosit yang mencapai metafase II dari oosit sapi PO lebih tinggi secara signifikan dibandingkan sapi SimPO dan LimPO.

Abstract

This study was conducted to explore the potential of adjuvant for the production of immunoglobulin Y (IgY) as antistreptococcosis in layer chicken with mass production orientation. Enterococcus faecalis which causes streptococcosis in the red tilapia was selected as a candidateantigen. The production of immunoglobulin Y (IgY) was carried out on Isa Brown layer chickens and aged around 20 weeks. Furthermore, thechickens were grouped into four groups (A, B, C, and D groups), each consisting of three chickens based on the type of adjuvant, while twochickens were used as a control group. Each group was treated by giving MONTANIDE ISA 71R VG adjuvant (A), Freund's adjuvant (B), aluminum potassium sulphate adjuvant (KAl(SO4)212H2O) concentration of 50 ppm in pH 7 (C), and only antigens without adjuvant (D). Chickens were kept for 35 days and each week was checked for presence the IgY antigen in the serum and egg yolk. Booster was conducted on 14th and 28th days of maintenance. The results showed that IgY in treatment group A was detected on day 28 in the serum and day 35 in the yolk. Whereas the treatment group B could be detected on day 35 in the serum. However, the IgY was not detected in the serum and yolk in C, D, and control groups until the end of the maintenance. Based on the results, it can be concluded that the appearance of IgY in serum and yolk in a relatively fast time is obtained in the combination of Enterococcus faecalis antigen with the emulsion of water-in-oil adjuvant (SEPPICMONTANIDE ISA 71R VG) compared to the other types of adjuvant that use in this study.

Abstract

The clinical symptom and anatomical pathology effect of sulphaquinoxalin administration were studied in 12 male -seven days-old Hubbard broiler chickens with the weight of 85.57.83 g. The chickens were randomly dividedinto 2 groups of 6 each. The P0 group as control group just orally received aquadest and the P groups were treated 1 orally with 0.1% sulphaquinoxalin which dissolved in drinking water. The sulphaquinoxalin was administrated forseven days. At the end of experiment all chickens were sacrificed for necropsy. The results indicated that less appetite and drinking water as well as decrease of body weight (P0.01). Ptechial haemorrhagic was seen in thorax andfemur muscle, mucosa of duodenum, liver, kidney, heart, lung, and spleen. The liver, kidney, heart, and spleen were swollen.

Abstract

Penelitian ini bertujuan mengetahui efek injeksi inhibin B sebagai kandidat kontrasepsi pria berbasis hormonpeptida terhadap berat badan, berat dan panjang testis, dan duktus epididimis. Sebanyak 24 ekor tikus (Rattusnovergicus) jantan strain Wistar berumur 4 bulan dengan berat badan 150-200 g dikelompokkan secara acak ke dalam 4 kelompok, yaitu kontrol (KO), KI, KII, dan KIII. Kelompok kontrol, tikus hanya diinjeksi dengan 0,1 ml PBS tanpa inhibin B; Kelompok KI, KII, dan KIII tikus diinjeksi dengan 25, 50, dan 100 pg inhibin B/ekor. Injeksi dilakukan secara intra peritoneal sebanyak 5 kali dengan selang waktu 12 hari selama 48 hari. Injeksi pertama, isolat inhibin B dilarutkan dalam PBS sebanyak 0,05 ml dan diemulsikan dengan 0,05 ml Freud's complete adjuvant (FCA). Pada injeksi kedua, ketiga, keempat, dan kelima menggunakan inhibin B dalam PBS 0,05 ml dan diemulsikan dengan 0,05 ml Freud's incomplete adjuvant (FICA). Pada hari keenam setelah injeksi terakhir, tikus dikorbankan secara dislocatio cervicalis setelah terlebih dahulu dilakukan penimbangan berat badan. Berat testis dan duktus epididimis ditimbang dengan menggunakan timbangan elektrik, sedangkan diameter dan panjang testis dan duktus epididimis diukur dengan menggunakan jangka sorong. Hasil penelitian menunjukkan bahwa tidak terdapat perbedaan yang nyata (P0,05) antara kelompok kontrol dan perlakuan terhadap berat badan, berat, panjang, dan diameter testis dan duktus epididimis. Penelitian ini menyimpulkan bahwa inhibin B berpotensi dikembangkan sebagai kandidat kontrasepsi pria berbasis hormon peptida yang aman dan reversible.

