Abstract

Penelitian ini bertujuan mengetahui perbedaan respon antibodi yang ditimbulkan oleh vaksin AI dengan seed virus AI H5N1 Indonesia yang dicampur dengan adjuvant berbeda. Formula vaksin yang dicobakan pada penelitian ini adalah monovalen dan polivalen. Tiga isolat virus HPAI subtipe H5N1 yang digunakan adalah Chicken/Denpasar/Unud-01/2004, Chicken/Klungkung/Unud-12/2006, dan Chicken/Jembrana/Unud-17/2006. Adjuvant yang digunakan yaituFreund's complete dan incomplete adjuvant, aluminium hidroksida, dan immunostimulating complexs (Iscoms). Vaksin monovalen dibuat dengan cara masing-masing isolat virus AI yang telah diinaktivasi dicampur dengan masing-masing adjuvant. Vaksin campuran (polivalen) dibuat dengan mencampur ketiga isolat dengan masing-masing adjuvant. Vaksin disuntikkan secara subkutan pada ayam layer jenis Isa Brown umur 3 minggu dan diulang pada umur ayam 5 minggu masing-masing sebanyak 0,5 ml/ekor. Pengambilan serum untuk pengujian titer antibodi dilakukan setiap 1 minggu setelah vaksinasi. Pengujian antibodi poliklonal dilakukan dengan uji hambatan hemaglutinasi (HI). Hasil penelitian menunjukkan bahwa ayam percobaan yang divaksinasi dengan adjuvant aluminium hidroksida mempunyai GMT anti-H5 paling tinggi baik pada vaksin monvalen atau polivalen. Adjuvant aluminium hidroksida adalah adjuvant terbaik untuk pembentukan antibodi anti-AI subtipe H5N1 pada ayam.

Abstract

The aims of this research were to estimate the prevalence and to determine the risk factors related to Toxocara cati (T. cati) infection in pet cats in Bogor. This study was cross sectional study using two types of data; laboratory examination results of feces samples and interview results of the cat owner using a structured questionnaires including characteristics of pet owner and pet care management. The data obtained from this study was analyzed using chi-square test. The results showed that 85 of 243 cats were positively infected by T. cati with 35% prevalence. The significant risk factor associated with the T. cati infection were: sex, sand provided, deworming and type of feed. Pet care management in Kota Bogor were well managed, however the prevalence of T. cati infection was still high. Therefore it can be a potential public health problem especially on cat owners.

Abstract

Penelitian ini bertujuan mengetahui potensi kit deteksi cepat Anigen rapid test kit rabies Ag dalam mendeteksi virus rabies pada sampel otakanjing yang diperoleh dari lapangan yang meliputi batas deteksi, kecepatan reaksi, uji reaksi silang, uji sensitivitas, dan spesifisitas. Batas deteksi ditentukan dengan pengenceran secara serial kontrol positif virus rabies dan selanjutnya diuji dengan rapid test kit sesuai petunjuk produsen. Uji reaksi silang dilakukan dengan canine parvovirus, Escherichia coli, dan Salmonella sp. Uji sensitivitas dan spesifisitas dilakukan terhadap sampel otak yang telah dikonfirmasi positif rabies dengan uji fluorescent antibody technique. Konfirmasi uji rapid test tersebut dilakukan dengan reverse transcriptase polymerase chain reaction. Berdasarkan data yang diperoleh dalam penelitian ini dapat disimpulkan bahwa Anigen rapid test kit rabies Ag mampu mendeteksi sampel yang mengandung virus rabies dengan titer 0,5 x log 106,5/0,03 ml, dengan rata-rata kecepatan reaksi 1,8 menit 29,35 detik (kurang dari 2 menit). Di samping itu Anigen rapid test kit rabies menunjukkan tidak terdapat reaksi silang dengan canine parvovirus, Escherichia coli, dan Salmonella sp. serta mempunyai sensitivitas 92,30% dan spesifisitas 97,90%

Abstract

Penelitian ini bertujuan melakukan isolasi dan identifikasi serotipe lokal Escherichia coli (E. coli) O157:H7 dan uji profil hemolisisnya pada media agar darah. Isolasi bakteri dilakukan dengan media eosin methylene blue agar (EMBA), dilanjutkan dengan identifikasi pada media selektif sorbitol MacConkey agar (SMAC) dan uji konfirmasi menggunakan uji aglutinasi lateks O157 serta uji antiserum H7 sebagai konfirmasi akhir dari E. coli O157:H7. Gambaran hemolisis diuji dengan menumbuhkan isolat pada media agar darah domba. Hasil penelitian menunjukkan bahwa 7 isolat (8,54%) dari 82 sampel feses ayam teridentifikasi E. coli O157:H7 dan memperlihatkan profil enterohemolisis seperti halnya isolat kontrol ATCC 43894. Dari hasil penelitian disimpulkan bahwa isolat lokal E. coli O157:H7 hasil isolasi dari feses ayam diketahui memiliki patogenitas yang tinggi terkait dengan dihasilkannya enterohemolisin yang merupakan marka/penanda kemampuan dari isolat untuk menghasilkan faktor virulensi Shiga like toxin.

