Abstract

This study aimed to observe the inhibition capability of Rhizophora sp. leaf extract towards the Streptococcus agalactiae and Edwardsiella tarda bacteria. Rhizophora sp. leaf was extracted using ethanol. Inhibition action of Rhizophora sp. leaf extract towards Streptococcus agalactiae and Edwardsiella tarda was tested on TSA solid media. The concentration of Rhizophora sp. leaf extract used were 1000, 2000, 3000, 4000, 5000, 6000, 7000, 8000, 9000, 10000 ppm, and novobiocin antibiotic was used as a control. Each treatment was conducted three times. MIC (minimum inhibition concentration) was performed to determine the minimum dose that could inhibit the bacterial growth. The results showed that the inhibition value of Rhizophora sp. leaf extract at 2000 to 10000 ppm towards Streptococcus agalactiae was 8.60-16.30 mm, and 6.97-12.27 mm towards Edwardsiella tarda, whereas the inhibition value of novobiocin for both bacteria was 18.00-20.45 mm. The results of MIC value at dose of 2.000 ppm of Rhizophora sp. leaf extract was towards Streptococcus agalactiae with bacterial density of 165x108 CFU/mL, and towards Edwardsiella tarda 75x108 CFU/mL, respectively. In conclusion, Rhizophora sp. leaf extract had more bacteriostatic activity against Gram-positive bacteria (Streptococcus agalactiae) rather than the Gram-negative bacteria (Edwardsiella tarda).

Abstract

The study aims to reveal the morphology of the male sex accessory glands of the pangolin at macroscopic and microscopic levels. Macroscopic observation included measurement of length and thickness of each accessory gland while microscopic observation, sample of each accessory gland was processed by histology technique with paraffin method and sliced with 3-5 ?m thickness and stained with hematoxylin-eosin (HE) staining for general structural observation, coloration of alcian blue (AB) pH 2.5 and periodic acid Schiff (PAS) to observe the distribution of acid and neutral mucopolysaccharides in each glands. The results showed that the male sex accessory glands of the pangolin consist of vesicular gland and prostate, and bulbourethral gland which were not observed macroscopically. The average length and thickness of vesicular gland were 1.07 cm and 0.41 cm, while the prostate was 1.17 cm and 0.54 cm respectively. All accessory glands were lobulated and separated with a thick connective tissue into lobes and lobules. Acinar cells in the vesicular glands were a serous type, whereas acinar cells in the prostate and bulbourethral gland were the mucous types. Abstract

Former finding that avian influenza (AI) virus of H5N1 subtype from Indonesia shows reduced sensitivity against oseltamivir is critically reviewed trough molecular observation of the amino-acid sequence motive of neuraminidase protein (NA) of all H5N1 virus from human and animal in Indonesia available in GeneBank. Amino acid sequence of oseltamivir binding pocket of NA protein on all Indonesian viruses is typical for sensitive virus with a concerved motive of H274, E276, R292 dan N294. Resistance issue could not be explained based on available data.