Abstract

This research aimed to determine the optimum time for early pregnancy diagnosis in dwarf goat by trancutaneous ultrasonography and to determine ultrasonography imaging of dwarf goat pregnancy since insemination until embryo formation. Animal used in this research were 3 productive female goats with normal estrus cycle and 1male goat. The female goats were kept in separation pen from the male goat. The grasses were fed three times a day and additional feed were fed in the morning and afternoon with the water was given ad libitum. The goats were synchronized with intramuscular double injection of 1 ml estron/each female goat with 11 days interval. The observation of estrus was examined using male goat following the second injection of estron. The sign of estrus in female goats were characterized with standing immobile and allow the male goat to mount them. If the signs had been evident, the female goats were naturally mated. Pregnancy was determined by the presence of the embryonic vesicle by isoechogenic until hyperechogenic visualization surrounded by hypoechogenic of the embryonic fluid. The result of the transcutaneous ultrasonography in dwarf goat pregnancy showed the presence of embryonic vesicle on day 14 and embryo with isoechogenic visualization was detected on day 24 of pregnancy.Key words: dwarf goat, early pregnancy, transcutaneous ultrasonography

Abstract

Penelitian ini bertujuan mengetahui tingkat prevalensi Toxoplasma pada kucing dan menyusun basis epidemiologis pada kucing yang sangat penting dalam peranannya sebagai hospes definitif toksoplasmosis. Sampel serum darah dan feses kucing digunakan dalam penelitian ini. Metode pemeriksaan menggunakan card agglutination Toxoplasma test (CATT) Pastorex Toxo kit dan metode sentrifus. Data yang dip eroleh dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa tingkat prevalensi Toxoplasma dengan pemeriksaan serum darah kucing sebesar 6,8% dan pemeriksaan feses sebesar 9,4%. Infeksi toksoplasmosis yang terjadi pada kucing secara umum dari pemeriksaan klinis tidak mempunyai gejala yang spesifik.

Abstract

This study aimed to compare the effects of simvastatin, rosuvastatin, and fenofibrate therapies on the body weight and lipid profiles of the female rats receiving a combination of oral contraceptives (COC) and a high-fat diet (HFD). Twenty female Wistar rats (130-200 g) were divided equally into four groups. The rats received a standard diet for seven days, and their baseline lipid profiles were analyzed. All rats also received COC containing 15.1 g/kg levonorgestrel/3.1 g/kg estradiol with HFD for 60 days. Starting on day 31, the rats were given the respected treatment. The control group received the vehicle, whereas the others were treated with either simvastatin (2.1 mg/kg), rosuvastatin (0.5 mg/kg), or fenofibrate (8.2 mg/kg). Blood samples were taken on days 30 and 60. The results showed that the administration of COC+HFD and vehicle for 30 days increased the rats body weight and dyslipidemia, characterized by a significant decrease in Low-Density Lipoprotein (HDL) and an increase in total cholesterol (TC) and triglycerides (TG) levels (P0.05). Among the treatments, only fenofibrate was found to be able to prevent a significant weight gain in rats. Both fenofibrate and rosuvastatin inhibited a significant rise in TG and TC levels. Meanwhile, the simvastatin administration failed to do the same. Although statistically insignificant, all treatments increased rats HDL levels. Thus, it can be concluded fenofibrate was the most effective treatment among all in reducing weight gain and improving the lipid profile of dyslipidemic rats induced by oral contraceptives and a high-fat diet.

