Abstract

The growing demand for semisynthetic beta-lactams has directed attention towards enzymes, specifically Penicillin G Acylases (PGAs), for their potential in synthesizing these antibiotics. This study delves into the expression of Achromobacter xylosoxidans PGA (AxPGA) in Escherichia coli, with a focus on enhancing the yield of active PGA, often constrained by a complex maturation process. The optimization of PGA expression included variations in IPTG concentration and the addition of CaCl2. Furthermore, the study compared PGA expression in E. coli BL21 (DE3) with that in E. coli Arctic Express (DE3), capable of co-expressing chaperones (chaperonin Cpn60 and Cpn10). Induction with 0.5 mM IPTG resulted in the highest hydrolytic activity in both strains, with Arctic Express (AE) exhibiting significantly higher activity due to improved folding facilitated by cold-adapted chaperonins. Alongside optimal IPTG induction, the addition of 10 mM CaCl2 in the culture media significantly increased PGA activity in both strains, highlighting that Ca2+ supplementation is an effective strategy to enhance the yield of functional PGA. Subcellular fractionation demonstrates that the periplasmic fraction yielded higher volumetric and specific activities compared to the cytoplasmic fractions in both E. coli strains, highlighting the importance of periplasmic processing for PGA maturation. This suggests that extracting the periplasmic fraction is an effective strategy for recovering active PGA while avoiding or reducing contamination either from co-expressed cytoplasmic chaperones or other intracellular proteins. These findings emphasize that induction strategy, ionic stabilization, and host strain selection play synergistic roles in increasing active recombinant PGA expression.

Abstract

TaqMan qPCR analysis for detecting adulteration in processed meat products requires DNA with a high quantity, purity, and integrity. This study aims to evaluate the quantity, purity, and DNA integrity of various commercial pork products focusing on their ability to amplify the Cyt b gene. Fresh pork was used as a positive control, while five processed products - jerky, floss, meatball, canned corned, and smoked pork - were extracted for analysis. The DNA concentration and purity then measured, visualized using gel electrophoresis, and subjected to qPCR amplification. Statistical analysis of DNA concentration, purity, and ct value was carried out using one-way ANOVA followed by the Tukey post hoc test. The results showed significant differences in concentration, purity, and Ct values between all samples and positive controls (P˂0.05). Meanwhile, the Tukey test showed that all samples and fresh pork concentration differed significantly (P˃0.05) except between canned corned and fresh pork (P˂0.05). The purity was significantly different between the positive controls with pork meatball and smoked pork (P˂0.05), while the Ct value of all samples and positive controls were significantly different (P˂0.05) except for fresh pork and jerky (P˃0.05). All the commercially processed products experienced DNA fragmentation. Meat processing affects DNA concentration, purity, genomic integrity, and the amplification ability of qPCR.

Abstract

Sulistyaningsih LD, Abinawanto, Ardiyani M, Salamah A. 2018. Short Communication: Phylogenetic analysis and molecular identification of Canar (Smilax spp.) in Java, Indonesia Based on DNA Barcoding Analysis. Biodiversitas 19: 364-368. Smilax spp. (Smilacaceae) has long been used as medicinal herbs especially in East Asia and North America as they were known to be rich in steroidal saponin. Pharmacological study has been carried out in Indonesia. This genus is widespread in Indonesia and fairly abundant in Java and has been known either as edible fruit or medicinal plants. Characteristics of Smilax as a dioecious plant with high morphological variations make it thorny in species identification. Various molecular approaches have been devised to overcome identification problems such as DNA barcoding. This study, therefore was conducted to analyze the DNA barcoding application for phylogenetic and identification of Smilax in Java. A total of 31 samples were used in this study including 19 accession numbers from NCBI GeneBank. The genus Ripogonum was used as the out-group in phylogenetic reconstruction. Samples were successfully extracted by CTAB method with some modifications. rbcL region was used as the DNA barcode showed sufficient variation and conserved flanks. Two unidentified specimens have high similarity with S. leucophyla and lies in the same clade. The phylogenetic tree constructed by Maximum Likelihood analysis. The result showed that the monophyletic of Smilacaceae consisted of four clades. The genus Heterosmilax nested with Smilax though with low bootstraps value. It supports the monogeneric status of Smilacaceae.

Abstract

Abinawanto, Sriyani ED, Bowolaksono A. 2018. Characterization of Sentani gudgeon, Oxyeleotris heterodon (Weber, 1907) at Sentani Lake, Papua, Indonesia based on truss morphometric. Biodiversitas 19: 1013-1020. The study concerning the morphometry of Sentani gudgeon (Oxyeleotris heterodon, Weber 1907) was done from August 2016 until April 2017 in Sentani Lake, Jayapura District, Papua Province, Indonesia. The objectives of the study were to find out the diversity of the fish using truss morphometry approach. A total 56 individual sampled from three sampling sites were characterized for their 26 truss morphometric characters. The result showed that there were differences on morphometric characters of Sentani gudgeon. The highest correlation value found in character B2 and D5, which represented the dorsal portion and the tail fin of the fish body respectively. All variables showed significant different between each location (the p-value <c0.05). The canonical diagram resulted of 26 truss morphometric characters illustrated three separated clusters which discriminated the three populations based on sampling sites. A genetic distance analysis among Sentani gudgeon indicated that the fish originated from Putali Village was morphologically closed to the fish population from Sosiri. The truss morphometry technique can be used to identify the diversity of Sentani gudgeon in Sentani Lake.