Abstract

The aim of this study was to evaluate the deoxyribonucleic acid (DNA) status and characteristics of ram spermatozoa, maintained within epididymides stored at 4ºC. A total of 12 pairs of ram cauda epididymis were kept by means that one of the pair in a tube contain media (isotonic NaCl solution) and the other pair into a clean ziplock without medium then stored at 4ºC for three days. Following this, the motility and viability and DNA status of spermatozoa was observed using halomax sperm method, prior and post freezing process. The results showed a decreased in the percentage of motility and viability of spermatozoa from cauda epididymis which were kept in and without media, before or after cryopreservation (P <0.05). Based on the storage method, a significant difference in motility percentage occurs at day two and day three of the storage. Storage using media had a lower percentage of motility (23%; 10%) than without using media (50%; 37%) (P <0.05). The DNA status following storage at 4ºC for three days, descriptively showed impaired of spermatozoa DNA less than 1%. The percentage of impairment increased following the cryopreservation of spermatozoa due to the freezing process. This condition was especially observed on the third day of storage of cauda epididymis which were kept in media (10.16%). The characteristics of spermatozoa from cauda epididymis which were kept in the media were lower compared to without medium. The DNA status of cauda epididymis spermatozoa is not affected by the storage method. ABSTRAK Penelitian ini bertujuan untuk mengevaluasi status DNA dan karakterisik spermatozoa asal kauda epididimis domba yang disimpan pada suhu 4ºC. Sebanyak 12 pasang kauda epididimis domba disimpan dengan cara salah satu dari setiap pasang kauda epididimis dimasukan ke dalam tabung berisi media (NaCl fisiologis) dan pasangan yang lain ke dalam ziplock bersih (tanpa media). Penyimpanan dilakukan selama tiga hari pada suhu 4ºC kemudian dilihat motilitas dan viabilitasnya serta status DNA spermatozoa dengan metode sperm sus halomax, sebelum dan setelah pembekuan. Hasil penelitian menunjukkan bahwa persentase motilitas dan viabilitas spermatozoa asal kauda epididimis baik yang disimpan dengan atau tanpa media mengalami penurunan, sebelum atau setelah kriopreservasi (P<0,05). Berdasarkan metode penyimpanan, perbedaan persentase motilitas yang nyata terjadi pada hari ke dua dan ke tiga penyimpanan. Penyimpanan menggunakan media memiliki persentase motilitas yang lebih rendah (23%; 10%) dibandingkan dengan tanpa menggunakan media (50%; 37%) (P<0,05). Status DNA spermatozoa setelah dilakukan penyimpanan pada suhu 4ºC selama tiga hari, secara deskriptif memerlihatkan kerusakan DNA spermatozoa kurang dari 1%. Persentase kerusakan kemudian meningkat setelah kriopreservasi spermatozoa akibat dari pembekuan, terutama pada hari ke tiga penyimpanan kauda epididimis dengan media (10,16%). Karakteristik spermatozoa dari kauda epididimis yang disimpan dengan media lebih rendah dibandingkan tanpa media. Status DNA spermatozoa kauda epididimis tidak dipengaruhi oleh metode penyimpanan.

Abstract

A study was conducted to determine disease seroprevalence of egg drop syndrome (EDS) virus in layer ducks in Negara district, Jembrana, Bali. A total of 180 samples of laying duck serum with different management systems were used in this research. A number of 90 serum out of 180 samples were taken from ducks with an extensive management system (reared in the ricefield), and 90 other serum samples were taken from intensive ducks (caged) from community farms in six villages randomly taken from Nagara districts. Serum sampel were collected from EDS un-vaccinated ducks. Serum samples were tested using an hemagglutination/inhibition test (HA/HI test). Data were analyzed using chi-quadrat analysis (X2). The results showed that duck layers maintained in n Negara district had EDS antibody titer of 23 to 29 HI units. The results showed that 68 (75.56% ) of layer ducks with an extensive management system have EDS antibody titer, whereas only 18 (20%) of layer ducks with an intensive management system have EDS antibody titer. This indicates layer ducks in both management systems have antibody to EDS virus without EDS vaccination and that the layer ducks reared in ricefield has higher seroprevalence percentage than layer ducks reared in intensive cages.

