Abstract

This study aims to evaluate the expression of BAX, BCL-2, and BAX/BCL-2 ratio in maturation media of cow oocytes which supplemented with Urea in vitro because BAX and BCL-2 are the main regulators of apoptosis.A total of 263 oocytes from follicle aspirations originating from ovaries taken from slaughterhouses and were saturated with 3 addition of urea which was divided into three groups. The control group (P0) was control group without the addition of urea, P1 group was added with urea 20 mg/dL, while P2 group was added with urea 40 mg/dL.The results of in vitro oocyte maturation were continued with identification using immunocytochemical staining with the addition of BAX and BCL-2 antibodies.Positive results showed a brownish color on the oocyte and its cumulus.The results of this study indicated that there were significant differences (P0.05) in BAX and BCL-2 expression, although both curves were equally increase. The increase in BCL-2 was more significant than BAX, while the BAX BCL-2 ratio did not show a significant difference (P0.05) in whichthe curve of BAX/BCL-2 ratio was decreased.It can be concluded that the addition of urea does not affect the level of apoptosis.

Abstract

The objective of this research is to see the influence of weather on the prevalence of Newcastle Disease (ND) in chicken in Kabupaten Aceh Utara (North Aceh). Data used in this research were obtained from Dinas Peternakan North Aceh for the number of chicken suffered ND and from Badan Meteorologi dan Geofisika Lhokseumawe, North Aceh for the form of weather. Multiple Linear Regression Model with five independent variables (the average of rainfall per month, the average of maximum temperature, the average of minimum temperature, the velocity of the wind, and the average of humidity per month) was used to see the influence of wheather to the prevalence of Newcastle Disease. Proportion the number of chicken suffered from ND which is the ratio of the number of chicken suffered from ND to the total number of chicken was used as dependent variables. The result shows that the best model is = 120.529278 1.33 x wind humidity + 1.907 x wind velocity.

Abstract

Penelitian ini bertujuan menentukan efektivitas pentagamavunon-0 (PGV-0) terhadap penghambatan ekspresi siklooksigenase-2 (COX-2) pada kanker kolon tikus Wistar. Pada penelitian ini digunakan 20 ekor tikus Wistar jantan yang dibagi secara acak dalam 4 kelompok perlakuan. Kelompok I merupakan kontrol negatif, kelompok II kontrol positif, kelompok III diberi PGV-0 40 mg/kg BB selama 15 minggu, dan kelompok IV diberi PGV-0 40 mg/kg BB selama 25 minggu. Pemberian PGV-0 dilakukan secara oral dua kali seminggu. Induksi kanker kolon dilakukan dengan cara injeksi subkutan DMH 60 mg/kg BB, satu kali seminggu selama 15 minggu. Pada minggu ke-26, semua hewan coba dieutanasia, kolon difiksasi dalam formalin 10% untuk selanjutnya diamati perubahan makroskopik dan mikroskopik. Penilaian ekspresi COX-2 dilakukan dengan menggunakan metode Dukes stage dan skor imunoreaktivitas (IRS). Hasil penelitian ini memperlihatkan pemberian PGV-0 selama 25 minggu menurunkan jumlah nodul kanker kolon dari 5 ke 2 (berkurang 60%); diameter kanker kolon (pxl) dari 0,712 mm ke 0,0043 mm (berkurang 99,31%). Pemberian PGV-0 selama 15 minggu hanya menurunkan jumlah nodul 10% dan area kanker kolon dari 0,712 mm ke 0,0062 mm (99,07%). Skor imunoreaktivitas COX-2 diekspresikan oleh kelompok III dan IV adalah 4 dan 5. Gambaran histologis dari kolon mendukung hasil di atas. Pemberian PGV-0 efektif menurunkan jumlah dan area nodul kanker kolon melalui penghambatan ekspresi COX-2.

