Abstract

This study was conducted with aim to tend out morphology of oesophagus and stomach of the cave swiflets (Collocalia linchi) at macroscopic and microscopic levels. The data revealed that esophagus was 2.93 cm in length and possessed no crop. The stomach was small and the isthmus was not clear. Mucosa of the esophagus was lined by stratified squoamus epithelium. Esophageal gland was mucous type. The glands were well developed and distributed along the esophagus. The external muscle layer consisted of inner circular and outer longitudinal layers. The stomach could be distinguished into proventriculus and ventriculus with no clear isthmus between them. The mucosa of proventriculus was lined by single columnar epithelium. The ventricular gland area was divided into cardiac, fundic and pyloric gland areas. The surface of ventriculus was lined with cuticula. In general the esophagus and stomach of the cave swiftlets were simple with no crop in the esophagus and isthmus in the stomach. These might be related with the kind of food and feeding behavior of this species.

Abstract

Penelitian ini bertujuan mengkaji daerah nonkoding (D-loop) deoxyribonucleic acid (DNA) mitokondria kuda sumba. Tiga sampel darah kuda Sandel Wood dari Kabupaten Sumba Timur digunakan untuk isolasi DNA. Produk isolasi DNA diamplifikasi pada daerah D-loop parsial mitokondria dengan metode PCR, primer ECLOOP forward 5-CGCACATTACCCTGGTCTTG-3 dan primer ECLOOP reverse 5GAACCAGATGCCAGGTATG-3, pada kondisi predenaturasi 94 C selama 5 menit, denaturasi 94 C selama 30 detik, annealing 56 C selama 30 detik, elongation 72 C selama 45 detik, dan post elongation 72 C selama 5 menit sebanyak 35 siklus. Reaksi polymerase chain reaction (PCR) menghasilkan produk sepanjang 492 bp kemudian dilakukan pengurutan. Produk pengurutan D-loop dibandingkan dengan spesies kuda lain dari Genbank dan dianalisis menggunakan MEGA versi 4.1. Hasil analisis diperoleh 66 situs nukleotida yang berbeda. Urutan nukleotida D-loop yang dianalisis menggunakan Kimura 2- parameter ditemukan nilai paling kecil 0% dan nilai paling besar 2,9%. Konstruksi pohon filogenetika menggunakan metode neighbour joining dengan nilai bootstrap 1000 kali menunjukkan hubungan kekerabatan yang sangat dekat di antara kuda Sandel Wood. Artinya kuda Sandel Wood 2, 3, dan 4 berasal dari satu keturunan.

Abstract

This study aimed to evaluate three sets of B1 gene DNA primer for the diagnosis of Toxoplasma gondii. The DNA of Toxoplasma gondii that stored on liquid nitrogen was isolated using DNAzol reagent. The first step of Polymerase Chain Reaction (PCRs) was performed using external and internal primer sets, respectively, and then nPCR. PCR products sequencing was performed by Apical Science. All sequences were analysed using CLC Sequence Viewer Version 8.0 software and compared to sequence database that deposited in ToxoDB (Toxoplasma gondii genome database) using BLAST (https://toxodb.org/toxo/app). Each B1 gene primer was evaluated by performing single PCR (forward and reverse) and nested PCR reactions. Three sets of B1 gene primer have different amplification precision. According to the results of amplicon sequencing, the primer set #2 has the best amplification precision of B1 gene.

Abstract

Concentrations of lead (Pb), cadmium (Cd), and copper (Cu) were determined in muscle and liver tissue in the three commercially fish species, the Indian mackerel (Rastrellinger kanagurta), the tuna (Euthynnus affinis), and the tripletail (Lobotes surinamensis) collected from Aceh (Indonesia) and Penang (Malaysia). In this study, metal concentrations were found highest in the liver tissue compared to the muscle for all fish species (P0.05). Concentrations ofmetals in the liver were in the range of 0.05 to 0.4 0g/g dry wt for Pb, from 0.13 to 0.67 0g/g dry wt. for Cd, and 1.17 to 3.54 0g/g dry wt. for Cu. While in the muscle, Pb content were in the range of 0.02 to 0.14 0g/g dry wt.; Cd at below 0.12 0g/g dry wt.; and Cu at below 0.54 0g/g dry wt. On the average, among locations, Pb, Cd, and Cu contamination were found significantly higher (P0.05) in samples collected from Aceh (Indonesia). However, levels of metals in the fish studied were not exceeded the guideline values for food as recommended by Ministry of Agriculture, Fisheries, and Food, UK and the Ministry of Health Malaysia.