Abstract

The aim of this research was to study the safety application of xylazine-ketamine and xylazinepropofolrecurrent dosage combination as anesthesia for laparo-histerotomy surgery in cat. Thisresearch used 10 female cats, 12-18 months of age, followed randomly divided into two groups, P1:atropine 0,04 mg/kgBW/SC + xylazine 2 mg/kg BW/IM + ketamine 20 mg/kg BW/IM; P2 : atropine0,04mg/kg BW/SC + xylazine 2 mg/kg BW/IM + Propofol 20 mg/kg BW/IV. The blood of the allgroups was taken from vena femuralis at 0 minute (before treatment), 15, 30, 45 and 60 minutesduring anesthesia for measurement of blood gas value pH, pCO2 and HCO3. After all animals wereanesthetized, the animals were treated laparo-histerotomy surgery. The data were analyzed byusing Randomized Complete Block Design (RCBD). The result showed both of groups were notsignificantly difference (p>0,05) to blood gas values for pH, pCO2 dan HCO3. Besides, both groupsanaesthetic agent perfectly caused metabolic acidosis with respiratory alkalosis compensationperfectly, therefore it is relatively safe to use as anaesthetic agent for surgery that needs long timeprocedure, as laparo-histerotomy.

Abstract

This study was performed to elucidate the association effect on feeding thymolina powder in thecarcass characteristics and morphology of small intestine of Ross 308 broiler chickens. In the presentexperiment, 320 one day old Ross 308 broiler chickens were used by using completely randomized designwith 4 treatments, 4 replicates, in which each replicate contained 20 broiler chickens. Experimentaltreatments groups were: control, 0.5, 1, and 2% of Thymolina powder in chickens’ diet. Thymolina® is ananti-bacterial powder drug which is made by composing 4 medicinal plants. A significant difference observedamong treatments in each parameter of the intestinal morphology (villus height, villus width, crypt depthand epithelial thickness. The number of goblet cells were not affected. Results of this findings indicatedthat using Thymolina® in the diet of broiler chickens causes improvements in carcass characteristics.Therefore, it may improve the performance and creation of the favorable broiler chickens.

Abstract

A research aimed to evaluate nutrients digestibility in growing local male pig fed spontaneous bioconverted-tamarind seed has been done in animal feed technology laboratory of Kupang State Agricultural Polytechnic.The research used 25 local male grower pigs with body weight around 3-6 kg. The study used a randomized block design with five treatments, i.e. R0 = ration using tamarind seeds without bioconversion, R1 = ration using spontaneous bioconversion tamarinds for 24 hours, R2 = rations using spontaneous bioconversion tamarinds for 48 hours, R3 = rations using bioconversion tamarinds spontaneous for 72 hours, and R4 = rations using spontaneous bioconversion tamarinds for 96 hours. Each treatment was repeated five times in which each of them used five pigs. Parameters observed were nutrient digestibility (dry matter, crude protein, crude fat, crude fiber, and ash). The data were analyzed by using variance and continued by Duncan’s multiple range test to determine the differences between treatments. The results showed that the time of spontaneous bioconversion of tamarind seeds increased the digestibility of tamarind seed nutrients and it was concluded that the best time of spontaneous bioconversion was for 72 hours.