Abstract

The aim of the present study was to determine the survival of L3 populations in intestine ofchickens exposed to experimental Ascaridia galli infection. Nature female adult worm were obtained fromlumen of village chickens in a comercial abattoir in Bogor. The eggs (L1) obtained from uteri female adultworms were incubated in sterile aquadestilata at room temperature for 10-20 days developed embrionatedeggs (L2). Five groups (A-D) of 80 head chickens were infected with, 6000 L2 A. galli respectively. Thechickens of group A were infected six times with dose of each 1,000 L2 with an interval of one hour. Thechickens of group B were infected three times with dose of each 2,000 L2 with an interval of two hours.The chickens of group C were infected six times with dose of each 3,000 L2 with an interval of three hours. The chickens of group D were infected one time with single dose 6,000 L2. A. galli L3 were recovered from intestines of 80 heads chickens seven days after oesophagus inoculation with 6,000 L2.The result showed that total 702,000 L1 and 628,000 L2 collected from 124 A. galli female adult worms.The percentage of L1 developed L2 is 89.46% and L2 developed L3 is 11.27%. Significant survival of L3higher populations in intestine of chickens observed only in the group D. The results indicated thatchickens infected high dose of A. galli caused the decrease of host defence against ascaridiosis. Keywords: Ascaridia galli, embrionated eggs, larvae

Abstract

Penelitian ini bertujuan mendeteksi gen tst isolat Staphylococcus aureus (S. aureus) susu sapi perah dan susu kambing melalui amplifikasi gen 23S rRNA sebagai langkah awal pencegahan beberapa kasus keracunan susu. Metode penelitian meliputi reidentifikasi bakteri, preparasi deoxyribonucleic acid (DNA), amplifikasi gen 23S rRNA, amplifikasi gen tst, dan pengurutan DNA. Hasil reidentifikasi bakteri melalui pewarnaan Gram, uji katalase, uji koagulase, uji fermentasi MSA, uji VJA, dan uji VP adalah positif bakteri S. aureus. Hasil amplifikasi gen tst terhadap 3 isolat S. aureus menunjukkan hanya 2 isolat memberikan hasil positif. Hasil positif tersebut ditandai dengan munculnya fragmen DNA yang memiliki panjang spesifik (350 bp) sesuai dengan produk polymerase chain reaction (PCR) dari referensi dan database GeneBank. Disimpulkan bahwa dengan menggunakan metode PCR dapat terdeteksi adanya gen tst pada isolat S. aureus asal susu sapi perah dan susu kambing dari Bogor.

Abstract

The presence of glycoconjugates in caput, corpus, and cauda epididymis of the Javan muntjacs was performed using lectin histochemical method. The epididymis organs were obtained from two adult male Javan muntjacs in hard antler stage and then immediately fixed in Bouins solution and processed in histological manner. Six types of biotinylated lectins namely Peanut agglutinin (PNA), Soybean agglutinin (SBA), Wheat germ agglutinin (WGA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), and Ulex europaeus agglutinin I (UEA I) were applied to detect the specific glycoconjugates in the epididymal tissue of muntjacs. The result showed the presence of glycoconjugates were detected by five types of lectins except UEA I in caput, corpus, and cauda epididymis with specific distribution and also the intensity of lectins binding. Glycoconjugates -galactose, -glucose, mannose, N-acetyl-galactosamine, N-acetyl-glucosamine, and sialic acid were stained with various intensity in epithelial cells, basal cells, stereocilia, spermatozoa, and stroma of caput, corpus, and cauda epididymis. The presence of sialic acid on the surface of spermatozoa detected by WGA are thought to be play a role on sperm protection during their maturation and transit along epididymal lumen. In conclusion, the difference of glycoconjugates distribution pattern and lectin binding in the epididymal caput, corpus, and cauda of the Javan muntjac indicates the regional differences in epididymal function in spermatozoa maturation.

Abstract

The objective of this study to determine the effect of feeding fresh garlic to ducks on decrease the cholesterol level of duck eggs. This study composed of two treatment groups. The duck in the first group (control) was fed local feed without spplemente d with fresh garlic, while in the second group was fed with local feed and supplemented with 50 grams of fresh garlic/duck/day. Egg yolk cholesterol, high-density lipoprotein (HDL), low-density lipoprotein (LDL), and triglycerides (TG) analysis were carried out at the end of the study using the ether extract method and then with the Cholesterol Oksidase Para Amino Penazone (CHOD-PAP) or enzymatic color test. Data was analysed using one way Analysis of Variance (ANOVA) the results showed that fresh garlic reduced total cholesterol, LDL fraction, and triglyceride, but increased the HDL fraction of duck egg yolk. The total cholesterol, LDL fraction, and triglyceride of duck egg yolks dropped by around 15.98 mg per dL (8.06%), 10.40 mg per dL (12.97%), 64.92 mg per dL (12.71%), respectively, while the HDL fraction increased by 11.40 mg per dL (19.32%) The results of this study indicated that fresh garlic can be used to produce specific products such as low cholesterol and low fat duck eggs.