Abstract

The purpose of this study was to obtain Escherichia coli isolates from 11 sei meat sellers in Kupang City and to observe their resistance to13 types of antibiotics. Escherichia coli were isolated and identified based on SNI 2897: 2008. Antibiotic resistance of the Escherichia colii isolates was determined using the Kirby-Bauer method. The results showed that 13 isolates (39.39%) of Escherichia coli in sei meat samples had a high level of resistance to erythromycin (100%), tetracycline (76.92%), and doxycycline (61.54%). Isolates demonstrated increased resistance to streptomycin (46.15%), cephalothin (38.46%), trimethoprim-sulfamethoxazole (38.46%), amoxicillin (30.77%), chloramphenicol (30.77%), and choline sulfate (30.77%). The antibiotics nalidixic acid and ciprofloxacin demonstrated low Escherichia coli resistance (7.69%). Escherichia coli are sensitive to cefotaxime and gentamicin. A total of 12 isolates (92.31%) experienced MDR. The presence of non-MDR and MDR resistant Escherichia coli in sei meat can seriously threaten community health.

Abstract

This present project was carried out to determine the effect of apus bamboo (Gigantochloaapus )leaves infusion to mortality rate and morphometry of Haemonchus contortus adult worms in vitro. The formulated tools were collected through camera, 3 ml syringe, object glass, deck glass, mortar, drip dropper, waterbath, microscope, stopwatch, counter check, oven, erlenmeyer flask, electric scale, surgical scissor, and pipette. The allocated materials used in this studywere bamboo leaves infusion, Haemonchus contortus adult worms, aquadestilata, ethanol and NaCl 0.62%. The research was firstly concernedwith making 0.1% and 1% apus bamboo leaves infusion. The samples were obtained from Haemonchus contortus collection. Adult worms were obtained directly from abomasum of naturally infected goats that were cut at Animal Slaughter House (RPH). To investigate the matter, mortality rates of adult worms at various doses and times of observation as well as differences in Haemonchus contortus morphometry were analyzed using ANOVA. The findings disclosed that apus bamboo leaves infusion demonstrated a significant difference in the mortality rate of adult worms Haemonchus contortusat various doses and times of observation. This report presented the findings of research that the best dose to increase the mortality of worms was 1% bamboo leaves infusion. Moreover, apus bamboo leaves infusion affected the morphometry of Haemonchus contortus adult worms, especially in body length, cervical papilla width, and spicules length in males, and body length, cervical papilla width, and vulvar length in females.

Abstract

Tujuan penelitian ini adalah mengetahui kemampuan hemaglutinin pada Staphylococcus aureus (S. aureus) dalam proses adhesi. Uji adhesi pada sel epitel ambing dilakukan dengan menggunakan metode Velentine-Weiggand. Hasil penelitian ini menunjukkan bahwa S. aureus yang memiliki hemaglutinin menunjukkan kemampuan adhesi yang jauh lebih baik (380 bakteri/20 sel) dibandingkan dengan yang tidak memiliki hemaglutinin (56 bakteri/20 sel).

Abstract

This research was conducted to prove the ability of betel nut extract (Areca catechu) in causingdegradation of sperm motility on rats (Rattus norvegicus) strain Wistar. This research used 2-3 monthsage, 200-250 g body weight of male white rats Rattus norvegicus strain Wistar. The rats were dividedinto 5 groups in equal number, 3 rats each group. They were a control group without treatment and 4treatment groups which were given different doses of betel nut extract for 7 days, i.e., 1, 2, 3, and 4 g/200g body weight respectively. The result showed that increasing doses of betel nut extract can result indegradation of sperm motility significantly (P0.05).Keywords: Areca catechu, Rattus norvegicus, sperm motility

Abstract

Penelitian ini bertujuan mengetahui kontaminasi Aspergillus flavus dan konsentrasi aflatoksin B1 pada jenis makanan berbahan baku kacang tanah yang dipasarkan di Banda Aceh. Pengambilan sampel dilakukan di pasar tradisional dan swalayan di Banda Aceh. Sampel diisolasi dan diidentifikasi A. flavus serta dideteksi AFB1 dengan menggunakan metode enzym-linked immunosorbent assay (ELISA). Berdasarkan hasil penelitian yang dilakukan dari 15 sampel makanan berbahan baku kacang tanah, 73,33% (11 sampel) terkontaminasi aflatoksin B 1 dengan konsentrasi yang bervariasi. Walaupun terkontaminasi aflatoksin B1, namun sebanyak 86,67% sampel makanan berbahan baku kacang tanah masih aman dikomsumsi sesuai dengan persyaratan yang ditetapkan pemerintah yaitu maksimal 20 ppb.

Abstract

This study aimed to develop the use of thermographic analysis to evaluate wound healing for laparotomy surgery in animals. The experiment used nine adult dogs 5 males and 4 females, bodyweight 5-10 kg) that had undergone the same general anesthesia and laparotomy procedures. Evaluation of wound healing was performed at 24, 48, and 72 hours after surgery and 7 days after operations using a digital thermal camera. During each thermographic evaluation period, blood samples were taken for analysis of total leukocytes and leukocyte differential. The results of thermal imaging were compared with the values of inflammatory cells and the clinical condition of wound healing. Comparison of thermographic analysis with inflammatory status was evaluated using the regression equation and showed a strong correlation coefficient (Y= -0.4847 + 40.14, R2= 0.867), that is, the higher the temperature, the lower the inflammation. The conclusion of study is that thermographic analysis can be used to evaluate wound healing of laparotomy surgery in dogs.