Abstract

Telah dilakukan penelitian untuk mengetahui pemberian herbal serbuk biji pepaya matang secara oral pada babi ascariasis terhadap dayaberembrio telur cacing pada tinja dan pada uterus cacing yang keluar dari babi yang diobati tersebut. Pada percobaan ini sebanyak 24 ekor babi Landrace betina dengan bobot badan 10-15 kg, umur 15 minggu yang terinfeksi cacing Ascaris suum (A. suum) secara alami, dengan egg per gram (EPG) berkisar 250-2500 butir. Babi tersebut dibagi menjadi empat kelompok perlakuan (P0, P1, P2, dan P3) masing-masing terdiri dari 6 ekor. Kelompok P1 dan kelompok P2 mendapat perlakuan dengan herbal serbuk biji pepaya matang per oral dengan dosis masing-masing 1 dan 3 g/kg bobot badan (P2) selama 3 hari berturut-turut. Kelompok P3 mendapat perlakuan dengan albendazol dosis 0,5 mg/kg bobot badan (Zodalben 0,04 ml/kg bobot badan) sedangkan kelompok P0 bertindak sebagai kontrol tanpa pengobatan. Hasil penelitian ini menunjukkan bahwa pengobatan dengan herbal serbuk biji pepaya matang pada babi ascariasis ternyata mampu menurunkan daya berembrio telur cacing dalam tinja babi dan dalam uterus cacing secara efektif sehingga dapat digunakan untuk pengendalian askariasis pada babi.

Abstract

This study was conducted to assess the effectiveness of a diet contained fresh anchovies and calcitriol supplementation for 6 weeks to prevent osteoporosis in ovariectomized rats. Fifteen Wistar rats aged 8 weeks were divided randomly into 3 groups (normal/K, ovariectomized/Ov, and ovariectomized + calcitriol/OVD), 5 mice each. Group K and Ov rats were fed with fresh anchovies, while the OVD group was fed with fresh anchovies + calcitriol. At the age of 15 weeks, all mice were done for euthanasia, then left femur was collected for immunohistochemistry examination of tartrate resistant acid phosphatase5b (TRAP5b). The detection of (TRAP5b) was conducted using a monoclonal ant ibody anti TRAP5b, and detected using a streptavidin-biotin. The results showed that the metaphysis part of distal femur bone of mice group K, Ov, and OVD were positive TRAP5b stained with brown color on trabecular bone in bone marrow cavity and trabecular spiculum surface, but rats in group K and Ov had extensive bone marrow cavity and normal trabecular spiculum, whereas OVD group showed bone marrow cavity dilation, accumulation of adipocytes in the bone marrow cavity, and shorter the spiculum of trabeculae. It can be concluded that fresh anchovies diet and calcitriol supplementation for 6 weeks are not effective in preventing osteoporosis in ovariectomized rats.Key words: calcitriol, femur, ovariohisterectomy

Abstract

Penelitian ini bertujuan mengidentifikasi kandungan gonadotropin releasing hormone (GnRH) ekstrak jaringan otak sapi dewasa pada fase folikuler (ovari berfolikel) dan fase luteal (ovari berkorpus luteum). Pada penelitian ini digunakan 2 buah otak sapi perah peranakan Friesien Holstein (FH) dewasa dengan umur 7-8 tahun yang pada saat disembelih kondisi ovarinya menunjukkan fase folikuler (1 buah otak) dan fase luteal (1 buah otak). Analisis kandungan GnRH dilakukan dengan metode enzyme-linked immunosorbent assay (ELISA) indirect. Hasil penelitian menunjukkan bahwa konsentrasi GnRH bagian otak sapi pada fase folikuler vs fase luteal berturut-turut untuk bagian otak parietal lobe of cerebral hemisphere, corpus callosum, thalamus, hipothalamus, pituitary gland (hipofise), midbrain, cerebellum, pons, dan medulla oblongata masing-masing adalah 260 vs 270, 110 vs 170, 230 vs 320, 1590 vs 1310, 840 vs 1250, 530 vs 810, 200 vs 480, 230 vs 100, dan 220 vs 70 g/ml. Dari hasil penelitian disimpulkan bahwa semua bagian otak sapi yang diamati mengandung GnRH dengan konsentrasi yang berbeda antara fase folikuler dengan fase luteal.