Abstract

This study aimed to determine ultrasound of uterine involution and postpartum ovarian dynamics on Ongole Crossbreed Heifer (PO) associated with postpartum estrus signs. This study used 6 PO cows which were divided into primiparous group and pluripara group. The observation of uterine involution and ovarian dynamics was started from the first day postpartum using ultrasonography (USG) with a linear probe rectally, while the reproductive organs images were recorded every 2 days. The results showed that the time needed for all reproductive organs of 3 primiparous groups to complete the uterine involution after parturition was 37.331.15 days when the diameter of the vagina, cervix uterine, corpus uterine, left cornua, and right cornua uterine were 3.140.00 cm, 2.860.00 cm, 3.200.06 cm, 4.660.01 cm, and 4.660.01 cm, respectively. The time needed for all reproductive organs of 3 pluripara groups to complete uterine involution postpartum was 38.671.15 days with the diameter of the vagina, cervix uterine, corpus uterine, left cornua, and right cornua uterine 3.180.00 cm, 2.700.02 cm, 3.080.02 cm, 4.420.01 cm, and 4.420.01 cm, respectively. The average times of the first and second ovulation of primiparous cattle were 27.671.15 and 47.671.15 days postpartum, whereas in pluripara group was 28.331.15 and 48.331.15 days postpartum. At first ovulation all cows were not accompanied by signs of estrus, while at the second ovulation 2 primiparous cows and 1 pluripara cow showed less obvious signs of estrus, 1 primiparous cow and 1 pluripara cow showed signs of medium estrus, and 1 pluripara cow showed clear estrus signs.

Abstract

The objective of this research was to study the effectiveness of calcitriol and ethynil ethyl estradiol combinations to decrease osteoporosis and endometrial cancer risks in ovariectomized Wistar rats. Twenty five 8-week old female Wistar rats were randomly divided into five groups (normal controlled rats/NK, ovariectomized rats/OVK, ovariectomized rats + calcitriol supplementation/OVD, ovariectomized rats + ethynil ethyl estradiol supplementation/OVE, and ovariectomized rats + calcitriol + ethynil ethyl estradiol supplementation/OVDE). At the end of the study, all rats were weighed and euthanized with 10% ketamine and 2% xylazine. Uterus and left femur were taken and fixed in 10% formaldehyde solution for histopathological examination using hematoxylin and eosin stain. Uterus was weighed before the fixation. The results showed that the percentage of uterus weight in OVK was significantly decreased compared to NK. Meanwhile, the percentage of uterus weight in OVDE was significantly increased compared to OVK. Histopathological features of the uterus in OVK were atrophy, reduction of myometrial and endometrial layers thickness, and formation of cuboidal epithelium in the endometrial lumen. However, the thickness of myometrial and endometrial layers in OVDE were increased, and its endometrial lumen was lined with metaplastic and hyperplastic squamous cell. Histopathological features of distal femur epiphysis in OVK ras showed fewer trabecular spiculums and more adipocyte in the bone marrow compared to NK. However, OVDE had more trabecular bone spiculum and less adipocyte in the bone marrow compared to OVK. In conclusion, the combination of calcitriol and ethynil ethyl estradiol supplementation could reduce osteoporosis, but increased the endometrial cancer risk in ovariectomized Wistar rats.

Abstract

Mouse and vole embryos were transferred into pseudopregnant CD-1 and scid female mice. Cellular changes involved in the formation of decidua in the pregnant mouse uterus up to day 8 of pregnancy were examined by histological, electron microscopic, and histochemical techniques. On day 6 of pregnancy, the vole embryos were laid in interstitium of antimesometrial side of the uterus, as well as in intraspecific pregnancy. Compared with intraspesific pregnant mouse, blood vessels were numerous in the decidua around the vole embryos in interspecific pregnancy. Both distribution and dilation of the blood vessels were increased on day 8. A part of cells in the inner cell mass had not nuclei, suggesting damaged vole embryos on day 8. At the implantation site, the uterine decidua was invaded by extravillous trophoblast (EVT) cells whose function is to destroy the walls of the uterine spiral. Moreover, this decidua was infiltrated by a population of natural killer (NK) cells and macrophage. These cells were particularly numerous in the decidua basalis at the implantation site where they come into close contact with invading EVT cells. These results suggest that interaction between NK, macrophage, and EVT provides the controlling relationship of embryo-maternal in intraspecific and interspecific pregnancy.