Abstract

Burns are one of the health problems in modern society that are associated with tissue damage that is difficult to repair and affect patients, both physically and psychologically. This study was conducted to evaluate the potential of epigallocatechin gallate (EGCG ) ointment to the healing process of second degree skin burn induced by attaching 85ºC plate with 1 cm of diameter for 5 second on the skin of albino rat (Rattus norvegicus). Twenty-five rats were divided into 5 treatment groups. The P0 group was a group of rat that suffered burns and were treated with ointment base (PEG). The P1 group was a group of rat that suffered burns and were given standard therapy with silver sulfadiazine. P2, P3 and P4 groups are groups of rat that have burns and are treated with EGCG ointments with concentrations of 1%, 2%, and 4% respectively. At the end of the study, skin tissue excision was carried out to make histopathological preparations using HE staining. Evaluation of histopathological preparations was carried out on reepithelialization collagen deposition, PMN infiltration, and angiogenesis. The results of the study in group P4 showed that the highest collagen formation and re-epithelialization process was accompanied by a marked decrease in the inflammatory process and angiogenesis. This condition is significantly different from groups P0, P1, P2, and P3. Kesembuhan luka melibatkan beberapa fase yang berbeda dan saling tumpang tindih, yaitu fase inflamasi, granulasi, fibrogenesis, neo-vaskularisasi, kontraksi luka dan epithelialisasi (Robson, 1997). Penatalaksanaan luka bakar yang efektif memerlukan pemahaman proses kesembuhan luka normal dan mampu memilih intervensi yang tepat untuk mengoptimalkan proses kesembuhan luka (Snyder, 2005). Pada proses kesembuhan luka, inflamasi terjadi segera setelah jejas, diawali dengan vasokonstriksi yang berperan dalam proses hemostasis dan pelepasan mediator inflamasi. Fase proliferasi ditandai dengan terbentuknya jaringan granulasi oleh fibroblas dan proses angiogenesis. Reformulasi dan perbaikan kompartemen serabut kolagen yang disertai dengan penigkatan tensile strength menandai fase remodeling (Varoglu et al., 2010). Faktor yang memiliki peran penting pada tertundanya proses kesembuhan luka antara lain, trauma berulang, perfusi dan oksigenasi yang buruk serta inflamasi yang berlebihan (Harding et al., 2003)). Penggunaan bahan alamiah untuk pengobatan luka merupakan bagian penting dari penatalaksanaan kesehatan dan metode baik untuk menyediakan pilihan layanan kesehatan yang murah dan efektif (Gurung et al., 2009; Suntar et al., 2010). Beberapa penelitian menggunakan polifenol yang berasal dari teh hijau sebagai penyembuh alami sebagai agen anti penuaan, antiinflamasi, antikanker, antioksidan dan antidiabetes (Obaid et al., 2011). Beberapa penelitian dengan menggunakan Epigallocatechin gallate (EGCG), sebagai salah satu polifenol yang terkandung dalamteh hijau, membuktikan sejumlah efek biologis EGCG sebagai antioksidan, antimikrob, antiinflamasi, antialergi dan antineoplastik yang aktif (Hosnuter et al., 2015). Teh hijau memiliki manfaat yang beragam, antara lain mencegah kanker, meningkatkan kesehatan jantung dan pembuluh darah, melindungi kulit dari kerusakan yang disebabkan karena radiasi dan penyebab yang lain. Manfaat ini disebabkan karena the hijau yang mengandung EGCG memiliki aktivitas antioksidan, antiinflamasi dan antibakteri yang cukup kuat. Berdasarkan hal tersebut, tidak menutup kemungkinan bahwa EGCG dapat membantu percepatan proses kesembuhan luka bakar pada kulit (Nagle et al., 2006). Berdasarkan uraian tersebut, penelitian ini dilaksanakan untuk melihat pengaruh pemberian salep EGCG terhadap proses kesembuhan luka bakar pada tikus putih (Rattus norvegicus) jantan.METODE PENELITIAN Kelayakan Etik Penelitian ini dilaksanakan setelah mendapatkan persetujuan dari komisi etik Fakultas Kedokteran Hewan, Universitas Airlangga. Tempat Penelitian Penelitian ini dilakukan di Laboratorium Hewan Percobaan di Fakultas Kedokteran Universitas Airlangga, Surabaya. Persiapan dan pembuatan salep dilakukan di Fakultas Farmasi, Universitas Airlangga. Pembuatan dan pemeriksaan preparat histopatologis dilakukan di Laboratorium Patologi Anatomi, Fakultas Kedokteran Hewan, Universitas Airlangga. Hewan Eksperimental Sebanyak 25 ekor tikus putih (Rattus norvegicus) jantan, umur tiga bulan dengangroups. This is presumably because the compounds contained therein have antioxidant activity, antiinflammatory and antibacterial. These three activities will synergize in the process of healing wounds. Provision of 4% EGCG ointment for 14 days in second degree burns can accelerate the wound healing process which is characterized by improved re-epithelialization, collagen deposition, PMN infiltration in the wound area, and angiogenesis.
