Abstract

Abstract. Zakaria Z, Abinawanto, Angio MH, Supriatna J. 2022. Habitat preferences and site fidelity of Tarsius supriatnai in agricultural area and secondary forest of Popayato-Paguat Landscape (Gorontalo, Indonesia). Biodiversitas 23: 3844-3851. Tarsius supriatnai is a new species experiencing a declining population trend due to deforestation. This study aims to determine the microhabitat preferences and site fidelity of T. supriatnai in the agricultural areas and secondary forests of the Popayato-Paguat landscape, Gorontalo, Indonesia. Measuring important value index (IVI) conducted by vegetation analysis of the nest tree location found in the two habitats was carried out using a quadrat method in seedling, sapling, pole, and tree forms. In addition, temperature and light intensity around the nest tree was also measured. The results showed that in the habitat of agricultural areas, the plant with the highest IVI was not the nest tree. Meanwhile, in the secondary forest habitat, the plant with the highest IVI at the tree form (Ficus virens) was the nest tree used by T. supriatnai. The diversity value of the H’ and E index showed that the two habitats were in the high to moderate. There was no significant difference in temperature and light intensity between the two habitat types. The results also showed that T. supriatnai mostly used the nest tree of Bambusa vulgaris (26.32%) in the agriculture area and Schizostachyum lima and Calamus zollingeri (28.57%) in the secondary forest. A survey was conducted to evaluate site fidelity to several nest trees found in 2018. The result revealed that T. supriatnai still uses 42.9% of nest trees.

Abstract. Bowolaksono A, Mayasari A, Sundari AM, Simamora ATJ, Suryaningsih R, Suryawan A, Simamora ATJ, Abinawanto, Sjahfirdi L, Hasna A, Supriatna J, Kyes RC. 2022. Urine biochemical profile analysis and transabdominal ultrasonography for pregnancy detection in the endangered lowland anoa (Bubalus depressicornis). Biodiversitas 23: 6056-6061. The population of lowland anoas (Bubalus depressicornis) has decreased significantly due to illegal hunting and habitat loss. As such, the urgency in developing successful ex-situ conservation programs cannot be overstated. Ex-situ breeding programs of anoas, however, are hindered by difficulties in early pregnancy detection. The failure to detect pregnancy at an early stage often results in miscarriage due to repeated breeding attempts of unconfirmed pregnant females. Moreover, frequent false heat during 5 to 6 months of pregnancy corroborated the repeated breeding of pregnant females. Therefore, early detection techniques are needed to improve breeding success. The present study evaluated the use of urine biochemical profiles as a minimally invasive approach for early pregnancy detection and transabdominal ultrasonography as a confirmation tool of false heat in four lowland anoas. Statistical analysis showed that glucose and protein level were significantly elevated during pregnancy since the 0 days of post-mating (p<0.05) and consistently higher compared to the non-pregnant period. Meanwhile, transabdominal ultrasonography successfully identified organ structures and development by the 155th, 176th, 180th, and 191st days of pregnancy. The present study suggests that urine biochemical data provide a promising, minimally invasive approach to early pregnancy detection in anoas. Incorporating urine data and ultrasonography can significantly help to maintain pregnancy by validating false heat.

Abstract

Abstract. Abinawanto A, Lestari S, Bowolaksono A, Dwiranti A, Lestari R, Gustiano R, Kristanto AH. 2023. Title The effect of different honey concentrations on the ultrastructure profile of spermatozoa in Dewa Mahseer (Neolissochilus soro). Biodiversitas 24: 1025-1031. Dewa Mahseer (Neolissochilus soro) is one of Indonesia's endemic freshwater fish at risk of population decline due to water pollution and asynchronous maturation of gonads. One of the efforts to conserve this species is preserving their spermatozoa using honey as a protector at certain temperatures. Therefore, this study aims to evaluate the effect of multiflora honey concentrations on ultrastructure spermatozoa profile of Dewa Mahseer. The temperature was maintained at 4°C and stored for 48 hours, followed by the cement dilution (1:10) in a stock solution containing fish ringer and honey. Transmission electron microscopy (TEM) examined the spermatozoa profiles at a magnification of ×1,500. Furthermore, ANOVA one-way analysis of variance was conducted to test the effect of head length, width, and area of spermatozoa on honey. The results showed a statistically significant difference in head width (F=13.929, p=0.001). Also, there is a difference in area between honey spermatozoa, fresh spermatozoa, and without honey at a cold of 4°C. The fresh spermatozoa stored at cold temperatures without honey observed considerable membrane damage. The novel aspect is an investigation of the effect of honey as a natural protector in the storage of Neolissochilus spermatozoa at 4°C for 48 hours. This study can be applied as a guide for the 48 hours of spermatozoa in a cold temperature in fish farming.

Abstract

Our study aims to scrutinize the hormonal and laboratory profile of women with PCOS visiting a fertility center in Indonesia. A computerized database of 120 PCOS women and 120 controls was retrospectively reviewed and analyzed. According to analysis, the concentration of AMH and basal LH were significantly more elevated in the PCOS group than in the control group (6.77 vs. 2.05, P<0.001; and 6.8 vs. 5.3, P<0.001, respectively). The level of basal FSH was significantly lower in the PCOS group (5.83 vs. 7.86; P<0.001). After an IVF cycle, PCOS women had significantly higher retrieved oocytes and mature oocytes compared to controls (18 vs. 8, P<0.001; 12 vs. 6, P<0.001). The number of fertilizations, cleavages, and top-quality cleavages were also higher in the PCOS group than in the control group (10 vs. 4, P<0.001; 10 vs. 4, P<0.001; 4 vs. 2, P<0.001). At the blastocyst stage, patients with PCOS had a higher number of blastocysts and top-quality blastocysts than control (9 vs. 4, P<0.001); 4 vs. 1, P<0.01). However, the clinical pregnancy was significantly higher in the control group than in the PCOS group (52.6% vs. 16.7%; P = 0.045).