Abstract

Penelitian ini bertujuan melakukan isolasi dan penentuan aktivitas mikrobial senyawa bioaktif reduktiomisin dari bakteri Streptomyces sp. Ank181 daratan (terrestrial). Penelitian ini dilaksanakan di laboratorium kimia organik, Institute of Organic and Biomoleculare Chemistry, University of Goettingen, Germany. Isolat dan subkultur agar bakteri Streptomyces sp. Ank181 diperoleh dari koleksi sampel genus Streptomyces Professor Dr. H. Anke, Institute for Biotechnology and Drug Research, Kaiserslautern, Germany. Struktur reduktiomisin dalam penelitian ini dielusidasi berdasarkan data spektroskopi dan spektrometri massa. Tiga senyawa metabolit sekunder telah berhasil diisolasi dan diidentifikasi dari bakteri tanah genus Streptomyces sp. Ank181, yaitu reduktiomisin, asam 2,3-dihidroksibenzoat, dan asam indole-3-karboksilat. Hasil uji antimikroba menunjukkan bahwa reduktiomisin bersifat bioaktif terhadap bakteri, jamur dan sitotoksik terhadap Artemia salina.

Abstract

The aim of this study was to describe the role of diagnostic imaging and endoscopy to define the diagnose of the upper gastrointestinal tract disorder of the Schnauzer. The information from the owner stated that the dog has been vomiting for a year, sometimes containing blood. There might also be a possibility that it had eaten a corpus alienum. This case study covered physical examination, hematology and blood chemical analysis, diagnostic imaging radiography and ultrasonography as well as endoscopy performed. Physical examination showed weight loss and anorexia. Hematology and blood chemical analysis showed an increase in hemoglobin, hematocrit, lymphocyte, ureum, creatinine, alanine aminotransferase (ALT), and alkaline phosphatase (ALP) values. Abdominal radiograph showed no abnormality in the abdominal organs. Abdominal ultrasonography showed a hiperechoic elongated mass attached on the stomach mucosal surface which forms an acoustic shadowing at the ventral. Endoscopy showed pathological lesions that is inconsistency of stomach mucosa surface, foamy fluid in the stomach, ulcers and erosion of the stomach mucosal surface. Based on the diagnostic imaging and endoscopy performed, the animal was clearly diagnosed with chronic gastritis accompanied by stomach ulcer and erosion.

Abstract

Penelitian ini bertujuan mengarakterisasi marmut sebagai hewan model untuk penyakit Alzheimer dengan mengamati histopatologis otak dan aktivitas seluler glial fibrillary acidic protein (GFAP) pada otak yang diakibatkan oleh deplesi hormon testosteron. Dua belas marmut dibagi dua kelompok berdasarkan umur, yaitu enam marmut umur 16-32 bulan dan enam marmut umur 32-48 bulan. Deplesi testosteron dilakukan dengan cara kastrasi. Dua marmut dari setiap kelompok dinekropsi untuk koleksi sampel otak pada waktu satu, tiga, dan lima bulan setelah kastrasi. Bagian otak yang diambil adalah korteks, lobus parietalis, temporalis, dan hipokampus. Sampel otak dilakukan evaluasi patologis dengan pewarnaan hematoksilin dan eosin dan immunohistokimia dengan antibodi GFAP. Hasil penelitian menunjukkan bahwa deplesi testosteron dapat menyebabkan kerusakan otak yang ditandai oleh kematian sel neuron, peningkatan aktivitas sel-sel glia dan ekspresi GFAP pada jaringan otak.Kesimpulan penelitian adalah penurunan kadar testosteron dalam plasma darah menyebabkan terjadinya kematian sel neuron dan peningkatan aktivitas sel-sel glia pada otak.