Abstract

This study aims to determine the effectiveness of Areca catechu Crude Aqueous Extract (ACCAE) as an anthelmintic, the number ofAscaridia galli worms in the duodenum, jejunum, and ileum after treatment, and an economic analysis. In this study, 50 female chickens wereused. The chickens were divided into 5 groups: Treatment 1, negative control (P1), which was given aquadestylates; treatment 2 (P2), which wasgiven infusion of ACCAE at a dose of 26 mg/mL; treatment 3 (P3), which was given infusion of ACCAE at a dose of 53 mg/mL; treatment 4(P4), which was given ACCAE with a dose of 79 mg/mL; and treatment 5 (P5) (positive control with pyrantel pamoat). Each group consisted of10 chickens. An examination was carried out that included weighing the population of adult worms according to their predilection 14 days aftertreatment. Weighing was carried out at the beginning before treatment, on the 7th day after treatment and on the 14th day after treatment. Thechickens were then euthenized, the intestinal worms were counted, and the economic analysis was calculated. Based on the results of the study, itcan be concluded that the best dose for removing A. galli worms is 79 mg/mL, and the location of the predilection of the intestine where the mostworms are found is the jejunum and the least is in the ileum. Economically, the highest income was obtained from treatment 4, namely the groupwith infusion of areca nut at a dose of 79 mg/mL.

Abstract

This research was conducted to identify the estrogen hormone profile and estrus response in the thin tailed ewes synchronized with controlled internal drug release (CIDR) implant. This research was carried out by employing 8 thin tailed ewes, CIDR, and estrogen kit. The ewes were distributed into: 3 ewes as control group and 5 ewes as treated group (with 12 days-CIDR implantation). Ewes were raised in the Sidomukti farm group, Sleman Regency, Yogyakarta. The data obtained were estrogen hormone profile during estrus examined by using ELISA method and estrus response marked with the reddening and oedema of vulva, mucus vaginal discharge, behavioral changes, vaginal pH, and superficial cells population. The data were analyzed by applying independent sample T-test. The result did not show any significant difference (P0.05) in the normal range between the control and CIDR group in estrogen hormone profile, estrus response, and pH level, but showed significant differences with positive correlation (P0.05) in population of superficial cells. In conclusion, CIDR was an effective tool for estrus synchronization at the farmer group, and gave positive estrus response towards thin tailed ewes with estrogen hormone profile, pH level, and superficial cell percentage on the normal range.

Abstract

This study aimed to determine the role of guava fruit filtrate (GFF) on maintaining the pre-freezing and post-thawed quality of Bali bullspermatozoa in tris-egg yolk (TEY) extender. The bovine semen was collected using artificial vagina twice a week (n= 10). Ejaculate sampleswere divided into four tubes and each tube was respectively added the following diluents: TEY, TEY + 5% GFF, TEY + 10% GFF, and TEY +15% GFF. The diluted semen was filled into the straws (0.25 mL) and cooled in refrigerator at 4 C for 2.5 hours. The samples were kept onvapor phase of liquid nitrogen (N2) (-120 C) for 10 minutes prior to be stored in liquid phase (-196 C). The quality of pre-freezing and postthawedspermatozoa in straw sample (i.e. motility, viability, and abnormality) was evaluated under light microscopy at 400x magnification. Datawere analysed statistically. The percentage of motility and viability both on pre-freezing and post-thawed process were significantly higher insemen diluted with TEY + 10% GFF compared to control (TEY), TEY + 5% GFF, and TEY + 15% GFF. Addition of 10% GFF into tris-egg yolkextender play role for maintaining the quality of pre-freezing and post-thawed Bali bull spermatozoa.
